Abstract
本发明涉及一种三唑酮缓释体系及其制备方法,包括以下步骤:(1)制备聚多巴胺修饰的介孔二氧化硅:混合介孔二氧化硅、盐酸多巴胺,搅拌溶解,然后过滤,所得固体即为聚多巴胺修饰的介孔二氧化硅;(2)制备金属离子与聚多巴胺修饰的介孔二氧化硅:在聚多巴胺修饰的介孔二氧化硅中加入金属离子溶液,搅拌,然后过滤,所得固体即为金属离子与聚多巴胺修饰的介孔二氧化硅;(3)将步骤(2)获得的金属离子与聚多巴胺修饰的介孔二氧化硅真空活化后,浸入三唑酮溶液中,然后过滤,得到固体,即为三唑酮缓释体系。本发明制备的三唑酮高效缓释体系提高了样品的吸附量。

技术领域
本发明涉及一种三唑酮缓释体系及其制备方法,具体涉及一种采用聚多巴胺包覆的介孔二氧化硅,然后与金属离子络合,形成三唑酮高效缓释体系。
背景技术
在过去的十年中,介孔二氧化硅材料(MCM-41)已经引起了全球制药研究人员的关注。我们知道,介孔二氧化硅材料具有许多独特的性质,如无毒性,大表面积和孔体积,可调孔径,以及化学惰性和易于改性的表面性质。经过十多年的蓬勃发展,介孔二氧化硅纳米粒子被认为是治疗,诊断,预后和组合应用中最有前景的生物医学平台之一。得益于它们稳定的介孔结构,大表面积,可调节的孔径,易于表面官能化和良好的生物相容性,中孔二氧化硅纳米粒子不仅可以进行微调以实现理想的物理化学特性以适应多种货物分子,例如治疗药物,蛋白质,基因和显像剂,可以单独使用或组合使用,也可以设计用于促进按需药物释放和多模态成像。同时,各种类型的这种材料已经发展成药物输送系统。
药物主要通过吸附进入介孔二氧化硅,因此后者必须进行表面修饰以防止药物泄漏并控制药物释放。聚多巴胺(PDA)是一种仿生聚合物,它可以在弱碱性条件下(pH8.0-8.5)通过氧化聚合在各种材料上形成,包括陶瓷,共聚物和半导体。PDA涂层作为介孔二氧化硅表面的优秀守门员,对pH值极其敏感。使用PDA包衣,药物分子在中性条件下很容易在介孔二氧化硅纳米颗粒(MSN)中被阻断,并在较低的pH值下释放。
基于上述研究,我们提出了pH响应高效聚多巴胺修饰的三唑酮/MCM-41缓释体系。与金属离子络合,其性能进一步提高。通过金属离子的桥效应,PDA与三唑酮通过配位键相互作用。最后,开发了pH敏感性高,对农药输送具有较大吸附能力的高效缓释系统。
发明内容
本发明的目的在于克服上述现有技术的不足之处而提供一种三唑酮缓释体系及其制备方法。
为实现上述目的,本发明采用的技术方案为:一种三唑酮缓释体系的制备方法,包括以下步骤:
(1)制备聚多巴胺修饰的介孔二氧化硅:混合介孔二氧化硅、盐酸多巴胺,搅拌溶解,然后过滤,所得固体即为聚多巴胺修饰的介孔二氧化硅;
(2)制备金属离子与聚多巴胺修饰的介孔二氧化硅:在聚多巴胺修饰的介孔二氧化硅中加入金属离子溶液,搅拌,然后过滤,所得固体即为金属离子与聚多巴胺修饰的介孔二氧化硅;
(3)将步骤(2)获得的金属离子与聚多巴胺修饰的介孔二氧化硅真空活化后,浸入三唑酮溶液中,然后过滤,得到固体,即为三唑酮缓释体系。
作为本发明所述的三唑酮缓释体系的制备方法的优选实施方式,所述步骤(1)中介孔二氧化硅与盐酸多巴胺的重量比为5:1。
作为本发明所述的三唑酮缓释体系的制备方法的优选实施方式,所述步骤(1)中将所述介孔二氧化硅与盐酸多巴胺溶解的溶液为pH值=8.5的三羟甲基氨基甲烷-盐酸缓冲溶液。
作为本发明所述的三唑酮缓释体系的制备方法的优选实施方式,所述步骤(2)中,聚多巴胺修饰的介孔二氧化硅的重量与金属离子溶液的物质的量的比例为2000:1。
作为本发明所述的三唑酮缓释体系的制备方法的优选实施方式,所述步骤(2)中所述金属离子溶液为金属硝酸盐溶液。
作为本发明所述的三唑酮缓释体系的制备方法的优选实施方式,所述金属硝酸盐溶液为硝酸铜、硝酸锌或硝酸铁。
作为本发明所述的三唑酮缓释体系的制备方法的优选实施方式,所述步骤(3)中,金属离子和聚多巴胺修饰的介孔二氧化硅与三唑酮溶液中的三唑酮的重量比为3:2。

