Abstract

Description
本发明的目的是用于皮肤的美容或药物治疗的包含曲法罗汀(Trifarotène)的呈泡沫形式的免冲洗局部产品。曲法罗汀是一种新型类维生素A,对应于3"-叔丁基-4'-(2-羟基乙氧基)-4"-吡咯烷-1-基[1,1';3',1"]三联苯-4-甲酸,具有与他扎罗汀(类胡萝卜素(aroténoïdes))相同的药理学类别。
本发明还涉及根据本发明的产品用于治疗痤疮的用途。
尽管取得了所有进展,但消费者仍在寻找快速且更实用的用于局部施用的组合物。泡沫允许通过更好地控制剂量,凭借其铺展性质和低密度来克服耐受性问题。

目前市场上存在泡沫或发泡组合物。但是,它们都有一些缺点:

- 膨化霜剂,其中通过特定的制造方法将气泡引入产品中。这种方法具有在工业水平上受到限制并且需要与包装设备有关的大量投资的缺点;
- 较少含发泡表面活性剂的发泡制剂,但它们被包装在配备有机械泡沫发生系统(带有Pulvorex型格栅的泵)的包装中。这种类型的制剂需要使用发泡表面活性剂,其可以导致免冲洗产品中的刺激。
因此,仍然需要开发一种药物组合物,其盖仑制剂形式不同于已知的盖仑制剂形式,尤其以便提供旨在局部施用的组合物,该组合物在用于人类局部施用(特别是免冲洗施用(即,组合物在其施用后不通过冲洗被去除))的耐受性优良的组合物中含有曲法罗汀。
因此,本发明的目的是提出满足这些需求的组合物。
申请人因此开发了用于免冲洗的局部施用的新型药物组合物,其以有利地不含任何发泡表面活性剂的泡沫形式存在。术语"发泡表面活性剂"定义了这样的表面活性剂,其,根据本领域技术人员熟知的测试,与水混合时产生大体积的、稳定的且滑腻的泡沫。
以下构成发泡表面活性剂:阴离子表面活性剂,阳离子表面活性剂,两性表面活性剂和烷基多葡糖苷家族和葡糖酰胺家族的非离子表面活性剂。
根据本发明的盖仑制剂形式具有确保曲法罗汀的优良稳定性的优点。此外,这种制剂有利地引起获得完全耐受性的且无刺激性的温和泡沫,其凭借泡沫的铺展性能和低密度允许更好地覆盖要治疗的区域并且允许通过更好地控制剂量来克服耐受性问题。
最后,有利地,这种盖仑制剂形式在施用时不需要使用推进剂气体或气溶胶。因此,"气溶胶"或"喷射"泡沫被排除在本发明的范围之外。同样,需要泡沫生成机械系统(Pulvorex型)的膨化霜剂和/或泡沫制剂类型的现有技术的泡沫也排除在本发明之外。
最后,本发明的目的是根据本发明的组合物通过将这种组合物局部施用于皮肤上的美容用途,以及包含这种组合物的用于在皮肤上局部施用的药物。
本发明的目的还是根据本发明的组合物,其用于治疗痤疮。
本发明将在下面的描述和实施例中并参考本申请的附图进行更详细的描述。

图2说明了实施例5,并且表示根据本发明的泡沫组合物和含有0.01重量%曲法罗汀的霜剂形式的参照样品的穿透-渗透到角质层,表皮和真皮中的比较结果,所述根据本发明的泡沫组合物含有0.01重量%的曲法罗汀,由在实施例1中描述的中间组合物A2和B3的混合物(以50/50重量比)组成。
图3还说明了实施例5,并且以放大的比例尺显示穿透-渗透到图1的真皮中的比较结果。
根据本发明的组合物能够仅凭借其组成而产生泡沫形式,并且还可以被定义为用于局部施用的自发泡组合物。
因此,本发明的第一个目的是一种含有曲法罗汀的组合物,该组合物用于免冲洗局部施用,其以泡沫形式存在,有利地具有半固体稠度,有利地不含任何发泡表面活性剂并且包含与免冲洗局部施用,特别是在皮肤和表皮性组织上的免冲洗局部施用在药学上相容的介质。
