本发明公开了淀粉‑胆碱复合体系,所述体系用淀粉与胆碱或胆碱衍生物通过物理或化学的复合作用制得,所述物理或化学的复合作用起到制孔作用,所述制孔作用可提高所述体系的孔隙率。淀粉‑胆碱复合体系具有良好的生物相容性和吸收性,可以作为具有生物相容性的可吸收生物医用材料应用于止血、吸收渗液、伤口封闭,组织粘结以及组织防粘连等用途。

淀粉-胆碱复合体系及其制备方法和应用
技术领域
本发明属于生物医用材料领域,涉及淀粉-胆碱复合体系及其制备方法和应用。
背景技术
淀粉是禾本科植物玉蜀薯的硕果或大豆科植物木薯的块根中制的多糖类颗粒。淀粉具有良好的生物安全性、生物相容性及可吸收性,在药剂中主要用做稀释剂、填充剂、粘合剂等。淀粉作为吸水剂时,依靠其粗糙的表面以及表面的微孔所形成的较大的比表面积所产生的物理所用。当淀粉遇水时,淀粉上大量的羟基亲水基团于水分子进行水合作用,同时淀粉的高分子网络及多孔结构内外形成离子浓度差,进而在淀粉颗粒的内外形成渗透压,在渗透压的作用下产生吸液能力。胆碱属于B族维生素,在食品工业中常用作营养增补剂,无毒性,一般公认是安全的。胆碱及胆碱衍生物可通过与淀粉的物理及化学复合作用的一种或其组合起到制孔作用,从而提高淀粉-胆碱复合体系的孔隙率,增强吸液能力。因此,淀粉-胆碱复合体系具有优良的生物相容性、可吸收性及优良的吸液能力,可以作为生物医用材料应用。使用具有生物相容性的可吸收的淀粉-胆碱复合体系可满足外伤、急救、家庭自救、外科手术、军需战备的迫切要求,对开发止血效果更佳、绿色、环保、可降解、无毒、无副作用、无刺激性、具有良好的生物相容性的医用材料具有重要意义。
发明内容
有鉴于此,本发明提供淀粉-胆碱复合体系及其制备方法和应用。
本发明具体提供了如下的技术方案:
1、淀粉-胆碱复合体系,所述体系用淀粉与胆碱或胆碱衍生物通过物理或化学的复合作用制得,所述物理或化学的复合作用起到制孔作用,所述制孔作用可提高所述体系的孔隙率。
进一步,所述孔隙率的调节是通过改变胆碱或胆碱衍生物所含有的基团、用量或反应条件中的一种或几种实现。
进一步,所述物理复合作用包括静电作用、氢键作用或范德华力作用的一种或几种;所述化学复合作用包括自由基反应、离子型反应、亲电反应,亲核反应、硝化反应、卤化反应、磺化反应、氨化反应、酰化反应、氰化反应、加成反应、开环反应、消去反应、取代反应、加聚反应或缩聚反应的一种或几种。
进一步,所述体系能够在水中溶解或溶胀形成粘性胶膜。
进一步,所述体系具备可调节的吸水性及溶胀性,所述体系具备多微孔结构。
进一步,所述淀粉为玉米淀粉、木薯淀粉、红薯淀粉、马铃薯淀粉、小麦淀粉、糯米淀粉、变性淀粉、预糊化淀粉、糊精、酸变性淀粉、氧化淀粉、醚化淀粉、酯化淀粉、氨化淀粉、交联淀粉和复合变性淀粉中的一种或几种。
进一步,所述胆碱衍生物为氢氧化胆碱、氨化胆碱、氯化胆碱、溴化胆碱、碘化胆碱、磷酸胆碱、醋酸胆碱、巯基胆碱、氨基胆碱、叠氮基胆碱、烯基胆碱、炔基胆碱、环氧基胆碱、羧基胆碱或酯基胆碱中的一种或几种。
进一步,按重量份数计,淀粉1~10000份,胆碱或胆碱衍生物1~1000000份;所述淀粉分子量为10000-3000000道尔顿。
2、淀粉-胆碱复合体系的制备方法,将淀粉溶于去离子水中,50-100℃加热搅拌0.1-72小时,将胆碱或胆碱衍生物溶于去离子水中,缓慢滴加到淀粉溶液中,搅拌0.1-300小时后,常规乳化成型,干燥后得到淀粉-胆碱复合体系。
3、淀粉-胆碱复合体系作为生物医用材料在止血、吸收渗液、伤口封闭,组织粘结以及组织防粘连上的应用;所述生物医用材料包括粉剂材料、液体材料、凝胶材料、海绵材料和纤维材料。
