本发明公开了一种主动靶向特定肿瘤细胞的竹红菌素纳米制剂及其制备方法和应用,所述制剂为转铁蛋白修饰的聚乳酸‑羟基乙酸/羧甲基壳聚糖包封竹红菌素的制剂,制剂中有效成分竹红菌素的最终质量体积比为0.2‑0.6mg/mL、转铁蛋白的最终质量体积比为0.5‑2mg/mL,溶剂为超纯水。本发明的主动靶向特定肿瘤细胞的竹红菌素纳米制剂具有主动靶向过量表达转铁蛋白受体肿瘤细胞的功能,其具有水分散效果好,粒径均一,包封率高,稳定性好,材料的安全性能高等优点。同时制备工艺简单方便,制备得到的纳米制剂是一种主动靶向递送竹红菌素类光敏剂到多种肿瘤细胞的药物制剂。

技术领域
本发明属于纳米医药技术领域,具体涉及一种能够靶向过量表达转铁蛋白受体肿瘤细胞的转铁蛋白修饰的竹红菌素纳米制剂粒及其制备方法和应用。
背景技术
光动力疗法(photodynamic therapy,PDT)是一种新型的医疗技术,主要用于肿瘤和其他疾病的治疗。光动力疗法被认为是临床上继手术、放疗、化疗后的第4种癌症治疗方法。自Dousherty等在20世纪70年代将光动力疗法推向临床研究以来,光动力疗法取得了令人瞩目的成绩。目前,国内外研究光动力疗法仍集中于它的抗肿瘤方面的应用。此外,光动力疗法在治疗类风湿性关节炎,黄斑变性病,抗菌,抗病毒等方面也有了更深入的研究。光动力疗法过程中不可缺少的三个组成部分分别是光,光敏剂和氧。光动力疗法的过程主要是光敏剂吸收光子的能量,从稳态跃迁到激发态,激发态的光敏剂再将能量传递给氧,生成活性氧(Radical oxygen species,ROS)。机体内是有一定ROS水平,但是过高的活性氧水平则会损伤脂质,蛋白质和核酸等生物大分子,从而导致细胞死亡。在光动力过程中,光敏剂是整个过程的核心。整个PDT的发展都是随着光敏剂的发现,发展和应用而推进并完善的。
与传统的三种癌症治疗手段相比较,PDT具有以下优点:(1)暗毒性低,光活性高。可以通过选择性的进行局部照射的方式,控制光敏剂的作用范围,从而降低正常组织的光损伤。(2)副作用小。相比于传统的化疗和放疗,PDT的毒副作用较小,对组织的损伤小。(3)药物在体内代谢快。药物在体内快速代谢,将不会在体内进行循环,造成不必要的治疗负担。虽然PDT的研究已经取得可观的进展,但是PDT也存在一些问题。就临床批准的光敏剂而言,仍然存在许多的缺陷,特别是长期的皮肤毒性,使接受治疗后的病人不能长时间在日光下暴露。而且临床使用的光敏剂并没有表现出一定的细胞选择能力,所以研究更为合适的光敏剂靶向成为了新的目标。
竹红菌素(Hypocrellin)是我国民间药物竹黄的主要活性成分,是一种天然的苝醌类光敏色素,包括竹红菌甲素、竹红菌乙素、竹红菌丙素、竹红菌丁素。竹红菌素拥有杰出的抗肿瘤活性主要是由于它们的光敏特性决定的。其具有以下优点:(1)杰出的量子氧产率,包括产生单线态氧,超氧阴离子及羟基自由基。细胞内本身具有一定的活性氧水平,但是当大量的活性氧产生时,细胞由于无法及时清除这些额外的过氧化物,从而死亡。(2)此外,竹红菌素拥有较快的体内代谢速度,这能够避免病人在接受治疗后长时间光照带来的皮肤毒性。(3)易于分离纯化。可以从竹黄分离菌株中分离纯化得到大量的竹红菌素。因此竹红菌素被认为是一种非常有应用前景的光疗药物,具有潜在的临床应用价值。