术语"以泡沫形式存在的组合物"(以下也称为自发泡组合物)是指具有与泡沫相似的充气形式的半固体稠度的组合物。
根据本发明的自发泡组合物包含可变比例的两种中间组合物或制剂,特别地以下成分:
- 至少一种中间组合物或制剂A,其包含用于下面定义的气体发生剂的激活剂;
- 至少一种中间组合物或制剂B,其包含气体发生剂,和
- 曲法罗汀或其在药学上可接受的盐的一种,其被包含在所述中间制剂A和B中的至少一种中。
根据本发明,组合物是自发泡的,即通过简单混合中间组合物A和B而发泡。本发明的目的还是由所述中间组合物A和B的混合得到的呈泡沫形式的组合物。
根据本发明,每种中间组合物(或制剂)可以具有1cP-500000cP,有利地500cP-350000cP的粘度(在25℃和大气压下测量),该粘度使用Brookfield RV dv-II类型常规方法进行测量:转子6,转速2。
根据本发明,由气体发生剂产生的气体可以是允许获得泡沫的并在生理上相容的任何气体,例如二氧化碳(CO2)或氧气(O2)。优选地,由气体发生剂产生的气体是二氧化碳(CO2)。
根据本发明,由于气体浓度可以变化,所以在组合物中气泡的量可以变化并因此产生可以从低充气到强充气的组合物。
根据本发明,术语"用于气体发生剂的激活剂"是指通过与气体发生剂的化学反应释放气体的成分。优选地,它涉及酸/碱反应。
因此,根据本发明,自发泡组合物优选可以是从充气形式到高度膨胀泡沫的任何形式。
根据本发明的组合物适用于局部施用,并且可以还包含在生理上可接受的介质,即与皮肤和表皮组织相容的介质。它优选是在药学上可接受的介质。
另外,组合物可以包含任何活性剂,其可以具有活性,任选的治疗活性。这些活性剂尤其可以选自润肤剂,保湿剂,自由基清除剂,抗炎剂,维生素,脱色素剂,抗痤疮剂,抗皮脂溢剂,抗真菌剂,角质层剥落剂,防晒剂,瘦身剂和皮肤着色剂。
根据本发明,呈泡沫形式的组合物(即,即时施用)可具有2至8之间,优选4至8之间的pH。
由于一种或多种中间组合物(或制剂)由于成分的稳定性原因需要储存在至少两个隔室中,本发明或者涉及包含隔室(每个隔室接收一种中间制剂),优选包含两个或三个隔室的单个容器,或者涉及包含彼此独立储存并物理分隔的每种中间制剂的试剂盒。
中间制剂的临时亲密混合(直接在皮肤上或在任何其它载体上)允许获得根据本发明的呈泡沫形式的组合物。
更具体地说,中间组合物(或制剂)A可以是呈溶液,乳液(洗剂,霜剂,无乳化剂的霜剂,乳或流体霜剂)或凝胶的形式。这种组合物有利地含有足够量的用于气体发生剂的激活剂(优选酸)(其可以在酸性pH下为酸/碱缓冲液的形式),作为非限制性实例,其可以是柠檬酸/柠檬酸钠对。
制剂B可以是呈溶液,凝胶或乳液(洗剂,霜剂,无乳化剂的霜剂,乳或流体霜剂)的形式。这种组合物有利地含有足够量的气体发生剂,该气体发生剂特别地可以是碳酸氢钠。
因此,本发明的目的还是如先前所定义的试剂盒或具有多隔室的单个容器,其允许即时制备根据本发明的呈泡沫形式的组合物,其分开地包含至少两种中间制剂(或中间组合物):
- 中间制剂A,其包含至少一种用于气体发生剂的激活剂;和
- 中间制剂B,其包含至少一种气体发生剂;
- 曲法罗汀或其在药学上可接受的盐的一种,其被包含在所述中间制剂A和B中的至少一种中。
优选地,曲法罗汀被包含在中间组合物A中。
气体激活剂:
用于气体发生剂的激活剂(也称为"气体激活剂")是通过释放气体的化学反应(优选酸/碱反应)与气体发生剂反应的化合物。
它有利地是酸,部分成盐的多元酸盐或弱酸和共轭碱的缓冲溶液,或这些化合物的混合物。
