现代医学认为，类风湿关节炎和强直性脊柱炎都是慢性全身性自身免疫性疾病。医学学者们通过研究发现，引起此类关节炎发病的“元凶”是一种名叫“肿瘤坏死因子”的炎症因子，它能引发关节滑膜炎、活化破骨细胞造成骨破坏和关节畸形。类风湿关节炎具有隐匿性高、难确诊、受损不可逆和致残率高等特点。它可以发病于身体的任何一个关节，常发病于指、掌、腕部小关节。于是，医学学者们开发了生物制剂，通过抑制肿瘤坏死因子来治疗这类疾病。目前用得最多、副作用也相对小的是肿瘤坏死因子α(TNF-α)拮抗剂，与传统的改善病情的抗风湿药相比，其主要特点是起效快、抑制骨破坏的作用明显、患者总体耐受性好。该制剂主要有依那西普、阿达木单抗。西药作用明显，但是副作用相对而言还是比较大。

本发明所要解决的技术问题在于提供一种高澄清度的生物制剂及其应用，该生物制剂澄清度高，副作用小，对类风湿性关节炎有较好的治疗作用。
本发明所要解决的技术问题采用以下的技术方案来实现：
一种高澄清度的生物制剂，由以下重量份的原料组成：黄柏10-20份、艾叶5-8份、柴胡1-2份、菖蒲6-9份、鱼腥草12-15份、半枝莲8-10份、五味子3-5份、土荆芥15-20份、大青叶7-10份、岩黄连16-22份、蒲公英18-25份、藏红花7-10份、土茯苓10-13份、黄芩8-10份、金银花4-6份、青蒿5-8份、穿山甲2-3份、牛筋草16-20份、骆驼蓬1-2份、苍术2-5份、活化助剂3-5份。
所述活化助剂由以下重量份的原料组成：茶多糖25-35份、碳酸氢钠12-15份、天冬氨酸18-26份、山栀苷12-16份、香芹酚8-12份、泛醌6-10份、4-甲氧基香豆雌醇4-7份、西红花苷0.02-0.06份。
所述活化助剂的制备方法为：
(1)将碳酸氢钠和天冬氨酸混合均匀，放入反应釜中，向反应釜中加入去离子水至混合溶液体积达到反应釜体积的85％，将反应釜放入60-80℃烘箱中反应3-4h，反应后自然冷却到室温；
(2)将茶多糖、山栀苷、香芹酚混合均匀，在60-80r/min转速下搅拌30-40min，放入-10℃冷冻1-2h，自然升温到常温后，和泛醌、4-甲氧基香豆雌醇、西红花苷混合均匀，加入混合物重量2-3倍的去离子水，在32-36℃水浴中120-150r/min转速下搅拌15-20min，搅拌后静置水浴2-3h，静置后离心，取上层清液；
(3)将上述步骤制备的上清液和步骤(1)反应后的溶液混合均匀，在100-120r/min转速下搅拌10-15min，用巴氏杀菌法进行瞬时灭菌，制得所述活化助剂。
所述生物制剂的制备方法为：
(1)将除活化助剂以外的其它原料混合均匀，放入蒸笼中在60-80℃、0.3-0.4MPa条件下蒸制40-50min，蒸制后采用紫外灯照射进行杀菌，杀菌后用破碎机破碎成粉末，向粉末中加入粉末重量12-15％的去离子水，放入-5℃下冷冻3-4h，冷冻后自然升温到常温，将粉末晾干；
(2)向上述粉末中加入粉末重量12-15倍的乙醇，浸泡2-3h，回流提取3-4h，离心过滤，收集第一滤液和第一沉淀物，将第一沉淀物加入其重量8-10倍的乙醇，回流提取1-2h，离心过滤，收集第二滤液，将第一滤液和第二滤液合并，减压浓缩回收乙醇，制得初级药水；
(3)将初级药水采用热压灭菌法灭菌50min，灭菌后和活化助剂混合均匀，继续采用热压灭菌法灭菌40min，向混合液中加入混合液重量50倍的注射用水，用1200r/min转速搅拌60-80min，搅拌后离心过滤，收集上清液，将上清液用药用活性炭吸附，吸附后采用巴士杀菌法进行杀菌，制得所述生物制剂。
