1. Claims (7)
1. A preparation method and application of a bacterial liquid of bacillus methylotrophicus LW-6 for biological control of cotton fusarium wilt and verticillium wilt,
the preparation method is characterized by comprising the following steps:
(1) inoculating Bacillus methylotrophicus LW-6 stock seeds on an agar starch plate culture medium, culturing for 72 hours at 35 ℃, detecting under a microscope, and selecting large colonies with uniform formed central spores and clearly colored two ends as fermentation starting colonies;
(2) inoculating the fermentation starting bacterial colony in the step (1) to a liquid seed culture medium, wherein the liquid loading amount is as follows: shaking 500ml of the culture medium to fill 100ml of seed liquid, culturing at 35 ℃ and 200rpm for 72h to obtain fermented seed liquid;
(3) inoculating the fermentation seed liquid obtained in the step (2) into a fermentation culture medium according to the inoculation amount of 9%, and performing fermentation culture in a fermentation tank under the culture conditions that: the fermentation is terminated when the pressure of the fermentation tank is 0.07Mpa, the air volume is 1:0.8vvm, the rotating speed is 260rpm, the culture temperature is 35 ℃ and the number of live spores in the fermentation liquid in the fermentation tank reaches more than 200 hundred million/ml and the amount of spores is more than or equal to 92 percent,
(4) and (4) carrying out ultrafiltration concentration on the fermentation liquor obtained in the step (3) to remove 40% of water, wherein the fermentation liquor is characterized in that: : 3 ultrafiltration membrane assemblies with the aperture of 0.005um, 0.001um and 1nm are used in series, fermentation liquor is sequentially and simultaneously subjected to continuous ultrafiltration treatment at the temperature of 35 ℃ and the pressure of 0.3MPa for 4 hours, and the methylotrophic bacillus LW-6 liquid microbial inoculum for biological control of cotton fusarium wilt is obtained, wherein the number of live spores is more than or equal to 200 hundred million/ml.

The bacterial liquid of bacillus methylotrophicus LW-6 according to claim 1, wherein the bacillus methylotrophicus deposit accession number in step (1) is: CGMCC No.6793, the preservation organization is: bacillus methylotrophicus of the general microbiological center of China Committee for culture Collection of microorganisms.

The methylotrophic bacillus liquid as claimed in claim 1, wherein the agar starch plate culture medium in step (1) comprises the following components by weight percent: 15 percent of agar, 7 percent of starch, 1.8 percent of yeast extract powder, 01.3 percent of peptone, 0.1 percent of magnesium sulfate, 0.2 percent of sodium carbonate, 0.8 percent of sodium chloride and the balance of water, and the pH value is 7.2-7.3.

The bacillus methylotrophicus liquid according to claim 1, wherein the liquid seed culture medium in the step (2) comprises the following components in percentage by weight: peptone 1.0%, glucose 2.6%, yeast extract 1.3%, NaCl 0.8%, magnesium sulfate 0.1%, sodium carbonate 0.2%, and the balance of water, wherein the pH value is 7.0-7.1.

The bacillus methylotrophicus liquid according to claim 1, wherein the fermentation medium in the step (3) comprises the following components in percentage by weight: 0.7% of corn steep liquor, 0.2% of corn starch, 1.4% of bran powder, 0.8% of glucose, 0.5% of yeast extract, 0.2% of peptone, KH2PO40.005%, (NH4)2SO 40.009%, MnSO40.006%, CaCO 30.007%, MgSO4.7H20.008%, 0.1% of magnesium sulfate, 0.2% of sodium carbonate and the balance of water, wherein the pH value is 7.4-7.5.
6. The preparation and application of the Bacillus methylotrophicus LW-6 bacterial liquid as claimed in claim 1.
7. Use according to claim 6, characterized in that the steps are as follows:
soaking a bacillus methylotrophicus LW-6 liquid microbial inoculum for biological control of cotton fusarium wilt for 3h before sowing, and mixing with water 1: 800 after 4 hours of mixing (after thorough mixing and filtration through a 200 mesh sieve), drip irrigation was carried out, repeated every 7 days for a total of 3 treatments.