竹红菌素作为具有应用前景的光敏剂仍然存在着一些缺陷,如竹红菌素属于脂溶性化合物,在水溶液中的溶解度是极低的,不能达到有效的药物浓度,而且本身在血液中容易发生自发的聚集,造成血栓。此外,竹红菌素在合适的激发光下即可被激发形成活发态,传递能量给周围的氧形成活性氧,引起细胞的死亡。这种性质是没有肿瘤特异性的。所以竹红菌素的选择性不足也是需要解决的问题。为了克服竹红菌素的水溶性问题,主要采用化学修饰的方法合成水溶性的竹红菌素衍生物及制备水溶性的竹红菌素纳米载体。化学修饰的方法获得了一些水溶性竹红菌素的衍生物,但是往往得到的化合物量子氧产率不理想,或者是竹红菌素衍生物在细胞中的应用效果还未得到证实。所以通过化学修饰的途径是否能够达到理想的光动力治疗效果仍需进一步的验证。此外,使用载体包封竹红菌甲素是一个非常有前景方法。关于应用材料包封竹红菌素的资料:专利申请号200810024636.4公开了一种水溶性竹红菌素二氧化硅纳米粒的制备方法,选择的包封材料为二氧化硅。专利申请号200710025279.9公开了一种水溶性竹红菌素二氧化钛纳米粒的制备方法,所采用的包封材料为二氧化钛,包封药物为竹红菌素,形成的纳米粒径为150nm。利用上述载体对竹红菌素进行包裹,虽然得到了水溶性良好的纳米药物,但仍然存在缺陷,如该类药物载体的材料属于无机材料,药物经注射进入机体后,由于缺乏良好的生物相容性,在体内不能很好地进行代谢,可能给机体带来代谢负担。专利申请号200610001677.2公开了一种竹红菌素微乳液及其制备方法,采用天然油脂和表面活性剂为包封材料,其粒径为20-100nm。专利申请号200710120727.3公开了一种竹红菌素脂质体制剂及其制备方法,采用磷脂和胆固醇作为包封材料。这类脂质体纳米粒适合于静脉注射,但是在保存稳定性上仍然存在着不足,长期稳定性较低。专利申请号03148147.7公开了一类竹红菌素水溶性纳米及其用途,采用的包封材料为天然生物蛋白,多糖类大分子及可生物降解的高分子聚合物,制备成水溶性的超微粒,粒径为20-200nm。专利申请号201410198946.3公开了一种水溶性竹红菌素PLGA纳米粒及其制备方法,采用的包封材料是PLGA,纳米粒径为20-200nm之间。这类纳米粒采用可生物降解的高分子聚合物对竹红菌素进行包封,具有良好的生物相容性,但是形成的纳米粒粒径均匀度分布指数还不够理想,包封率较低,为55%。聚乳酸-羟基乙酸及羧甲基壳聚糖均属于生物可降解的高分子化合物,应用两种化合物对一些药物进行包封的文献已有报道,如胡云霞等报道的(高分子通报,2004(5):62-68)中采用两种化合物对抗癌药物5-氟尿嘧啶进行包封。但是两种化合物共同对竹红菌素进行包封来解决水溶性和包封率的问题还未见报道。
一些肿瘤细胞,如肺腺癌A549和乳腺癌MDA-MB-231等细胞株由于快速生长而需要大量的铁离子,而铁离子的摄取依赖于细胞膜转铁蛋白受体介导的胞吞途径进行内化,所以这些细胞表面均过量表达转铁蛋白受体。所以这个有效的细胞摄取途径被用于位点特异性的递送抗肿瘤药物。例如,2014年,中国的Han等人也利用转铁蛋白修饰的脂质体纳米粒共递送DNA和多柔比星,并在过表达转铁蛋白受体的A549细胞中得到验证(InternationalJournal of Nanomedicine,2014:4107-4116)。2015年,新加坡的Muthu等人采用流式细胞术分析MDA-MB-231细胞表面相比于NIH-3T3细胞有过量表达的转铁蛋白受体,于是用转铁蛋白与维生素E TPGS聚合共递送多烯紫杉醇和超亮金纳米粒,实现靶向递送化疗药物多柔比星(Biomaterials,2015,39:234)。但是使用转铁蛋白靶向修饰聚乳酸-羟基乙酸/羧甲基壳聚糖纳米粒用于递送竹红菌素类光敏剂进行光动力治疗的资料未见公开报道。同时在该靶向纳米粒中用聚乳酸-羟基乙酸和羧甲基壳聚糖制备纳米粒包封竹红菌素以提高包封率的资料未见公开文件。
