Abstract

本发明属于生物材料切片的包埋剂及制作方法。目前,世界上还没有切实可行的组织包埋技术适用于昆虫体壁(含外骨骼)的连续切片。本发明针对昆虫体壁的特质性,采用优质松香、石蜡为原料,加入少量丙酮、叔丁醇,其配比由切片时室温、材料硬度、大小而定。本发明广泛适用于各种生物材料的软、硬组织。具有支持强度高、韧性好、溶点低、切片不受室温限制、操作简便、易于推广、价格低廉等优点。

Description

松香包埋剂及制作方法
本发明属于生物材料切片的包埋剂。
目前,世界上常用的生物材料切片所用的包埋剂有:石蜡、火棉胶、炭蜡等,包括超薄切片用的环氧树脂及冰冻切片技术等,都不适于昆虫体壁组织(含外骨骼)的连续切片。由于昆虫的外骨骼坚硬、性脆、易碎,采用以上任何一种包埋剂,都不能有效地对昆虫体壁的外骨骼进行连续切片观察。如石蜡,即便是溶点高的(如58℃~62℃),也支持不住成虫发达的外骨骼,而且渗透温度较高(65℃左右),对昆虫材料有较强的破坏作用;环氧树脂,也只是对那些骨化程度不高,微小的器官有效。由于没有一种适合于昆虫体壁组织(含外骨骼)连续切片的包埋剂,长期以来,有关昆虫体壁组织显微结构的研究,很少有人问津。
为了解决昆虫体壁组织(含外骨骼)连续切片这一世界性的难题,特研制本发明,试制出目前对昆虫体壁组织特效的包埋剂。
本剂主要是由松香(Colophony)及石蜡(Paraffin)加其它辅剂配制而成。确切的配比,要随切片时的室温,生物材料的硬度、大小而定。我们试制出六个型号的包埋剂适于在18~36℃条件下进行连续切片。
该剂的优点:①支持强度高,对各种成虫的外骨骼均有效;②韧性好,切出的带弹性好,明显优于石蜡(在同等条件下,石蜡性脆,蜡带易断),保证了切片的连续性,一条带可长达60cm,连续标本片可达百余个(如触角横切);③切片薄,采用该包埋技术,可切出1~12μ的连续切片,保证了对一些特殊构造(如各种感器)的连续切片观察;④溶点低,采用该剂渗透时,温度可在50℃~53℃一进行,避免了高温对生物组织的破坏作用;⑤取材方便,不用解剖,保证了器官的完整性及系统性,甚至可以进行整体切片(如小型个体)。该剂不但适于昆虫体壁的硬组织,也同时用于各种生物的软组织。
本发明采取优质松香,石蜡为原料,按比例松香∶石蜡=1∶5~1∶0.15称重,入容器加热(120-150℃),使混合剂溶解,再加入少量丙酮,叔丁醇,待溶液呈透明状,固化或入恒温箱备用。固化后呈黄褐色或棕褐色。如在室温为21~23℃条件下切片时,松香∶石蜡=1∶1.5;24~26℃时,两者配比为1∶1。

Claims (3)

1、一种松香包埋剂,其特点是主要由优质松香、石蜡加入少量丙酮、叔丁醇混合而成,其中重量比松香∶石蜡=1∶0.15-1∶5。
2、一种松香包埋剂的制作方法,其特征是主要由优质松香、石蜡加入少量丙酮、叔丁醇制作而成,其中重量比是松香∶石蜡=1∶0.15-1∶5,混合入容器加热至120-150℃溶解,再加入少量丙酮、叔丁醇,使溶液呈透明状,固化后呈黄褐色或棕褐色,固化后放入恒温箱备用。
3、按权利要求2所述的松香包埋剂的制作方法,其特征是所说的松香、石蜡重量比在室温21-23℃条件下切片时为1∶1.5,在室温为24-26℃条件下切片时为1∶1。
