本发明公开了一种神经细胞网络化学信号和电信号同步监测方法,首先在经预处理后的微电极阵列上原位培养神经细胞网络;后对神经元囊泡进行染色处理,后加入缓冲液;采用全内反射荧光显微镜观察神经细胞网络,将拍摄相机、微电极阵列的控制器与脉冲信号发生器相连;对神经细胞网络进行外界刺激,同时开启脉冲信号发生器,拍摄相机和微电极阵列的控制器分别同步收集荧光信号和电信号。本发明的同步监测方法背景信号低,信噪比高,具有时空分辨响应性,可以满足神经细胞网络监测要求;本方法信号稳定、灵敏度高、可针对多种神经细胞及网络实现成像测量,并可以实现高通量分析。

技术领域
本发明涉及一种神经细胞的检测方法,特别是涉及一种神经细胞网络化学信号和电信号同步监测方法。
背景技术
探索神经系统的结构与信息处理机制,揭示高级神经活动的本质具有较为深远的一一。神经系统的所有高级功能实现都离不开信号的产生、传递、处理和储存。神经元,又称神经细胞,是构成神经系统结构和功能的基本单位,其相互作用连接构成了一个极其复杂的网络系统,通过电信号和化学信号传递条件生物体的行为。神经元集群间信息传递机制的研究接近生命存成的本质,具有重大科学意义和应用价值,其难度也非常大。
神经元细胞网络的信号传递研究包括电生理机制研究及神经递质传递研究。电生理学研究主要依靠微机电制造技术(MEMS)。目前结合MEMS技术的半导体芯片最具代表性的是场效应管和微电极阵列。场效应管虽然灵敏度高,但制备工艺复杂,成品率低,使用寿命短。应用MEMS技术设计制造的微电极阵列在细胞电生理的研究中应用更为广泛。神经递质传递研究主要应用微电极技术和荧光显微成绩技术。微电极电化学检测灵敏度和时间分辨率高,但缺乏空间分辨率。而荧光成像可提供更高的空间分辨率。
目前现有技术对于细胞网络电信号和化学信号传递过程的研究是相对独立的。然而要了解神经元集群间信息传递机制,只有将两种信息合并,才可获得神经活动的全貌。然而,目前未见有电信号与化学信号同步监测的报导。
发明内容
发明目的:本发明的目的是提供一种神经细胞网络化学信号和电信号同步监测方法,能够同步监测神经细胞网络化学信号和电信号,监测的空间分辨率高;通过联用超薄微电极阵列与全内反射荧光显微镜,实现了对神经细胞网络动作电位和神经递质传输的同步测量。
技术方案:本发明的神经细胞网络化学信号和电信号同步监测方法,包括如下步骤:
(1)在经预处理后的微电极阵列上原位培养神经细胞网络;
(2)对神经元囊泡进行染色处理,后加入缓冲液;缓冲液可以采用磷酸盐缓冲液;
(3)采用全内反射荧光显微镜观察神经细胞网络,将拍摄相机、微电极阵列的控制器与脉冲信号发生器相连;
(4)对神经细胞网络进行外界刺激,同时开启脉冲信号发生器,拍摄相机和微电极阵列的控制器分别同步收集荧光信号和电信号。
其中神经细胞为分化后生成的细胞网络;为了对两种信息进行同步监测,本发明采用一台脉冲信号发生器,同时发出两路TTL电平信号对微电极阵列控制器和拍摄相机进行触发。通过脉冲信号发生器的设置,以信号发生器触发微电极阵列(MEA)和拍摄相机,使得同步记录过程延时小于5ps。
进一步地,为了将收集到的信号用以研究神经信息传输机制;还包括采用低通滤波器对电信号进行处理,收集动作电位信号;分析荧光假神经递质释放信号与动作电位信号的同步响应性。
优选地,所述步骤(2)的染色处理采用的荧光分子为荧光假神经递质FFN102。
优选地,所述步骤(1)的预处理包括对微电极阵列进行化学修饰处理;使得神经细胞可在微电极阵列的表面进行生长。
优选地,所述外界刺激为化学刺激或电刺激;可以采用向微电极阵列储液池中加入厌氧缓冲液、高K+溶液或采用电脉冲等激发方式。
优选地,所述微电极阵列的厚度为微米级。
优选地,为了根据电极和神经细胞的不同,选取可是的滤波器频率进行处理;采用3000~5000Hz低通滤波器对电信号进行处理,阈值设定为50~200μV;取所有幅值大于50μV的电信号事件进行统计分析。
优选地,在微电极阵列的电极表面涂布纤维粘连蛋白、层粘连蛋白、玻连蛋白中的任意一种。
优选地,所述化学刺激为加入厌氧溶液。在原溶液的基础上外加厌氧溶液,使厌氧溶液和原溶液的体积比为1∶1、2∶1、3∶1,使其形成50%~75%的厌氧比率环境;其中厌氧溶液是在普通缓冲溶液中通氮气除氧得到。
优选地,所述拍摄相机为科研级互补金属氧化物半导体相机。