发明内容
发明目的:针对现有技术存在的缺陷,本发明提供一种主动靶向特定肿瘤细胞的竹红菌素纳米制剂,该制剂为转铁蛋白修饰的聚乳酸-羟基乙酸/羧甲基壳聚糖包封竹红菌素的制剂,能够主动靶向过量表达转铁蛋白受体的肿瘤细胞,具有靶向性高,水分散效果好,粒径均一,包封率高,稳定性好,材料的安全性能高等优点。
本发明还提供该主动靶向特定肿瘤细胞的竹红菌素纳米制剂的制备方法和应用。
技术方案:为了实现上述目的,如本发明所述的一种主动靶向特定肿瘤细胞的竹红菌素纳米制剂,所述制剂为转铁蛋白修饰的聚乳酸-羟基乙酸/羧甲基壳聚糖包封竹红菌素的制剂,制剂中有效成分竹红菌素的最终质量体积比为0.2-0.6mg/mL、转铁蛋白的最终质量体积比为0.5-2mg/mL,溶剂为水。
其中,所述竹红菌素选自竹红菌甲素、竹红菌乙素、竹红菌丙素和竹红菌丁素中任意一种。
本发明所述的主动靶向特定肿瘤细胞的竹红菌素纳米制剂的制备方法,包括如下步骤:
(1)将含有羧甲基壳聚糖和聚乙烯醇的水相及含有聚乳酸-羟基乙酸和竹红菌素的有机相,通过常规水包油挥发法制备聚乳酸-羟基乙酸/羧甲基壳聚糖包封的竹红菌素纳米制剂;
(2)将步骤(1)聚乳酸-羟基乙酸/羧甲基壳聚糖包封的竹红菌素纳米制剂采用偶联法交联转铁蛋白,形成主动靶向特定肿瘤细胞的竹红菌素纳米制剂。
步骤(1)的具体方法为将按比例称取的羧甲基壳聚糖和聚乙烯醇溶解在超纯水中制成水相;按比例称取聚乳酸-羟基乙酸和竹红菌素溶于丙酮中作为有机相;向高速搅拌的水相中逐滴滴加有机相,滴加结束后,超声分散,搅拌以挥去有机溶剂,剩下的溶剂再离心去除未包封的竹红菌素,未反应的聚乳酸-羟基乙酸和羧甲基壳聚糖,以及多余的乳化剂,得到的沉淀即为聚乳酸-羟基乙酸/羧甲基壳聚糖包封的竹红菌素纳米制剂。
步骤(2)的具体方法为将步骤(1)得到的聚乳酸-羟基乙酸/羧甲基壳聚糖包封的竹红菌素水溶性纳米粒离心然后重悬到氯化钠活化缓冲液中,加入1-(3-二甲氨基丙基)-3-乙基碳二亚胺盐酸盐/N-羟基丁二酰亚胺活化,然后加入β-巯基乙醇终止反应,离心收集纳米粒并重悬到磷酸盐缓冲液中,加入转铁蛋白反应,离心收集沉淀,采用分子截留透析袋纯化,得到转铁蛋白修饰聚乳酸-羟基乙酸/羧甲基壳聚糖包封的竹红菌素纳米制剂,即主动靶向特定肿瘤细胞的竹红菌素纳米制剂。
其中,步骤(1)所述水相中羧甲基壳聚糖初始质量体积比为2-10mg/mL,聚乙烯醇初始质量体积比为2-10mg/mL,溶剂为超纯水;所述有机相中聚乳酸-羟基乙酸初始质量体积比为40-200mg/mL,竹红菌素初始质量体积比为2.5–7.5g/mL,溶剂为丙酮。
步骤(2)所述转铁蛋白按与步骤(1)中水相的初始质量体积比为0.5–2.5mg/mL加入。
本发明所述的主动靶向特定肿瘤细胞的竹红菌素纳米制剂制备用于靶向递送竹红菌素类光敏剂到过量表达转铁蛋白受体的多种肿瘤细胞药物中的应用。
作为优选,所述肿瘤细胞包括肺腺癌细胞和乳腺癌细胞。
