Abstract
本发明是微生物研究领域新疆地区结核病人痰标本中结核分枝杆菌临床分离株结核分枝杆菌PhoP的天然突变株。通过采集多个新疆地区的传染科结核病人的痰标本得到438例临床分离株,提取总DNA、普通PCR、荧光定量PCR,发现临床分离株中PhoP的天然突变株。

技术领域
本发明是微生物研究领域结核分枝杆菌的基因突变。
背景技术
随着交通越来越便利,人口流动性逐步增加,临床免疫抑制剂的广泛应用,目前结核病疫情目前已再次拉响警钟。结核病的主要病原菌是结核分枝杆菌,关于结核分枝杆菌的研究目前机制尚不完全明确。
PohP属于双组分信号传导系统,一般认为普遍存在于原核生物中,由一个组氨酸激酶和一个反应调节剂构成,有文献认为其和细菌对宿主的适应能力及细菌毒力相关。目前对结核分枝杆菌标准株(H37Rv) 的功能展开大量研究。
新疆位于中国西北部,干旱少雨的气候特点更有利于结核分枝杆菌的传播,加之新疆多民族聚居,一些民族结核的易感性,使新疆的结核病疫情更严重。
采集喀什市人民医院,库尔勒市人民医院,石河子疾控中心、皮山县人民医院感染科结核病人的痰标本,2%的氢氧化钠消化痰标本,同时可以杀灭其他杂菌,液化后的痰标本接种到罗氏培养基中,37℃培养箱中孵育3周,得到临床分离株438株,普通PCR扩增PhoP,发现临床分离株中存在PhoP的天然缺失株。北京天根有限公司的DNA提取试剂盒分别提取临床分离株的DNA,跑电泳检测是否提取成功,如图1。根据PohP引物F:T C C A G C T G TT A G C A G A C T A C T G G C A A C G A G;R:T C C A A G C T T T G C C G A T CC G A T T A A C T A C C扩增如图2,对无法跑出条带的样本,做副空,重复试验。还是无法用普通PCR得到条带的样本,我们采用实时荧光定量PCR分子水平检测,发现分子水平也无法检测出PhoP的存在。
我们认为在结核分枝杆菌传播的过程中,不知是环境因素还是宿主因素,病菌发现了突变,这种突变是对其自身或菌群,是有利还是有害的,缺失后菌株依然得以存活,细菌有没有其他替代机制目前尚无报道。
