槐橘香精油所治消化不良多由饮食不节,起居不适等所致,病位在脾胃、或与肝相关。脾主运化,胃主受纳,脾胃不和则胃脘不适,肝主疏泄,脾主运化,肝失疏泄则易导致脾失健运。治宜健脾和胃、理气疏肝。方中橘核理气散结,丁香温中降逆、行气止痛,槐花清肝泻火,三药合用,利用精油芳香走窜的功效,共凑温中降逆,醒脾开胃,疏肝散结之功,治疗消化不良。

1.3 仪器
电子称,上海民桥医疗器械有限公司,型号:JY601;电子分析天平,奥豪斯(上海)公司,型号:AR1140/C;高速台式离心机,上海安亭科学仪器厂,型号:TGL-168;可调式移液器,上海雷勃分析仪器有限公司;游标卡尺。
实验方法
选取SPF级SD大鼠40只,体重控制在130-150g,分为4组,分别为空白对照组,槐橘香精油大、小剂量组(0.5mL/鼠、0.25mL/鼠;每鼠涂精油0.25mL/鼠,涂在脱毛后的大鼠腹部及大鼠口鼻处,大剂量在涂精油后1小时再同法同量涂一次),建胃消食片灌服(1g/kg),连续5d给予受试物。于最后一次给药后1小时检测结果。实验前禁食不禁水12h。
胃液采集:试验时采用腹腔注射水合氯醛麻醉大鼠,将大鼠固定后,从幽门向胃内插入一直径约为2mm的塑料管,在胃靠近幽门球部结扎固定,用以收集胃液,并用塑料管插入前胃与食管一起结扎,以PH为7.0,35℃的生理盐水用蠕动泵按15ml/h的速度运行胃内灌流,收集1h流出液,离心后收集上清液。
胃液总酸度酸测定:利用NaOH滴定测胃液酸度,取胃液1mL,置于小烧杯中,加入酚酞指示剂2滴,混匀,于小烧杯下衬一白纸,用微量滴管徐徐滴入0.01mol/L氢氧化钠(NaOH)标准溶液,随滴随摇,直至出现微红色,30s内红色不退,记录NaOH溶液的消耗量,即为总酸度终点量,按下式计算总酸度:
总酸度(mmol/L)=总酸度终点量×10
胃蛋白酶活性测定:采用Mett毛细玻管法。首先用内径1mm长10cm的毛细玻璃管吸满蛋清(纱布过滤),然后于75℃热水中使蛋白凝固,制成长2.5cm的蛋白管。取胃液1ml加入50ml的三角烧瓶中,再加0.05mol/L盐酸溶液15ml摇匀,再放入新鲜制作的蛋白管2根,塞好瓶口,在37℃恒温孵箱中孵育24h,取出蛋白管,用游标卡尺测量蛋白管两端透明部分的长度(mm),以两端之值求其平均值,计算胃蛋白酶活性和胃蛋白酶的排出量。
胃蛋白酶活性单位(μ/ml)=四端蛋白管透明部分长度均值2×16
3.实验结果
对大鼠胃液总酸度、胃蛋白酶活性的影响,见表1

