本发明涉及一种金银花精油-β-环糊精包合物的制备方法。
背景技术
金银花为忍冬科植物忍冬的花蕾,含有绿原酸、木樨草苷和金银花精油等多种天然活性成分,具有清热解毒、凉散风热、抗菌、抗病毒、增强免疫及抗氧化等功效,是银黄颗粒、银黄口服液和柴黄口服液等多种制剂的主要配方成分。金银花精油为金银花花蕾经亲脂性有机溶剂萃取、水蒸气蒸馏、超临界流体萃取或分子蒸馏等技术制备的淡黄色油状液体,具有特异花香气味和抗真菌等功效,在中药制剂、食品、卷烟、香氛和日用化妆品中应用广泛。金银花精油极易挥发,稳定性差,严重限制了其应用。β-环糊精是由7个葡萄糖基以1,4一糖苷键连接而成的筒状分子,具有环状中空圆筒形结构,能够通过分子问的非共价键将客体分子包嵌在其空穴结构内形成包合物,从而增加客体分子的稳定性、改善客体分子的溶解性,并使液态的客体分子固态化。因此,将金银花精油制成β-环糊精包合物能够防止有效成分的快速挥发,并使其固态化以便于广泛应用。

本发明所述一种金银花精油-β-环糊精包合物的制备方法,其包括以下制备步骤:
(1) 取500g金银花新鲜花蕾,置于10L圆底烧瓶中,加入5 L去离子水,用水蒸气蒸馏法,收集馏出液,约1 L;
(2) 将35g/LNaCl加至馏出液中,溶解,静置1h,用等体积元水乙醚萃取,萃取液用无水硫酸钠干燥处理后,减压蒸馏得到淡黄色金银花精油,置于5℃冰箱待用;
(3) 称取一定量金银花精油,溶解于少量无水乙醇中,在一定温度下,慢速滴至100 mL相同温度的β-环环糊精饱和溶液中,以300r/min恒温搅拌一定时间后,取出冷却至室温,5℃冰箱中静置12 h;
(4) 取步骤(3)产物过滤,用少量无水乙醇快速洗涤沉淀,收集滤饼;
(5) 将β-环糊精包合金银花精油形成的滤饼经一定温度烘干至恒重,即得到金银花精油-β-环糊精包合物的干燥粉末,存储于5℃环境中。