本发明的有益效果在于:胆碱或胆碱衍生物通过与淀粉的物理或化学作用的一种或其组合能够起到制孔作用,从而提高淀粉-胆碱复合体系的孔隙率。较大的孔隙率能够使得淀粉-胆碱复合体系的三维空间网络结构具有较高的空隙,使得淀粉-胆碱复合体系的内外形成较高的渗透压,达到较快的吸液速率。同时淀粉-胆碱复合体系的三维空间网络结构能够有效的限制水分子的运动,因此具备良好的吸水倍率以及吸水性能。
通过改变胆碱或胆碱衍生物所含有的基团、用量、反应条件调节淀粉-胆碱复合体系的孔隙率,从而使淀粉-胆碱复合材料体系具有可调节的吸水及溶胀特性,可以作为吸液材料。
胆碱或胆碱衍生物通过与淀粉的物理复合作用包括静电作用,氢键作用和范德华力作用。胆碱或胆碱衍生物通过与淀粉的化学作用包括有自由基反应,离子型反应、亲电反应、亲核反应、硝化反应、卤化反应、磺化反应、氨化反应、酰化反应、氰化反应、加成反应、开环反应、消去反应、取代反应、加聚反应、缩聚反应等。
淀粉-胆碱复合体系的三维空间网络结构不但具有较好的吸水性能,同时还具有良好的吸附性能,在应用于出血伤口时能够将血液中的血红细胞、血小板、血红蛋白快速的富集到淀粉-胆碱复合体系的表面,激活内源性凝血机制,发生凝血反应,并保持促凝血活性,起到快速止血的效果,促进伤口愈合。
淀粉-胆碱复合体系具有良好的系内的亲和作用,在水中溶解或溶胀时能够依靠复合体系表面原子化合价产生的吸附力或分子间的氢键作用产生良好的亲和力形成粘性胶膜。该胶膜可以有效封闭伤口和组织、血管断面,防止出血和组织液渗出,防止外界感染,促进组织修复。由于淀粉-胆碱复合体系良好的生物形容性和可吸收性,所形成的胶膜也可以用于组织间的隔层,起到组织间防粘连的作用。
淀粉-胆碱复合体系材料具有良好的生物相容性和吸收性,可以作为具有生物相容性的可吸收生物医用材料应用于止血、吸收渗液、伤口封闭,组织粘结以及组织防粘连等用途。

具体实施方式
下面结合附图,对本发明的优选实施例进行详细的描述。
实施例1
淀粉-胆碱复合体系的制备:将1g玉米淀粉(分子量10000-3000000道尔顿)与100mg胆碱溶于200mL去离子水中,90℃加热搅拌4小时。常规乳化成型,干燥后得到淀粉-胆碱复合体系。
实施例2
淀粉-胆碱衍生物复合体系的制备:将1g马铃薯淀粉(分子量10000-3000000道尔顿)溶于200mL去离子水中,90℃加热搅拌4小时。将50mg氯化胆碱溶于20mL去离子水中后缓慢滴加到淀粉溶液中,搅拌2小时后,常规乳化成型,干燥后得到淀粉-胆碱复合体系。
实施例3
淀粉-胆碱衍生物复合体系的制备:将1g氨化淀粉(分子量10000-3000000道尔顿)溶于200mL的去离子水溶液中;将100mg酯基胆碱溶于20mL去离子水中后滴加到氨化淀粉溶液中,搅拌12小时后,常规乳化成型,干燥后得到淀粉-胆碱复合体系。
实施例4
淀粉-胆碱衍生物复合体系的制备:将1g氧化淀粉(分子量10000-300000道尔顿)溶于200mL去离子水中,搅拌均匀后,加入100mg氨化胆碱,搅拌24小时后,常规乳化成型,干燥后得到淀粉-胆碱复合体系。
从图1为为淀粉-胆碱复合体系的电镜图片图,从图1中可以看出淀粉-胆碱复合体系结构完整,表面具有丰富的孔洞,具有较高的比表面积,能够快速吸液。
从图2为淀粉-胆碱复合体系对血细胞的富集电子显微镜图片图,从图2中可以看出淀粉-胆碱复合体系的边缘富集了大量新鲜兔全血中的血细胞,所富集的血细胞能够快速激活内源性凝血机制,实现快速止血。
从图3为淀粉-胆碱复合体系在新西兰白兔股动脉剪断后的快速止血效果以及形成的粘胶膜效果图,从图3中可以看出在将新西兰白兔股动脉剪断并施予淀粉-胆碱复合体系后,能够在伤口快速形成体系内亲和力介导的粘胶膜,该粘胶膜能够有效封闭伤口,止血并吸收渗液,和阻隔外部细菌。