由表1可看出,与空白组比,槐橘香精油大、小剂量组和健胃消食片组均能显著升高大鼠的胃液总酸度(P<0.01),显著提高大鼠胃蛋白酶活性(P<0.01)。
(二)、槐橘香精油对小鼠小肠蠕动的影响实验
材料
1.1动物
KM小鼠,SPF级,12-15g,雌雄各半,提供单位:济南朋悦实验动物繁育有限公司。合格证号:37009200018683
1.2 药品与试剂
本发明实施例1产品,由河南中医药大学提供;复方地芬诺酯,安阳市华安药业有限责任公司,批号:171201;炭末,无锡市亚泰联合化工有限公司,批号:20180301;健胃消食片,江中药业股份有限公司,批号:18090081。
1.3 仪器
电子称,上海民桥医疗器械有限公司,型号:JY601;尺子、手术剪。
2.实验方法
KM小鼠50只按体重大小随机分为空白对照组、模型、健胃消食片组(阳性对照组)和槐橘香精油大、小剂量组。槐橘香精油大、小剂量组(0.3mL/鼠、0.15mL/鼠;每鼠涂精油0.15mL/鼠,涂在脱毛后的小鼠腹部及小鼠口鼻处,大剂量在涂精油后1小时再同法同量涂一次),建胃消食片灌服(1.35g/kg,给药体积0.2mL/10g),连续3d给予受试物。实验前禁食不禁水12h。于测定当天各给药组、模型及空白对照组再给予一次受试样品或蒸馏水,30min后各样品组和模型对照组灌复方地芬诺酯混悬液0.2mL/10g(即5mg·kg-1,0.025%),空白对照组灌予蒸馏水。30min后,各组再给予墨汁指示剂,25min后断颈处死动物,打开腹腔分离肠系膜,剪取上端自幽门、下端至回盲部的肠管,置于平板上,轻轻拉成直线,分别测量从幽门到回盲部和从幽门到墨汁前沿的距离,计算墨汁推进率:
墨汁推进率(%)=墨汁推进长度/小肠总长度×100
3.结果
对小鼠小肠蠕动的影响,结果见表2:

由表2可知,与空白组比,模型组小鼠小肠墨汁推进率显著减小(P<0.01)。与模型组比,槐橘香精油大、小剂量组和健胃消食片组均能显著增加小鼠小肠墨汁推进率(P<0.01)。
二、临床试验
在动物实验确保安全的情况下,经临床试用,效果非常好,有关资料如下:
选择脾失健运型小儿厌食症患儿 20 例,其中男 11 例,女 9 例;年龄为 0.7-4.5岁;病程为1-3个月。 将槐橘香精油涂在患儿神阙穴周围及口鼻周围,0.5ml/人(可根据涂用情况,将此量精油分作多次涂用),每天给上述剂量精油一次。连用 1 周为 1 个疗程,统计疗效。
痊愈:临床症状消失,恢复食欲,进食量增多;
有效:患儿临床症状得到改善;
无效:患儿临床症状无改善。
临床症状是指长期的食欲减退或消失、以食量减少为主要症状。
统计学处理结果:痊愈5 例,占比25%;有效13例,占65%;无效2例,占10%,总有效率90%。
在对实施例1做上述实验的同时,还对实施例2、3也做了同样的实验,均取得了相同和相近似的结果,表明产品质量好,疗效稳定可靠,这里不再一一列举。
以上实验表明,本发明具有改善胃肠功能,治疗小儿厌食症之功效,有效用于治疗小儿厌食,制备方法简单,原料丰富,易生产制备,疗效确切,是治疗小儿厌食药物上的创新,有显著的经济和社会效益。
Claims (4)
1.一种外用改善小儿消化功能治疗厌食症的槐橘香精油,其特征在于,由以下原料:槐花精油4-6mL、丁香精油2.5-3.5mL和橘核精油5-7mL混匀制成;其中:
所述的槐花精油,是将槐花粉碎成过10~20目筛粗粉,超临界CO2流体萃取,液料重量体积比1:10~20,重量体积比是指固体以g计,液体以mL计,萃取温度42~50℃,萃取压力20~25MPa,分离温度50~60℃,分离压力4.5~5.5MPa,萃取时间1.5~2.5h,得槐花精油;
所述的丁香精油、橘核精油的制备方法均同槐花精油。
2.根据权利要求1所述的外用改善小儿消化功能治疗厌食症的槐橘香精油,其特征在于,由以下原料:槐花精油5mL、丁香精油3mL和橘核精油6mL混匀制成。
3.根据权利要求1所述的外用改善小儿消化功能治疗厌食症的槐橘香精油,其特征在于,由以下原料:槐花精油4.5mL、丁香精油3.3mL和橘核精油5.5mL混匀制成。
4.根据权利要求1所述的外用改善小儿消化功能治疗厌食症的槐橘香精油,其特征在于,由以下原料:槐花精油5.5mL、丁香精油2.8mL和橘核精油6.5mL混匀制成。
