(6) removing impurities: removing impurities of the wall-broken homogenate by filtering or centrifuging, and collecting filtrate or supernatant after centrifuging to obtain ceramide solution.
In this example, the ceramide solution obtained is concentrated and crystallized to obtain a ceramide solid, which is specifically prepared by: mixing the prepared ceramide solution with an ethanol solution with the mass fraction of 95% at a weight ratio of 1:5 at normal temperature, uniformly stirring, standing overnight, filtering out precipitates, concentrating under reduced pressure at 30-40 ℃ under the pressure of 0.001-0.01 MPa until the solution is turbid, cooling and crystallizing under normal pressure to obtain a ceramide solid, wherein the mass fraction of the ceramide in the obtained ceramide solid is 10%.
Example 2
A method for preparing ceramide from rice bran by a fermentation method specifically comprises the following steps:
(1) raw material treatment: drying rice bran in an oven at 80 deg.C, removing impurities, pulverizing, and sieving with 20-40 mesh sieve;
(2) activating strains: cutting Ganoderma leucocontextum (G.leucocotextum) with inoculating hook to 1cm from slant2Inoculating slant mycelia with culture medium into the middle of solid culture medium, and culturing at 28 deg.C for 5 days;
(3) preparing a seed solution: mashing the solid culture medium with Ganoderma with inoculating hook, and adding appropriate amount of sterile water to obtain Ganoderma seed solution;
(4) fermentation culture: mixing the ganoderma lucidum seed liquid and the rice bran raw material powder according to the mass ratio of 3:1, putting the mixture into a fermentation tank, culturing for 5 days at the temperature of 28 ℃, and controlling the oxygen concentration of the space in the fermentation tank to be 0.02 mol/L;
(5) homogenizing and breaking walls: after fermentation, adding distilled water with equal weight into the fermented product, mixing, and homogenizing for 5 times at room temperature and 70MPa with a high-pressure homogenizer to obtain wall-broken homogenate;
(6) removing impurities: removing impurities of the wall-broken homogenate by filtering or centrifuging, and collecting filtrate or supernatant after centrifuging to obtain ceramide solution.
(7) Concentrating the ceramide solution under reduced pressure to obtain ceramide stock solution with total ceramide content of 0.5%.
According to the method, the rice bran is fermented by the ganoderma lucidum, so that the dissolution of ceramide in the rice bran can be remarkably improved, and the ganoderma lucidum can also generate a certain amount of ceramide, so that any derivatization process is not needed, the operation is simple, and the extraction is convenient; the fermentation period is short, the effect is quick, and the method is not limited by regional climate; the process for producing the ceramide can greatly reduce the dosage of the organic solvent, reduce the environmental influence and reduce the economic cost.
Claims (8)
1. A method for preparing ceramide from rice bran by fermentation is characterized in that the rice bran is used as a raw material, ganoderma lucidum is adopted to ferment the rice bran so as to degrade cell walls of the rice bran, and meanwhile, the ganoderma lucidum can propagate in a large quantity to generate a certain amount of ceramide; homogenizing and breaking the wall after fermentation to obtain wall-broken homogenate, and removing impurities to obtain a ceramide solution;
when the rice bran is fermented by adopting the ganoderma lucidum, the fermentation conditions are controlled so that the ganoderma lucidum is subjected to submerged fermentation;
in order to make ganoderma lucidum carry out submerged fermentation on rice bran, the specific fermentation conditions are as follows: mixing the ganoderma lucidum seed liquid and the rice bran raw material powder according to the mass ratio of (1-5) to 1, putting the mixture into a fermentation tank, fermenting for 2-5 days at the temperature of 28 +/-2 ℃, and controlling the oxygen concentration of the space in the fermentation tank to be 0.010-0.025 mol/L.
2. The method for preparing ceramide from rice bran by fermentation according to claim 1, wherein the ganoderma lucidum seed solution is prepared by the method comprising:
activating strains: inoculating Ganoderma mycelia with culture medium cut from the slant with an inoculating hook into the middle of solid culture medium, and culturing at 28 + -2 deg.C for 3-5 days;
preparing a seed solution: mashing the solid culture medium with Ganoderma with inoculating hook, and adding appropriate amount of sterile water to obtain Ganoderma seed solution, wherein the amount of sterile water is 2-5 times of the volume of the solid culture medium.

The method for preparing ceramide from rice bran by fermentation according to claim 1, wherein the homogenate is broken after fermentation by the following specific method: after fermentation, adding distilled water with equal weight into the fermented product, mixing, and homogenizing for 1-5 times at 70-120Mpa at normal temperature by high pressure homogenizer to obtain wall-broken homogenate.

The method for preparing ceramide from rice bran through fermentation according to claim 1, wherein the impurities are removed by: removing impurities in the wall-broken homogenate by filtering or centrifuging, and collecting filtrate or supernatant after centrifuging to obtain ceramide solution.
5. The method of claim 1, wherein the rice bran is treated by a fermentation process prior to fermentation with the ganoderma lucidum, the fermentation process comprising: removing impurities from testa oryzae, pulverizing, and sieving.
6. The method for preparing ceramide from rice bran according to any one of claims 1-5, wherein the Ganoderma lucidum is selected from Ganoderma lucidum, Ganoderma sinensis, Ganoderma sinense, and Ganoderma tsugae.
7. The method for preparing ceramide from rice bran by a fermentation method according to claim 1, wherein the prepared ceramide solution and 95% ethanol solution are mixed at a weight ratio of 1:5 at normal temperature, are uniformly stirred and placed overnight, are filtered and precipitated, are concentrated under reduced pressure at 30-40 ℃ under the condition of 0.001-0.01 MPa until the solution is turbid, and are cooled and crystallized under normal pressure to obtain a ceramide solid, wherein the ceramide mass fraction in the ceramide solid is 0.1-1.0%.
8. The method for preparing ceramide from rice bran through fermentation according to claim 1, which comprises the following steps:

homogenizing and breaking walls: after fermentation, adding distilled water with equal weight into the fermented product, mixing, and homogenizing for 1-5 times at 70-120Mpa at normal temperature by high pressure homogenizer to obtain wall-broken homogenate;
removing impurities: removing impurities of the wall-broken homogenate by filtering or centrifuging, and collecting filtrate or supernatant after centrifuging to obtain ceramide solution.
