Abstract
The invention belongs to the technical field of longan seedling raising, and particularly relates to a method for improving the germination rate of longan seeds stored at room temperature in a short term. According to the method, longan seeds stored at room temperature in a short period are soaked in the Indian pyricularia indica fermentation liquor, and then are taken out for sowing, so that the germination rate is remarkably improved and the seedlings grow better compared with longan seeds which are not soaked in the Indian pyricularia indica fermentation liquor. The fermentation liquor of Piriformospora indica is easy to prepare, has strong feasibility, is simple to operate when used for treating seeds, and has higher application and economic values.
Description
Technical Field
The invention belongs to the technical field of longan seedling raising, and particularly relates to a method for improving the germination rate of longan seeds stored at room temperature in a short term.
Background
Seeds can be classified into normal seeds and recalcitrant seeds according to storage characteristics. Wherein, the recalcitrant seeds still have higher water content when mature, are highly sensitive to dehydration and are not resistant to drying and low temperature, so the recalcitrant seeds are not resistant to storage and have shorter service life. The longan seeds belong to recalcitrant seeds and have a service life of only 8-10 days under common conditions. In order to improve the germination rate of stored seeds, low-temperature or ultralow-temperature storage at 15 ℃ is often used, but specific storage equipment or low-temperature environment is required for the storage methods, and due to the fact that longan seeds are large, the storage method is high in use cost under the condition that the number of seeds is large, and fresh seed sowing or short-term room-temperature storage sowing is adopted in production rarely. The germination rate of longan seeds stored at room temperature in a short period is obviously reduced, and a plurality of difficulties are brought to production and related scientific research work. Therefore, the improvement of the germination rate of longan seeds stored at room temperature in a short period is significant.
Piriformospora indica (B)Piriformospora indica) Have been found to increase the germination rate of various plant seeds under stress conditions (e.g., drought, salinity, low temperature, high temperature, etc.). 2) preparation of Piriformospora indica fermentation broth
Activating the Piriformospora indica strains, selecting aerial mycelia at the edge part of a bacterial colony, inoculating the aerial mycelia on a new PDA solid culture medium, placing the PDA solid culture medium in an incubator at 28 ℃ for dark culture for 5 d, taking a bacterial block by using a puncher with the diameter of 5mm in a triangular flask filled with 100 ml of sterilized PDA liquid culture medium, culturing the bacterial block in a shaking table at the temperature of 28 ℃ and the rotating speed of 120 r/min for 2 days, filtering the bacterial block by using single-layer gauze, and collecting filtrate to obtain Piriformospora indica fermentation liquor;
3) longan seed storage and seed soaking
Storing longan seeds for 3-5 days at room temperature, and soaking in an Indian pyricularia indica fermentation liquor for 2-4 hours for sowing.
The invention has the following advantages:
1) the Piriformospora indica fermentation liquor used in the invention is easy to prepare and has strong feasibility;
2) the method can improve the germination rate of longan seeds stored at room temperature for a short time, and the seedlings grow better and have higher application and economic values;
3) the method is simple to operate and high in feasibility.

