The effects of the external preparation of the present invention will be described below by taking the formulations of examples 1 to 5 as examples.
First, bacteriostasis experiment
Staphylococcus aureus, staphylococcus epidermidis and diphtheroid bacillus are used as experimental strains, and the strains are all from a microorganism preservation center CGMCC. Respectively preparing culture solution of Staphylococcus aureus, Staphylococcus epidermidis and diphtheroid bacillus, and diluting thallus concentration to 106cfu/ml, soaking filter paper disc with diameter of 0.5cm in Chinese medicinal preparation (different Chinese medicinal preparation for each group) for 10 min; preparing beef extract peptone plates, coating culture solutions for respectively preparing staphylococcus aureus, staphylococcus epidermidis and diphtheroid bacillus on the beef extract peptone plates under the condition of no bacteria, then placing the soaked filter paper in the center of the plates, culturing for 14h at 37 ℃, and measuring the size of a bacteriostatic zone.
Wherein, the traditional Chinese medicine preparation of the group of the examples 1 to 5 adopts the formula of the example 1 to 5 and the preparation method of the example 6, the traditional Chinese medicine preparation of the group of the comparison 1 adopts the formula of the example 1 and the preparation method of the example 9, and the traditional Chinese medicine preparation of the group of the comparison 2 adopts the penicillin solution of 35mg/100 ml. The results are shown in Table 1.

The results in table 1 show that the bacteriostatic effect of the traditional Chinese medicine prepared by the formula of examples 1-5 and the preparation method of example 6 on staphylococcus aureus, staphylococcus epidermidis and diphtheroids is equivalent to that of a 35mg/100ml penicillin solution, but the bacteriostatic effect of the control group 1 is poor. The reason is that the preparation method of the embodiment 6 firstly adopts the sodium bicarbonate solution to soak the medicinal materials, modifies the medicinal materials, accelerates the dissolution of active substances in the medicinal materials, and then neutralizes the medicinal materials by the hydrochloric acid solution. The control group 1 had no soaking step with sodium bicarbonate solution, so the bacteriostatic effect was poor.
Second, the influence on the microcirculation of the eye conjunctiva of the rabbit
30 rabbits are selected by a rabbit eye conjunctiva microvascular fluorometry, the male and female clapboards weigh 1.8-2.5kg and are randomly divided into 3 groups, wherein in the gastric perfusion traditional Chinese medicine in the example 1 group, the traditional Chinese medicine is prepared by weighing each medicinal material in the formula of the example 1, grinding the medicinal materials into 100 meshes of medicinal powder, and the gastric perfusion amount is 1.5g/kg, and continuously administering for 7 days; example 2 group of gavage traditional Chinese medicine, the traditional Chinese medicine is prepared by weighing the medicinal materials of the formula of example 2, grinding into 100-mesh powder, the gavage amount is 1.5g/kg, and continuously administering for 7 days; the control group was administered with saline of equal quality by gavage for 7 days. 1h after the last gastric lavage, 10% sodium fluorescein normal saline solution is injected into the ear vein of the rabbit at 1ml/kg (equivalent to that of fluorescein being injected at 100mg/kg), and the time of fluorescence appearing in eye conjunctiva of the rabbit is recorded, so that the result shows that the fluorescence appearing time of the group in example 1 is 7.58s (the average value of 10 rabbits), the fluorescence appearing time of the group in example 2 is 7.70s (the average value of 10 rabbits), the fluorescence appearing time of the control group is 12.79s (the average value of 10 rabbits), and the result shows that the microcirculation blood flow of eye conjunctiva of the rabbit is accelerated remarkably and the blood flow supply is improved remarkably in the groups in example 1 and example 2, and the difference from the control group is remarkable (P is less than 0.05).
Third, clinical cases
Shen Shi, male, 32 years old, clinically diagnosed as Staphylococcus aureus conjunctivitis with symptoms of staphylococcal blepharitis etc., the external traditional Chinese medicine preparation of example 1 was used 4 times a day, 3 drops each time, after 1 week the symptoms disappeared, recovered to health, without side effect.
Plum, female, 29 years old, clinically diagnosed as gonococcal conjunctivitis, the external traditional Chinese medicine preparation of example 1 was used once an hour and 4 drops each time, and symptoms disappeared after 4 days, so that the health was recovered without side effects.
The externally-applied traditional Chinese medicine preparation of the example 1 is used for 4 times a day, 3 drops each time, and the symptoms disappear after 10 days, so that the health is recovered, and no side effect is caused.
The externally applied traditional Chinese medicine preparation in the example 1 is used for 4 times a day and 3 drops each time, meanwhile, the formula in the example 1 (each weight part represents 1 g, for example 8 parts of angelica are weighed) is decocted and filtered according to a conventional traditional Chinese medicine method, the traditional Chinese medicine liquid is taken 3 times a day and 100ml each time, and after 6 days, symptoms disappear, and the health is recovered without side effects.
The clinical cases show that the traditional Chinese medicine prepared by the medicine formula and the preparation method provided by the invention can effectively treat viral or bacterial ocular surface diseases. In addition, 100 cases of herpes simplex keratitis patients are selected, the patients are administrated by referring to the administration mode of the above-mentioned scholar, the curative effect is observed after 1 week, wherein 80 patients are cured, 10 patients are effectively relieved (the eye irritation symptom is basically completely disappeared, the fluorescein staining is punctiform or the dendritic corneal epithelium pathological change part is slightly spotted), and the effective rate of the traditional Chinese medicine preparation is 90%.