本发明以微电极阵列(microelectrode arrays)联用全内反射荧光显微镜(total-internal reflection fluorescence microscopy)系统为检测平台,在微阵列电极表面利用神经生长因子培育神经细胞网络,采用荧光假神经递质标记神经细胞囊泡;当神经细胞受到外界刺激时,胞体和突触产生的电脉冲信号通过微阵列电极进行收集,神经递质释放经由全内反射荧光显微镜进行成像,可获得同步测量结果;可以实现对同一神经细胞网络中的动作电位脉冲与神经递质的同步测量。
有益效果:与现有技术相比:
(1)本发明采用荧光假神经递质染色神经细胞网络,通过全内反射荧光显微成像收集信号,极大的提高了监测的空间分辨率;而现有技术中的神经递质传输监测方法,主要采用微电极伏安法,这种方法缺乏空间分辨率;
(2)本发明联用超薄微电极阵列与全内反射荧光显微镜,以脉冲信号发生器触发,同步记录过程延时小于5ps,首次实现了神经网络电信号与化学信号的同步监测;并且在联用这两种技术的同时保持了其灵敏度和分辨率,获取精准的神经传递信息,并同步信息收集过程;
(3)本发明的同步监测方法背景信号低,信噪比高,具有时空分辨响应性,可以满足神经细胞网络监测要求;本方法信号稳定、灵敏度高、可针对多种神经细胞及网络实现成像测量,并可以实现高通量分析。

具体实施方式
本实施例中使用的水均为二次蒸馏水,所用试剂包括荧光假神经递质FFN102、多聚赖氨酸、神经生长因子(NGF)、细胞培养液、纤维粘连蛋白、层粘连蛋白等均为市售;所用细胞为鼠嗜铬细胞瘤细胞、细胞实验所用缓冲溶液为pH值为7.4的磷酸盐缓冲液均为市售。
实施例1:
本实施例采用厚度为180μm的超薄8×8微电极阵列,型号为60ThinMEA200/30iR-ITO,布局如图1所示;图1所示的中部位置为电极,REF为参比电极,环绕在四周的是引脚,其中电极编号第一位为列数,第二位为行数;其中单个电极直径10μm,间距30μm,电极电阻250~400kΩ,电极材料为TiN;微电极阵列的表面设有储液池,内径为24mm,高度为12mm。
本实施例的检测装置如图2所示,将微电极阵列1安装于全内反射荧光显微镜的载物台上,使微电极阵列1位于全内反射物镜3的正上方;微电极阵列1采集的电信号经由微电极阵列的控制器2收集,神经递质释放信息通过科研级互补金属氧化物半导体(SCMOS)相机4进行拍摄。为了对两种信息进行同步,采用一台脉冲信号发生器5,同时发出两路TTL电平信号对微电极阵列控制器2和SCMOS相机4进行触发。
采用上述微电极阵列联用全内反射荧光显微镜装置对神经细胞网络化学信号和电信号进行监测的具体步骤如下:
(1)对微电极阵列进行预处理:首先将微电极阵列在乙醇溶液中润洗20分钟,随后空气中风干;后采用75%的乙醇进行消毒处理,自然风干后用紫外灯照射6小时;在微电极储液池中加入0.1mg/ml多聚赖氨酸溶液,在37℃,5%二氧化碳环境中静置1小时;然后将微电极阵列用蒸馏水彻底清洗;最后,采用化学修饰法,将纤维粘连蛋白置于储液池中,4℃下静置过夜,则该电极阵列可用于细胞培养;
(2)在微电极阵列表面构建神经网络模型:将浓度为105cells/ml的未分化的鼠嗜铬细胞瘤细胞(PC12)置于微电极阵列储液池,加入50ng/ml神经生长因子(NGF)培育6天,使其生长成网络结构;
(3)染色处理:在培育好神经细胞网络的微电极阵列储液池中加入10μM荧光假神经递质(FFN102),孵育30分钟,随后水洗3次备用;
(4)在微电极阵列储液池中加入pH值为7.4的磷酸盐缓冲液,启动微电极阵列控制器,记录细胞网络自发动作电位,获得结果如图3所示;图3中每个小图的左上角的数字为电极编号,第一位为列数,第二位为行数;
(5)将微电极阵列安装于全内反射荧光显微镜载物台,通过全内反射模块采用405nm荧光光源激发。在微电极阵列储液池中加入厌氧溶液刺激神经细胞,厌氧溶液与原溶液的体积比为3∶1;其中厌氧溶液是在普通缓冲溶液中通氮气除氧30min后得到的;同时启动脉冲信号发生器,同步触发SCMOS相机与微电极阵列控制器,记录荧光信号与电信号;所得神经递质释放图像如图4所示;图4中的右下角数字均为时间,单位是秒,均以左上角的5μm为标尺;
