本发明涉及金线莲的培育领域,特别是一种富硒金线莲的培养基及其培育方法。
背景技术
金线莲又名金线兰,金丝草,是一种具有很高药用价值的中草药,在生活中的使用范围广。金线莲中含有多种糖类,牛磺酸,生物碱,以及多种微量元素和多种氨基酸等对人体有益的物质。
农家肥是很好的有机肥,但一定要堆放腐熟一年以上才能施用。可用这种肥液兑水10-20倍浇施,尽量施在盆沿处,不接触根部,不沾兰叶。施用化肥时,要注意氮、磷、钾的平衡,并严格控制浓度,叶艺苗要控制叶绿素的合成,不宜施氮肥,也不可施含锰、镁的肥料(如钙镁磷肥等),因为锰、镁元素能加速叶绿素的合成对花艺兰,如果培育大瓣型花,可适量增施氮肥。
以叶面喷肥为主,叶面喷肥既可通过兰叶吸收补充兰株的营养,又不会使肥接触根部引起肥害,对兰花来说是一种最好的施肥方式。只要在生长发育期,坚持10天左右进行一次叶面喷肥,完全可以养好兰花,不需要往盆里施肥。可用1∶1000的磷酸二氢钾和1∶1000的硼肥,也可用芦荟液。因为芦荟中含有大量的多糖和氨基酸等营养物质,并含有二十多种矿物质,可满足兰花所需要的养分,且芦荟里含有遥芦荟酊几乎可杀死所有的病菌和病毒,施用后无需再施农药防病。
施肥要勤施薄施,少吃多餐:兰花在发育生长期需要的肥料不多,对生长旺盛的健壮兰株,可每隔10-15天施一次稀薄液。
炭疽杆菌是一种很大的革兰阳性杆菌,产芽孢,可在普通培养基上需氧或厌氧条件下培养,在土壤中寄居于世界各地。大蒜油:具有高仿类硫醚化合物,真菌、细菌通杀,气味对空气中病菌孢子有直接杀灭作用,对粉虱、蛾类等害虫有趋避作用。

一种富硒金线莲的培养基,按照质量份数包括:MS培养基50-60份,大蒜油0.5-1份,黄芩提取物2-5份,硒营养剂0.3-0.6份,碱性化肥1-2份。

富硒金线莲的培养基按照质量份数包括:MS培养基50-60份,大蒜油0.5-1份,黄芩提取物2-5份,硒营养剂0.3-0.6份,碱性化肥1-2份;
在温度为16-24℃,湿度≥70%,光照度≤3000勒克斯的环境下,培育120天;
移栽到移栽基质中30-100天,得到含硒金线莲。
前述的一种富硒金线莲的培育方法,组培培养基的配方包括:86%MS培养基,6%马铃薯,8%香蕉。
前述的一种富硒金线莲的培育方法,移栽基质按照质量百分比配方包括:54%炭泥土,42%珍珠岩,2%发酵后的农家肥,1.4%硒营养剂,0.6%硫酸亚铁。
本发明的有益之处在于:
本发明的富硒培养基通过黄芩提取物和大蒜油的配合能够达到灭菌驱虫效果,提高金线莲的产量;
富硒培养基的碱性化肥的选用促进金线莲初期根的生长;
富硒培养基的的硒营养剂的选用提高硒转化率,产出的金线莲含硒量高,质量好。
移栽基质中添加硫酸亚铁,使得基质偏酸性,适合金线莲生长。
具体实施方式
以下结合具体实施例对本发明作具体的介绍。
一种富硒金线莲的培育方法,包括如下内容:
将金线莲茎在组培培养基中培育成苗,待长出根后培育到富硒金线莲的培养基;
作为一种实施例,组培培养基的配方包括:86%MS培养基,6%马铃薯,8%香蕉。组培培养基的配方不受限制,一般组培培养基都可以使用,还可以在培养基中加入生长因子等。
富硒金线莲的培养基,按照质量份数包括:MS培养基50-60份,大蒜油0.5-1份,黄芩提取物2-5份,硒营养剂0.3-0.6份,碱性化肥1-2份。作为一种实施例,硒营养剂包括:亚硒酸盐,硒蛋氨酸;可以是单独使用或者是混合使用。作为一种实施例,碱性化肥包括:氧化钙,硝酸钙,硝酸钠,碳酸氢铵、硫酸钾;富硒培养基的碱性化肥的选用促进金线莲初期根的生长。
黄芩提取物的提取方法为:取黄芩药材,除去杂质,洗净,隔水蒸制半小时,切片,用乙醇煮,蒸馏提取得到黄芩提取物。
在温度为16-24℃,湿度≥70%,光照度≤3000勒克斯的环境下,培育120天;
移栽到移栽基质中30-100天,得到含硒金线莲。金线莲中有机硒结合到氨基酸中,取代含S氨基酸中的S,以蛋白态有机硒形态存在,包括Se-甲基-硒半胱氨酸(MSeC)和硒胱氨酸。
作为一种实施例,移栽基质按照质量百分比配方包括:54%炭泥土,42%珍珠岩,2%发酵后的农家肥,1.4%硒营养剂,0.6%硫酸亚铁;移栽基质中添加硫酸亚铁,使得基质偏酸性,适合金线莲生长。
以下实验验证本发明的效果;
实验一,
实施例1:培育方法,包括如下步骤:
将金线莲茎在组培培养基中培育成苗,待长出根后培育到富硒金线莲的培养基;
组培培养基的配方包括:86%MS培养基,6%马铃薯,8%香蕉;
富硒金线莲的培养基,包括:MS培养基50g,大蒜油0.5g,黄芩提取物2g,硒营养剂0.3g(质量比为:2亚硒酸盐:1硒蛋氨酸),硝酸钙1g。
在温度为18℃,湿度70%,光照度3000勒克斯的环境下,培育120天;
移栽到移栽基质中30-100天,得到含硒金线莲。移栽基质配方包括:54%炭泥土,42%珍珠岩,2%发酵后的农家肥,1.4%硒营养剂,0.6%硫酸亚铁。培育环境为:温度为26℃,湿度80%,光照度3000勒克斯。
实施例2:只有富硒金线莲的培养基与实施例1有区别,其他都一样;
富硒金线莲的培养基,包括:MS培养基54g,大蒜油0.8g,黄芩提取物4g,硒营养剂0.5g(质量比为:2亚硒酸盐:1硒蛋氨酸),碳酸氢铵1g。
实施例3:只有富硒金线莲的培养基与实施例1有区别,其他都一样;
富硒金线莲的培养基,包括:MS培养基56g,大蒜油0.7g,黄芩提取物4g,硒营养剂(质量比为:2亚硒酸盐:1硒蛋氨酸)0.5g,硝酸钙1.4g。
实施例4:只有富硒金线莲的培养基与实施例1有区别,其他都一样;
富硒金线莲的培养基,包括:MS培养基60g,大蒜油1g,黄芩提取物5g,硒营养剂0.6g(质量比为:2亚硒酸盐:1硒蛋氨酸),碳酸氢铵2g。
对比实施例1:只有富硒金线莲的培养基与实施例3有区别,其他都一样,配方区别在于没有大蒜油;
富硒金线莲的培养基,包括:MS培养基56g,黄芩提取物4g,硒营养剂(质量比为:2亚硒酸盐:1硒蛋氨酸)0.5g,硝酸钙1.4g。
对比实施例2:只有富硒金线莲的培养基与实施例3有区别,其他都一样,配方区别在于没有黄芩提取物;
富硒金线莲的培养基,包括:MS培养基56g,大蒜油0.7g,硒营养剂(质量比为:2亚硒酸盐:1硒蛋氨酸)0.5g,硝酸钙1.4g。
每个实施例为一列7颗金线莲,一共6列,按照上面的方法培育后,摘取叶片,称取重量,观察虫眼严重程度(0-1个为低,2-8为中,9-15为高)。