根据本发明,所述酸的酸/碱缓冲剂可以是弱酸的任何酸/碱缓冲剂,例如柠檬酸/柠檬酸钠缓冲剂或酒石酸/酒石酸钠缓冲剂。优选地,将提及的是α-羟基酸,它们是优选具有2-6的pKa的弱酸,如柠檬酸,酒石酸,苹果酸或乳酸,以及磷酸和焦磷酸,以及它们的任选部分盐化的盐,如焦磷酸二钠或磷酸二氢钠(也称为磷酸一钠)。
优选地,根据本发明,气体激活剂选自酒石酸/酒石酸盐(例如酒石酸钠)缓冲液;单独的柠檬酸/柠檬酸钠缓冲液;磷酸,磷酸一钠,焦磷酸二钠,其是单独的或与柠檬酸/柠檬酸钠缓冲液的混合物。
根据一个非常优选的实施方案,气体激活剂是单独的或与磷酸一钠和/或焦磷酸二钠混合的柠檬酸/柠檬酸钠缓冲液。
在用于敏感性皮肤或受损皮肤(例如患痤疮皮肤)的组合物中,相对于中间组合物A的总重量,柠檬酸/柠檬酸钠的含量优选小于或等于2.4%,以限制任何的刺痛风险。为了改善耐受性并避免刺痛感,优选地柠檬酸/柠檬酸钠缓冲液作为与焦磷酸二钠或磷酸二氢钠的混合物进行使用。
根据本发明,所述气体激活剂可以以相对于中间组合物A的总重量的0.001重量%至95重量%的量存在于中间制剂A中。
气体发生剂:
术语"气体发生剂"是指具有通过化学反应产生气体的性质的任何试剂。在这方面将提到任何化合物,当它与弱酸混合时,可以通过等效于以下的化学反应形成气体:
NaHCO3+RCOOH→RCOONa+H2O+CO2 。
根据本发明,由存在于中间组合物B中的气体发生剂产生的气体优选为含碳气体或二氧化碳(CO2),更优选地二氧化碳(CO2)。
根据本发明,气体发生剂优选地选自碳酸氢钠,碳酸氢钾,碳酸钠和碳酸钾,以及它们的混合物。
优选地,根据本发明,中间制剂B包含产生含碳气体的发生剂,其特别优选为碳酸氢钠。
相对于中间组合物B的重量,所述气体发生剂可以以1重量%至10重量%,优选2重量%至8重量%的量存在于中间制剂B中。
根据本发明,中间制剂A可以具有酸性pH,有利地为1.0至6.0的pH,并且中间制剂B可以具有碱性pH,有利地为7至12的pH。
根据本发明,所述一种或多种中间制剂包含原样形式或盐形式的曲法罗汀,其量对应于0.00001重量%至20重量%酸形式的曲法罗汀(即3"-叔丁基-4'-(2-羟基乙氧基)-4"-吡咯烷-1-基[1,1';3',1"]三联苯-4-甲酸),相对于该总组合物的总重量。
优选地,总组合物(中间制剂A与中间制剂B的混合物)含有原样形式或盐形式的曲法罗汀,其含量相当于0.00001重量%至1重量%,优选0.0001重量%至0.1重量%,更优选0.001重量%至0.1重量%的为酸形式的曲法罗汀,相对于总组合物的重量。
在本说明书中,术语"总组合物"或"总制剂"是指所述在中间组合物混合后呈泡沫形式的产品组合物。优选地曲法罗汀被包含在具有与活性剂更好相容性的中间组合物A中。
事实上,申请人已经发现,在某些情况下,曲法罗汀可与构成制剂B的成分具有较差的相容性,最特别是与碳酸氢钠具有较差的相容性。
中间制剂A可以是呈与对于通过将制剂A与制剂B混合而获得的最终组合物所希望的盖仑制剂形式相容的任何盖仑制剂形式。
中间制剂B可以是呈与通过将制剂B与制剂A混合而获得的最终组合物所希望的盖仑制剂形式相容的任何盖仑制剂形式。有利地,制剂B可以是凝胶,溶液,悬浮液或乳液(霜剂,无表面活性剂的霜剂,洗剂,乳或流体霜剂),优选乳剂。
根据本发明的一个实施方案,两种中间制剂之一(即中间制剂A或中间制剂B)呈凝胶形式。在这种实施方案中,其它中间制剂优选不是呈凝胶形式。
如前面所定义的根据本发明的试剂盒或多隔室容器的每种中间制剂包含在生理学上可接受的介质,其输送所述一种或多种化合物并进行选择使得化合物能够彼此反应以在使至少中间制剂A和B混合期间形成自发泡组合物。