所述生物制剂用于治疗类风湿性关节炎。
本发明提供了一种高澄清度的生物制剂，其有益效果为：
(1)生物制剂配方中加入活化助剂，能够激发中药配方中其它原料的治疗活性，缩短治疗周期，制备的生物制剂见效快，副作用小，不仅能够治疗类风湿性关节炎，而且能够补充气血，增强食欲。
(2)活化助剂采用特定配方，能够激发中药的药用活性，使药性发挥更快和更强，且活化助剂本身无副作用，在其制备方法中，首先将碳酸氢钠和天冬氨酸混合，利用天冬氨酸的两性，天冬氨酸中的羧基和碳酸氢钠反应生成羧酸盐和水，给活化助剂提供盐溶液基础，增强其它原料的稳定性，有利于活化助剂的耐久性。
(3)生物制剂制备方法中，首先将原料进行低温、高压蒸制，初步分解原料纤维组织，再杀菌，杀灭原料带有的细菌，破碎成粉末后冷冻，进一步分解原料纤维组织，然后用乙醇回流提取两次，合并提取液灭菌后，和活化助剂混合均匀，该混合方式既能保持原料的药性，又能发挥活化助剂的作用，使得制备的生物制剂药效快、副作用小。
具体实施方式：
为了使本发明实现的技术手段、创作特征、达成目的与功效易于明白了解，下面结合具体实施例，进一步阐述本发明。
实施例1：
一种高澄清度的生物制剂，由以下重量份的原料组成：黄柏10份、艾叶5份、柴胡1份、菖蒲6份、鱼腥草12份、半枝莲8份、五味子3份、土荆芥15份、大青叶7份、岩黄连16份、蒲公英18份、藏红花7份、土茯苓10份、黄芩8份、金银花4份、青蒿5份、穿山甲2份、牛筋草16份、骆驼蓬1份、苍术2份、活化助剂3份。
所述活化助剂由以下重量份的原料组成：茶多糖25份、碳酸氢钠12份、天冬氨酸18份、山栀苷12份、香芹酚8份、泛醌6份、4-甲氧基香豆雌醇4份、西红花苷0.02份。
所述活化助剂的制备方法为：
(1)将碳酸氢钠和天冬氨酸混合均匀，放入反应釜中，向反应釜中加入去离子水至混合溶液体积达到反应釜体积的85％，将反应釜放入60℃烘箱中反应3h，反应后自然冷却到室温；

(3)将上述步骤制备的上清液和步骤(1)反应后的溶液混合均匀，在100r/min转速下搅拌10min，用巴氏杀菌法进行瞬时灭菌，制得所述活化助剂。
所述生物制剂用于治疗类风湿性关节炎。
实施例2：
一种高澄清度的生物制剂，由以下重量份的原料组成：黄柏20份、艾叶8份、柴胡2份、菖蒲9份、鱼腥草15份、半枝莲10份、五味子5份、土荆芥20份、大青叶10份、岩黄连22份、蒲公英25份、藏红花10份、土茯苓13份、黄芩10份、金银花6份、青蒿8份、穿山甲3份、牛筋草20份、骆驼蓬2份、苍术5份、活化助剂5份。
所述活化助剂由以下重量份的原料组成：茶多糖35份、碳酸氢钠15份、天冬氨酸26份、山栀苷16份、香芹酚12份、泛醌10份、4-甲氧基香豆雌醇7份、西红花苷0.06份。
所述活化助剂的制备方法为：
(1)将碳酸氢钠和天冬氨酸混合均匀，放入反应釜中，向反应釜中加入去离子水至混合溶液体积达到反应釜体积的85％，将反应釜放入80℃烘箱中反应4h，反应后自然冷却到室温；
(2)将茶多糖、山栀苷、香芹酚混合均匀，在80r/min转速下搅拌40min，放入-10℃冷冻2h，自然升温到常温后，和泛醌、4-甲氧基香豆雌醇、西红花苷混合均匀，加入混合物重量3倍的去离子水，在36℃水浴中150r/min转速下搅拌20min，搅拌后静置水浴3h，静置后离心，取上层清液；
(3)将上述步骤制备的上清液和步骤(1)反应后的溶液混合均匀，在120r/min转速下搅拌15min，用巴氏杀菌法进行瞬时灭菌，制得所述活化助剂。
