Abstract
本发明公开了基于太赫兹衰减全反射模式的早期癌症检测方法,建立全反射面‑样品池底面‑细胞层‑缓冲液层的多界面与倏逝波相互作用的物理模型,由于全反射面和样品池底面为同一材质,所以全反射发生在样品池底面‑细胞层界面。本发明基于细胞的太赫兹特征指纹谱的检测将会比基于细胞形态的检测具有更高的灵敏度和准确度。

技术领域
本发明属于早期癌症检测技术领域,具体涉及基于太赫兹衰减全反射模式的早期癌症检测方法。
背景技术
癌症是我国城乡居民的第一位死因,我国每年新发癌症病例达429万,占全球新发病例的20%,死亡281万例。癌症防治已成为我国的重要公共卫生问题。大量研究表明,对早期癌症的治疗效果明显优于中、晚期癌症。如果能够在癌症初期对其准确诊断,则可能根治、有效控制或延长癌症的发病期,从而改善病人的生存质量、提高生存年限。因此,包括我国在内的许多国家把癌症的早期诊疗作为最主要的防治癌症的手段。
准确检测早期癌变是癌症早期治疗的先决条件。目前,检测癌症的方法可分为影像法、免疫组织化学法、穿刺病理法等。影像法是运用电子计算机断层扫描或是核磁共振技术,根据不同密度的组织对射线反射强度不同,形成不同的影像,并对照正常组织来判断癌变组织。影像法多用于肿瘤组织层面的检测,适合于癌症中、晚期的检测。免疫组织化学法是基于免疫组织化学、分子生物学、流式细胞术等检测肿瘤标记物的新的病理学检测方法,适合于癌症的早期检测。但是,该方法存在肿瘤标记物的灵敏度和特异性不理想,容易出现假阳性和假阴性的问题,因为许多肿瘤标志物不仅在发生癌变时产生,在正常和良性肿瘤的情况下也有不同程度表达。准确检测癌症的“金标准”是要求在显微镜下观测到癌细胞,穿刺病理法是利用光学显微镜对可疑部位的组织切片或对脱落细胞进行直接观测,根据细胞的形态,如:核浆比、细胞核的形态及核内染色体形态等判断是否有癌细胞,从而判断病人是否患有癌症,这种方法是确认癌症最可靠的方法,适合肿瘤的早、中期检测。然而,这种基于细胞形态观测的病理学检测方法受到包括样品处理过程、光学显微镜分辨率、医生主观经验多种因素影响,导致准确度不理想,比如:美国的乳腺癌的误诊率最高可达43%。可见,目前早期癌症的检测存在灵敏度和特异性不理想的问题。因此,我们必须发展新方法来实现对早期癌变细胞的准确检测,以达到癌症早期准确诊断的目的。
近些年,随着太赫兹生物医学的发展,太赫兹生物检测技术在癌症诊断领域凸显出非常大的潜力。太赫兹波是频率范围为0.1-10THz的电磁波,其波长(0.03-3mm)介于微波与远红外之间(如图1所示)。由于太赫兹波与生物分子低频振动(如生物分子骨架的振动、蛋白质中残基的转动和扭动等)的能量水平具有很大的交界,对生物分子间的弱相互作用(如氢键、范德华力等)以及生物体中的水含量非常敏感,因此,可通过生物体在太赫兹频段内的特征波谱(即复介电谱、特征吸收谱),实现对生物样本的定性及定量分析。不同生物分子、同一生物分子的不同构象、不同细胞及胞内化学成、同一细胞的不同生理阶段、不同细菌的含水量在太赫兹波段均有不同的响应;同时太赫兹光子能量低,对生物几乎无电离损伤,非常安全。因此,太赫兹诊断已公认是下一代极具竞争力的生物医学技术。目前,国际上已有应用太赫兹技术对癌症进行诊断的报道:在组织检测层面,癌变组织的太赫兹检测结果与病理学检测非常一致,如脑胶质瘤[13]和胃癌组织[14]等;在细胞检测层面,不同种类的癌细胞有着明显的太赫兹介电差异[10];在分子检测层面,通过太赫兹特征波谱可以灵敏地检测到肿瘤标志物和其特异性配体的结合作用,其检测灵敏度达到1pmol/μL。相对于正常细胞,癌变细胞所含物质的化学成分和比例的变化早于光学显微镜观测到的细胞形态的变化,并且细胞的这些变化会敏感地影响细胞的物化特性。
将太赫兹技术应用于癌症的早期检测,利用自主研发的衰减全反射探测模式,尝试将太赫兹生物医学检测从实验室推向临床应用,属于临床医疗先进技术,对增强我国数字诊疗装备的技术竞争力,推动医疗器械产业的发展具有一定的现实意义。现有癌细胞检测主要依赖标记技术,样本制备复杂,细胞损害性较强。太赫兹因其独特的频谱优势,能探测其它电磁波段和现有活细胞检测技术无法解析的胞内生物分子和化学成分的信息,能够通过细胞内所含物质的化学成分和比例的变化来鉴别早期癌变细胞,检测灵敏度优于光学显微镜,有望成为现有癌症早期检测不可或缺的全新手段。
目前太赫兹技术在癌症检测的组织-细胞-分子三个层面均已凸显出明显的优势,尤其在细胞层面能比传统光学显微镜更早的发现初期癌变细胞,是一种极具潜力的早期癌症检测技术。传统的太赫兹生物检测一般采用透射模式,鉴于活性生物样本具有丰富的水含量,样本中的水含量以及样本的厚度都会对太赫兹检测结果产生较大的影响,因此,人们通常采用合适的液体样品池来装载生物样本,以控制样本的厚度。因此,透射式检测存在以下几个问题:(1)样品前处理过程复杂而精细、对实验条件和操作技术要求高,限制了太赫兹诊断技术在临床环境的直接运用。(2)检测结果受样品厚度误差的影响,降低了检测精度。(3)透射式检测会在检测结果中带来驻波谐振的干扰,增加了数据的后处理难度。因此,开发合适的探测模式,快速、精准的探测生物样本的太赫兹波谱,是太赫兹技术在实际临床运用中亟需突破的问题。