因此,至少两种制剂(例如制剂A和制剂B)的即时混合产生根据本发明的呈泡沫形式的组合物。
在两种配制剂A和B混合期间,气体发生剂如碳酸氢钠与气体激活剂如酸反应,并由此特别产生与酸对应的盐,水和CO2气体。正是这种被捕获在组合物的气泡中的气体产生了表征本发明自发泡组合物的泡沫。
因此,通过使至少中间制剂A和中间制剂B混合,获得根据本发明的被称为总组合物的泡沫组合物。
在使至少制剂A和B混合之后获得的组合物中,当然可以仍然存在未反应的气体激活剂和/或气体发生剂。
有利地,根据本发明的试剂盒或具有多隔室的单个容器可以进行设计使得在制备根据本发明的组合物期间,中间制剂A和B可以以0.5至2,优选0.5至1.5,更优选接近1(即0.9至1.1),甚至更优选为1的A/B重量比进行混合。这意味着试剂盒可以进行设计以同时释放这样剂量(以重量计)的中间组合物A和B,其剂量可以是为2剂量B/1剂量A至2剂量A/1剂量B,优选2剂量B/1剂量A至3剂量A/2剂量B的重量比。根据本发明的优选实施方案,对该试剂盒进行设计以同时释放1重量剂量A和1重量剂量B。
根据本发明,试剂盒可以是一方面与中间制剂A和B的分开储存以及另一方面进行A和B的即时混合的能力相容的任何形式。
例如,中间制剂A和B可以被包装在具有至少两个分开的隔室的盒子中,每个隔室包含A或B。
根据另一方面,所述试剂盒可以以具有至少两个分开的主体的注射器的形式存在,每个主体配备有活塞,所述两个主体包含分别的制剂A和B并且进行设计以通过在活塞上施加力量同时释放所需剂量的制剂A和B。
本发明还涉及制备根据本发明的组合物的方法,其特征在于,为了获得呈泡沫形式的组合物,将如上定义的试剂盒的中间制剂A和中间制剂B临时混合,其中按重量计的相对比例A/B可以为0.5-2,优选0.5-1.5,更优选1。
为了获得最佳泡沫(最终组合物),本发明人通过实验研究了气体发生剂(优选碳酸氢钠)和气体激活剂(优选柠檬酸和/或焦磷酸二钠和/或磷酸二氢钠或磷酸一钠)的最佳含量。
因此,已通过实验确定,当气体激活剂是柠檬酸时,柠檬酸/碳酸氢钠重量比有利地在0.1和2之间,优选在0.5和1之间,非常优选等于0.7。
类似地,已通过实验确定,当气体激活剂是焦磷酸二钠时,焦磷酸二钠/碳酸氢钠重量比在0.5和5之间,优选在1和3之间,非常优选等于2.4。
类似地,已通过实验确定,当气体激活剂是磷酸二氢钠时,磷酸二氢钠一水合物/碳酸氢钠重量比在0.5和5之间,优选在1和3之间,非常优选等于2。
在实施例4中描述了碳酸氢钠/柠檬酸,碳酸氢钠/焦磷酸钠和碳酸氢钠/磷酸氢钠的比例的举例。
令人惊讶的是,柠檬酸/柠檬酸钠,焦磷酸二钠或磷酸二氢钠和与盖仑制剂形式相容的胶凝体系的组合允许获得具有非常稳定的物理化学性质的制剂,在其中曲法罗汀是特别稳定的,其不会产生任何对皮肤不愉快的感觉,并允许释放气体并由此产生泡沫。
下面的实施例2B显示了根据本发明的组合物具有优异的物理和化学稳定性。
当在不同温度的条件下:室温(RT或TA)和40℃,其感官特征,其pH,其粘度和曲法罗汀的均匀性不随时间变化时,组合物被认为是物理稳定的。
根据本发明,室温对应于15℃至25℃的温度。
当组合物含有的活性成分的含量在不同温度条件(RT和40℃)下不随时间变化时,该组合物被认为是化学稳定的。
根据本发明,当曲法罗汀的含量(以相对于中间制剂的重量进行表示)被包括在从90%至110%的规格中时,该组合物被认为是稳定的。