所述生物制剂的制备方法为：
(1)将除活化助剂以外的其它原料混合均匀，放入蒸笼中在60℃、0.3MPa条件下蒸制40min，蒸制后采用紫外灯照射进行杀菌，杀菌后用破碎机破碎成粉末，向粉末中加入粉末重量12％的去离子水，放入-5℃下冷冻3h，冷冻后自然升温到常温，将粉末晾干；
(2)向上述粉末中加入粉末重量12倍的乙醇，浸泡2h，回流提取3h，离心过滤，收集第一滤液和第一沉淀物，将第一沉淀物加入其重量8倍的乙醇，回流提取1h，离心过滤，收集第二滤液，将第一滤液和第二滤液合并，减压浓缩回收乙醇，制得初级药水；
(3)将初级药水采用热压灭菌法灭菌50min，灭菌后和活化助剂混合均匀，继续采用热压灭菌法灭菌40min，向混合液中加入混合液重量50倍的注射用水，用1200r/min转速搅拌60min，搅拌后离心过滤，收集上清液，将上清液用药用活性炭吸附，吸附后采用巴士杀菌法进行杀菌，制得所述生物制剂。
所述生物制剂用于治疗类风湿性关节炎。
实施例3：
一种高澄清度的生物制剂，由以下重量份的原料组成：黄柏15份、艾叶6份、柴胡1份、菖蒲8份、鱼腥草13份、半枝莲9份、五味子4份、土荆芥18份、大青叶9份、岩黄连19份、蒲公英21份、藏红花8份、土茯苓12份、黄芩9份、金银花5份、青蒿6份、穿山甲2份、牛筋草18份、骆驼蓬2份、苍术4份、活化助剂4份。
所述活化助剂由以下重量份的原料组成：茶多糖30份、碳酸氢钠14份、天冬氨酸22份、山栀苷14份、香芹酚10份、泛醌8份、4-甲氧基香豆雌醇6份、西红花苷0.04份。
所述活化助剂的制备方法为：
(1)将碳酸氢钠和天冬氨酸混合均匀，放入反应釜中，向反应釜中加入去离子水至混合溶液体积达到反应釜体积的85％，将反应釜放入70℃烘箱中反应3h，反应后自然冷却到室温；
(2)将茶多糖、山栀苷、香芹酚混合均匀，在70r/min转速下搅拌35min，放入-10℃冷冻1h，自然升温到常温后，和泛醌、4-甲氧基香豆雌醇、西红花苷混合均匀，加入混合物重量2倍的去离子水，在34℃水浴中130r/min转速下搅拌18min，搅拌后静置水浴3h，静置后离心，取上层清液；
(3)将上述步骤制备的上清液和步骤(1)反应后的溶液混合均匀，在110r/min转速下搅拌12min，用巴氏杀菌法进行瞬时灭菌，制得所述活化助剂。
所述生物制剂的制备方法为：
(1)将除活化助剂以外的其它原料混合均匀，放入蒸笼中在80℃、0.4MPa条件下蒸制50min，蒸制后采用紫外灯照射进行杀菌，杀菌后用破碎机破碎成粉末，向粉末中加入粉末重量15％的去离子水，放入-5℃下冷冻4h，冷冻后自然升温到常温，将粉末晾干；
(2)向上述粉末中加入粉末重量15倍的乙醇，浸泡3h，回流提取4h，离心过滤，收集第一滤液和第一沉淀物，将第一沉淀物加入其重量10倍的乙醇，回流提取2h，离心过滤，收集第二滤液，将第一滤液和第二滤液合并，减压浓缩回收乙醇，制得初级药水；
(3)将初级药水采用热压灭菌法灭菌50min，灭菌后和活化助剂混合均匀，继续采用热压灭菌法灭菌40min，向混合液中加入混合液重量50倍的注射用水，用1200r/min转速搅拌80min，搅拌后离心过滤，收集上清液，将上清液用药用活性炭吸附，吸附后采用巴士杀菌法进行杀菌，制得所述生物制剂。
所述生物制剂用于治疗类风湿性关节炎。
对比例：
一种治疗风湿性关节炎与类风湿性关节炎的中药组合物，
提供如下组分及其重量：
