Whether the tobacco branch ointment (hereinafter referred to as the ointment) can inhibit the growth of escherichia coli, staphylococcus aureus and bacillus subtilis is examined.
1. Principle of experiment
The effect of the ointment on the growth of bacteria was examined by using the continuous dissolution of the ointment and the formation of different concentration gradients through diffusion of agar. Whether the bacteriostatic ring has bacteriostatic ability is judged by measuring the diameter of the bacteriostatic ring, when the diameter is more than or equal to 7mm, the sample has bacteriostatic action, and if the diameter is less than 7mm, the sample has no bacteriostatic action.
2. Experimental methods
2.1 Experimental groups
Sample group: weighing 5g of tobacco ointment, adding 3ml of methanol, and mixing to obtain methanol extract of the ointment.
Blank control group: methanol
2.2 preparation of Experimental Filter paper sheet
5 pieces of sterile dried filter paper were taken, wherein 4 pieces of the sample group and 1 piece of the blank control group were used. Dripping 20 μ l of methanol extract of ointment into the sample group, dripping 20 μ l of methanol into the blank control group, then placing the filter paper sheet in a clean sterile plate, and opening the cover to place in an ultra-clean workbench for natural drying for later use.
2.3 inoculation
Taking the concentration of 5X 10 by using a pipettor5cfu/ml~5×106cfu/ml Escherichia coli, Staphylococcus aureus, Bacillus subtilis suspension 200 ul in tryptone soy agar medium plate surface, immediately using the coating rod evenly spread. The plate was covered and dried at room temperature for 5 min.
2.4 paste-on ointment
Each test is stuck with 1 infectious bacterium plate, and each plate is stuck with 4 test sample plates and 1 blank reference sample plate, and the number of the test sample plates and the blank reference sample plates is 5. A sample was taken with sterile forceps and placed on the surface of the plate. 3. Results of the experiment
The tobacco ointment has the effects of inhibiting growth of Escherichia coli, Staphylococcus aureus, and Bacillus subtilis.
The experimental results are statistically as follows:
(1) the natural bactericidal ointment results in colibacillus resisting experiment, where a, b, c and d are ointment sample groups and e is blank control group
(2) The natural bactericidal ointment has the test result of resisting staphylococcus aureus, wherein a, b, c and d are ointment sample groups, and e is a blank control group
(3) The experimental result of the natural bactericidal ointment for resisting bacillus subtilis shows that a, b, c and d are ointment sample groups, and e is a blank control group

Claims (6)
1. The preparation method of the natural bactericidal ointment is characterized by comprising the following steps:

(2) in the step (2) (a) of the specification, the ointment raw materials are added or reduced or are mixed according to any proportion by using tobacco branch extract or plant extract, PEG-4000, PEG-400, glycerol, borneol, 95% ethanol, sorbic acid, benzoic acid and the like;
(3) the filling in the step (2) (b) is conventional filling or intelligent filling.

The method for preparing the natural bactericidal ointment by taking the fine extract of the tobacco branches as the raw material according to claim 1, which is characterized by comprising the following steps: the centrifugation treatment in the step (1) (a) is to place the concentrated extracting solution after temperature reduction treatment in a centrifuge bottle, wherein the centrifugation time is 30min, and the rotation speed is 3500 rpm/min.

The method for preparing the natural bactericidal ointment by taking the fine extract of the tobacco branches as the raw material according to claim 1, which is characterized by comprising the following steps: in the step (1) (a), the volume ratio of the added extraction solvent is V/V-1: 2, the extraction temperature is 90 ℃, the extraction time is 30min, and the centrifugation temperature is 8 ℃.

The method for preparing the natural bactericidal ointment by taking the fine extract of the tobacco branches as the raw material according to claim 1, which is characterized by comprising the following steps: the dipping extraction temperature in the step (1) and the step (b) is 4 ℃, the extraction time is 8h, the centrifugation time is 4 ℃, and the rotary evaporation time is 60 ℃.

The method for preparing the natural bactericidal ointment by taking the fine extract of the tobacco branches as the raw material according to claim 1, which is characterized by comprising the following steps: the stirring and dissolving temperature in the step (2) is 80 ℃, and the dissolving time is 10 min.
6. The method for preparing the natural bactericidal ointment by taking the fine extract of the tobacco branches as the raw material according to claim 1, which is characterized by comprising the following steps: the solvent in the step (2) is 95% ethanol, and the water bath temperature is 70 ℃.