根据本发明的组合物还可以包含一种或多种试剂,其选自:分散剂,稳定剂,防腐剂,脂肪物质,增稠剂,染料,香料,表面活性剂,胶凝剂,络合剂,中和剂,非发泡乳化剂,填料,螯合剂,还原剂,气味掩蔽剂,增塑剂,软化剂,增湿剂,颜料,粘土,矿物填料,矿物胶体,聚合物,蛋白质,珠光剂,蜡,油例如石蜡,硅酮,脂肪酸,或脂肪酸或脂肪醇的固体酯,树胶和润湿剂。
水溶性染料如FD&C Blue 1(具有经验式C37H34N2Na2O9S3)和脂溶性染料如RougeSoudan III或Red Nil具有染色中间制剂之一的优点。这种染色允许控制两种中间制剂令人满意的混合并突出泡沫的形成。这种染色特别地在实施例和图1中示出。
用于包含气体激活剂的中间制剂的胶凝剂
有利地含有至少一种气体激活剂的中间组合物A优选含有至少一种胶凝剂和/或悬浮剂。
制剂A可以含有大量的酸和电解质。这些制剂的粘度和悬浮能力往往随着时间的推移难以得到确保。

用于含有气体发生剂的中间制剂的胶凝剂

保湿剂
在发挥使皮肤保湿并促进配制剂施用的功能的保湿剂和/或润肤剂中,任选地使用这样的化合物,如在室温(25℃)下可与水混溶的多元醇,尤其选自特别地含有2至20个碳原子,优选含有2至10个碳原子,优选含有2至6个碳原子的多元醇,如甘油,二醇衍生物如丙二醇,丁二醇,戊二醇,己二醇,二丙二醇,二甘醇及它们的混合物,以及糖(例如葡萄糖或乳糖),聚乙二醇(PEG)(例如Lutrol E400®),尿素和氨基酸(例如丝氨酸,瓜氨酸,精氨酸,天冬酰胺或丙氨酸),该名单是非限制性的。
作为优选的保湿剂和/或润肤剂,可以提及甘油和丙二醇。
保湿剂可以单独或组合进行使用,相对于总制剂的重量,其优选浓度为0.001%至30%,更优选0.01%至10%重量。
螯合剂
在螯合剂中,作为非限制性实例,可以提及乙二胺四乙酸(EDTA),二亚乙基三胺五乙酸(DTPA),乙二胺双(O-羟基苯基乙酸)(EDDHA),羟基-2-乙二胺三乙酸(HEDTA),乙基二胺-双(O-羟基-对-甲基苯基)乙酸(EDDHMA)和乙二胺-双(5-羧基-2-羟基苯基)乙酸(EDDCHA)。
作为优选的螯合剂,可以提及特别地以名称Titriplex III®出售的乙二胺四乙酸(EDTA),其可以相对于总制剂重量的0.001%至1%,更优选0.05%至0.1%的优选浓度进行使用。
具有补充性质的赋形剂
根据本发明的组合物可以含有一种或多种美容活性剂,例如,作为非限制性实例,具有抗刺激性质的尿囊素,抗痤疮的葡萄糖酸锌,甘草酸二钾(由于其抗炎特性),或者瘢痕形成剂α-红没药醇或二甲基异山梨醇(由于其促渗性质)。
填料和颗粒
可以使用填料和/或颗粒以使泡沫稳定和增强。它们中的某些具有位于水/空气界面的特定性质并由此稳定该界面。作为填料,可以提及滑石,金属氧化物如氧化锌,由Merck公司以名称Eusolex® E-2000出售的二氧化钛TiO2 T2000,粘土如合成锂皂石,皂土或膨润土,以及纤维素醚如由Dow公司出售的Methocel® K100 LV,由Evonik公司出售的二氧化硅如Aerosil® R972或由Wacker出售的Silice HDK® H13L;它们可以以相对于总制剂重量的0.01重量%至10重量%的浓度进行使用。
脂肪相的油
根据本发明的组合物还可以包含脂肪相。这种脂肪相可以存在于中间组合物A和B中的一种和/或另一种中。根据中间制剂的盖仑制剂形式,脂肪相可以占相对于每种中间制剂的重量的0重量%至95重量%。
根据本发明的组合物的脂肪相可以包含例如植物油,矿物油,动物油或合成油,硅油及它们的混合物。
