本发明公开了一种负载生长因子的复合缓释微粒的制备方法,包括以下步骤:由胶原蛋白和壳聚糖组成胶原蛋白‑壳聚糖支架,由壳聚糖和弹性蛋白重组体组成包裹生长因子的微型胶囊,将所述微型胶囊嵌合在所述胶原蛋白‑壳聚糖支架上制备得到产品。本发明提供一种负载生长因子的复合缓释微粒的制备方法,首先制备得到具有三维折叠结构的胶原蛋白‑壳聚糖支架,该支架的孔隙中嵌合包裹生长因子的多层微型胶囊,对其中包裹的生长因子具有长时匀速的缓释作用,从而实现生长因子主动诱导缺损处组织修复。本发明还提供了一种负载生长因子的复合缓释微粒,具有良好的生物相容性、生物降解性和极低的免疫原性,使生长因子具有长时间,均匀的释放。

技术领域
本发明涉及一种复合缓释微粒,尤其涉及一种负载生长因子的复合缓释微粒及其制备方法。
背景技术
目前,富血小板血浆(Platelet-rich plasma,简称PRP),是自体全血经离心后得到的血小板浓缩物,PRP中含有大量生长因子及蛋白质,广泛应用于临床:骨关节的修复、大面积烧伤皮肤的修复以及医美整形等领域中。
胶原蛋白是细胞外基质(ECM)的主要成份,属于结构蛋白质,约占哺乳动物蛋白质总质量的1/3,大量存在于皮肤、韧带、软骨、肌键等结缔组织或器官中。胶原具有良好的生物相容性,植入体内无毒副作用和刺激性,不易引起机体免疫反应,而且也有利于细胞的黏附、增殖和分化,加快创面愈合,并可降解而为新生组织提供足够的空间,广泛应用于生物材料的研究和开发利用中。但是单一材料存在着生物活性、强度等问题。
壳聚糖是一种天然碱性聚合电解质多糖,具有良好的生物相容性和抗菌活性,并且价格低廉。壳聚糖表面带有正电荷,能够吸附带负电的红细胞而诱导其聚集,进一步促进血小板凝血,因此被广泛应用于伤口止血和抗菌领域。
现有的负载生长因子的胶原蛋白-壳聚糖微粒作用于人体,被激活后1小时内达到90%的释放,在体外作用3天左右,在体内5天左右凋亡,作用时效短且集中,不利于持续调动和补充生长因子对创面进行修复。
发明内容
为了克服现有技术的不足,本发明的目的之一在于提供一种负载生长因子的复合缓释微粒的制备方法,制备工艺条件温和,原料易得。
本发明的目的之二在于提供一种负载生长因子的复合缓释微粒,具有良好的生物相容性、生物降解性和极低的免疫原性,对其中包裹的生长因子具有长时匀速的缓释作用,从而实现生长因子主动诱导缺损处组织修复。
本发明的目的之一采用如下技术方案实现:
一种负载生长因子的复合缓释微粒的制备方法,包括以下步骤:由胶原蛋白和壳聚糖组成胶原蛋白-壳聚糖支架,由壳聚糖和弹性蛋白重组体组成包裹生长因子的微型胶囊,将所述微型胶囊嵌合在所述胶原蛋白-壳聚糖支架上制备得到产品。
进一步地,所述微型胶囊的制备过程如下:
(1)制备弹性蛋白重组体和壳聚糖的混合溶液:取弹性蛋白重组体和中分子量壳聚糖按1:1的比例进行混合,中分子量壳聚糖的分子量为300-400KDa,脱乙酰度75-85%,用氯化钠溶液溶解;
(2)制备包裹生长因子的碳酸钙微粒:在生长因子溶液中先加入碳酸钠溶液,再加入标记生长因子的荧光素,最后加入氯化钙溶液,搅拌后沉淀一段时间,去除上清液,洗涤后得到包裹生长因子的碳酸钙微粒;
(3)将上述步骤(2)的包裹生长因子的碳酸钙微粒浸入到上述步骤(1)的混合溶液中,重复进行浸泡、洗涤过程,采用悬浮沉淀法去除上清液,得到由弹性蛋白重组体和壳聚糖包裹生长因子的微型胶囊。
进一步地,所述胶原蛋白-壳聚糖支架的制备过程如下:将胶原蛋白和低分子量壳聚糖分别配置成溶液,混合后搅拌至溶液不透明,超声去除气泡,将混合溶液冷冻后冻干,将冻干后的固体放入交联剂中浸泡交联,洗去交联剂,冷冻后冻干,即得胶原蛋白-壳聚糖支架。
