Abstract
本发明提供一种白细胞的分离纯化方法,所述分离纯化方法包括步骤:(1)收集血液样本;(2)离心分离出白细胞粗提物;(3)在白细胞粗提物中加入红细胞裂解液裂解红细胞,离心去上清;(4)重复步骤(3)至少一次,分离获得白细胞。本发明方法操作方便,时间短,成本低,可以大规模处理样本。

技术领域
本发明涉及生物医药领域,具体的涉及细胞的分离方法。
背景技术
白细胞来源于骨髓中的造血干细胞,在骨髓中发育后进入血液和淋巴液循环,也存在于血管和淋巴管外的组织中,它的功能是使机体免受病原体(细菌和病毒)、癌细胞、异物侵入,是人体的守卫者。人体中白细胞有八种类型,包括巨噬细胞、树突状细胞、B淋巴细胞、T淋巴细胞、自然杀伤细胞、嗜中性粒细胞、嗜酸性粒细胞以及嗜碱性粒细胞。
巨噬细胞是个头最大的白细胞,巨噬细胞由单核细胞分化发育而来。巨噬细胞对细胞残体和病原体进行吞噬和消化,参与适应性免疫的专职抗原提呈,促进生殖细胞发育、性激素、骨组织的再吸收以及血络的形成。树突状细胞也是一种单核细胞,通过摄取、加工处理和递呈抗原到淋巴结和淋巴器官中的淋巴细胞帮助机体识别病原体,启动特异性免疫应答。B细胞是淋巴细胞的成分之一。B细胞受抗原刺激后,会合成和分泌抗体对抗病原体。抗体与抗原结合帮助机体识别病原体,并使之成为靶细胞被其他免疫细胞清除。被称为记忆细胞的B细胞可以保持对病原菌生物分子标记物的记忆,使得同一种抗原再次进入机体时免受感染。T细胞也是淋巴细胞之一,包括细胞毒性T细胞、辅助性T细胞、抑制性T细胞、自然杀伤T细胞和记忆T细胞。细胞毒性T细胞直接杀伤靶细胞,辅助性T细胞协助B细胞产生抗体,抑制性T细胞抑制B细胞和其他T细胞对抗原的免疫应答。自然杀伤细胞是分布于血液循环的淋巴细胞,它能识别被感染细胞和衰老细胞,通过释放化学颗粒吞噬破坏病变细胞。嗜中性粒细胞是血液循环中数量最多的粒细胞,机体遭遇病原体入侵时能迅速出现在感染和损伤的部位。嗜酸性粒细胞是具有吞噬作用的白细胞,当过敏反应和寄生虫感染时嗜酸性粒细胞会明显增多。嗜酸性粒细胞具有粗大的嗜酸性颗粒,可释放化学物质消灭病原体。嗜碱性粒细胞是颗粒内含有组胺和肝素等物质的粒细胞(有粒白细胞),肝素能稀释血液,抑制血凝块的形成,组胺能扩张毛细血管、增加血流量。
在先前的实验中,一般利用淋巴细胞分离液分离外周血液中白细胞。血液中白细胞其体积、形态和密度与其他细胞不同。红细胞和粒细胞密度较大,为1.090g/ml左右,而淋巴细胞和单核细胞密度为1.075~1.090g/ml,血小板为1.030~1.035g/ml。淋巴细胞分离液是一种密度介于1.075~1.090g/ml之间而近似于等渗的溶液,用它做密度梯度离心,使一定密度的细胞按相应密度梯度分布,从而将各种血细胞加以分离。
利用淋巴细胞分离液进行全血中白细胞的分离的体操作步骤为:先将淋巴细胞分离液恢复到室温,4ml淋巴细胞分离液置于15ml离心管中,将4ml全血用移液器慢慢沿管加入离心管,可以看到血液在上层,分离液在下层。放入离心机,常温,400g离心30分钟;将离心管从离心机取出,此时管内可以分为4层:上层为血浆,第二层为白膜层,第三层浑浊层为淋巴细胞分离液层,下层为红细胞层;用移液器吸取第二层浑浊层白细胞,置于15ml离心管中,加入8ml磷酸盐缓冲液洗涤细胞,将白细胞轻轻弹起混匀,常温离心,250g,10分钟;去除上清,加入1ml磷酸盐缓冲液洗涤细胞,将白细胞轻轻混匀,转移全部溶液至2ml离心管中,250g,离心5分钟;吸取上清,将白细胞放入-80℃冰箱保存。
