Abstract

The invention relates to a decolouring process for preparing a sheep casing in a sheep intestine surgical suture, which comprises the steps of putting the sheep casing into mixed treatment liquid consisting of an oxidation-type decolouring agent, a stabilizing agent and an alkaline solution for ultrasonic oscillation, and then putting the mixed treatment liquid consisting of a reduction-type decolouring agent, a stabilizing agent and an alkaline solution for ultrasonic oscillation. The process is green and environment-friendly, simple to operate and low in cost, and not only is the efficiency of removing the pigment in the sheep sausage skin high, but also the mechanical strength of the sheep sausage skin is maintained.

Technical Field
The invention relates to a novel process for removing pigment in a sheep intestine casing, belonging to the technical field of processing of sheep intestine sutures.
Background
The catgut is an absorbable medical operation suture line, is mainly used for department of digestive and urinary systems surgery, and has wide raw material sources, simple manufacturing process, no toxic or side effect and good human body compatibility. It can provide temporary support or barrier for body, and can be degraded into absorbable matter for elimination after the completion of mission, so as to avoid inflammation reaction and other adverse effects caused by long-term existence of foreign body in body, and avoid secondary operation, thereby being widely applied.
The sausage skin is prepared by removing mucosa from sheep intestine, defatting, decolorizing, treating with chromium, and drying to obtain absorbable suture. Because the sausage casing contains natural pigments, the colors of the pigments are darkened after the pigments are oxidized, and the appearance of the catgut is influenced, so that the decoloration of the sausage casing is an essential process in the preparation process of the catgut. At present, the following two main methods are commonly used for chemical decolorization:
(1) the principle of the oxidation bleaching method is that a bleaching agent having a strong oxidizing property destroys a chromophore of a pigment, for example, decomposes an azo group, and thus cannot develop a color any more. The oxidation decoloring method has the advantages that the decolored substance is stable and does not return under the action of heat and illumination. The defects are that the strong oxidation decolorant can destroy sheep intestine collagen molecules, so that the molecular weight of protein fibers is reduced, and the strength of the sheep intestine threads is reduced.
(2) The principle of the reduction decoloring method is that a bleaching agent and an organic pigment are combined into an unstable colorless substance. The method has the advantages that the bleached fiber has high whiteness, small molecular damage and high strength retention degree. The defects are that the bleaching is reversible, the bleaching whiteness is high in heat fading rate, and if the bleached product is heated, illuminated and the like, the original color of the bleached product can be recovered.
Due to the complex distribution of pigments in the sheep intestines, complete removal is difficult with only one method. At present, the high-concentration hydrogen peroxide oxidation method is mainly used for decoloring the sheep intestine casing, so that the decoloring effect is not ideal, and the strength of the sheep intestine casing is lost. The composite decoloring process combining the sodium hydrosulfite and hydrogen peroxide oxidation reduction is already applied to the wool decoloring field, and the effect is better than that of a single sodium hydrosulfite reduction method or a hydrogen peroxide oxidation method. Therefore, the development of the efficient decolorizing process for the sheep intestine casing by combining various decolorizing methods is an urgent problem to be solved in the field of sheep intestine decolorization.
Disclosure of Invention
The invention aims to overcome the defects, and provides the decolorizing process for preparing the sheep intestine casing in the sheep intestine surgical suture, which is green and environment-friendly, simple to operate and low in cost, has high efficiency of removing pigment in the sheep intestine casing, and maintains the mechanical strength of the sheep fat casing.
The technical scheme adopted by the invention is as follows:

(1) ultrasonic oxidation type decolorizing agent treatment: soaking the casing of sheep with the attachments such as chorion, serosa and the like removed in a container filled with mixed treatment liquid consisting of an oxidation-type decolorizing agent, a stabilizing agent and an alkaline solution, placing the container in ultrasonic equipment, and keeping the temperature at 20-40 ℃ for ultrasonic oscillation for 1-4 h;
(2) ultrasonic reduction type decolorant treatment: and (2) soaking the sheepskin casing treated in the step (1) in a container filled with mixed treatment liquid consisting of a reduction-type decolorizing agent, a stabilizing agent and water, placing the container in ultrasonic equipment, and keeping the temperature at 20-40 ℃ for ultrasonic oscillation for 1-4 hours.
In the process, the oxidation-type decolorant in the step (1) is one or more of hydrogen peroxide, sodium peroxide and sodium percarbonate; hydrogen peroxide or sodium percarbonate is preferred; the concentration of the oxidation-type decolorant in the mixed treatment liquid is 0.1-1.5 wt%; the stabilizer is one or more of sodium silicate, trisodium phosphate and sodium polyphosphate; the concentration of the stabilizer in the mixed treatment fluid is 0.5-5 wt%; the alkaline solution is prepared from one or more of sodium carbonate, sodium bicarbonate and potassium carbonate and deionized water, and is used for adjusting the pH value of the mixed treatment liquid to be 7.5-9.
The reduction-type decolorizing agent in the step (2) is one or more of sodium sulfite, sodium bisulfite, sodium hydrosulfite and thiourea dioxide; preferably thiourea dioxide; the concentration of the reduction-type decolorizing agent in the mixed treatment liquid is 0.2-2 wt%; the stabilizer is one or more of sodium silicate, trisodium phosphate and sodium polyphosphate; the concentration of the stabilizer in the mixed treatment fluid is 0.5 wt% -5 wt%.
The ultrasonic oscillation frequency in the steps (1) and (2) is 20-70 kHz.
In the decoloring process, the casing is swelled in alkalescent liquid, so that the oxidized decoloring agent and the reduced decoloring agent can be favorably diffused into the casing to react with chromophoric groups of protein, and the reaction speed of the decoloring agent and the chromophoric groups is accelerated by ultrasound.
The invention has the beneficial effects that:
1) the decoloring treatment process adopted by the invention not only greatly improves the decoloring efficiency, but also has the pigment removal rate of more than 70% in 2h and the decoloring rate of 85% in 4 h; meanwhile, the damage of the oxidation decolorant to the sheep intestine collagen molecules is reduced, and the mechanical strength of the sheep intestine is kept to the maximum extent.
2) The invention belongs to a green environment-friendly treatment process, does not use an organic solvent, and has simple operation and low cost.
Detailed Description
The following is a further description with reference to specific examples.
Example 1
A decolorization process for preparing a sheep casing in a sheep intestine operation suture comprises the following steps:
(1) treating the sheep sausage casing by using an ultrasonic oxidation type decolorizing agent: soaking a sheepskin casing with attachments such as chorion, serosa and the like removed in a container filled with a mixed treatment liquid containing 0.25 wt% of hydrogen peroxide, 1 wt% of sodium silicate and a sodium carbonate aqueous solution, wherein the pH of the mixed treatment liquid is 8, placing the container in ultrasonic equipment, and keeping the ultrasonic frequency at 40kHz, ultrasonic oscillation for 2h and the temperature at 30 ℃;
(2) treating the sheep sausage casing by using an ultrasonic reduction type decolorizing agent: soaking the sheepskin casing treated in the step (1) in a container containing an aqueous solution containing 0.5 wt% of thiourea dioxide and 1 wt% of trisodium phosphate, and placing the container in an ultrasonic device, wherein the ultrasonic frequency is 40kHz, the ultrasonic vibration is carried out for 2 hours, and the temperature is kept at 30 ℃.
Example 2
A decolorization process for preparing a sheep casing in a sheep intestine operation suture comprises the following steps:
(1) treating the sheep sausage casing by using an ultrasonic oxidation type decolorizing agent: soaking a sheep sausage casing with attachments such as chorion, serosa and the like removed in a container containing a mixed treatment liquid containing 0.25 wt% of sodium percarbonate, 1.5 wt% of sodium silicate and a sodium carbonate aqueous solution, wherein the pH of the mixed treatment liquid is 7.5, and placing the container in an ultrasonic device, wherein the ultrasonic frequency is 40kHz, the ultrasonic vibration is carried out for 4 hours, and the temperature is kept at 30 ℃;
(2) treating the sheep sausage casing by using an ultrasonic reduction type decolorizing agent: soaking the prepared sheep intestine with the oxidized decolorizing agent in a container containing 0.75 wt% thiourea dioxide and 1 wt% sodium polyphosphate, and placing the container in an ultrasonic device with ultrasonic frequency of 40kHz and ultrasonic oscillation for 4h at 30 ℃.
Example 3
A decolorization process for preparing a sheep casing in a sheep intestine operation suture comprises the following steps:
(1) treating the sheep sausage casing by using an ultrasonic oxidation type decolorizing agent: soaking a sheepskin casing with attachments such as chorion, serosa and the like removed in a container filled with a mixed treatment liquid containing 0.25 wt% of hydrogen peroxide, 1 wt% of sodium silicate and a sodium carbonate aqueous solution, wherein the pH of the mixed treatment liquid is 8, and the container is placed in ultrasonic equipment, the ultrasonic frequency is 60kHz, the ultrasonic oscillation is carried out for 3 hours, and the temperature is kept at 40 ℃;

Comparative example 2
Treating the casing of the sheep with a reduction-type decolorizing agent: the prepared sheep intestine with the attachments such as chorion and serosa removed is soaked in a container containing 0.75 wt% sodium dithionite and 1 wt% trisodium phosphate solution for 4h, and the temperature is kept at 40 ℃.

(2) ultrasonic reduction type decolorant treatment: and (2) soaking the sheepskin casing treated in the step (1) in a container filled with mixed treatment liquid consisting of a reduction-type decolorizing agent, a stabilizing agent and water, placing the container in ultrasonic equipment, and keeping the temperature at 20-40 ℃ for ultrasonic oscillation for 1-4 hours.

9. The decolorization process for preparing casing of sheep intestine in surgical suture of sheep intestine as claimed in claim 1, wherein said stabilizer in step (2) is one or more of sodium silicate, trisodium phosphate, and sodium polyphosphate.

The decolouring process for preparing the sheep intestine casing in the sheep intestine surgical suture line as claimed in claim 1, wherein the concentration of the reduced decolouring agent in the step (2) in the mixed treatment solution is 0.2 wt% -2 wt%, and the concentration of the stabilizing agent in the mixed treatment solution is 0.5 wt% -5 wt%.