进一步地,将胶原蛋白-壳聚糖支架溶解后,加入由弹性蛋白重组体和壳聚糖包裹生长因子的微型胶囊进行浸泡,冻干,即得产品。
进一步地,所述弹性蛋白重组体的制备过程如下:采用基因重组技术制备重组大肠杆菌BLR(DE3)表达弹性蛋白重组体,其中诱导温度为25℃,pH为7.0,溶氧为30%;以5g·5L-1·5h-1流速流加葡萄糖,收获培养物,通过超声破碎进行裂解,离心除去不溶性残渣,并将澄清的裂解物进行冷热循环离心,然后冷冻干燥,即得弹性蛋白重组体。弹性蛋白重组体(ELR)是经过基因工程改造的多肽,可模仿天然弹性蛋白并表现出对温度敏感性,本发明使用ELR的一级结构为通过包含L-精氨酸-甘氨酸-L-天冬氨酸(RGD)残基来表现出生物活性,同时利用负天门冬氨酸部分实现与壳聚糖的自组装。
进一步地,将胶原蛋白和壳聚糖分别溶解在3%的乙酸水溶液中,形成质量浓度分别为2-5%的胶原蛋白溶液和2-5%的壳聚糖溶液,混合时,胶原蛋白溶液和壳聚糖溶液的体积比为1:2-5。
进一步地,所述低分子量壳聚糖的分子量为100-200KDa,脱乙酰度为80-90%。
进一步地,所述生长因子为转化生长因子-β和血小板衍生生长因子-AB中的至少一种。
进一步地,所述胶原蛋白-壳聚糖支架的平均孔径为140-160um,包裹生长因子的壳聚糖-弹性蛋白重组体微型胶囊的平均直径为7-10um。
进一步地,在构建碳酸钙微粒的过程中、将包裹生长因子的碳酸钙微粒浸泡在壳聚糖弹性蛋白重组体制备微型胶囊的过程中、包裹生长因子的微型胶囊嵌合到胶原蛋白-壳聚糖支架的过程中,把每一步骤中的上清液收集起来进行荧光检测,以确定在构建和嵌合过程中生长因子蛋白的损失情况。
本发明的目的之二采用如下技术方案实现:
一种负载生长因子的复合缓释微粒,由上述方法制备得到。
相比现有技术,本发明的有益效果在于:本发明提供一种负载生长因子的复合缓释微粒的制备方法,首先制备得到具有三维折叠结构的胶原蛋白-壳聚糖支架,该支架的孔隙中嵌合包裹生长因子的多层微型胶囊,对其中包裹的生长因子具有长时匀速的缓释作用,从而实现生长因子主动诱导缺损处组织修复。将生长因子包裹在微型胶囊中,由于微型胶囊的直径更小,作用面积更大,相较于平面的凝剂对创面的作用效果更好,将制备的微型胶囊嵌合在胶原蛋白-壳聚糖支架上,一方面,微型胶囊的多层结构可以降低聚合物的流动性;另一方面,采用的弹性蛋白重组体对人体温度敏感,可以使生长因子的释放更加缓慢。本发明还提供了一种负载生长因子的复合缓释微粒,具有良好的生物相容性、生物降解性和极低的免疫原性,使生长因子具有长时间,均匀的释放。

具体实施方式
下面,结合附图以及具体实施方式,对本发明做进一步描述,需要说明的是,在不相冲突的前提下,以下描述的各实施例之间或各技术特征之间可以任意组合形成新的实施例。
实施例1
一种负载生长因子的复合缓释微粒的制备方法,包括以下步骤:
(1)制备胶原蛋白-壳聚糖支架:将胶原蛋白和分子量为100KDa,脱乙酰度为80%的壳聚糖分别溶解在3%的乙酸水溶液中,配置成质量浓度为2%的胶原蛋白溶液和2%的壳聚糖溶液,将胶原蛋白溶液搅拌过夜,壳聚糖溶液搅拌30min,以胶原蛋白溶液和壳聚糖溶液的体积比为1:2混合,搅拌至溶液不透明,超声去除气泡,将混合溶液倒入分离的培养皿中,在-40℃,冷冻24h,然后冻干,将冻干后的固体放入0.5%(w/v)的戊二醛溶液的交联浴中浸泡2小时,用去离子水冲洗,在-40℃,冷冻24h,然后冻干,即得胶原蛋白-壳聚糖支架;
(2)制备由壳聚糖和弹性蛋白重组体包裹生长因子的微型胶囊:
A:制备弹性蛋白重组体:采用基因重组技术制备重组大肠杆菌BLR(DE3)表达弹性蛋白重组体,其中诱导温度为25℃,pH为7.0,溶氧为30%;以5g·5L-1·5h-1流速流加葡萄糖,收获培养物,通过超声破碎进行裂解,离心除去不溶性残渣,并将澄清的裂解物进行冷热循环离心,冷冻干燥,即得弹性蛋白重组体;
B:制备弹性蛋白重组体和壳聚糖的混合溶液:取弹性蛋白重组体和分子量为300KDa,脱乙酰度为75%的壳聚糖按1:1的比例进行混合后,用0.15mol/L的氯化钠溶液溶解;
C:制备包裹生长因子的碳酸钙微粒:将转化生长因子-β溶于pH为7.4的磷酸盐缓冲液中,浓度为1mol/L,取上述含有转化生长因子-β的溶液先加入1mol/L碳酸钠溶液100ul,再加入标记生长因子的异硫氰酸荧光素100mg,最后加入1mol/L氯化钙溶液100ul,迅速搅拌30s后沉淀15min,去除上清液,采用超纯水洗涤后得到包裹生长因子的碳酸钙微粒;
D:将上述步骤(2)的包裹生长因子的碳酸钙微粒浸入到上述步骤B的混合溶液中,重复进行浸泡10次,每分钟最少一次,然后用0.15mol/L的氯化钠溶液(pH=5.5)洗涤,洗涤过程,采用悬浮沉淀法去除上清液,重复上述过程以增加微型胶囊层数,得到由弹性蛋白重组体和壳聚糖包裹生长因子的微型胶囊;
(3)将胶原蛋白-壳聚糖支架溶于pH为7.4的磷酸盐缓冲液中,溶解后浓度为1mol/L,然后加入1g由弹性蛋白重组体和壳聚糖包裹生长因子的微型胶囊进行浸泡,浸泡24h,冻干,即得产品。
实施例2
一种负载生长因子的复合缓释微粒的制备方法,包括以下步骤:
(1)制备胶原蛋白-壳聚糖支架:将胶原蛋白和分子量为150KDa,脱乙酰度为86%的壳聚糖分别溶解在3%的乙酸水溶液中,配置成质量浓度为3%的胶原蛋白溶液和3%的壳聚糖溶液,将胶原蛋白溶液搅拌过夜,壳聚糖溶液搅拌30min,以胶原蛋白溶液和壳聚糖溶液的体积比为1:3混合,搅拌至溶液不透明,超声去除气泡,将混合溶液倒入分离的培养皿中,在-38℃,冷冻27h,然后冻干,将冻干后的固体放入0.5%(w/v)的戊二醛溶液的交联浴中浸泡2小时,用去离子水冲洗,在-38℃,冷冻27h,然后冻干,即得胶原蛋白-壳聚糖支架;
(2)制备由壳聚糖和弹性蛋白重组体包裹生长因子的微型胶囊:
A:制备弹性蛋白重组体:采用基因重组技术制备重组大肠杆菌BLR(DE3)表达弹性蛋白重组体,其中诱导温度为25℃,pH为7.0,溶氧为30%;以5g·5L-1·5h-1流速流加葡萄糖,收获培养物,通过超声破碎进行裂解,离心除去不溶性残渣,并将澄清的裂解物进行冷热循环离心,冷冻干燥,即得弹性蛋白重组体;
B:制备弹性蛋白重组体和壳聚糖的混合溶液:取弹性蛋白重组体和分子量为350KDa,脱乙酰度为80%的壳聚糖按1:1的比例进行混合后,用0.15mol/L的氯化钠溶液溶解;
C:制备包裹生长因子的碳酸钙微粒:将血小板衍生生长因子-AB溶于pH为7.4的磷酸盐缓冲液中,浓度为1mol/L,向上述含血小板衍生生长因子-AB的溶液中先加入1mol/L碳酸钠溶液100ul,再加入标记生长因子的异硫氰酸荧光素100mg,最后加入1mol/L氯化钙溶液100ul,迅速搅拌30s后沉淀15min,去除上清液,采用超纯水洗涤后得到包裹生长因子的碳酸钙微粒;
D:将上述步骤(2)的包裹生长因子的碳酸钙微粒浸入到上述步骤B的混合溶液中,重复进行浸泡10次,每分钟最少一次,然后用0.15mol/L的氯化钠溶液(pH=5.5)洗涤,洗涤过程,采用悬浮沉淀法去除上清液,重复上述过程以增加微型胶囊层数,得到由弹性蛋白重组体和壳聚糖包裹生长因子的微型胶囊;