本发明还提供一种由上述三唑酮缓释体系的制备方法制备而成的三唑酮释缓体系。
与现有技术相比,本发明具有如下有益效果:
聚多巴胺膜包覆的介孔硅提高了样品的吸附量。聚多巴胺膜包覆后,在介孔空的表面形成复合膜,复合膜与基体之间存在缝隙,使得载体的比表面积增加,有助于增强载体对三唑酮的吸附能力。金属离子在聚多巴胺膜表面形成络合物后进一步提高了载体介孔硅的吸附能力。聚多巴胺膜包覆的介孔硅的缓释速率较未修饰介孔硅的要高,而通过金属离子络合载体的药物缓释速率要更快。

为了更加简洁明了的展示本发明的技术方案、目的和优点,下面结合具体实施例对本发明做进一步的详细描述。
实施例1
本实施例的一种三唑酮缓释体系的制备方法包括以下步骤:
(1)制备聚多巴胺修饰的介孔二氧化硅(PDA-MCM-41):将0.05g介孔二氧化硅(MCM-41),10mg盐酸多巴胺和80mL三羟甲基氨基甲烷-盐酸(THAM-HCl)缓冲溶液(pH=8.5)加入烧瓶中,在25℃下搅拌溶解24小时,然后过滤,将所得的固体洗涤并干燥后获得PDA-MCM-41,备用。
(2)制备金属离子与聚多巴胺修饰的介孔二氧化硅(M-PDA-MCM-41):在30℃,搅拌条件下,向100mg PDA-MCM-41中加入50mL硝酸铜溶液(0.1mol/L)24小时。然后过滤将所得的固体洗涤,90℃烘箱干燥24h,得到金属铜离子与聚多巴胺修饰的介孔硅,记为Cu-PDA-MCM-41,备用。
(3)将步骤(2)获得的Cu-PDA-MCM-41,在80℃真空下活化6h。将300mg样品在35℃下搅拌24小时浸入20mL三唑酮乙醇溶液(10mg/mL)中,然后过滤将所得的固体洗涤并干燥,得到金属铜离子与聚多巴胺修饰的介孔硅三唑酮释缓体系,记为Tri/Cu-PDA-MCM-41。同时,制备不同载体的三唑酮样品,进行性能比较分析,分别记为Tri/MCM-41,Tri/PDA-MCM-41。
实施例2
本实施例的一种三唑酮缓释体系的制备方法包括以下步骤:
(1)制备聚多巴胺修饰的介孔二氧化硅(PDA-MCM-41):将0.05gMCM-41,10mg盐酸多巴胺和80mL THAM-HCl缓冲溶液(pH=8.5)加入烧瓶中,在25℃下搅拌溶解24小时,然后过滤,将所得的固体洗涤并干燥后获得PDA-MCM-41,备用。
(2)制备金属离子与聚多巴胺修饰的介孔二氧化硅(M-PDA-MCM-41):在30℃,搅拌条件下,向100mg PDA-MCM-41中加入50mL硝酸锌溶液(0.1mol/L)24小时。然后过滤将所得的固体洗涤,90℃烘箱干燥24h,得到金属锌离子与聚多巴胺修饰的介孔硅,记为Zn-PDA-MCM-41,备用。
(3)将步骤(2)获得的Zn-PDA-MCM-41,在80℃真空下活化6h。将300mg样品在35℃下搅拌24小时浸入20mL三唑酮乙醇溶液(10mg/mL)中,然后过滤将所得的固体洗涤并干燥,得到金属锌离子与聚多巴胺修饰的介孔硅三唑酮释缓体系,记为Tri/Zn-PDA-MCM-41。
实施例3
本实施例的一种三唑酮缓释体系的制备方法包括以下步骤:
(1)制备聚多巴胺修饰的介孔二氧化硅(PDA-MCM-41):将0.05g MCM-41,10mg盐酸多巴胺和80mL THAM-HCl缓冲溶液(pH=8.5)加入烧瓶中,在25℃下搅拌溶解24小时,然后过滤,将所得的固体洗涤并干燥后获得PDA-MCM-41,备用。
(2)制备金属离子与聚多巴胺修饰的介孔二氧化硅(M-PDA-MCM-41):在30℃,搅拌条件下,向100mg PDA-MCM-41中加入50mL硝酸铁溶液(0.1mol/L)24小时。然后过滤将所得的固体洗涤,90℃烘箱干燥24h,得到金属铁离子与聚多巴胺修饰的介孔硅,记为Fe-PDA-MCM-41,备用。
(3)将步骤(2)获得的Fe-PDA-MCM-41,在80℃真空下活化6h。将300mg样品在35℃下搅拌24小时浸入20mL三唑酮乙醇溶液(10mg/mL)中,然后过滤将所得的固体洗涤并干燥,得到金属铁离子与聚多巴胺修饰的介孔硅三唑酮释缓体系,记为Tri/Fe-PDA-MCM-41。
将上述实施例的样品进行理化分析鉴定其理化性质:
1、吸附性能测试计算公式
使用来自UV-2550 UV-Vis分光光度计测量中孔二氧化硅吸附的三唑酮的量。进行不同浓度的三唑酮标准溶液在λ=223nm处的溶液浓度(C/(mg*L-1))和吸光度(A)的线性回归以获得标准曲线方程:C=40.00A+9.220,R2=0.9993。在三唑酮的乙醇溶液中进行吸附之前和之后,进行UV光谱测定该溶液的吸光度。吸附容量(AC)和加载量(LC)可以通过以下公式计算:

使用Bruker AXS D8 X射线衍射仪,在25℃,40kV和30mA下用Cu辐射和石墨单色仪分析样品。以0.02°步长以2°/分钟(角度范围2θ=0.5~10°)扫描测量结果。在2×10-7Pa的真空下,在ESCALAB250XI光谱仪(ThermoFisherScientific,AlKα,hν=1486.6eV)上记录X射线光电子能谱(XPS),通过调节主要C1s峰值到284.8eV的位置。通过使用KBr压片技术,通过Spectrum100傅里叶红外光谱仪(PerkinElmer Inc.,USA)分析颗粒的形态。在N2保护下将金颗粒喷在样品表面上,用S4800扫描电子显微镜观察SEM观察表面形貌。在FEI Tecnai G2F20传输选择显微镜上进行TEM观察。样品的BET表面积通过在77K下的N2吸附等温线测定,在Quadrasorb SI吸附设备上操作。在N2吸附实验之前,将样品在200℃真空脱气12小时。通过电感耦合等离子体原子发射光谱法(Agilent 725)确认金属离子的负载量。
4、介孔材料的结构表征
采用傅立叶红外光谱仪(FTIR)比较MCM-41,PDA-MCM-41,Zn-PDA-MCM-41,Cu-PDA-MCM-41和Fe-PDA-MCM-41的不同组成。如图1所示,MCM-41分别在3420cm-1和950cm-1出现了两个谱带,分别归因于Si-OH的拉伸和弯曲振动。800cm-1处的谱带归因于SiO2骨架上Si-O-Si的特征峰。对于PDA-MCM-41的谱图,780cm-1出现新的谱带,属于邻位双取代芳香环的特征峰,PDA的羟基组分带3420cm-1明显增强证实样品被PDA封装。与金属离子配位后发生的伸缩振动带从1631cm-1到1625cm-1的C=O蓝移。
图2显示了MCM-41和PDA-MCM-41的小角度X射线衍射XRD图谱。有三个特征峰归属于(100),(110)和(200)晶面,分别表明这些颗粒具有规则的六方孔结构。经PDA封装后,XRD峰仍然存在,表明样品中仍然存在正六边形孔结构。PDA封装后,(110)晶面和(200)晶面的XRD峰强度降低,证明PDA被引入体系并降低其有序性。
如图3所示,进行了X射线光电子能谱(XPS)分析以确定表面元素的化学状态。在通过PDA包封后,氮的新结合能(BE)出现如图3a所示,表明介孔二氧化硅已被PDA封装。对于如图3b和图3d,三价铁和锌从710.535电子伏特降低到710.478eV和装载三唑酮后分别从1021.645电子伏特至1021.831eV的增加,由于所述的Fe-PDA-MCM-41和Zn-PDA-MCM-41电子在金属离子和三唑酮之间转移,这意味着噻唑酮和金属离子之间形成配位相互作用,增强了介孔二氧化硅载体与三唑酮的相互作用。而对于Cu-PDA-MCM-41,如图3c所示,加载三唑酮表明铜离子和三唑酮之间的强相互作用后,铜的BE具有从934.585eV至933.893eV的显著蓝移。
如图4a所示,MCM-41和PDA-MCM-41的N2吸附/解吸等温线属于LangmuirⅣ(斜率下降)和H4滞后环(迟滞环在p/po=0.4),这证实了他们的介孔结构根据以前的报告。如图4b所示,所有样品的孔径分布从3nm到4nm符合IUPAC对介孔材料的推荐。更重要的是,如表1所示,由于PDA和介孔二氧化硅之间形成的空隙面积和新的孔隙,PDA包封将显着增加介孔二氧化硅的BET表面积和孔体积,但对孔径略有影响。

