Abstract
本发明提供一种快速筛查水环境中重金属的方法,将待测水样与碱性条件下的硫胺素溶液混合后,使用荧光光谱仪或荧光分光光度计检测混合液在激发波长为365 nm,发射波长为440 nm的荧光信号,记录荧光强度F,根据荧光强度F值判断待测水样中是否含重金属;本发明克服传统电感耦合等离子体-质谱法(ICP‑MS)和阳极溶出伏安法监测水中重金属污染技术价格昂贵、运维成本高、检测时间长的缺点,利用硫胺素在碱性条件下被某些重金属氧化后荧光信号会发生特征性变化的原理,使用荧光光谱仪或荧光分光光度计实现了水中重金属的快速、低成本筛查及预警。

技术领域
本发明主要涉及水环境重金属监测技术领域,特别是涉及一种快速筛查水环境中重金属的方法。
背景技术
随着我国经济社会发展的不断深入,水污染问题日趋严重。其中,重金属类污染物因其具有长期性、累积性、潜伏性和不可逆性等特点,成为了水污染的重点防治对象。重金属不但会通过径流和淋洗作用污染地表水,还会通过食物链的方式进入人体内,对于重金属的富集人体难以代谢,最终直接或间接危害人体器官的健康,近年来,我国由重金属超标而引发的水污染事件不胜枚举,例如2010年的紫金矿业铜酸水渗漏事件以及2012年的柳江镉污染事件。水中的重金属污染不仅严重威胁生态环境和人民群众身体健康,还给我国带来了极大的经济损失。《水污染防治行动计划》提出要加强“水环境监控预警”,严格“环境风险控制”,即加强污染物泄露的“源头监管”,提高突发水污染事件的应急处置能力,将环境污染扼杀在源头。
目前,在实际的环境监管工作中,针对水中重金属污染的在线监测技术主要为电感耦合等离子体-质谱法(ICP-MS)和阳极溶出伏安法。ICP-MS技术结合了电感耦合等离子体高温电离特性与双重四级杆质谱仪技术特点,可对多种重金属同时进行定性与定量分析,具有检测精度高的优点,但是其存在设备昂贵、预处理过程繁琐、检测时间长、二次污染风险高等缺点;阳极溶出伏安法具有灵敏度较高、选择性较好、可同时分析多种元素等优点,但是其存在重复性差、维护成本高、二次污染风险大等缺点。
上述两种检测方法均存在检测时间长的缺点,当重金属污染事件发生时,环境监管人员往往不能立刻获悉事故的发生,从而使重金属污染态势扩大的风险加剧。此外,高昂的设备成本及运行成本也限制了这两种技术的应用。
重金属检测是一项长期的工作,要求检测手段向更高灵敏度、更高选择性、更方便快捷的方向发展,荧光光谱是一种能够描述荧光强度与激发波长和发射波长变化关系的光谱。荧光技术具有灵敏度高、检测成本低、扫描速度快,预处理过程简单等优点。某些荧光有机物与重金属离子发生氧化还原反应后,其荧光信号会发生特征性的改变。根据特征荧光信号的荧光强度的变化,可以定性、定量地判断水中重金属的污染程度。目前在水环境重金属监测领域还没有将荧光光谱技术应用其中实现水环境重金属监测。
发明内容
本发明为了克服背景技术中的不足提供一种快速筛查水环境中重金属的方法,克服传统电感耦合等离子体-质谱法(ICP-MS)和阳极溶出伏安法监测水中重金属污染技术价格昂贵、运维成本高、检测时间长的缺点,利用硫胺素在碱性条件下被某些重金属氧化后荧光信号会发生特征性变化的原理,使用荧光光谱仪或荧光分光光度计实现了水中重金属的快速、低成本筛查及预警。
为实现上述目的,本发明提供如下技术方案:
一种快速筛查水环境中重金属的方法,将待测水样与碱性条件下的硫胺素溶液混合后,使用荧光光谱仪或荧光分光光度计检测混合液在激发波长为365 nm,发射波长为440nm的荧光信号,记录荧光强度F,
混合液在激发波长和发射波长分别为365和440 nm 处的荧光强度F与重金属离子浓度c(μmol/L)满足如下线性正相关关系:
根据荧光强度F值判断待测水样中是否含重金属;
当F值大于等于荧光光谱仪或荧光分光光度计最大量程的3%-8%时,则说明样品中含有污染。

S1:检测试剂的制备:配置碱性条件的硫胺素溶液;
S2:样品的预处理:待测样品过滤,常温贮存;
S3:样品的检测:在环境温度下将检测试剂与样品按照1:0.8~1.5的体积比混合均匀,使用荧光光谱仪或荧光分光光度计,检测混合液激发波长为365 nm,发射波长为440 nm的荧光信号,记录荧光强度F;
S4:若检测混合液在激发波长和发射波长分别为365和440 nm的荧光强度F大于等于荧光光谱仪或荧光分光光度计的最大量程的3%-8%时,则说明样品中含有污染,且含有铜、锌、汞、镉、铬、铅、铁、钼、锰、钴、锑、镍、钒、钛、铊、银的一种或多种重金属。

本发明的一种快速筛查水环境中重金属的方法,利用荧光有机物与重金属离子发生氧化还原反应后,其荧光信号会发生特征性的改变,根据特征荧光信号的荧光强度的变化,可以定性、定量地判断水中重金属的污染程度的原理,采用硫胺素作为荧光有机物,利用硫胺素在碱性条件下被某些重金属氧化后荧光信号会发生特征性变化的原理,可以达到快速筛查水中重金属并实现预警的目的。
本发明的利用荧光光谱技术的水环境中重金属检测方法,灵敏度高、检测成本低、扫描速度快,预处理过程简单,同时,本方法因其可以同时筛查多种重金属,检测成本低、时间快、可重复性好、二次污染风险小,可应用于在线监测,检测精度可达到μmol/L级;本发明有利于大范围推广,对水质监测及水污染防治具有重要意义。
具体实施方式
为了更好的解释本发明,以便于理解,下面结合具体实施例,对本发明作详细描述。
实施例1,一种快速筛查水环境中重金属的方法,将待测水样与碱性条件下的硫胺素溶液混合后,使用荧光光谱仪或荧光分光光度计检测混合液在激发波长为365 nm,发射波长为440 nm的荧光信号,记录荧光强度F,
混合液在激发波长和发射波长分别为365和440 nm 处的荧光强度F与重金属离子浓度c(μmol/L)满足如下线性正相关关系:
其中,k为与金属种类相关的常数,荧光强度F越大,样品中重金属离子浓度c越高,证明样品中重金属污染程度越严重;
根据荧光强度F值判断待测水样中是否含重金属;
当F值大于等于荧光光谱仪或荧光分光光度计最大量程的
3%-8%时,则说明样品中含有污染;其中,选择最大量程的
3%-8%的补偿系数是考虑到检测仪器如荧光光谱仪或荧光分光光度计自身有噪声信号,在检测过程中在365/440 nm处有时会出现杂峰信号;抑或待测样品本身在365/440nm的位置上有时也会出现杂峰信号,导致实验测得F值与实际F值之间存在一些偏差,综合考虑上述因素后,通过补偿系数来确定待测样品在365/440 nm处的实际F值。
实施例2,快速筛查水环境中重金属的方法包括如下步骤:
S1:检测试剂的制备:配置碱性条件的硫胺素溶液;
S2:样品的预处理:待测样品过滤,常温贮存;
S3:样品的检测:在环境温度下将检测试剂与样品按照1:1.2的体积比混合均匀,使用荧光光谱仪或荧光分光光度计,检测混合液激发波长为365 nm,发射波长为440 nm的荧光信号,记录荧光强度F;
S4:若检测混合液在激发波长和发射波长分别为365和440 nm的荧光强度F大于等于荧光光谱仪或荧光分光光度计的最大量程的6%时,则说明样品中含有污染,且含有铜、锌、汞、镉、铬、铅、铁、钼、锰、钴、锑、镍、钒、钛、铊、银的一种或多种重金属。
通过步骤S2中对待测样品过滤,可以去除水样中的杂质及沉淀物,避免水样中的沉淀物影响光谱扫描。