(3)将胶原蛋白-壳聚糖支架溶于pH为7.4的磷酸盐缓冲液中,溶解后浓度为1mol/L,然后加入1g由弹性蛋白重组体和壳聚糖包裹生长因子的微型胶囊进行浸泡,浸泡24h,冻干,即得产品。
实施例3
一种负载生长因子的复合缓释微粒的制备方法,包括以下步骤:
(1)制备胶原蛋白-壳聚糖支架:将胶原蛋白和分子量为200KDa,脱乙酰度为90%的壳聚糖分别溶解在3%的乙酸水溶液中,配置成质量浓度为5%的胶原蛋白溶液和5%的壳聚糖溶液,将胶原蛋白溶液搅拌过夜,壳聚糖溶液搅拌30min,以胶原蛋白溶液和壳聚糖溶液的体积比为1:5混合,搅拌至溶液不透明,超声去除气泡,将混合溶液倒入分离的培养皿中,在-35℃,冷冻30h,然后冻干,将冻干后的固体放入0.5%(w/v)的戊二醛溶液的交联浴中浸泡2小时,用去离子水冲洗,在-35℃,冷冻30h,然后冻干,即得胶原蛋白-壳聚糖支架;
(2)制备由壳聚糖和弹性蛋白重组体包裹生长因子的微型胶囊:
A:制备弹性蛋白重组体:采用基因重组技术制备重组大肠杆菌BLR(DE3)表达弹性蛋白重组体,其中诱导温度为25℃,pH为7.0,溶氧为30%;以5g·5L-1·5h-1流速流加葡萄糖,收获培养物,通过超声破碎进行裂解,离心除去不溶性残渣,并将澄清的裂解物进行冷热循环离心,冷冻干燥,即得弹性蛋白重组体;
B:制备弹性蛋白重组体和壳聚糖的混合溶液:取弹性蛋白重组体和分子量为400KDa,脱乙酰度为85%的壳聚糖按1:1的比例进行混合后,用0.15mol/L的氯化钠溶液溶解;
C:制备包裹生长因子的碳酸钙微粒:将转化生长因子-β和血小板衍生生长因子-AB溶于pH为7.4的磷酸盐缓冲液中,浓度为1mol/L,取上述溶液先加入1mol/L碳酸钠溶液100ul,再加入标记生长因子的异硫氰酸荧光素100mg,最后加入1mol/L氯化钙溶液100ul,迅速搅拌30s后沉淀15min,去除上清液,采用超纯水洗涤后得到包裹生长因子的碳酸钙微粒;
D:将上述步骤(2)的包裹生长因子的碳酸钙微粒浸入到上述步骤B的混合溶液中,重复进行浸泡10次,每分钟最少一次,然后用0.15mol/L的氯化钠溶液(pH=5.5)洗涤,洗涤过程,采用悬浮沉淀法去除上清液,重复上述过程以增加微型胶囊层数,得到由弹性蛋白重组体和壳聚糖包裹生长因子的微型胶囊;
(3)将胶原蛋白-壳聚糖支架溶于pH为7.4的磷酸盐缓冲液中,溶解后浓度为1mol/L,然后加入1g由弹性蛋白重组体和壳聚糖包裹生长因子的微型胶囊进行浸泡,浸泡24h,冻干,即得产品。
试验例
在15kv的加速电压下,使用扫描电子显微镜(SEM;Philips XL30)来测量实施例1中制备的胶原蛋白-壳聚糖的平均孔径,结果如图1所示,胶原蛋白-壳聚糖支架的平均孔径为140-160um。
使用溅射涂布机(Enitech K450X;英国)在胶原蛋白-壳聚糖支架上涂上一层薄薄的金,再次使用扫描电子显微镜观察支架的横截面的孔径,可以看到具有三维折叠结构的胶原蛋白-壳聚糖支架,结果如图2所示。
在10kv的加速电压下,使用扫描电子显微镜(SEM;Philips XL30)来测量包裹生长因子的壳聚糖-弹性蛋白重组体的微型胶囊的平均粒径,结果如图3所示,平均粒径为7-10um。
使用阳离子染料若丹明B(10-6M)对微型胶囊进行染色,在Olympus FV1000共聚焦显微镜(德国)下检测各层微型胶囊在胶原蛋白-壳聚糖支架的孔隙中排列方式,以及形态嵌合。所有样品均稳定在25或37℃并在水合条件下观察。结果如图4所示。