为了解决上述问题,本发明提供了基于太赫兹衰减全反射模式的早期癌症检测方法,为早期癌症的检测提供革命性的技术手段,为癌细胞、肿瘤标记物的检测研究提供一种定性、定量检测的新技术与新方法,所构建的针对不同组织学分类的癌细胞的太赫兹波谱指纹库有望成为癌症检测的新指标,能有效提升肿瘤疾病的诊治与监测能力,对癌症的早期诊断及预防具有重要的临床意义。
为了实现上述目的,本发明的技术方案为:
基于太赫兹衰减全反射模式的早期癌症检测方法,建立全反射面-样品池底面-细胞层-缓冲液层的多界面与倏逝波相互作用的物理模型,由于全反射面和样品池底面为同一材质,所以全反射发生在样品池底面-细胞层界面,根据下式可得该接触面发生内反射的临界角θc,关系如下:
上式中nSam、nSi分别为样本和硅棱镜的折射率,倏逝波在细胞层-缓冲液层的穿透深度dp与波长λ的关系如下:
上式表征了倏逝波以指数型衰减穿透细胞层和缓冲液层,dp为太赫兹波的电场强度衰减至表面处的1/e时的特征穿透深度,λ为入射波波长,θ为入射角。
进一步,将临床样本置于样品池的样品区,将样品池装载在全反射面上,转动样品池使得倏逝波与样品池的参考区域完全作用,此时输出的电场幅值ERef为参考信号,将临床样本置于样品池的样品区,将样品池装载在全反射面上,转动样品池使得倏逝波与样品池的参考区域完全作用,再次转动样品池使得倏逝波与样品池的样品区域完全作用,此时太赫兹出射信号会明显发生改变,此时输出的电场幅值为样本信号。若入射太赫兹波的电场幅值为EIn,则根据反射定律,可得

与现有技术相比,本发明的有益效果:相对于正常细胞,癌变细胞所含的生物分子和化学成分的变化早于光学显微镜能观测到的细胞形态的变化。胞内生物分子、化学分子的分子间相互作用力(氢键、范德华力等)和分子内作用力(分子骨架振动和偶极子旋转等)正好处在太赫兹频段。当细胞发生癌变时,胞内水含量、生物分子、化学物质的变化会敏感地反映在太赫兹特征波谱上。因此,基于细胞的太赫兹特征指纹谱的检测将会比基于细胞形态的检测具有更高的灵敏度和准确度。根据衰减全反射物理光学原理,倏逝波与样品之间的作用距离与样本的厚度无关,该技术有效避免了活性生物样本检测中厚度引起的误差,提高了检验精度。另外,倏逝波首先射入活细胞层,其作用深度与细胞厚度相匹配,出射的太赫兹信号主要反映细胞层的太赫兹物化性质,提高了检验的灵敏性和特异性。最后,该技术简化了样本的前处理过程,能满足临床检测的要求,实现了一种无需预处理、无需试剂、实时快速检测癌症的新技术。

