Abstract
The invention relates to a method for producing 2, 3-butanediol by utilizing carbohydrate waste, which comprises the following steps: s1, obtaining sludge containing soluble protein SPN, and digesting and pretreating the sludge by microwaves, wherein the treatment conditions are as follows: heating to 100 deg.C for 15min, and maintaining for 0-20 min; s2, mixing the carbohydrate waste and the pretreated sludge according to different mass ratios, placing the mixture into a reaction container, adding water to enable the mass concentration of a substrate to be 2% -5%, stirring and reacting at the constant temperature of 35 ℃, and adjusting the pH of reactants in the reaction container every 12 hours to enable the pH to be 6. The method promotes the carbohydrate garbage to produce the 2, 3-butanediol by adding the sludge, thereby solving the problem of organic solid waste in the environment and carrying out resource utilization.
Description
Technical Field
The invention belongs to the technical field of fermentation in bioengineering, and particularly relates to a method for producing 2, 3-butanediol by using carbohydrate waste.
Background
2, 3-butanediol is a chemical raw material and liquid fuel with great value and is widely applied to the fields of chemical industry, food, fuel and aerospace; the chemical synthesis method for producing the 2, 3-butanediol has the disadvantages of complicated process and high cost, and fossil resources are increasingly in short supply on the earth, and the petroleum resource supply is unstable, so that the large-scale production is difficult to realize. The method for producing the 2, 3-butanediol by using the biological fermentation method is a promising production mode, uses renewable biomass resources as raw materials, and is concerned by countries in the world.
The raw materials used in the biological fermentation method consist of a carbon source and a nitrogen source. The production method of cheap substrate is developed and utilized, so that the production cost can be greatly reduced. With the widespread use of the activated sludge process in sewage treatment plants, the amount of excess sludge increases year by year, which has become a great burden in the urbanization process of China. The protein content in the residual sludge is rich and can be used as a nitrogen source. With the rapid development of economic society and the increasing living standard of people, the amount of domestic garbage is also increased. The household garbage has various types, wherein the household garbage belongs to a part capable of being biologically utilized and does not consist of three parts of protein, carbohydrate and grease. Both of these materials are organic solid wastes that are urgently needed to be disposed of in the environment.
Therefore, based on the problems, the method has important practical significance for promoting the carbohydrate waste to produce the 2, 3-butanediol by adding the sludge, thereby solving the problem of organic solid waste in the environment and carrying out resource utilization.
Disclosure of Invention
The invention aims to overcome the defects of the prior art and provide a method for promoting carbohydrate garbage to produce 2, 3-butanediol by adding sludge, thereby solving the problem of organic solid waste in the environment and carrying out resource utilization.
The technical problem to be solved by the invention is realized by adopting the following technical scheme:

s1, obtaining sludge containing soluble protein SPN, and digesting and pretreating the sludge by microwaves, wherein the treatment conditions are as follows: heating to 100 deg.C for 15min, and maintaining for 0-20 in;
s2, mixing the carbohydrate waste and the pretreated sludge according to different mass ratios, placing the mixture into a reaction container, adding water to enable the mass concentration of a substrate to be 2% -5%, stirring and reacting at the constant temperature of 35 ℃, and adjusting the pH of reactants in the reaction container every 12 hours to enable the pH to be 6.
Further, the mass ratio of the carbohydrate waste to the pretreated sludge is 1-11: 1.
Further, in the step S2, the reaction process is carried out at 150 r/min.
Further, the carbohydrate waste may be carbohydrate household waste.
Further, the carbohydrate household garbage is potato carbohydrate household garbage.
The invention has the advantages and positive effects that:
1. the method utilizes the sludge and the carbohydrate garbage to produce the 2, 3-butanediol, and can greatly reduce the production cost of the 2, 3-butanediol; and, compared to the use of these substrates in research for the production of acetic acid, ethanol, the value of producing 2, 3-butanediol is much higher than them;
2. the microwave digestion treatment of the sludge can quickly increase the concentration of SPN in the sludge to be used as a nitrogen source; and when the mass ratio of the potatoes to the digested sludge is 8: at 1 hour, the yield of 2, 3-butanediol was 6.9 times that of the potato group alone; the method can reduce and stabilize the organic solid waste in the environment and simultaneously produce products with high added value.

