Experimental example 2 selection of extraction conditions
2.1 orthogonal test factors and horizontal design
The traditional Chinese medicine meridian prescription granule follows the theory of traditional Chinese medicine, and is prepared according to the characteristics of traditional Chinese medicine decoction, the traditional Chinese medicine meridian prescription granule needs to be heated and extracted by taking water as an extraction solvent in principle, so the experiment follows that the water is used as the extraction solvent, in the decoction operation, the factors influencing the extraction effect mainly comprise the water adding quantity, the decoction time, the decoction times and the like, 3 factors of the water adding quantity (A), the decoction time (B) and the decoction times (C) and 3 levels of the factors are selected, the content and the dry extract rate of quercetin-3-O- β -D-glucose-7-O- β -D-gentiobioside in roasted lepidium seeds are used as investigation indexes, and the L content and the dry extract rate of the quercetin9(34) Orthogonal tables were designed experimentally, with the optimal leaching process being preferred. The level of each factor is designed as shown in Table 1.
TABLE 1 level table of orthogonal test factors for decoction extraction
Figure BDA0002419279010000041
2.2 optimized leaching process by multi-index weighting method
In each index component, the weight of the active component quercetin-3-O- β -D-glucose-7-O- β -D-gentiobioside content is 0.8 because stir-fried lepidium seed is the monarch drug in the formula, and the dry extract yield is an auxiliary investigation index, and the weight is 0.2.
The test adopts a multi-index test total probability method to analyze data, the total probability evaluation method is carried out on the 2 indexes of the content of quercetin-3-O- β -D-glucose-7-O- β -D-gentiobioside and the dry extract yield, and the two indexes are considered to be mutually incompatible, and the weights are different, and P is calculated according to the content weight of quercetin-3-O- β -D-glucose-7-O- β -D-gentiobioside, namely the weight of dry extract yield index which is 8: 2iP of 2 indicesi=Xj/∑Xj(X is each index value, i is 1, 2, 3, j is 1, 2, 3.. 9), and the composite score P is 0.8P1+0.2P2
Weighing 9 parts of crude drug decoction pieces (i.e. 90g of parched semen Lepidii and 100g of fructus Jujubae) in 10 times of the formula amount, placing in 5L round bottom flask, and performing 3-factor 3-level L according to factor level table9(33) Orthogonal test, adding water for decocting for a certain time, filtering with 100-mesh filter cloth or electric spinning to obtain Chinese date liquid medicine, discarding Chinese date dregs, taking fried semen lepidii, placing into a gauze bag for tightening, placing into a round-bottomed flask containing the Chinese date liquid medicine, wrapping and decocting for a certain time, filtering with 100-mesh filter cloth or electric spinning to obtain decoction, concentrating under reduced pressure to a certain volume (about 100mL) to obtain No. 1-9 leaching liquor samples, respectively measuring quercetin-3-O- β -D-glucose-7-O- β -D-gentiobioside and dry extract in the samples, carrying out comprehensive scoring on the samples, wherein the results are shown in table 2, and carrying out variance analysis on the comprehensive scoring, wherein the results are shown in table 3.
TABLE 2L9(33) Results of orthogonal experiments

TABLE 3 analysis of composite score variance
Figure BDA0002419279010000052
Note, F0.05(2,2)=19,F0.01(2,2)=99
2.3) analysis of the results
As can be seen from the visual analysis in Table 3, the optimal extraction process of the product is A3B2C3The influence degree of each factor on the comprehensive score is A>B>C, the most significant influence is the amount of water added, the time for decoction and the times for decoction.
Analysis of variance was performed on the orthogonal experiments and the results are shown in table 3. As can be seen from Table 3, the factor A has significant difference, the factor B has no significant difference from the factor C, and the important order of the influence of the factors is that the water adding amount is more than the date decocting time and more than the pepperweed seed decocting time. By combining practical requirements of production benefit ratio, energy conservation, consumption reduction and the like in large-scale production, the preferable extraction conditions are as follows: adding 20 times of water, extracting fructus Jujubae for 1.5 hr, and extracting semen Lepidii by parching for 1.5 hr.
Experimental example 3 selection of concentration, drying conditions and granulation method
The traditional Chinese medicine granules are breakthrough and innovation of traditional Chinese medicine decoction. The concentration of the extracting solution is one of the key operations of the modern Chinese medicinal preparation, and the concentration process adopts reduced pressure concentration by combining the large production condition and the economical efficiency, the practicability and the feasibility of the production operation. The method has the advantages of simple process, continuous production, low heating temperature of effective components, short time and the like. Concentrating the extracting solution to an extract with the relative density of 1.10-1.20 at the temperature of 60-70 ℃ by adopting a double-effect concentrator under the condition of vacuum degree of-0.06-0.08 Mpa, filtering (100-mesh sieve), and collecting the extract.
In consideration of the economy, the practicability and the feasibility of large-scale production operation, the belt type vacuum drying is adopted in the drying process. The inventor creatively selects medical auxiliary materials with large specific surface area and good adsorptivity, such as dextrin and cyclodextrin, as dispersing aids, namely, a certain amount of auxiliary materials such as dextrin are weighed according to the material amount of the extract in proportion, and are uniformly stirred and mixed with the extract, and then the mixture is loaded onto a belt type vacuum drying device for reduced pressure drying, so that the problems of difficult drying, time consumption and material waste are well solved.
The granulation process selects dry granulation technology, dry extract is weighed and crushed into fine powder, sugar powder (sucrose powder, lactose powder and the like) and medical auxiliary materials such as dextrin, cyclodextrin and the like are added to 1000g, and the mixture is uniformly mixed and pressed into granules, thus obtaining the product.
