Abstract

一种水溶性氨基酸复配防腐剂,由月桂基二亚乙二氨基甘氨酸或月桂基二亚乙二氨基甘氨酸盐0.1‑3份,桃叶珊瑚苷0.05‑2份,烷基甜菜碱1‑3份,碱助剂0.01‑0.5份,抗氧化剂2‑5份,去离子水70‑80份混合制得。本发明将月桂基二亚乙二氨基甘氨酸或月桂基二亚乙二氨基甘氨酸盐与具有天然抑菌效果的萜类桃叶珊瑚苷和烷基甜菜碱组成复配增效体系,不仅抑菌效果可观,毒性低,同时具有优秀的水溶性,在日化领域的水性体系产品和保湿产品中具有很好的适配性。

Description

一种水溶性氨基酸复配防腐剂
技术领域
本发明涉及日用领域的防腐体系,尤其涉及一种水溶性氨基酸复配防腐剂及其制备方法。
背景技术
目前我国大量使用的还是苯甲酸及其盐、山梨醇及其盐、对羟基苯甲酸酯等化学合成防腐剂。经长期的应用和研究发现一些合成防腐剂有诱癌性、致畸性和易引起人体中毒问题。三氯生、尼泊金酯、异噻唑啉酮、甲醛缓释剂等的使用已经被禁止或者限制。化学合成防腐剂的使用条件越来越严格。
微生物防腐剂是通过微生物发酵技术获得的可以杀灭或抑制微生物的一种天然防腐剂。目前,公认的微生物防腐剂有:乳酸链球菌素、纳他霉素、食用乳酸菌、抗菌肽、噬菌体制品等。
现有的日化领域或保湿产品用防腐剂,化学合成防腐剂存在着毒性高,使用越来越受到限制的趋势。大部分天然防腐剂其抑菌效果有限,成分复杂,效果不稳定,难以持久,需与化学合成防腐剂复配使用,仍需要大量的化学合成防腐剂。随着对防腐剂使用要求的提高,迫切需要一种安全无毒,防腐效果稳定持久的防腐剂体系。
发明内容
为解决上述防腐剂体系中的问题,本发明提供一种水溶性氨基酸复配防腐剂,具体如下:
一种水溶性氨基酸复配防腐剂,含有月桂基二亚乙二氨基甘氨酸或月桂基二亚乙二氨基甘氨酸盐0.1-3份,桃叶珊瑚苷0.05-2份,烷基甜菜碱1-3份,碱助剂0.01-0.5份,抗氧化剂2-5份,去离子水70-80份。
优选的,所述水溶性氨基酸复配防腐剂,含有月桂基二亚乙二氨基甘氨酸或月桂基二亚乙二氨基甘氨酸2.5份,桃叶珊瑚苷1.2份,烷基甜菜碱1.5份,碱助剂0.25份,抗氧化剂4.5份,去离子水75份。
桃叶珊瑚苷,其抗菌活性很高,对于金黄色葡萄球菌的最小抑菌浓度仅为6.25μg·mL-1。但其化学性质不稳定,常规条件下易发生化学反应而分解。
烷基甜菜碱
烷基甜菜碱能与各种类型染料、表面活性剂及化妆品原料配伍,在酸性及碱性条件下均具有优良的稳定性,配伍性良好。无毒,对皮肤刺激性低,生物降解性好,易溶于水,具有优良的增稠性、杀菌性、抗静电性,可用于杀灭包括结核菌在内的多种细菌。
优选的,所述烷基甜菜碱为十二烷基二羟乙基甜菜碱,十二烷基二甲基胺乙内酯,十四烷基二甲基胺乙内酯中的任意一种或两种以上的混合物。
优选的,所述碱助剂为三乙醇胺,三异丙醇胺,二异丙醇胺,氨甲基丙醇中的任意一种或两种以上的混合物。
优选的,所述抗氧化剂为壳聚糖,绿原酸,茶多酚中的任意一种或两种以上的混合物。
优选的,所述月桂基二亚乙二氨基甘氨酸盐为月桂基二亚乙二氨基甘氨酸钠,月桂基二亚乙二氨基甘氨酸盐酸盐中的任意一种或两种以上的混合物。
本发明水溶性氨基酸复配防腐剂的制备可采用如下方法:将月桂基二亚乙二氨基甘氨酸或月桂基二亚乙二氨基甘氨酸盐、桃叶珊瑚苷、烷基甜菜碱、碱助剂、抗氧化剂溶于去离子水中,形成混合液,将上述混合液在均质温度为20-40℃,工作压力为0.5-2MPa的条件下,均质10-20min,而制备得到水溶性氨基酸复配防腐剂。
本发明还要求保护所述的水溶性氨基酸复配防腐剂在日化领域或保湿产品中的应用。
特别的本发明还要求保护所述的水溶性氨基酸复配防腐剂在湿巾浸液或湿巾中的应用。
本发明将月桂基二亚乙二氨基甘氨酸或月桂基二亚乙二氨基甘氨酸盐与具有天然抑菌效果的萜类桃叶珊瑚苷和烷基甜菜碱组成复配增效体系,不仅使月桂基二亚乙二氨基甘氨酸或月桂基二亚乙二氨基甘氨酸盐、桃叶珊瑚苷和烷基甜菜碱能充分发挥抗菌、防腐效果,实现了在低浓度下月桂基二亚乙二氨基甘氨酸或月桂基二亚乙二氨基甘氨酸盐的抗菌广谱性,同时克服了萜类桃叶珊瑚苷化学性质不稳定的问题,对霉菌等抑菌效果可观,同时具有毒性低和优秀的水溶性,效果持续长久,在日化领域的水性体系产品和保湿产品中具有很好的适配性。
具体实施方式
实施例1
将月桂基二亚乙二氨基甘氨酸0.1份,桃叶珊瑚苷0.05份,十二烷基二羟乙基甜菜碱1份,三异丙醇胺0.01份,壳聚糖2份溶于80份去离子水中,形成混合液,将上述混合液在均质温度为20-40℃,工作压力为0.5MPa的条件下,均质10min,而制备得到水溶性氨基酸复配防腐剂。