实施例3,其它步骤同实施例2,其中步骤S1中:配置浓度为15 μmol/L硫胺素溶液,调节pH至11.00。
浓度为10 μmol/L以上的硫胺素溶液作为检测试剂,适用于重金属强度不够强的待检测样品,辨识度更高,避免硫胺素浓度过低导致辨识度低、检测不准确;且硫胺素相对于其它荧光有机物具有特征性,即只有硫胺素在365/440nm处检测荧光强度会出峰,当硫胺素溶液浓度选用10 -20μmol/L以内时,可避免因浓度过高导致检测出的荧光强度易超量程,进而导致检测范围过窄。
实施例4,其它步骤同实施例3,其中步骤S3中:检测试剂与样品按照1:1的体积比混合均匀。
经过反复实验,数据表明,检测试剂与样品混合的体积比对于检测结果具有一定影响,二者体积比在1:0.8~1.5的范围内检测效果好。
实施例5,其它步骤同实施例4,其中步骤S3中:检测试剂与样品混合后静置3 min以上,再使用荧光光谱仪或荧光分光光度计检测混合液激发波长为365 nm,发射波长为440nm的荧光强度。
检测试剂与样品混合后通过静置,可以保证硫胺素和待测样品充分反应,避免二者混合后短时间内进行检测导致荧光光谱的信号过弱,检测试剂与样品混合后静置时间在3 min以上为宜。
实施例6,其它步骤同实施例5,其中步骤S4中:若检测混合液在激发波长和发射波长分别为365和440 nm的荧光强度F大于等于荧光光谱仪或荧光分光光度计的最大量程的5%时,则说明样品中含有污染,且含有铜、锌、汞、镉、铬、铅、铁、钼、锰、钴、锑、镍、钒、钛、铊、银的一种或多种重金属。
本发明的快速筛查水环境中重金属的方法,用于检测样品中重金属离子与其还原态组成的氧化还原电对的标准电位大于-1.36 V时的重金属离子,该重金属离子可被硫胺素试剂检测,适用于本方法。

S3:样品的检测:在环境温度下将检测试剂与样品按照1:0.8~1.5的体积比混合均匀,使用荧光光谱仪或荧光分光光度计,检测混合液激发波长为365 nm,发射波长为440 nm的荧光信号,记录荧光强度F;
S4:若检测混合液在激发波长和发射波长分别为365和440 nm的荧光强度F大于等于荧光光谱仪或荧光分光光度计的最大量程的3%-8%时,则说明样品中含有污染,且含有铜、锌、汞、镉、铬、铅、铁、钼、锰、钴、锑、镍、钒、钛、铊、银的一种或多种重金属。
3.根据权利要求1所述的快速筛查水环境中重金属的方法,其特征是:步骤S1中:配置浓度为10 μmol/L以上的硫胺素溶液,调节pH至11.00。
4.根据权利要求3所述的快速筛查水环境中重金属的方法,其特征是:步骤S1中:配置浓度为10-20 μmol/L的硫胺素溶液,调节pH至11.00。
5.根据权利要求1所述的快速筛查水环境中重金属的方法,其特征是:步骤S3中:检测试剂与样品按照1:1的体积比混合均匀。
6.根据权利要求1所述的快速筛查水环境中重金属的方法,其特征是:步骤S3中:检测试剂与样品混合后静置3 min以上,再使用荧光光谱仪或荧光分光光度计检测混合液激发波长为365 nm,发射波长为440 nm的荧光强度。
7.根据权利要求2所述的快速筛查水环境中重金属的方法,其特征是:步骤S4中:若检测混合液在激发波长和发射波长分别为365和440 nm的荧光强度F大于等于荧光光谱仪或荧光分光光度计的最大量程的5%时,则说明样品中含有污染,且含有铜、锌、汞、镉、铬、铅、铁、钼、锰、钴、锑、镍、钒、钛、铊、银的一种或多种重金属。
8.根据权利要求1-7任一项所述的快速筛查水环境中重金属的方法,其特征是:用于检测样品中重金属离子与其还原态组成的氧化还原电对的标准电位大于-1.36 V时的重金属离子。