结果分析:由此可知,大蒜油和黄芩提取物针对驱虫除菌具有协同作用,最优实施例为实施例3。

对比实施例3:只有富硒金线莲的培养基与实施例3有区别,其他都一样,区别在于硒营养剂不同;
富硒金线莲的培养基,包括:MS培养基56g,大蒜油0.7g,黄芩提取物4g,硒营养剂(亚硒酸盐)0.5g,硝酸钙1.4g。
对比实施例4:只有富硒金线莲的培养基与实施例3有区别,其他都一样,区别在于硒营养剂不同;
富硒金线莲的培养基,包括:MS培养基56g,大蒜油0.7g,黄芩提取物4g,硒营养剂(硒蛋氨酸)0.5g,硝酸钙1.4g。
对比实施例5:
只有富硒金线莲的培养基与实施例3有区别,其他都一样,区别在于硒营养剂不同;

只有富硒金线莲的培养基与实施例3有区别,其他都一样,区别在于硒营养剂不同;
富硒金线莲的培养基,包括:MS培养基56g,大蒜油0.7g,黄芩提取物4g,硒营养剂(质量比为:1亚硒酸盐:2硒蛋氨酸)0.5g,硝酸钙1.4g。
每个实施例为一列7颗金线莲,一共9列,按照上面的方法培育后,摘取叶片烘干,检测金线莲干品(DW)硒(有机硒)的含量,计算平均每株的有机硒的含量。

结果分析:由此可知,亚硒酸盐和硒蛋氨酸配合使用具有更好的硒转化率,最优质量比为:2亚硒酸盐:1硒蛋氨酸。作为一种优选,富硒金线莲的培养基,按照质量份数包括:MS培养基56份,大蒜油0.7份,黄芩提取物4份,硒营养剂0.5份,碱性化肥1.4份。

1.一种富硒金线莲的培养基,其特征在于,按照质量份数包括:MS培养基 50-60份,大蒜油 0.5-1份,黄芩提取物 2-5份,硒营养剂 0.3-0.6份,碱性化肥 1-2份;所述硒营养剂按照质量比为:2亚硒酸盐: 1硒蛋氨酸。
2.根据权利要求1所述的一种富硒金线莲的培养基,其特征在于,所述碱性化肥为氧化钙、硝酸钙、硝酸钠、碳酸氢铵或硫酸钾。
3.根据权利要求1所述的一种富硒金线莲的培养基,其特征在于,按照质量份数包括:MS培养基 56份,大蒜油 0.7份,黄芩提取物 4份,硒营养剂 0.5份,碱性化肥 1.4份。
4.一种富硒金线莲的培育方法,其特征在于,包括如下内容:
将金线莲茎在组培培养基中培育成苗,待长出根后培育到富硒金线莲的培养基;
所述富硒金线莲的培养基按照质量份数包括:MS培养基 50-60份,大蒜油 0.5-1份,黄芩提取物 2-5份,硒营养剂 0.3-0.6份,碱性化肥 1-2份;
所述硒营养剂按照质量比为:2亚硒酸盐: 1硒蛋氨酸;
在温度为16-24℃,湿度≥70%,光照度≤3000勒克斯的环境下,培育120天;
移栽到移栽基质中30-100天,得到含硒金线莲。