党参25g、制川乌55g、制草乌55g、何首乌25g、木瓜25g、牛膝14g、威灵仙14g、附子7g、桂枝14g、麻黄14g、荆芥25g、防风14g、羌活14g、天麻14g、红花14g、杜仲14g、五加皮14g、细辛7g、苍术14g、当归25g、川芎14g、木香14g、高良姜25g、丁香25g以及陈皮14g，共计495g；
将上述组分中的硬药打碎，然后将全部组分切成碎片，并加入陶瓷缸中；
向陶瓷缸中加入3.465～4.950Kg的55～60度白酒，浸没各组分，加盖密封陶瓷缸；
根据本实施例中药组合物的重量，优选放置三个月后，倒出陶瓷缸中的药液A，并过滤药液A得到药液B备用；
采用纱布包裹陶瓷缸中的各组分，并挤压，得到药液C，过滤药液C得到药液D备用；
混合药液B和药液D得到药液E，取出药液E并装瓶密封备用。
在使用时，将瓶装的药液E倒入杯中，每次2～5毫升，患者日服2次，早晚各一次，一月为一疗程，一疗程未愈可连续服用，孕妇、年老体弱者及十二岁以下儿童禁用。也可外用擦洗患处，每日一次，一月为一疗程。
对以上各实施例和对比例制备的制剂进行澄清度检测，选用散射光浊度仪进行检测浊度，结果如下：

选取临床200例类风湿性关节炎患者，选择在冬季-4～3℃温度期间进行实验，在此期间，类风湿性关节炎患者病情最严重，常表现为关节胀痛、酸痛、无法下蹲、腿关节无法直立，将200例患者平均分成4组，前3组分别注射本发明实施例1-3制备的生物制剂，每次注射5毫升，每天注射2次，第4组服用对比例所述药液E，每天服用两次，治疗1月后，观察治疗效果，结果如下：

其中，显效指病状完全消失，有效指病状得到缓解，无效指病状没有任何改变。

1.一种高澄清度的生物制剂，其特征在于，由以下重量份的原料组成：黄柏10-20份、艾叶5-8份、柴胡1-2份、菖蒲6-9份、鱼腥草12-15份、半枝莲8-10份、五味子3-5份、土荆芥15-20份、大青叶7-10份、岩黄连16-22份、蒲公英18-25份、藏红花7-10份、土茯苓10-13份、黄芩8-10份、金银花4-6份、青蒿5-8份、穿山甲2-3份、牛筋草16-20份、骆驼蓬1-2份、苍术2-5份、活化助剂3-5份。
2.根据权利要求1所述的高澄清度的生物制剂，其特征在于，所述活化助剂由以下重量份的原料组成：茶多糖25-35份、碳酸氢钠12-15份、天冬氨酸18-26份、山栀苷12-16份、香芹酚8-12份、泛醌6-10份、4-甲氧基香豆雌醇4-7份、西红花苷0.02-0.06份。
3.根据权利要求2所述的高澄清度的生物制剂，其特征在于，所述活化助剂的制备方法为：
(1)将碳酸氢钠和天冬氨酸混合均匀，放入反应釜中，向反应釜中加入去离子水至混合溶液体积达到反应釜体积的85％，将反应釜放入60-80℃烘箱中反应3-4h，反应后自然冷却到室温；
(2)将茶多糖、山栀苷、香芹酚混合均匀，在60-80r/min转速下搅拌30-40min，放入-10℃冷冻1-2h，自然升温到常温后，和泛醌、4-甲氧基香豆雌醇、西红花苷混合均匀，加入混合物重量2-3倍的去离子水，在32-36℃水浴中120-150r/min转速下搅拌15-20min，搅拌后静置水浴2-3h，静置后离心，取上层清液；
(3)将上述步骤制备的上清液和步骤(1)反应后的溶液混合均匀，在100-120r/min转速下搅拌10-15min，用巴氏杀菌法进行瞬时灭菌，制得所述活化助剂。
4.根据权利要求3所述的高澄清度的生物制剂，其特征在于，所述生物制剂的制备方法为：