具体实施方式
下面将对本发明实施例中的技术方案进行清楚、完整地描述,显然,所描述的实施例仅仅是本发明一部分实施例,而不是全部的实施例。基于本发明中的实施例,本领域普通技术人员在没有作出创造性劳动前提下所获得的所有其他实施例,都属于本发明保护的范围。
实施例1:
拟选用Menlo Tera SYNC太赫兹时域光谱仪与衰减全反射模块集成,搭建参考光路如图1所示。调节太赫兹波发射天线和接收天线的位置,使太赫兹波为P偏振,并将发射天线、衰减全反射模块、接收天线调至同一水平线,调节太赫兹时域光谱仪的时间延迟,获得太赫兹信号。太赫兹波束在衰减全反射进样模块中的传播如下所述:模块水平放置,太赫兹波发射端将太赫兹波束水平投射到棱镜的一个耦合面上,太赫兹波经过氮气-棱镜界面折射后进入棱镜内,并投射到全反射面上的样品承载池,太赫兹波在样品承载池底面发生全内反射。在较高频段上,界面上的倏逝波完全与样本中的细胞作用;在较低频段上,界面上的倏逝波与细胞相互作用后,又穿透细胞与细胞上层的液体作用,如图5所示。最后太赫兹波束反射到对侧的耦合面上,经棱镜-氮气界面折射后,水平射出到检测器上。此时,出射的太赫兹波束主要携带样本中细胞成分的太赫兹物化信息。
实施例2:
通过太赫兹波谱检测适配体和靶分子结合反应的无标记方法。利用太赫兹波谱和分子动力学模拟,研究了肿瘤标记物MUC1多肽和其对应适配体Anti-MUC1发生特异性结合反应时,溶液太赫兹吸收波谱的变化以及溶液中氢键网络的变化。结果表明,太赫兹波谱可灵敏地反映MUC1和Anti-MUC1的结合反应,该方法的最小检测浓度为1pmol/μL,如图2所示。其中,(a)太赫兹时域光谱仪原理图及频域动态范围示意图;(b)微流控芯片三维模型示意图;(c)分子动力学模拟中观察到的MUC1肿瘤标记物(红色)和其适配体(蓝色)结合过程的快照;(d)太赫兹波谱检测MUC1肿瘤标记物和其适配体结合反应的浓度依赖性,最小检测浓度为1pmol/μL。
实施例3:
将超材料生物传感芯片于太赫兹波谱检测相结合,对3’端5碱基突变的DNA寡核苷酸进行了无标记研究。研究表明太赫兹超材料生物传感芯片的灵敏度足够区别不同突变序列的DNA寡核苷酸链。进一步,项目申请人通过分子动力学模拟,探讨了寡核苷酸与其周围水分子之间形成氢键的数量,这些氢键对生物溶液的太赫兹吸收起着关键作用,如图3所示。该工作为太赫兹波谱结合超材料技术对DNA的无标记检测的研究奠定了基础,为基于太赫兹波谱的基因突变的研究奠定了基础。其中,(a)本研究中涉及的DNA寡核苷酸序,末尾5个碱基发生了突变;(b)四种寡核苷酸在溶液中与周围水分子形成的氢键数,该数量排序与四种寡核苷酸溶液的吸收系数排序相一致;(c)超材料传感芯片中谐振环单元的光学显微镜图像;(d)寡核苷酸在超材料传感芯片上的太赫兹透射光谱,根据太赫兹透射光谱可清晰的分辨出核酸的突变。
实施例4:
运用分子动力学结合准谐振近似(Quasi-Harmonic Approximation),研究了溶液中丙氨酸多肽链长和构象对太赫兹吸收谱(10~40cm-1)的影响。研究表明,随着链长增长,大部分吸收峰位置改变;并从波谱中解析到Alan多肽的指纹峰39cm-1。其次,研究了Ala15多肽在线团和螺旋构象下太赫兹谱的不同,研究表明可以通过太赫兹谱区分生物分子的不同构象,并从多肽与水分子形成氢键的平均数目,解释了线团构象的吸收强度大于螺旋构象的原因,如图4所示。其中,(a)、太赫兹吸收强度(b)、多肽分子与周围水分子形成的氢键数量(c);Ala15丙氨酸多肽在水溶液下的线团和螺旋构象(d)、太赫兹吸收强度(e)、不同构象下的Ala15分子与周围水分子形成的氢键数量(f)。
对于本领域技术人员而言,显然本发明不限于上述示范性实施例的细节,而且在不背离本发明的精神或基本特征的情况下,能够以其它的具体形式实现本发明。因此,无论从哪一点来看,均应将实施例看作是示范性的,而且是非限制性的,本发明的范围由所附权利要求而不是上述说明限定,因此旨在将落在权利要求的等同要件的含义和范围内的所有变化囊括在本发明内。

Claims (2)
1.基于太赫兹衰减全反射模式的早期癌症检测方法,其特征在于,建立全反射面-样品池底面-细胞层-缓冲液层的多界面与倏逝波相互作用的物理模型,由于全反射面和样品池底面为同一材质,所以全反射发生在样品池底面-细胞层界面,根据下式可得该接触面发生内反射的临界角θc,关系如下:

2.根据权利要求1所述的基于太赫兹衰减全反射模式的早期癌症检测方法,其特征在于,将临床样本置于样品池的样品区,将样品池装载在全反射面上,转动样品池使得倏逝波与样品池的参考区域完全作用,此时输出的电场幅值ERef为参考信号,将临床样本置于样品池的样品区,将样品池装载在全反射面上,转动样品池使得倏逝波与样品池的参考区域完全作用,再次转动样品池使得倏逝波与样品池的样品区域完全作用,此时太赫兹出射信号会明显发生改变,此时输出的电场幅值为样本信号。若入射太赫兹波的电场幅值为EIn,则根据反射定律,可得