FIG. 1 is a schematic diagram illustrating the influence of microwave digestion pretreatment on the concentration of SPN in sludge according to an embodiment of the present invention; FIG. 2 is a schematic diagram illustrating the effect of changing the mixing ratio of carbohydrate waste and pretreated sludge on the production of 2, 3-butanediol by fermentation according to an embodiment of the present invention; as shown in fig. 1-2, the method for producing 2, 3-butanediol by using carbohydrate waste provided in this embodiment includes the following steps:
obtaining sludge containing soluble protein SPN, adopting a secondary sedimentation tank of a sewage treatment plant in Tianjin city in the embodiment, and digesting and pretreating the sludge by using microwaves, wherein the treatment conditions are as follows: after the temperature is raised to 100 ℃ for 15min, and then the temperature is maintained for 15min, as shown in figure 1, the concentrations of SPN are respectively 28.4, 31.2, 36.8 and 37.6 times of those of the untreated group 1 through the treatment of conditions 2,3, 4 and 5, which shows that the concentration of SPN can be greatly improved by pretreating the sludge with microwaves and the sludge can be used as a nitrogen source substance;
mixing carbohydrate waste and pretreated sludge according to different mass ratios, in the embodiment, the carbohydrate waste is household garbage potatoes, the mixed household garbage potatoes and the pretreated sludge are placed in a reaction vessel, water is added to enable the mass concentration of a substrate to be 2.26%, stirring reaction is carried out at the constant temperature of 35 ℃, and the PH of reactants in the reaction vessel is adjusted every 12 hours to enable the PH to be 6.
In the embodiment, the mass ratio of the domestic garbage potatoes to the pretreated sludge is adjusted to 1:1, 4:1, 8:1, 9:1 and 11: 1; and setting a single domestic garbage potato whitening experiment, taking 10ml of fermentation supernatant at 1, 2,3 and 5d, centrifuging at 11000r/min, filtering by a 0.45um fiber filter membrane, and detecting the obtained filtrate by using a FID (flame ionization detector) of gas chromatography to determine the content of the 2, 3-butanediol.
Domestic garbage potato: digesting the sludge to be 8: the yield of the group 1 is 6.9 times of that of a single potato group, and the digestion of protein in sludge plays a key role.
The invention reduces and stabilizes the organic solid waste in the environment and simultaneously produces products with high added value.

1. A method for producing 2, 3-butanediol by utilizing carbohydrate waste is characterized by comprising the following steps: the method comprises the following steps:
s1, obtaining sludge containing soluble protein SPN, and digesting and pretreating the sludge by microwaves, wherein the treatment conditions are as follows: heating to 100 deg.C for 15min, and maintaining for 0-20 in;
s2, mixing the carbohydrate waste and the pretreated sludge according to different mass ratios, placing the mixture into a reaction container, adding water to enable the mass concentration of a substrate to be 2% -5%, stirring and reacting at the constant temperature of 35 ℃, and adjusting the pH of reactants in the reaction container every 12 hours to enable the pH to be 6.
2. The method for producing 2, 3-butanediol using carbohydrate waste as claimed in claim 1, wherein: the mass ratio of the carbohydrate waste to the pretreated sludge is 1-11: 1.
3. The method for producing 2, 3-butanediol using carbohydrate waste as claimed in claim 1, wherein: in the step S2, the reaction is carried out at 150 r/min.
4. A method for producing 2, 3-butanediol using carbohydrate waste according to any one of claims 1 to 3, wherein: the carbohydrate garbage can be carbohydrate household garbage.
5. The method for producing 2, 3-butanediol using carbohydrate waste as claimed in claim 4, wherein: the carbohydrate household garbage is potato carbohydrate household garbage.