本发明在采用的高分子化合物聚乳酸-羟基乙酸优点的基础上,在纳米粒包埋材料中增加引入了羧甲基壳聚糖和转铁蛋白制备了主动靶向特定肿瘤细胞的竹红菌素纳米制剂。羧甲基壳聚糖的加入明显降低了制剂中的纳米粒的粒径,并增加了纳米粒粒径均一度,同时提高了竹红菌素的包封效率。选用的光敏剂是稀有的竹黄真菌上获得的竹红菌素光敏剂。制备得到的纳米制剂中竹红菌素包封率高,具有良好的水溶性,并能够靶向肿瘤组织,以减少竹红菌素光敏中带来的毒副作用,增加了光动力治疗的靶向性和成功率。此外,本发明通过水包油乳化溶剂挥发法,在自然挥发的过程中形成了粒径均一且水分散性较好的聚乳酸-羟基乙酸/羧甲基壳聚糖包封的竹红菌素纳米制剂,包封了优良的天然光敏剂竹红菌素。然后在通过碳二亚胺偶联法,对聚乳酸-羟基乙酸/羧甲基壳聚糖包封的竹红菌素纳米粒上添加了靶向配体,获得了主动靶向特定肿瘤细胞的竹红菌素纳米制剂,得到的纳米制剂对肺腺癌(A549)和乳腺癌(MDA-MB-231)等肿瘤细胞具有靶向性,水分散效果较好,粒径均一,粒径分布在96-156nm,包封率高,达80%以上。本发明合成的纳米粒表面带有肿瘤位点过量表达受体的靶向配体,能够实现配体和受体的特异性结合,增加肿瘤部位的药物摄取,最大限度的降低药物对正常组织的毒副作用,并保留了竹红菌素光敏剂暗毒性低,光活性高的优点。本发明所提供的制备方法采用的材料均为生物可降解的材料,能够在机体中正常降解,材料的安全性能较高。
本发明中的试剂都是市售可得,转铁蛋白购买自北京索莱宝科技有限公司,本发明中所使用的水均为超纯水。
有益效果:与现有技术相比,本发明具有如下优点:
1、具有肿瘤位点靶向性。由于纳米粒表面添加了肿瘤位点过量表达受体转铁蛋白受体的靶向配体转铁蛋白,本发明制备的纳米制剂可以实现特异性的靶向配体与受体的结合,增加肿瘤部位的药物吸收,减少药物在正常组织的积累,从而降低副作用。
2、安全性能高。本发明所提供的制备方法采用的材料均为生物可降解的材料,不会引起机体的免疫反应,并能够在机体中正常降解。
3、暗毒性低光活性高。通过本发明制备的纳米粒保留了光敏剂的优点,在暗条件下几乎不具有毒性作用,但是在光处理情况下能够产生非常强的光毒性。
4、水分散性好。通过本发明制备的纳米制剂能够在水溶液中形成均匀分散的介质,而且通过在表面增加水溶性的的蛋白进一步增加了纳米粒在水溶液中的稳定性。
5、包封率高,粒径均一。本发明的制备的纳米制剂相对于已经公开的文件中(201410198946.3)的水溶性竹红菌素PLGA纳米率,包封率提高到80%以上;且纳米粒粒径均一,粒径分布在96-156nm。
6、制备工艺简单。此方法采用的仪器常见,操作步骤简单,易重复,且原料易得;制备得到的纳米制剂是一种主动靶向递送竹红菌素类光敏剂到过量表达转铁蛋白受体的多种肿瘤细胞的药物制剂。

具体实施方式
以下结合附图对本发明作进一步说明。
实施例1
称取300mg的羧甲基壳聚糖(CMC),500mg的聚乙烯醇(PVA)溶解在50mL超纯水中,制成水相;分别称取300mg的聚乳酸-羟基乙酸(PLGA)、20mg的竹红菌甲素(HA)溶于5mL丙酮中,作为有机相,向10000rpm/min高速搅拌的水相中逐滴滴加有机相。滴加结束后,50W超声分散10min,然后置于磁力搅拌器上1000rpm/min搅拌12h以挥去有机溶剂。剩下的溶剂在13000rpm/min条件下离心去除未包封的HA,未反应的PLGA和CMC,以及多余的乳化剂,得到的沉淀即为聚乳酸-羟基乙酸/羧甲基壳聚糖包封的竹红菌素纳米粒。将纳米粒洗涤,重悬到50mL 0.5M NaCl活化缓冲液中,加入20mg的1-(3-二甲氨基丙基)-3-乙基碳二亚胺盐酸盐和30mg的N-羟基丁二酰亚胺活化15min,然后加入20μLβ-巯基乙醇终止活化。1000rpm/min离心5min收集沉淀,沉淀重悬于50mL反应缓冲液中(0.1M磷酸缓冲液(PBS)),然后称取50mg的转铁蛋白加入PBS缓冲液中室温反应2h,加入反应终止剂,采用10000mW分子截留透析袋纯化24h,得到主动靶向特定肿瘤细胞的竹红菌素纳米制剂。通过透射电镜检测纳米粒表面形态及团聚状况,如图1所示,主动靶向特定肿瘤细胞的竹红菌素纳米粒粒径均一,形态为圆形,且均匀分散没有团聚。
