Abstract
The invention discloses a preparation method for extracting total flavonoids from guava leaves, belonging to the technical field of traditional Chinese medicine processing. The specific process is as follows: drying guava leaves, crushing, immersing in 60-70% ethanol solution, extracting with the aid of an ultrasonic extraction method to obtain filtrate, removing impurities with activated carbon, collecting with a macroporous resin column, eluting with ethanol solution to obtain eluent, and concentrating and drying the eluent to obtain the total flavone extract of the guava leaves. The preparation method is suitable for industrial production, and the prepared total flavone has high content and high purity and has the effects of eliminating dampness, strengthening spleen, clearing heat and detoxicating.
Description
Technical Field
The invention relates to a preparation method for extracting total flavonoids from guava leaves, belonging to the technical field of traditional Chinese medicine processing.
Background
Guava leaf, name of traditional Chinese medicine. Is leaf of Psidium guajava L of Myrtaceae. Is distributed or cultivated in Fujian province, Taiwan province, Guangdong province, Guangxi province, Sichuan province, Yunnan province and the like. Has the effects of eliminating dampness, strengthening spleen, clearing away heat and toxic materials. It is commonly used for diarrhea, dysentery, abdominal distention, gingival swelling and pain, rheumatic arthralgia, eczema, ecthyma, furuncle, toxic swelling, traumatic injury, traumatic hemorrhage, and snake and insect bite.
The flavone is a flavonoid compound, is a large natural product, widely exists in the plant world, and is an effective component of a plurality of Chinese herbal medicines. The most common in nature are flavones and flavonols, others include dihydroflavons (alcohols), isoflavones, biflavones, flavanols, chalcones, aurones, anthocyanins, neoflavonoids, and the like. In the end of the 60 s, people find that the flavonoid compounds have the effects of resisting inflammation, resisting virus, benefiting gallbladder, strengthening heart, calming, relieving pain and the like. In the 70 s, they were found to have antioxidant, antiaging, immunoregulatory and antitumor effects.
The prior flavone extraction method mostly adopts a heating reflux extraction method, so that the damage and the loss of effective components are easily caused, the existing ultrasonic extraction process is widely applied, the condition is avoided, and the operation is simpler and more convenient.
Disclosure of Invention
The invention aims to provide a method for extracting total flavonoids from guava leaves, which is convenient for industrial production and is simple and easy to operate.
A preparation method for extracting total flavonoids from guava leaves is characterized by comprising the following steps:
1) removing impurities from guava leaves, drying the guava leaves in a 50 ℃ oven, crushing the guava leaves, and sieving the crushed guava leaves with a 50-mesh sieve;
2) soaking guava leaf powder into 60-70% ethanol solution, extracting for 30-60min by using an ultrasonic-assisted extraction method with the power of 200-600w, mixing the extracting solutions after 3-5 times of extraction, and filtering to obtain filtrate;
3) adsorbing the filtrate with macroporous resin column, slowly eluting with distilled water at flow rate of 2-3ml/min, and removing impurities;
4) eluting with 70-90% ethanol solution, collecting eluate, adsorbing with active carbon, concentrating, and drying to obtain folium Psidii Guajavae flavone extract.
The method of claim 1, wherein the volume/mass ratio of the ethanol solution used for extracting the guava leaf powder to the raw material powder is 10ml/g-50 ml/g.
The method of claim 1, wherein the distilled water used in the extraction step 3 is 8-10 times the volume of the filtrate of the guava leaves.
