Description

Method for carrying out half-environment accommodation capacity expanding culture on brachionus calyciflorus
Technical Field
The invention belongs to the technical field of functional relationship between a fishery biological community structure and a food net.
Background
Rotifers belong to the phylum rotifer (rotifer), most species are 100-500 μm in body size and are a very abundant group in pseudocoelomates. The number of species is second only to nematode animals, and there are about 2000 species worldwide. Most (about 1600 species) of rotifers belong to the mononidae class, the rest belong to the bisnidae class (another classification method is divided into the leech morphea class and the coccinea class). The rotifers have many species distributed widely throughout the world, most living in freshwater environments.
The brachylostomus calyciformis (Brachionus calyciflorus) has a very important position in fresh water rotifers. The brachylostomus calyciformis belongs to the order Mononidae (monogonta), the order swim (Plioma), the family brachyloideae (Brachinonidae), the genus brachylostomus (Brachinus). The brachionus calyciformis has the advantages of rapid growth, wide distribution in China, easy acquisition, easy culture and the like, and is an important model organism in experimental research. How to carry out rapid propagation in a laboratory and keep the population of the plant growing stably has great significance in adapting to experimental requirements.
In the artificial propagation culture with space and food limitation, the population density of the rotifers can quickly reach a peak value due to the high propagation speed of the rotifers, so that the density is too high, the dissolved oxygen in a water body is quickly consumed, and the rotifers are dead due to the accumulation of metabolic wastes, so that the population density is quickly reduced. This drastic fluctuation in population density is not conducive to the development of experimental studies by researchers.
How to find a solution to overcome the current problems is a problem that researchers in the field can not solve.

Detailed Description
Example 1
The invention aims to provide a method for carrying out half-environment volume expanding culture on brachionus calyciflorus.
The method adopts EPA culture solution to culture the brachionus calyciformis. The day-night period of illumination is 12h:12h, and the illumination intensity is 1500 Lx. The food is Chlorella cultured by BG-11 (institute of aquatic organism #1067 of Chinese academy of sciences), and the addition amount is 106cell/ml。
The method of the invention performs transfer culture and propagation culture when the rotifer fertility is the most vigorous, is beneficial to maintaining the continuous culture of the rotifer population and ensures the stability and reliability of the rotifer as the source of experimental materials.
In order to achieve the above object, the method comprises the steps of:
1. determining the growth rate K point of brachionus calyciflorus culture
(1) Hatching the dormant eggs of brachionus calyciflorus:
absorbing bottom culture solution generated by culturing Brachionus calycinus in a light incubator, selecting resting eggs, placing in a refrigerator at 4 ℃ for 4 weeks, taking out, and placing in a special small glass cup of 10ml for incubation.
(2) Culturing and rapidly propagating the calyx flower brachiarius in a food-rich environment:
selecting 9 larvae from hatched rotifers, dividing into three groups (A, B, C), placing 3 larvae in 10ml small glass cup, adding EPA culture solution, and adding Chlorella (FACHB #1067) to 10%6cell/ml, after the number of rotifers is increased to 20 or more, transferring the rotifers to a 250ml beaker, adding 200ml of EPA culture solution for culturing, and keeping the chlorella concentration at 106cell/ml。
(3) Determining a growth rate K point;
and uniformly mixing the rotifers cultured in 200ml of culture solution every 24 hours, taking 8 ml of the rotifers under a microscope for counting, and determining the K point and the corresponding time according to the relation between the change of the number and the time.
The date of the first batch of rotifers is calculated from the beginning of changing to a 250ml beaker, the rotifers are cultured until the rotifer population begins to decline, a rotifer density change curve is drawn, and a point with the maximum growth rate is found.
The K point is an inflection point where the number no longer increases and a drop occurs.

(2) Culturing and rapidly propagating the calyx flower brachiarius in a food-rich environment:

(3) Re-culture was performed when the population reached half the environmental capacity, i.e. K/2 point:
the K and K/2 values found in the first step are labeled (see FIG. 1). Transferring and expanding culture when the population multiplication rate is maximum; mixing the original 200ml of rotifer and culture solution, transferring 100 ml of rotifer into a new beaker, and adding the culture solution to 200 ml. The rotifer remained growing at a high rate after propagation (see fig. 2).