本实施例最后得到的主动靶向特定肿瘤细胞的竹红菌素纳米制剂中,竹红菌素0.36mg/mL、转铁蛋白0.90mg/mL,溶剂为水,竹红菌素的包封率为90%,转铁蛋白的装载量为90%。
实施例2
实施例2与实施例1的原料和制备方法相同,不同之处在于:采用100mg的CMC,300mg的PVA、100mg的PLGA、12mg的竹红菌乙素以及125mg的转铁蛋白。
本实施例得到的主动靶向特定肿瘤细胞的竹红菌素纳米粒中竹红菌乙素0.2mg/mL、转铁蛋白2mg/mL,溶剂为水,竹红菌素的包封率为83%,转铁蛋白的装载量为80%。
实施例3
实施例3与实施例1的原料和制备方法相同,不同之处在于:采用500mg的CMC,100mg的PVA、500mg的PLGA、37.5mg的竹红菌丙素以及25mg的转铁蛋白。
本实施例得到的主动靶向特定肿瘤细胞的竹红菌素纳米制剂中竹红菌丙素0.6mg/mL,转铁蛋白0.5mg/mL,溶剂为水。竹红菌素的包封率为82%。
实施例4
实施例4与实施例1的原料和制备方法相同,不同之处在于:采用竹红菌丁素。
本实施例得到的主动靶向特定肿瘤细胞的竹红菌素纳米制剂中竹红菌丁素0.36mg/mL、转铁蛋白0.9mg/mL,溶剂为水,竹红菌素的包封率为89%。
实施例5
细胞存活率计算:
将处于对数生长期的肺腺癌细胞株(A549)和乳腺癌细胞株(MDA-MB-231)细胞用胰酶消化,然后调整浓度,每孔接种100μL至96孔板中,使每孔细胞浓度为1×104个,培养24h使细胞贴壁。移除培养液,用PBS冲洗细胞2次,加入不同浓度本发明制备的纳米粒,以培养基处理为对照,继续培养6h。然后移去药物,PBS冲洗细胞两次,加入100μL培养基后,将96孔板置于LED灯(470nm)下,光照处理15min或不做光照处理,然后放回培养箱继续培养12h。然后移除培养液,PBS冲洗细胞,加入100μL稀释的细胞活性检测试剂后放入培养箱孵育1h,使用多孔酶标仪检测450nm波长处的吸光值。细胞生存率计算公式如下:
试验例1评估有/无转铁蛋白修饰的竹红菌甲素纳米制剂对A549细胞光毒性。
将转铁蛋白受体阳性的A549细胞接种到96孔板上,置于37℃,5%CO2细胞培养箱中培养12h贴壁,采用实施例1制备的转铁蛋白修饰的靶向纳米制剂和未经转铁蛋白修饰的纳米制剂用培养基稀释到不同的浓度,使竹红菌甲素的含量分别为5.46、10.92、16.38、21.84、27.30ng/mL,每孔加入100μL稀释后的纳米制剂,与A549细胞在培养箱中孵育8h,然后用470nm的LED灯照射15min,继续置于细胞培养箱中培养16h。最后加入细胞活性检测试剂共孵育1h,450nm下检测吸收值,计算细胞的存活率。转铁蛋白修饰的靶向纳米制剂不同浓度(分别含竹红菌甲素5.46、10.92、16.38、21.84、27.30ng/mL)处理的细胞存活率分别为70.41%,43.21%,37.46%,12.76%,1.40%;未经转铁蛋白修饰的纳米制剂不同浓度(分别含竹红菌甲素5.46、10.92、16.38、21.84、27.30ng/mL)处理的细胞存活率分别为106.31%,82.66%,58.98%,43.71%,13.02%。