作为矿物油的例子,可以提及例如具有不同粘度的石蜡油,例如由Esso公司出售的Primol 352®,Marcol 82®和Marcol 152®。
作为植物油或其衍生物,可以提及甜杏仁油,例如Sictia提供的PrunusAmygdalus Dulcis(甜杏仁油),棕榈油,大豆油,芝麻油,葵花籽油和橄榄油,杏仁油及其酯,如产品Apricot Apricot Kernel Oil PEG-6 Ester (Labrafil M1944CS)。
作为动物油或其植物来源的替代物,可以提及的是羊毛脂,角鲨烯,鱼油,与,作为衍生物,由Sophim公司以名称Sophider®出售的全氢角鲨烯。
作为合成油,可以提及酯,如异壬酸鲸蜡酯,例如由Cognis France公司以名称Cetiol SN PH®出售的产品,异壬酸异壬基酯,如由Stéarineries Dubois公司出售的DubININ®,己二酸二异丙酯,例如由Croda公司以名称Crodamol DA®销售的产品,棕榈酸异丙酯,例如由Croda公司以名称Crodamol IPP®销售的产品,以及辛酸/癸酸甘油三酯,例如由Univar公司销售的Miglyol 812®。作为氢化聚异丁烯,可以提及由Rossow公司出售的Parleam®,由Croda提供的PPG-15硬脂基醚(Arlamol PS 15 E),由Gattefossé提供的PPG-11硬脂基醚(Arlamol PS11E-LQ)。
作为硅油,可以提及聚二甲基硅氧烷,例如由Dow Corning公司以名称Q7-9120Silicone Fluid®出售的产品,其具有为20cSt至12500cSt的粘度,或环甲硅油,例如也由Dow Corning公司以名称ST-Cyclomethicone 5NF®出售的产品。
这些油可以单独或组合存在,其含量为相对于总组合物的重量的0.5重量%至50重量%,优选2重量%至30重量%。
非液体脂肪物质
根据本发明的组合物还可以包含固体脂肪物质,例如天然或合成蜡,脂肪酸如硬脂酸,脂肪醇如由Cognis公司销售的Speziol C18® Pharma或Speziol C16® 以及三山萮酸酯类型的结构改进剂,例如由Gattefossé公司销售的Compritol 888®或由Cognis公司出售的氢化蓖麻油例如Cutina HR®或者由Gattefossé公司出售的Geleol®或由Dow Corning公司出售的DC 9045 Elastomer Blend®。

非离子乳化剂
根据本发明的组合物还可以包含一种或多种非离子乳化剂。

防腐剂

实施例2A:泡沫密度的测量

实施例3:刺激性测量的比较研究
研究方案
该研究根据现行的TG439 OCDE协议(用于短施用时间(RHE/产品接触时间15分钟))进行。该方案进行调整用于长的施用时间(RHE/产品接触时间18小时)。
这种研究的目的是评估在重建人表皮(RHE,Episkin模型)上完整制剂和中间制剂的载体的耐受性,通过:
- 评估MTT(细胞活力)的降低
- 测量IL-1α的释放(刺激性标记物)。
测试的制剂是:
- 酸性制剂的中间组合物:安慰剂实施例A7(即不含曲法罗汀),
- 碱性制剂的中间组合物:实施例B7,
- 由混合物组成的完整制剂:A7安慰剂+B7(50/50重量比),
- 霜剂形式的商业参照样品。
研究结果:
根据现行的OCDE协议的MTT测量结果表明,所有测试的制剂都是非刺激性的。
在短暴露时间和长暴露时间后,根据本发明的完整制剂的IL-1a配量显示刺激性标记物的含量低于在施用商业参照样品之后的含量。