利用淋巴细胞分离液分离白细胞操作过程复杂、繁琐,操作时间长,不能大规模处理样本。因此,本领域迫切需要开发操作简单,时间短,成本低的白细胞的分离方法。
发明内容
本发明提供了一种操作简单,时间短,成本低的白细胞的分离方法。
本发明第一方面提供一种白细胞的分离纯化方法,其特征在于,所述分离纯化方法中采用红细胞裂解液进行分离纯化。
本发明第二方面提供一种白细胞的分离纯化方法,其特征在于,所述分离纯化方法包括步骤:
(1)收集血液样本;
(2)离心分离出白细胞粗提物;
(3)在白细胞粗提物中加入红细胞裂解液,充分裂解红细胞,离心去上清;
(4)重复步骤(3)至少一次,分离获得白细胞成品。
上述步骤(3)中,加入红细胞裂解液后充分混匀,并静置5-10分钟,每3分钟上下颠倒容器多次。
上述步骤(3)中,加入红细胞裂解液后充分混匀,静置10分钟,每3分钟上下颠倒容器5次。
上述步骤(1)中采用EDTA抗凝管收集血液样本。
上述分离纯化方法还包括步骤:
(5)在白细胞成品中加入PBS,将白细胞轻轻混匀,吸取上清后冻存。
上述步骤(2)中,离心参数为800g,20分钟。
上述步骤(3)(4)中白细胞与红细胞裂解液的体积比为1:3-4。该处白细胞为白细胞粗提物。
上述步骤(3)(4)中,离心参数为400g,5分钟。
本发明方法操作方便,时间短,成本低,可以大规模处理样本。市售的红细胞裂解液均可以用于本分离纯化方法中,且获得的白细胞符合质量要求。

具体实施方式
以下,参照实施例对本发明进行更详细和具体地描述,但下述实施例并不意在限制本发明。
实施例1不同方法分离白细胞的比较
分别比较红细胞裂解液,淋巴细胞分离液的分离血液样本中白细胞的结果,以纯细胞作为分离结果的对照。利用rRNA比率(28S/18S)以及RNA完整度指数,作为分离的细胞的质量指标,当rRNA比率大于1.5,RNA完整度指数大于7,则质量合格。
具体操作方法如背景技术部分和图1所示。具体测试结果如图2所示,红细胞裂解液得到的结果符合质检要求,操作时间短,方法简单。
该分离纯化方法包括步骤:
(1)收集血液样本;
(2)离心分离出白细胞粗提物;
(3)在白细胞粗提物中加入红细胞裂解液,充分裂解红细胞,离心去上清;
(4)重复步骤(3)至少一次,分离获得白细胞;
(5)加入PBS,将白细胞轻轻混匀,吸取上清后冻存。
加入红细胞裂解液时,白细胞与红细胞裂解液的体积比在1:2-4。优选三倍以上。
实施例2采用红细胞裂解液大规模分离白细胞
本专利是利用红细胞裂解液分离血液中白细胞。血液红细胞表面上有红细胞专属的表面抗原,当裂解液中含可以攻击特定红细胞表面细胞抗原的酶的时候,就只会造成红细胞的变形,生物通道扩大、膨胀、裂解,而不会攻击其他细胞。另外红细胞有自己的电负性、渗透脆性、悬浮稳定性,这些特点使得红细胞区别于血液的其他细胞。红细胞裂解液利用这些特性裂解红细胞。