由细胞存活率结果发现:在光照处理的条件下,转铁蛋白修饰的靶向纳米制剂比没有靶向修饰的纳米制剂具有更高的细胞毒性,且具有统计学差异。这是由于转铁蛋白的修饰使纳米粒能够与A549细胞表面过量表达的转铁蛋白受体相结合,促进细胞对纳米粒的内化作用,从而使更多的药物能够进入到细胞内,诱导更多的细胞凋亡。而且在此浓度下,随着药物浓度的增加,细胞的存活率呈现浓度依赖的降低。
试验例2评估有/无转铁蛋白修饰的竹红菌甲素纳米制剂对A549细胞暗毒性
将转铁蛋白受体阳性的A549细胞接种到96孔板上,置于37℃,5%CO2细胞培养箱中培养12h贴壁,采用实施例1制备的转铁蛋白修饰的靶向纳米制剂和未经转铁蛋白修饰的纳米制剂用培养基稀释到不同的浓度,使竹红菌甲素的含量分别为5.46、10.92、16.38、21.84、27.30ng/mL,每孔加入100μL稀释后的纳米制剂,与A549细胞在培养箱中孵育24h。最后加入细胞活性检测试剂共孵育1h,450nm下检测吸收值,计算细胞的存活率。转铁蛋白修饰的靶向纳米制剂不同浓度(分别含竹红菌甲素5.46、10.92、16.38、21.84、27.30ng/mL)处理的细胞存活率分别为109.02%,111.01%,99.56%,106.89%,111.72%;未经转铁蛋白修饰的纳米制剂不同浓度(分别含竹红菌甲素5.46、10.92、16.38、21.84、27.30ng/mL)处理的细胞存活率分别为109.77%,99.26%,107.98%,106.73%;107.23%。由细胞存活率结果发现:在未经光照处理的条件下,不同浓度纳米制剂处理的A549细胞存活率没有表现出浓度的差异,不同类型纳米制剂处理A549细胞存活率也为表现出差异。所有的组处理均未表现出明显的细胞毒性。A549细胞维持较好的细胞活力,未发生细胞凋亡现象,证明药物和载体在暗培养的条件下是安全的,不会引起任何细胞毒性反应。
试验例3评估有/无转铁蛋白修饰的竹红菌甲素纳米制剂对MDA-MB-231细胞光毒性。
将转铁蛋白受体阳性的MDA-MB-231细胞接种到96孔板上,置于37℃,5%CO2细胞培养箱中培养12h贴壁,采用实施例1制备的转铁蛋白修饰的靶向纳米制剂和未经转铁蛋白修饰的纳米制剂用培养基稀释到不同的浓度,使竹红菌素的含量分别为5.46、10.92、16.38、21.84、27.30ng/mL,每孔加入100μL稀释后的纳米,与MDA-MB-231细胞在培养箱中孵育8h,然后用470nm的LED灯照射15min,继续置于细胞培养箱中培养16h。最后加入细胞活性检测试剂共孵育1h,450nm下检测吸收值,计算细胞的存活率。转铁蛋白修饰的聚乳酸/羟基乙酸包封的竹红菌甲素纳米粒不同浓度(分别含竹红菌甲素0.01、0.02、0.03、0.04、0.05μM)处理的细胞存活率分别为80.24%,64.32%,46.57%,24.11%,9.84%;未经转铁蛋白修饰的聚乳酸/羟基乙酸包封的竹红菌甲素纳米粒不同浓度(分别含竹红菌甲素0.01、0.02、0.03、0.04、0.05μM)处理的细胞存活率分别为103.22%,90.56%,74.18%,56.35%,31.74%。由细胞存活率结果发现:在光照处理的条件下,转铁蛋白修饰的纳米制剂比没有靶向修饰的纳米制剂具有更高的细胞毒性,且具有统计学差异。这是由于转铁蛋白的修饰使纳米粒能够与MDA-MB-231细胞表面过量表达的转铁蛋白受体相结合,促进细胞对纳米粒的内化作用,从而使更多的药物能够进入到细胞内,诱导更多的细胞凋亡。而且在此浓度下,随着药物浓度的增加,细胞的存活率呈现浓度依赖的降低。