图5描绘了MCM-41和PDA-MCM-41的SEM和TEM图像。如图所示,两个样品的正六边形孔结构保持良好而没有团聚,与XRD结果一致。在图5a和c中,MCM-41的表面粗糙。PDA封装后,样品表面检测到薄层,如图5b和d所示。
5、吸附性能分析
表2列出了各种介孔二氧化硅的AC。PDA包容将显著增加介孔二氧化硅的BET表面积和孔体积。此外,PDA层与三唑酮之间的π-π相互作用改善了三唑酮与介孔二氧化硅载体之间的相互作用。因此,用PDA封装后介孔二氧化硅的交流电明显增加。根据XPS分析,由于PDA层-金属离子-三唑酮之间形成的配位相互作用,加强了三唑酮与介孔二氧化硅载体之间的相互作用,与PDA络合的铁离子和锌离子进一步增加了样品的吸附能力。例如,Fe-PDA-MCM-41的AC增加到173mg/g,比MCM-41多160%。如Cu-PDA-MCM-41,由于铜离子和三唑酮之间的强相互作用,AC减少,如XPS分析所示。PDA和金属离子之间的配位可能会因致密化而减弱,PDA层和三唑酮之间的金属离子的桥接效应将失去作用。

6、不同载体的三唑酮在不同pH值条件下缓释速率
由图6可知,MCM-41在酸性或者碱性的条件下释放都比中性条件下的快,随着酸性的加强,药物的释放也逐渐加快。碱性条件下的三唑酮释放比酸性条件稍慢。MCM-41在不同pH条件下缓释十天时间,缓释率都达到了20%至25%。由于酸性或碱性条件都可能对介孔结构造成破坏使药物析出,所以达到缓释平衡时中性条件下的样品的缓释率最低。
图7表明PDA-MCM-41在5小时内的缓释速率pH=1条件下最为迅速。pH=4与pH=7条件下速率接近,为中等速率。pH=10的碱性条件下释放速率较慢。缓释平衡后,pH=1与pH=7条件下的样品能达到较高的缓释率,在75%左右。pH=4条件下最低,为45%。酸性缓释体系中的氢离子容易对聚多巴胺膜造成破坏,相反碱性条件对聚多巴胺膜有保护作用,所以酸性与中性条件下的药物释放速率都比碱性条件下的快。
从图8可以看出Fe-PDA-MCM-41的释放行为在pH=1的条件下与模型拟合度不好,其他pH值条件下的拟合相关性则较高。同样,样品的释放速率为酸性体系>中性体系>碱性体系。缓释平衡时,pH=1条件下的样品达到了45%的缓释率,其余的样品缓释率在25%左右。
图9为载药Zn-PDA-MCM-41样品在不同pH条件下的药物缓释曲线。酸性和碱性的缓释体系都加快了Zn-PDA-MCM-41样品的药物缓释速率,酸性体系的加速效果较碱性体系更为明显。达到缓释平衡后,酸性和碱性条件下样品的缓释率较中性条件的高,分布在30%至35%之间。pH=7条件下样品缓释率较低,仅达到了19%。
图10为载药Cu-PDA-MCM-41样品在不同pH条件下的药物缓释曲线。样品的药物缓释速率为酸性体系>中性体系>碱性体系,且随着酸性的加强,药物的析出也加快。由于Cu离子具有很强的还原性,经过Cu离子修饰的样品可以达到较高的缓释率,分布在50%至60%之间。