Example 1
A method for improving the germination rate of longan seeds stored at room temperature comprises the following specific steps:
1. longan seed collection and cleaning
Taking freshly picked longan fruits, removing peels and pulps, putting seeds into clear water, removing malformed, germinated, small and light seeds, quickly cleaning to remove the pulps adhered to the seeds, and draining;
2. preparation of Piriformospora indica fermentation broth
The Piriformospora indica strain is activated, and then aerial hyphae at the edge part of a colony are picked and inoculated on a new PDA solid culture medium. Placing in an incubator at 28 deg.C for dark culture for 5 d, taking out the bacterial blocks with a punch with diameter of 5mm, placing in a 250 ml triangular flask containing 100 ml sterilized PDA liquid culture medium, culturing in a shaking table with temperature of 28 deg.C and rotation speed of 120 r/min for 2 days, filtering with single-layer gauze, and collecting filtrate to obtain Piriformospora indica fermentation broth;
3. longan seed storage and seed soaking
Storing the longan seeds of Dongding into 2 groups after 3 days at room temperature, soaking one group in Piriformospora indica fermentation liquor for 2-4 h (treatment group), taking the other group without treatment as a control group, wherein each group contains 50 seeds, and each group is repeated for at least 3 times;
4. observation and statistics of sowing and seed germination conditions
Sowing the seeds of the Dongbing longan into a hole tray (the length, width and height of each hole are 4 cm multiplied by 4 cm), adding 2/3 nutrient soil into the hole tray, placing the seeds on a substrate after watering thoroughly, then covering the substrate with about 1 cm thick nutrient soil, and placing the substrate in a greenhouse for culturing at 25 +/-2 ℃ after watering thoroughly;
seed germination was counted at 2 weeks after sowing (fig. 1, fig. 2). The germination rate of the control group seeds was 54.2% + -13.0%, while the germination rate of the Piriformospora indica treated group was 95.8% + -3.6%. The reduction of the germination rate of the longan seeds after 3 days of room temperature storage is demonstrated, and the germination rate can be obvious by using the pyricularia indica treatment. In addition, as can be seen from fig. 1 and 2, the treated longan seedlings tend to grow better than the control seedlings.
Example 2
A method for improving the germination rate of longan seeds stored at room temperature comprises the following specific steps:
1. longan seed collection and cleaning
Taking freshly picked 'mid-autumn first-number' longan fruits, removing peels and pulps, then placing seeds in clear water, removing malformed, germinated, small and light seeds, quickly cleaning to remove pulps adhered to the seeds, and draining;
2. preparation of Piriformospora indica fermentation broth
The Piriformospora indica strain is activated, and then aerial hyphae at the edge part of a colony are picked and inoculated on a new PDA solid culture medium. 4. observation and statistics of sowing and seed germination conditions
The longan seeds of 'mid-autumn No. I' are sowed in hole trays (the length, width and height of each hole are 5 cm multiplied by 5 cm). Adding 2/3 nutrient soil into the plug, watering, placing the seeds on the substrate, covering with nutrient soil with thickness of about 1 cm, watering, and culturing in a greenhouse at 25 + -2 deg.C.
Seed germination was counted 2 weeks after sowing (fig. 3). The germination rate of the control group seeds was 66.7% + -9.4%, while the germination rate of the Piriformospora indica treated group was 88.9% + -10.2%. The result shows that the germination rate of the longan seeds in mid-autumn period I is reduced after 5 days of room temperature storage, and the germination rate of the seeds is obviously promoted by using the Piriformospora indica treatment. As can be seen from FIG. 3, the longan seedlings in the treated group grew slightly better than those in the control group.

A method for improving the germination rate of longan seeds stored at room temperature is characterized by comprising the following steps: soaking longan seeds stored at room temperature for a short time in an Indian pyricularia indica fermentation liquor for 2-4 h, and taking out the longan seeds for sowing;
the preparation method of the Piriformospora indica fermentation liquor comprises the following steps: activating an Indian pyricularia pyricularis strain, selecting aerial hyphae at the edge part of a bacterial colony, inoculating the aerial hyphae on a new PDA solid culture medium, placing the culture medium in an incubator at 28 ℃ for dark culture for 5 days, taking a bacterial block by using a puncher with the diameter of 5mm in a triangular flask filled with 100 ml of sterilized PDA liquid culture medium, culturing the bacterial block in a shaking table at the temperature of 28 ℃ and the rotating speed of 120 r/min for 2 days, filtering the bacterial block by using single-layer gauze, and collecting filtrate to obtain Indian pyricularia pyricularis fermentation liquor;
the longan seeds stored at the room temperature for a short time are as follows: taking fresh picked longan fruits, removing peels and pulps, putting seeds into clear water, removing malformed, germinated, small and light seeds, quickly cleaning to remove pulps adhered to the seeds, draining, and storing the longan seeds for 3-5 days at room temperature.
