Abstract
本发明提供了一种聚乙二醇改性胶原膜,制备方法如下:以培养板为模具,取胶原蛋白2ml,加入浓度为10%w/v醋酸溶液中制备胶原膜;按照1000mg:1ml的比例将制备的300mg胶原膜置于1%w/v聚乙二醇溶液中交联,清洗,干燥,即得。本发明的优点在于:本发明胶原膜拥有更好的能力诱导颅顶骨缺损的修复,提高了以前胶原膜的生物性能,具有很好的应用前景,为引导再生术等相关增量手术提供了新选择。
Description
技术领域
本发明属于生物材料领域,具体涉及一种聚乙二醇改性胶原膜及其应用。
背景技术
牙科手术中常需要应用引导骨再生手术进行骨增量。胶原是细胞外基质的一种成分,已经广泛应用于引导骨再生术中。胶原膜有较好的生物兼容性,并能促进成骨细胞黏附、增殖、迁移和分化,也能有阻挡成纤维细胞快速入侵骨组织生长位点、隔开软组织与硬组织的生长等屏障功能。
然而,作为一种外源性移植物,胶原仍然会引起一定的炎症反应,同时也因为它的易降解性而缺乏较好的机械性能。本领域常用采用戊二醛与胶原进行交联,从而提高它的机械性能,但容易引发炎症和产生细胞毒性。因此,胶原膜长期的保存和使用,不仅仅需要提高机械性能,更要抗炎和提高生物兼容性。
发明内容
本发明的目的是针对现有技术中存在的不足,提供一种聚乙二醇改性胶原膜及其应用。
为了达到上述目的,本发明采用了下列技术方案:一种聚乙二醇改性胶原膜,制备方法如下:
A、以培养板为模具,取胶原蛋白2ml,加入浓度为10%w/v醋酸溶液中制备胶原膜;
B、按照1000mg:1ml的比例将制备的300mg胶原膜置于1%w/v聚乙二醇溶液中交联,清洗,干燥,即得。
作为优选,所述胶原蛋白为牛来源I型胶原。
作为优选,所述步骤A中,在-26℃温度下冷冻过夜,在-53℃、1Pa条件下,真空冷冻干燥24h。
作为优选,所述步骤B中,交联时间为12h。
作为优选,所述步骤B中,将制备好的胶原膜置于-20℃条件下冷冻干燥过夜。
作为优选,所述步骤B中,清洗采用1XPBS缓冲液漂洗3次。
所述改性胶原在医疗领域的应用。
本发明的优点在于:聚乙二醇是一种低毒性的材料,不会引起严重的排异反应。许多研究已经利用聚乙二醇改性材料表面并植入人体,得到了良好的结果。美国食品与药品监督管理局也批准了相应的材料可以在临床上使用。
本发明将胶原膜以聚乙二醇改性,进而得到促骨再生性能良好的聚乙二醇胶原复合膜,有望于应用在引导骨组织再生手术(GBR)中。
本发明胶原膜拥有更好的能力诱导颅顶骨缺损的修复,提高了以前胶原膜的生物性能,具有很好的应用前景,为引导再生术等相关增量手术提供了新选择。

具体实施方式
下面结合附图,对本发明作详细的说明。
为了使本发明的目的、技术方案及优点更加清楚明白,以下结合附图及实施例,对本发明进行进一步详细说明。应当理解,此处所描述的具体实施例仅仅用以解释本发明,并不用于限定本发明。
实施例1:一种聚乙二醇改性胶原膜,包括如下制备步骤:
1.胶原膜的制备
以直径为3.5cm/孔的培养板为模具,取2mL胶原蛋白加入浓度为10%(w/v)醋酸溶液。于-26℃低温冰箱冷冻过夜,在-53℃、1Pa条件下,冻干机真空冷冻干燥24h,制备胶原膜。
2.配制聚乙二醇(1%w/v)溶液。
3.聚乙二醇改性的胶原膜
将制备的胶原膜置于1%w/v聚乙二醇溶液中改性12h后,lXPBS缓冲液漂洗3次。
将制备好的聚乙二醇改性胶原膜于-20℃条件下冷冻干燥过夜
实施例2:一种聚乙二醇改性胶原膜,,制备方法如下:
A、以培养板为模具,取胶原蛋白2ml,加入浓度为10%w/v醋酸溶液中制备胶原膜;
B、按照1000mg:1ml的比例将制备的300mg胶原膜置于1%w/v聚乙二醇溶液中交联,清洗,干燥,即得。
所述胶原蛋白为牛来源I型胶原。
所述步骤A中,在-26℃温度下冷冻过夜,在-53℃、1Pa条件下,真空冷冻干燥24h。
所述步骤B中,交联时间为12h。
所述步骤B中,将制备好的胶原膜置于-20℃条件下冷冻干燥过夜。
所述步骤B中,清洗采用1XPBS缓冲液漂洗3次。
实施例3:分别取等量的胶原膜、聚乙二醇改性胶原膜,作为实验对比组。
1.扫描电子显微镜(SEM)检测表面形态
扫描式电子显微镜观察材料表面形态,分别为胶原膜、聚乙二醇改性胶原膜。如图1所示,聚乙二醇改性胶原膜表面变的更为平滑。
2.HE染色观察骨再生情况
如图2所示,聚乙二醇改性的胶原膜组别植入大鼠颅顶骨缺损(圆形,直径:5mm),明显促进骨再生。
3.Masson染色胶原沉积
如图3所示,聚乙二醇改性的胶原膜组别明显促进胶原的沉积。

Claims (7)
1.一种聚乙二醇改性胶原膜,其特征在于制备方法如下:
A、以培养板为模具,取胶原蛋白2ml,加入浓度为10%w/v醋酸溶液中制备胶原膜;
B、按照1000mg:1ml的比例将制备的300mg胶原膜置于1%w/v聚乙二醇溶液中交联,清洗,干燥,即得。
2.根据权利要求1所述的一种聚乙二醇改性胶原膜,其特征在于:所述胶原蛋白为牛来源I型胶原。
3.根据权利要求1所述的一种聚乙二醇改性胶原膜,其特征在于:所述步骤A中,在-26℃温度下冷冻过夜,在-53℃、1Pa条件下,真空冷冻干燥24h。
4.根据权利要求1所述的一种聚乙二醇改性胶原膜,其特征在于:所述步骤B中,交联时间为12h。
5.根据权利要求1所述的一种聚乙二醇改性胶原膜,其特征在于:所述步骤B中,将制备好的胶原膜置于-20℃条件下冷冻干燥过夜。
6.根据权利要求1所述的一种聚乙二醇改性胶原膜,其特征在于:所述步骤B中,清洗采用1XPBS缓冲液漂洗3次。
7.根据权利要求1-6任一所述的一种聚乙二醇改性胶原膜,其特征在于:所述改性胶原在医疗领域的应用。