Thus, if such modifications and variations of the present invention fall within the scope of the claims of the present invention and their equivalents, the present invention is also intended to include such modifications and variations.

Claims (5)

1. The application of the combination of angelica, long-noded pit viper and cassia seed in preparing the external traditional Chinese medicine for treating eye surface diseases is characterized in that the external traditional Chinese medicine is prepared from the following raw materials in parts by weight: 8-10 parts of angelica sinensis, 15-20 parts of long-noded pit viper, 10-15 parts of semen cassiae, 20-30 parts of radix isatidis and 15-20 parts of houttuynia cordata; the ocular surface disease is caused by bacteria or viruses.

The use of the combination of angelica, agkistrodon, cassia tora according to claim 1 in the preparation of a topical traditional Chinese medicine for the treatment of ocular surface diseases is characterized by consisting of the following raw materials in parts by weight: 8 parts of angelica, 15 parts of long-noded pit viper, 15 parts of cassia seed, 20 parts of isatis root and 20 parts of cordate houttuynia.

The use of the combination of angelica, agkistrodon and cassia seed according to claim 1 in the preparation of an external traditional Chinese medicine for treating ocular surface diseases, wherein the preparation method of the external traditional Chinese medicine for treating ocular surface diseases comprises the following steps:
s1, weighing the following raw materials in parts by weight: 8-10 parts of angelica sinensis, 15-20 parts of long-noded pit viper, 10-15 parts of semen cassiae, 20-30 parts of radix isatidis and 15-20 parts of houttuynia cordata;
s2, mixing the raw materials weighed in the S1, adding 0.6g/100ml of sodium bicarbonate solution which is 10 times of the total weight of the raw materials into the obtained mixture, soaking for 2d, boiling for 10-15 min, filtering out medicine residues, and collecting filtrate;
s3, adding 0.05mol/l hydrochloric acid solution into the filtrate of S2, and adjusting the pH to 7.0 to obtain neutral liquid medicine;
s4, boiling the neutral liquid medicine of S3 for 5-10 min, and performing decolorization treatment to obtain decolorized liquid medicine;
s5, rotationally evaporating the decolored liquid medicine of S4 to 1/10 of the volume of the sodium bicarbonate solution to obtain the external traditional Chinese medicine preparation for treating the ocular surface diseases.
4. The use of the combination of angelica, agkistrodon and cassia seed as claimed in claim 3, wherein the decolorizing treatment is carried out by using polar adsorbent and non-polar adsorbent, the polar adsorbent is silica gel, and the non-polar adsorbent is activated carbon or diatomite.

The use of the combination of angelica, agkistrodon and cassia seed as claimed in claim 4, in the preparation of a topical Chinese medicinal preparation for treating ocular surface diseases, wherein in S4, after the neutral medicinal liquid is boiled for 5-10 min, the neutral medicinal liquid is passed through a non-polar adsorbent column and a polar adsorbent column in sequence for decolorization, wherein the diameter of the non-polar adsorbent column is 50cm, and the height of the non-polar adsorbent column is 50 cm; the polar adsorbent column had a diameter of 40cm and a height of 60 cm.