实施例2
将月桂基二亚乙二氨基甘氨酸盐酸盐1份,桃叶珊瑚苷1份,十四烷基二甲基胺乙内酯2份,二异丙醇胺0.3份,茶多酚3份溶于79份去离子水中,形成混合液,将上述混合液在均质温度为20-40℃,工作压力为1MPa的条件下,均质10min,而制备得到水溶性氨基酸复配防腐剂。
实施例3
将月桂基二亚乙二氨基甘氨酸钠1.5份,桃叶珊瑚苷2份,十二烷基二羟乙基甜菜碱3份,三乙醇胺0.5份,茶多酚5份溶于78份去离子水中,形成混合液,将上述混合液在均质温度为20-40℃,工作压力为2MPa的条件下,均质10min,而制备得到水溶性氨基酸复配防腐剂。
实施例4
将月桂基二亚乙二氨基甘氨酸2份,桃叶珊瑚苷1.5份,十四烷基二甲基胺乙内酯2.5份,三乙醇胺0.35份,茶多酚4份溶于77份去离子水中,形成混合液,将上述混合液在均质温度为20-40℃,工作压力为1MPa的条件下,均质20min,而制备得到水溶性氨基酸复配防腐剂。
实施例5
将月桂基二亚乙二氨基甘氨酸2.5份,桃叶珊瑚苷1.2份,十四烷基二甲基胺乙内酯1.5份,三乙醇胺0.25份,抗氧化剂4.5份溶于75份去离子水中,形成混合液,将上述混合液在均质温度为20-40℃,工作压力为1MPa的条件下,均质15min,而制备得到水溶性氨基酸复配防腐剂。
实施例6
将月桂基二亚乙二氨基甘氨酸3份,桃叶珊瑚苷0.2份,十四烷基二甲基胺乙内酯2份,氨甲基丙醇0.25份,绿原酸4.5份溶于75份去离子水中,形成混合液,将上述混合液在均质温度为20-40℃,工作压力为1MPa的条件下,均质15min,而制备得到水溶性氨基酸复配防腐剂。
对比实施例1
将桃叶珊瑚苷0.2份,十四烷基二甲基胺乙内酯2份,氨甲基丙醇0.25份,绿原酸4.5份溶于75份去离子水中,形成混合液,将上述混合液在均质温度为20-40℃,工作压力为1MPa的条件下,均质15min,而制备得到水溶性复配防腐剂。
对比实施例2
将月桂基二亚乙二氨基甘氨酸0.2份,十二烷基二羟乙基甜菜碱1份,三异丙醇胺0.01份,壳聚糖2份溶于80份去离子水中,形成混合液,将上述混合液在均质温度为20-40℃,工作压力为0.5MPa的条件下,均质10min,而制备得到水溶性氨基酸复配防腐剂。
对实施例1-6制备的水溶性氨基酸复配防腐剂和对比实施例1-2制备的水溶性复配防腐剂按下列方法进行杀菌试验,测定其杀菌效力。
试验菌
细菌:大肠杆菌(8099或ATCC25922)、金黄色葡萄球菌(ATCC6538);酵母菌:白色念珠菌(ATCC10231)。
培养基制备
依据不同的微生物菌种配制不同的培养基。营养琼脂培养基:取一定量的蛋白胨、牛肉膏、氯化钠和蒸馏水混合溶解后,调pH至7.2~7.4,加入琼脂,加热溶解于121℃压力蒸汽灭菌20min,供大肠杆菌、金黄色葡萄球菌培养用。沙氏琼脂培养基:取一定量的葡萄糖、蛋白胨、琼脂和蒸馏水混合后,加热至完全溶解,调pH至5.6±0.2,于115℃压力蒸汽灭菌30min,供白色念珠菌培养用。
菌悬液制备
取试验菌24h斜面培养物用5mL0.03mol/L磷酸盐缓冲液(以下简称PBS)洗下,在用上述PBS稀释至所需浓度,制成菌悬液(要求的浓度为:用100μL滴于5mL样液内,回收菌数为1×104~9×104cfu/mL)。
杀菌试验
取被试样液(5mL)和对照样液(与试样同质材料,同等大小,但不含抗菌材料,且经灭菌处理)各4管。取上述菌悬液,分别在每个被试样液和对照样液内滴加100μL,用电动混合器混合均匀,开始计时,作用2、5、10、20min,分别将样液(0.5mL)投入含5mL相应中和剂的试管内,充分混匀,作适当稀释,然后取其中2~3个稀释度,分别吸取0.5mL,置于两个平皿,用凉至40~45℃的营养琼脂培养基(细菌)或沙氏琼脂培养基(酵母菌)15mL作倾注,转动平皿,使其充分均匀,琼脂凝固后翻转平板,35℃±2℃培养48h(细菌)或72h(酵母菌),作活菌菌落计数。试验重复3次,取平均值。
杀菌效力判定
培养48h或72h后,按以下公式计算抑菌率:X=(A-B)/A×100%。式中:X为抑菌率,A为对照样品平均菌落数,B为被试样品平均菌落数。评价标准:杀菌率≥90%,产品有杀菌作用。
运用以上的杀菌试验方法,分别对本实施例1-6的水溶性氨基酸复配防腐剂和对比实施例1-2的水溶性复配防腐剂进行杀菌试验,最后以未存活的微生物浓度与微生物的初始浓度的百分比为判断依据,判断本发明的防腐杀菌性能,其结果如下表所示:

虽然参照其中的几个优选实施方式对本发明进行了显示和描述,但是上述仅为本发明的几个具体实施方式,但本发明的设计构思并不局限于此,在不脱离本发明精神和范围的情况下可对其形式和细节进行各种改变、删除和增加。但是应该指出的是,可能的一些变型仍落入权利要求的保护范围内。

Claims (5)

1.一种水溶性氨基酸复配防腐剂,含有月桂基二亚乙二氨基甘氨酸或月桂基二亚乙二氨基甘氨酸盐0.1-3份,桃叶珊瑚苷0.05-2份,烷基甜菜碱1-3份,碱助剂0.01-0.5份,抗氧化剂2-5份,去离子水70-80份;
所述烷基甜菜碱为十二烷基二羟乙基甜菜碱,十二烷基二甲基胺乙内酯,十四烷基二甲基胺乙内酯中的任意一种或两种以上的混合物;所述碱助剂为三乙醇胺,三异丙醇胺,二异丙醇胺,氨甲基丙醇中的任意一种或两种以上的混合物;所述抗氧化剂为壳聚糖,绿原酸,茶多酚中的任意一种或两种以上的混合物;所述月桂基二亚乙二氨基甘氨酸盐为月桂基二亚乙二氨基甘氨酸钠,月桂基二亚乙二氨基甘氨酸盐酸盐中的任意一种或两种以上的混合物。
2.根据权利要求1所述的水溶性氨基酸复配防腐剂,其特征在于,含有月桂基二亚乙二氨基甘氨酸或月桂基二亚乙二氨基甘氨酸盐2.5份,桃叶珊瑚苷1.2份,烷基甜菜碱1.5份,碱助剂0.25份,抗氧化剂4.5份,去离子水75份。
3.水溶性氨基酸复配防腐剂的制备方法,其特征在于,包括以下步骤:按照权利要求1-2任一项所述的水溶性氨基酸复配防腐剂中含有的组份及其含量,将月桂基二亚乙二氨基甘氨酸或月桂基二亚乙二氨基甘氨酸盐、桃叶珊瑚苷、烷基甜菜碱、碱助剂、抗氧化剂溶于去离子水中,形成混合液,将上述混合液在均质温度为20-40℃,工作压力为0.5-2MPa的条件下,均质10-20min,而制备得到水溶性氨基酸复配防腐剂。
4.权利要求1所述的水溶性氨基酸复配防腐剂在日化领域或保湿产品中的应用。
5.权利要求1所述的水溶性氨基酸复配防腐剂在湿巾浸液或湿巾中的应用。