(6)采用5000Hz低通滤波器对电信号进行处理,阈值设定为50μV。取所有幅值大于50μV的电信号事件进行统计分析;
(7)统计在厌氧刺激下60秒内神经递质释放事件与动作电位发生事件,所得结果如图5所示,其中图5(a)为荧光假神经递质释放事件随时间变化的分布,图5(b)为动作电位发生事件在60秒内的统计分布。在加入刺激后2~5秒,SCMOS相机捕捉到明显的荧光释放事件,在30秒内以较高的频率发生,随后频率迅速下降。同时,微电极阵列在5秒左右开始收集到大量动作电位发生事件,电信号的发生频率在30秒之后也迅速下降。该监测结果说明在厌氧刺激作用下,神经递质释放与动作电位发生具有的明显关联机制。
实施例2:
本实施例采用与实施例1相同型号的微电极阵列,检测装置与实施例1相同;采用上述微电极阵列联用全内反射荧光显微镜装置对神经细胞网络化学信号和电信号进行监测的具体步骤如下:
(1)对微电极阵列进行预处理:首先将微电极阵列在乙醇溶液中润洗20分钟,随后空气中风干;后采用75%的乙醇进行消毒处理,自然风干后用紫外灯照射6小时;在微电极储液池中加入0.1mg/ml多聚赖氨酸溶液,在37℃,5%二氧化碳环境中静置1小时;然后将微电极阵列用蒸馏水彻底清洗;最后,采用化学修饰法,将层粘连蛋白置于储液池中,4℃下静置过夜,则该电极阵列可用于细胞培养;
(2)在微电极阵列表面构建神经网络模型:将浓度为105cells/ml的未分化的鼠嗜铬细胞瘤细胞(PC12)置于微电极阵列储液池,加入50ng/ml神经生长因子培育6天,使其生长成网络结构;
(3)染色处理:在培育好神经细胞网络的微电极阵列储液池中加入10μM荧光假神经递质(FFN102),孵育30分钟,随后水洗3次备用;
(4)在微电极阵列储液池中加入pH值为7.4的磷酸盐缓冲液,启动微电极阵列控制器,记录细胞网络自发动作电位;
(5)将微电极阵列安装于全内反射荧光显微镜载物台,通过全内反射模块采用405nm荧光光源激发。在微电极阵列储液池中加入厌氧溶液刺激神经细胞,厌氧溶液与原溶液的体积比为2∶1;其中厌氧溶液是在普通缓冲溶液中通氮气除氧30min后得到的;同时启动脉冲信号发生器,同步触发SCMOS相机与微电极阵列控制器,记录荧光信号与电信号;
(6)采用4000Hz低通滤波器对电信号进行处理,阈值设定为100μV;取所有幅值大于50μV的电信号事件进行统计分析;
(7)统计在厌氧刺激下60秒内神经递质释放事件与动作电位发生事件。

1.一种神经细胞网络化学信号和电信号同步监测方法,其特征在于包括如下步骤:
(1)在经预处理后的微电极阵列上原位培养神经细胞网络;
(2)对神经元囊泡进行染色处理,后加入缓冲液;

(4)对神经细胞网络进行外界刺激,同时开启脉冲信号发生器,拍摄相机和微电极阵列的控制器分别同步收集荧光信号和电信号。
2.根据权利要求1所述的神经细胞网络化学信号和电信号同步监测方法,其特征在于:还包括采用低通滤波器对电信号进行处理,收集动作电位信号;分析荧光假神经递质释放信号与动作电位信号的同步响应性。
3.根据权利要求1所述的神经细胞网络化学信号和电信号同步监测方法,其特征在于:所述步骤(2)的染色处理采用的荧光分子为荧光假神经递质FFN102。
4.根据权利要求1所述的神经细胞网络化学信号和电信号同步监测方法,其特征在于:所述步骤(1)的预处理包括对微电极阵列进行化学修饰处理。
5.根据权利要求1所述的神经细胞网络化学信号和电信号同步监测方法,其特征在于:所述外界刺激为化学刺激或电刺激。
6.根据权利要求1所述的神经细胞网络化学信号和电信号同步监测方法,其特征在于:所述微电极阵列的厚度为微米级。
7.根据权利要求2所述的神经细胞网络化学信号和电信号同步监测方法,其特征在于:采用3000~5000Hz低通滤波器对电信号进行处理,阈值设定为50~200μV;取所有幅值大于50μV的电信号事件进行统计分析。
8.根据权利要求4所述的神经细胞网络化学信号和电信号同步监测方法,其特征在于:所述化学修饰处理包括在微电极阵列的电极表面涂布纤维粘连蛋白、层粘连蛋白、玻连蛋白中的任意一种。
9.根据权利要求5所述的神经细胞网络化学信号和电信号同步监测方法,其特征在于:所述化学刺激为加入厌氧溶液。
10.根据权利要求1所述的神经细胞网络化学信号和电信号同步监测方法,其特征在于:所述拍摄相机为科研级互补金属氧化物半导体相机。