图11为各组样品在pH值7的体系的缓释曲线,其中a为MCM-41,b为PDA-MCM-41,c为Fe-PDA-MCM-41,d为Zn-PDA-MCM-41,e为Cu-PDA-MCM-41。在pH=7的缓释体系下,PDA-MCM-41和Cu-PDA-MCM-41能够达到较高的缓释率,分别为73%和57%。其余样品的缓释率较为接近,在20%左右。MCM-41、Cu-PDA-MCM-41短时间内的缓释率达到了35%,释放速率较快。三唑酮释放速率PDA-MCM-41>Cu-PDA-MCM-41>Fe-PDA-MCM-41>MCM-41>Zn-PDA-MCM-41。经过Fe离子和Zn离子修饰过的样品缓释性能得到了改善。Fe离子和Zn离子在聚多巴胺膜表面络合改变了膜的通透性,膜表面结构变得紧密。
7、持续释放测试
图12描述了Tri/MCM-41,Tri/PDA-MCM-41,Tri/Zn-PDA-MCM-41,Tri/Cu-PDA-MCM-41和Tri/Fe-41在不同的pH值下。在如图6a所示的自然条件下,释放速度顺序为Tri/PDA-MCM-41>Tri/Cu-PDA-MCM-41>Tri/Fe-PDA-MCM-41>Tri/MCM-41≈Tri/Zn-PDA-MCM-41表明由于三唑酮被介孔二氧化硅表面上的PDA层吸附而容易释放到溶液中,因此PDA包含增加了释放速度。金属离子的络合作为桥梁,通过配位键,特别是锌离子和铁离子,增强了PDA层与三唑酮的相互作用。此外,与金属离子复合后,PDA层变得致密,这增加了持续释放系统的空间位阻。结果,金属离子肤色后释放速度降低。
对于Tri/MCM-41,在不同pH值下的缓释性能没有显着差异。经PDA装饰后,该系统对pH敏感。在如图6b和c所示的酸性条件下,pH越低,用PDA包封的样品的释放速度越快。由于PDA膜在酸性条件下不稳定并倾向于水解。因此,被PDA层吸附的三唑酮会被释放并且释放速度加快。在图6d所示的碱性条件下,由于碱性条件对PDA膜的保护作用,释放速度变小以增加空间位阻。结果表明,根据不同的pH值,PDA包封的体系具有pH敏感性和不同的缓释性能。对于Tri/Cu-PDA-MCM-41和Tri/Fe-PDA-MCM-41,不同pH值下的性能趋势与Tri/PDA-MCM-41相似。而对于Tri/Zn-PDA-MCM-41,氢氧根离子与锌离子的配位作用强于PDA层-锌离子-三唑酮配位作用。因此,在碱性条件下,PDA层-锌离子-三唑酮配位键减弱并且加速释放速度加快。
8、缓释动力学研究
为了进一步了解缓释机制,将pH值为7的Tri/MCM-41,Tri/PDA-MCM-41和Tri/Cu-PDAMCM-41三唑酮的缓释数据拟合为零级模型,一级模型,Higuchi模型和Korsmeryer-Pappas模型。如表3所示,缓释颗粒的药物释放行为与Korsmeryer-Pappas动力学方程最为一致。根据Tri/MCM-41,Tri/PDA-MCM-41和Tri/Fe-PDAMCM-41的动力学方程计算时间(t)的扩散系数n次幂为0.0543,0.2880和0.0619,低于0.45。得到的数据表明,三唑酮从颗粒中的持续释放是通过Fickian扩散机制来控制的,这证明了浓度的差异是对释放过程的主要影响。换句话说,介孔二氧化硅的缓释机制并未因PDA装饰或金属离子肤色而改变。