试验例4评估有/无转铁蛋白修饰的竹红菌甲素纳米制剂对MDA-MB-231细胞暗毒性。
采用实施例5的方法将转铁蛋白受体阳性的MDA-MB-231细胞接种到96孔板上,置于37℃,5%CO2细胞培养箱中培养12h贴壁,采用实施例1制备的转铁蛋白修饰的靶向纳米制剂和未经转铁蛋白修饰的纳米制剂用培养基稀释到不同的浓度,使竹红菌素的含量分别为5.46、10.92、16.38、21.84、27.30ng/mL,每孔加入100μL稀释后的纳米制剂,与MDA-MB-231细胞在培养箱中孵育24h。最后加入细胞活性检测试剂共孵育1h,450nm下检测吸收值,计算细胞的存活率。转铁蛋白修饰的靶向纳米制剂不同浓度(分别含竹红菌甲素5.46、10.92、16.38、21.84、27.30ng/mL)处理的细胞存活率分别为106.45%,102.21%,109.32%,100.22%,103.20%;未经转铁蛋白修饰的纳米制剂不同浓度(分别含5.46、10.92、16.38、21.84、27.30ng/mL)处理的细胞存活率分别为103.56%,104.62%,101.24%,109.14%;103.92%。由细胞存活率结果发现:在未经光照处理的条件下,不同浓度纳米制剂处理的MDA-MB-231细胞存活率没有表现出浓度的差异,不同类型纳米制剂处理MDA-MB-231细胞存活率也未表现出差异。所有的组处理均未没有明显的细胞毒性。MDA-MB-231细胞维持较好的细胞活力,未发生细胞凋亡现象,证明药物和载体在暗培养的条件下不会引起MDA-MB-231任何的细胞毒性。
Claims (6)
1.一种主动靶向特定肿瘤细胞的竹红菌素纳米制剂,其特征在于,所述制剂为转铁蛋白修饰的聚乳酸-羟基乙酸/羧甲基壳聚糖包封竹红菌素的制剂,制剂中有效成分竹红菌素的最终质量体积比为0.2-0.6mg/mL、转铁蛋白的最终质量体积比为0.5-2mg/mL,溶剂为水。
2.根据权利要求1所述的主动靶向特定肿瘤细胞的竹红菌素纳米制剂,其特征在于,所述竹红菌素选自竹红菌甲素、竹红菌乙素、竹红菌丙素和竹红菌丁素中任意一种。
3.一种权利要求1所述的主动靶向特定肿瘤细胞的竹红菌素纳米制剂的制备方法,其特征在于,包括如下步骤:
(1)将含有羧甲基壳聚糖和聚乙烯醇的水相和含有聚乳酸-羟基乙酸和竹红菌素的有机相,通过水包油挥发法制备聚乳酸-羟基乙酸/羧甲基壳聚糖包封的竹红菌素纳米制剂;
(2)将步骤(1)聚乳酸-羟基乙酸/羧甲基壳聚糖包封的竹红菌素纳米制剂采用偶联法交联转铁蛋白,形成主动靶向特定肿瘤细胞的竹红菌素纳米制剂。
4.根据权利要求3所述的主动靶向特定肿瘤细胞的竹红菌素纳米制剂的制备方法,其特征在于,步骤(1)所述水相中羧甲基壳聚糖的初始质量体积比为2-10mg/mL,聚乙烯醇的初始质量体积比为2-10mg/mL,溶剂为超纯水;所述有机相中聚乳酸-羟基乙酸的初始质量体积比为40-200mg/mL,竹红菌素的初始质量体积比为2–7.5mg/mL,溶剂为丙酮。
5.根据权利要求3所述的主动靶向特定肿瘤细胞的竹红菌素纳米制剂的制备方法,其特征在于,步骤(2)所述的转铁蛋白在步骤(1)所述水相的初始质量体积比为0.5–2.5mg/mL。
6.一种权利要求1所述的主动靶向特定肿瘤细胞的竹红菌素纳米制剂在制备用于靶向递送竹红菌素类光敏剂到过量表达转铁蛋白受体的多种肿瘤细胞药物中的应用。