实施例4:
根据经验建立了柠檬酸,焦磷酸钠和磷酸二氢钠一水合物的理想含量以与5%碳酸氢钠反应。数值以相对于两种中间制剂中的每一种的重量的重量/重量百分比表示。

实施例5:释放-渗透曲线的比较研究
这种研究的目的是评价根据本发明的不同制剂在人体皮肤中的渗透和分布。
研究方案:
在这种研究中测试的制剂在32℃下以5mg/cm2的比率施用于安装在Franz池上的切除的完整的人类皮肤样品。
在施用16小时后,测定在以下部分中的曲法罗汀:未吸收部分,角质层,表皮,真皮和接受液。
这种研究允许研究制剂对活性成分的释放及其穿过皮肤的渗透的影响。目的是比较在施用熟知的制剂和施用呈化学泡沫形式的组合物的情况下曲法罗汀在皮肤的不同层中的分布。
测试的制剂是:
- 含有0.01%重量曲法罗汀的呈霜剂形式的参照样品,和
- 包含0.01重量%曲法罗汀的完整化学泡沫制剂,其由实施例1的中间制剂A2+B3的混合物(50/50重量比)组成。
扩散池(cellules de diffusion):
所使用的扩散池是基于Franz模型扩散池的静态扩散池,具有以下特征:
- 施用区域=2平方厘米
- 接收流体的隔室的容积=3毫升。
接收隔室用被加热到37℃±1℃的水套包围,以确保在皮肤表面的32℃±1℃的温度。接收隔室通过皮肤膜与供体隔室分开,表皮面位于供体侧。包含磁力搅拌棒的接收隔室充满了接收流体以防止任何气泡的形成。在扩散时间期间,将接收流体连续搅拌以确保均质化。
皮肤样品的制备:
在这种研究中使用了来自整容手术的腹部皮肤。样品到达后,使用镊子将皮下组织从该整体中分离出来,并将剩余部分轻轻地清洗并平放储存在铝箔中以在-20℃冻结。在实验当天,将皮肤样品解冻,然后切成片状以与扩散池的几何形状相适合。
将来自42、44和69岁供体的皮肤样品安装在具有PBS作为接受液的扩散池上。皮肤平均厚度为0.89±0.07毫米,最大值为1.39毫米,最小值为0.45毫米。所有样品的厚度。
在用接收液平衡至少45分钟后,皮肤完整性通过测量经皮无感水分流失(TEWL)进行评估。其中超出接受标准的TEWL测量值的所有池子都要仔细清洗,并在重新测量TEWL之前在延长时间期间进行平衡。平均TEWL值为5.51±1.63g/m2/h。
研究参数:
室温:21.7℃
相对湿度45.6%。
研究结果:
如图2所示,在角质层中,在制剂(根据本发明的泡沫和霜剂)之间,不存在皮肤渗透的显著差异。
如图2所示,在表皮中,化学泡沫制剂的渗透与霜剂参照样品(根据实施例A2+B3,为50:50混合物)是很小差异的。
如图3所示,在真皮中,化学泡沫组合物的渗透少于霜剂参照样品(根据实施例A2+B3,为50:50的混合物)。
无论测试的制剂为如何,接收流体样品都是不可定量的;这表明低的系统暴露。
本研究证实了获得了这样的泡沫,其保留在皮肤表面上,以避免不希望的效果,如刺激作用,并且获得适合于治疗痤疮的泡沫。
Claims (24)
1.旨在用于免冲洗局部施用的含有曲法罗汀的自发泡组合物,其包含:
- 至少一种中间组合物B,其包含气体发生剂,
- 至少一种中间组合物A,其包含用于气体发生剂的激活剂;
- 曲法罗汀或其在药学上可接受的盐的一种,其被包含在所述中间组合物A和B的至少一种中。
2.根据前述权利要求所述的组合物,其特征在于曲法罗汀被包含在中间组合物A中。
3.根据前述权利要求中任一项所述的组合物,其特征在于其不包含发泡表面活性剂,特别地不包含选自阴离子表面活性剂,阳离子表面活性剂,两性表面活性剂和烷基聚葡糖苷家族和葡糖酰胺家族的非离子表面活性剂的表面活性剂。