图13和14分别是MCM-41和PDA-MCM-41的Higuchi模型拟合曲线,表4为Higuchi模型拟合数据。从中可以看出MCM-41在pH=4、7、10的时候相关系数达到了0.95,与模型拟合度较好,所以MCM-41的释放模型为Higuchi模型。此时n≤0.45,释放机理为Fick扩散。PDA-MCM-41与模型拟合相关系数在0.9左右,但是释放指数n>1,不符合Higuchi模型的释放机理。

图15与16分别是MCM-41和PDA-MCM-41的Korsmeyer-Peppas模型拟合曲线,表5为Korsmeyer-Peppas模型拟合数据。由表可知,MCM-41的拟合相关系数均达到了0.9,拟合度高。PDA-MCM-41相关系数也达到0.8以上。但是两个样品的释放指数n均小于0.45,不符合Peppas模型的扩散与溶蚀。

图17与18分别是MCM-41和PDA-MCM-41的零级模型拟合曲线,表6为零级模型拟合数据。由数据可以得出,MCM-41相关系数较高,与零级模型拟合度较好,释放机理更接近恒速释放。
PDA-MCM-41在四个模型中与零级模型最为符合,所以
PDA-MCM-41的释放机理为恒速释放。

图19与20分别是MCM-41和PDA-MCM-41的一级模型拟合曲线,表7为一级模型拟合数据。两个样品的相关系数R均大于0.8,但是模型拟合的结果显示样品的释放指数n不存在。表明MCM-41和PDA-MCM-41的缓释数据不符合一级模型。

以上所述实施例仅表达了本发明的几种实施方式,其描述较为具体和详细,但并不能因此而理解为对发明专利范围的限制。应当指出的是,对于本领域的普通技术人员来说,在不脱离本发明构思的前提下,还可以做出若干变形和改进,这些都属于本发明的保护范围。因此,本发明专利的保护范围应以所附权利要求为准。
Claims (6)
1.一种三唑酮缓释体系的制备方法,其特征在于,包括以下步骤:
(1)制备聚多巴胺修饰的介孔二氧化硅:混合介孔二氧化硅、盐酸多巴胺,搅拌溶解,然后过滤,所得固体即为聚多巴胺修饰的介孔二氧化硅;
(2)制备金属离子与聚多巴胺修饰的介孔二氧化硅:在聚多巴胺修饰的介孔二氧化硅中加入金属离子溶液,搅拌,然后过滤,所得固体即为金属离子与聚多巴胺修饰的介孔二氧化硅;
(3)将步骤(2)获得的金属离子与聚多巴胺修饰的介孔二氧化硅真空活化后,浸入三唑酮溶液中,然后过滤,得到固体,即为三唑酮缓释体系;
所述步骤(1)中介孔二氧化硅与盐酸多巴胺的重量比为5:1;
所述步骤(2)中,聚多巴胺修饰的介孔二氧化硅的重量与金属离子的物质的量的比例为20:1 g/mol;
所述步骤(3)中,金属离子和聚多巴胺修饰的介孔二氧化硅与三唑酮溶液中的三唑酮的重量比为3:2;
所述金属离子为锌离子。
2.如权利要求1所述的三唑酮缓释体系的制备方法,其特征在于,所述步骤(1)中分散所述介孔二氧化硅与盐酸多巴胺的溶液为pH值=8.5的三羟甲基氨基甲烷-盐酸缓冲溶液。
3.如权利要求1所述的三唑酮缓释体系的制备方法,其特征在于,所述步骤(2)中所述金属离子溶液为金属硝酸盐溶液。
4.如权利要求1所述的三唑酮缓释体系的制备方法,其特征在于,所述步骤(3)中,所述三唑酮溶液为三唑酮乙醇溶液。
5.如权利要求1所述的三唑酮缓释体系的制备方法,其特征在于,所述步骤(3)中真空活化的温度为80℃,活化时间为6 h。
6.一种由权利要求1-5任一项所述制备方法制备而成的三唑酮缓释体系。