具体操作过程:利用5ml EDTA抗凝管收集血液。放入离心机,4℃,800g离心20分钟;将离心管从离心机取出,此时管内可以分为2层:上层为血浆,下层为红细胞层,血浆与红细胞中间白色为白细胞层;用移液器吸取300ul白细胞,置于1.5ml离心管中,加入1ml红细胞裂解液,用移液器吹打均匀;室温静置10分钟,每3分钟上下颠倒5次;400g,4℃离心5分钟,去除上清;加入1ml红细胞裂解液,用移液器吹打均匀,400g,4℃离心5分钟;除去上清,加入1ml 1X PBS,将白细胞轻轻混匀,吸取上清,将白细胞放入-80℃冰箱保存。
如图3、图4所示,利用红细胞裂解液分离了18个血液样本,并对分离的白细胞进行质检。结果显示,质检指标rRNA比率以及RNA完整度指数均符合质量标准。
如果只加入一次红细胞裂解液混匀后离心,去上清,则得到的白细胞指标达不到质量要求,因此本发明方法步骤(3)需至少重复一次。且较佳的,红细胞裂解液体积为白细胞体积的3倍以上,这样实验时间短,分离效果更好。
因此,分离白细胞至少需二次加入红细胞裂解液。可以使用任意市售红细胞裂解液。本发明红细胞裂解液购自天根生化科技有限公司,货号RT-122-02。
此外,应理解,以上实施例仅用以说明本发明的技术方案,而非对本发明保护范围的限制,尽管参照较佳实施例对本发明作了详细地说明,本领域的普通技术人员应当理解,在不脱离本发明的范围和实质的情况下,可以对本发明的技术方案进行各种修改或者等同替换。
Claims (10)
1.一种白细胞的分离纯化方法,其特征在于,所述分离纯化方法中采用红细胞裂解液进行分离纯化。
2.一种白细胞的分离纯化方法,其特征在于,所述分离纯化方法包括步骤:
(1)收集血液样本;
(2)离心分离出白细胞粗提物;
(3)在白细胞粗提物中加入红细胞裂解液,充分裂解红细胞,离心去上清;
(4)重复步骤(3)至少一次,分离获得白细胞成品。
3.如权利要求2所述白细胞的分离纯化方法,其特征在于,所述步骤(3)中,加入红细胞裂解液后充分混匀,并静置5-10分钟,每3分钟上下颠倒容器多次。
4.如权利要求2所述白细胞的分离纯化方法,其特征在于,所述步骤(3)中,加入红细胞裂解液后充分混匀,静置10分钟,每3分钟上下颠倒容器5次。
5.如权利要求1-4任一权利要求所述白细胞的分离纯化方法,其特征在于,所述步骤(1)中采用EDTA抗凝管收集血液样本。
6.如权利要求1-4任一权利要求所述白细胞的分离纯化方法,其特征在于,所述分离纯化方法还包括步骤:
(5)在白细胞成品中加入PBS,将白细胞轻轻混匀,吸取上清后冻存。
7.如权利要求5所述白细胞的分离纯化方法,其特征在于,所述分离纯化方法还包括步骤:(5)在白细胞成品中加入PBS,将白细胞轻轻混匀,吸取上清后冻存。
8.如权利要求5所述白细胞的分离纯化方法,其特征在于,所述步骤(2)中,离心参数为800g,20分钟。
9.如权利要求1-4或7所述白细胞的分离纯化方法,其特征在于,所述步骤(2)中,离心参数为800g,20分钟;步骤(3)(4)中白细胞与红细胞裂解液的体积比为1:2-4。
10.如权利要求8所述白细胞的分离纯化方法,其特征在于,所述步骤(3)(4)中,离心参数为400g,5分钟。