4.根据前述权利要求中任一项所述的组合物,其特征在于由所述气体发生剂产生的气体是二氧化碳(CO2)。
5.根据前述权利要求中任一项所述的组合物,其特征在于所述气体发生剂选自碳酸氢钠,碳酸氢钾,碳酸钠,碳酸钾及它们的混合物,优选地,所述气体发生剂为碳酸氢钠。
6.根据前述权利要求中任一项所述的组合物,其特征在于所述气体发生剂以1重量%至10重量%,优选2重量%至8重量%的量存在于中间组合物B中,相对于中间组合物B的重量。
7.根据前述权利要求中任一项所述的组合物,其特征在于所述中间组合物B具有碱性pH,有利地为7-12的pH。
8.根据前述权利要求中任一项所述的组合物,其特征在于用于气体发生剂的激活剂选自酸,部分成盐的多元酸盐,弱酸和其共轭碱的缓冲溶液,以及这些化合物的混合物。
9.如前一项权利要求所述的组合物,其特征在于用于气体发生剂的激活剂选自柠檬酸,酒石酸,苹果酸,乳酸,磷酸和焦磷酸,和这些酸的盐,更优选地,所述激活剂选自:
- 单独的柠檬酸/柠檬酸钠缓冲液;
- 磷酸,磷酸钠,焦磷酸二钠,其是单独的或与柠檬酸/柠檬酸钠缓冲液混合。

12.根据前述权利要求中任一项所述的组合物,其特征在于所述中间组合物A具有酸性pH,有利地为1.0-6.0的pH。
13.根据前述权利要求中任一项所述的组合物,其特征在于所述中间组合物A以溶液,凝胶或乳液的形式存在,优选以凝胶形式存在。
14.根据前述权利要求中任一项所述的组合物,其特征在于所述中间组合物B以溶液,凝胶或乳液的形式存在,优选以乳液形式存在。
15.呈泡沫形式的组合物,其特征在于它由根据前述权利要求任一项的所述中间组合物A和B的混合所产生。
16.根据前述权利要求中任一项所述的组合物,其特征在于它还含有一种或多种活性剂,其选自润肤剂,保湿剂,自由基清除剂,抗炎剂,维生素,脱色素剂,抗痤疮剂,抗皮脂溢剂,抗真菌剂,角质层剥落剂,防晒剂,瘦身剂和皮肤着色剂。
17.根据前述权利要求中任一项所述的组合物,其特征在于其还含有一种或多种试剂,其选自分散剂,增溶剂,稳定剂,防腐剂,脂肪物质,增稠剂,染色剂,香料,胶凝剂,络合剂,中和剂,非发泡乳化剂,填料,螯合剂,还原剂,臭味掩蔽剂,增塑剂,软化剂,保湿剂,颜料,粘土,矿物填料,矿物胶体,聚合物,蛋白质,珠光剂,推进剂,蜡,油例如石蜡,脂肪酸,脂肪醇或脂肪酸的固体酯,树胶和润湿剂。
18.根据前述权利要求中任一项所述的组合物的美容用途,通过将这种组合物局部施用于皮肤上。
19.用于在皮肤上局部施用的药物,其包含如在权利要求1至17中任一项中所定义的组合物。
20.根据权利要求1至17中任一项所述的组合物,其用于治疗痤疮。
21.一种试剂盒或具有多隔室的单个容器,其分开地包含至少两种中间组合物:
- 如在权利要求1至17任一项中所定义的中间组合物A,其包含至少一种用于气体发生剂的激活剂;和
- 如在权利要求1至17任一项中所定义的中间组合物B,其包含至少一种气体发生剂;
- 曲法罗汀或其在药学上可接受的盐的一种,其被包含在所述中间组合物A和B的至少一种中。
22.根据权利要求21所述的试剂盒或具有多隔室的单个容器,其特征在于,其进行设计以按照2剂量B/1剂量A至2剂量A/1剂量B,优选2剂量B/1剂量A至3剂量A/2剂量B,更优选地1剂量A/1剂量B的重量比同时释放中间组合物A和B的剂量。
23.根据权利要求21和22中任一项所述的试剂盒或具有多隔室的单个容器,其特征在于,其进行设计以按照0.5至2,优选0.5至1.5,更优选0.9至1.1,甚至更优选1的A/B重量比混合中间组合物A和B。