将实施例1的产品重悬于25℃和37℃的PBS中,缓慢搅拌进行释放实验,每隔24h将PBS吸出,加入新的PBS,从第一天开始计算,每隔48h使用酶标仪检测回收PBS中荧光,来确定生长因子释放量,结果如图5所示,由图5可以看出,在25℃的环境中,第10天,生长因子的释放量可以达到60%;在第21天,生长因子的释放量可以达到80%,进一步说明本申请的产品,一方面,微型胶囊的多层结构可以降低聚合物的流动性;另一方面,采用的弹性蛋白重组体对人体温度敏感,可以使生长因子的释放更加缓慢。
上述实施方式仅为本发明的优选实施方式,不能以此来限定本发明保护的范围,本领域的技术人员在本发明的基础上所做的任何非实质性的变化及替换均属于本发明所要求保护的范围。
Claims (10)
1.一种负载生长因子的复合缓释微粒的制备方法,其特征在于,包括以下步骤:由胶原蛋白和壳聚糖组成胶原蛋白-壳聚糖支架,由壳聚糖和弹性蛋白重组体组成包裹生长因子的微型胶囊,将所述微型胶囊嵌合在所述胶原蛋白-壳聚糖支架上制备得到产品。
2.根据权利要求1所述一种负载生长因子的复合缓释微粒的制备方法,其特征在于,所述微型胶囊的制备过程如下:
(1)制备弹性蛋白重组体和壳聚糖的混合溶液:取弹性蛋白重组体和中分子量壳聚糖按1:1的比例进行混合,中分子量壳聚糖的分子量为300-400KDa,脱乙酰度75-85%,用氯化钠溶液溶解;
(2)制备包裹生长因子的碳酸钙微粒:在生长因子溶液中先加入碳酸钠溶液,再加入标记生长因子的荧光素,最后加入氯化钙溶液,搅拌后沉淀一段时间,去除上清液,洗涤后得到包裹生长因子的碳酸钙微粒;
(3)将上述步骤(2)的包裹生长因子的碳酸钙微粒浸入到上述步骤(1)的混合溶液中,重复进行浸泡、洗涤过程,采用悬浮沉淀法去除上清液,得到由弹性蛋白重组体和壳聚糖包裹生长因子的微型胶囊。
3.根据权利要求2所述一种负载生长因子的复合缓释微粒的制备方法,其特征在于,所述胶原蛋白-壳聚糖支架的制备过程如下:将胶原蛋白和低分子量壳聚糖分别配置成溶液,混合后搅拌至溶液不透明,超声去除气泡,将混合溶液冷冻后冻干,将冻干后的固体放入交联剂中浸泡交联,洗去交联剂,冷冻后冻干,即得胶原蛋白-壳聚糖支架。
4.根据权利要求3所述一种负载生长因子的复合缓释微粒的制备方法,其特征在于,将胶原蛋白-壳聚糖支架溶解后,加入由弹性蛋白重组体和壳聚糖包裹生长因子的微型胶囊进行浸泡,冻干,即得产品。
5.根据权利要求1所述一种负载生长因子的复合缓释微粒的制备方法,其特征在于,所述弹性蛋白重组体的制备过程如下:采用基因重组技术制备重组大肠杆菌BLR(DE3)表达弹性蛋白重组体,其中诱导温度为25℃,pH为7.0,溶氧为30%;以5g·5L-1·5h-1流速流加葡萄糖,收获培养物,通过超声破碎进行裂解,离心除去不溶性残渣,并将澄清的裂解物进行冷热循环离心,然后冷冻干燥,即得弹性蛋白重组体。
6.根据权利要求3所述一种负载生长因子的复合缓释微粒的制备方法,其特征在于,将胶原蛋白和壳聚糖分别溶解在3%的乙酸水溶液中,形成质量浓度分别为2-5%的胶原蛋白溶液和2-5%的壳聚糖溶液,混合时,胶原蛋白溶液和壳聚糖溶液的体积比为1:2-5。
7.根据权利要求3所述一种负载生长因子的复合缓释微粒的制备方法,其特征在于,所述低分子量壳聚糖的分子量为100-200KDa,脱乙酰度为80-90%。
8.根据权利要求1所述一种负载生长因子的复合缓释微粒的制备方法,其特征在于,所述生长因子为转化生长因子-β和血小板衍生生长因子-AB中的至少一种。
9.根据权利要求1所述一种负载生长因子的复合缓释微粒的制备方法,其特征在于,所述胶原蛋白-壳聚糖支架的平均孔径为140-160um,包裹生长因子的壳聚糖-弹性蛋白重组体微型胶囊的平均直径为7-10um。
10.一种负载生长因子的复合缓释微粒,其特征在于,由权利要求1至9任一项所述方法制备得到。
