Description

Medicament for increasing wax secretion amount of fraxinus chinensis and treatment method thereof
Technical Field
The invention belongs to the technical field of biology, and particularly relates to a medicament for increasing wax secretion amount of wax insect and a treatment method thereof.
Background
White wax insectEricerus pela(channels) are classified in the order Hemiptera (Hemiptera), the family Lecanidae (Coccoidea) the genus Ericerus (Hemiptera)Ericerus). The Ceriporia alba is a resource insect with great economic value, and the Ceriporia alba secreted by 2-instar male larva is a widely used pure natural polymer compound, and contains hexacosanoic acid hexacosanyl ester (C)25H51COOC26H53) The white wax is mainly produced in China, and the white wax is utilized and produced in ancient China (more than 1000 years ago), and is mainly used for candles, traditional Chinese medicines, wax printing and engraving stamping. In traditional Chinese medicine, the traditional Chinese medicine is often used as a wound healing agent, a hemostatic agent, a tranquilizer and the like, and modern traditional Chinese medicine also uses white wax for treating uterine inflammation, chronic gastritis, rheumatism and the like. The white wax has the excellent characteristics of high melting point, good gloss, stable physicochemical property, moisture resistance, lubrication, gloss and the like, and is widely applied to the industries of chemical industry, medicine, food, agriculture and the like at present. White wax productThe method has the advantages that the areas are mostly distributed in poor mountainous areas in China, the improvement of the yield of the white wax can drive local economic development, meanwhile, a large number of host plants are needed for the development of the white wax, the method plays a positive promoting role in improving the local environment, and the method has important ecological, economic and social benefits in the development of the white wax industry in China.
The life cycle of the wax insect is 1 generation in 1 year, and there are amphoterism and parthenogenesis. The male wax insect grows through nymph, pupa and adult stages, and belongs to complete metamorphosis, the nymph grows on host plants twice and transfers in a lifetime, the nymph grows out from eggs for the first time, a proper leaf is searched on the host plants, the nymph grows on the back of the leaf and is fixed in an aggregation manner, and the nymph is called as 'fixed leaf' in the production of wax; the second time is that the nymphs of 1 year enter 2 years after molting, and the nymphs of 2 years are transferred from leaves to branches to live, are intensively distributed in the middle of host plants, and secrete white wax to cover worm bodies, as shown in figure 1, the nymphs are called 'fixed poles' in white wax production. The growth, development, reproduction and other behaviors of insects are influenced by various environmental factors such as temperature, humidity, illumination and the like, wax secretion of male white wax insects comprises complex ecological and physiological problems, wax secretion and other physiological phenomena are generated in the process of adapting to the complex ecological environment, the wax secretion and other physiological phenomena are mainly influenced by humidity and illumination, and the wax secretion tendency of the white wax insects in high-humidity and low-illumination areas is presented.
At present, the white wax is produced in Yunnan, Guizhou, Sichuan, Hunan, Shaanxi and other provinces, and the annual yield is more than 1000 tons at the highest. The white wax contains 26 alkanol, 28 alkanol and other higher alkanol, and has wide application in medicine, agriculture, health product and other products, and the annual output of the white wax and related products reaches over 20 hundred million. However, the white wax is limited by the quality, variety and potential energy deficiency factors of the seed insects and the later-period management, so that the yield of the white wax in the white wax producing area generally tends to decline year by year, and the production enthusiasm of wax farmers is greatly influenced. Therefore, how to improve the yield of the white wax by a convenient, effective and low-cost medicament treatment and forest management method becomes an important technical problem for solving the development bottleneck of the white wax industry in China.

Figure DEST_PATH_IMAGE004
The first four groups are respectively treated by spraying, drying and injecting three methods to treat male larva population of Ceriporiopsis chinensis 2 of Kunming area, and are compared with wax secretion (CK) of 2-instar male larva in nature state (as shown in figure 2). Under the spraying treatment, JH3 and JH2 have significant difference from CK, while JH0 and JH1 have insignificant difference from CK; under the coating dry treatment, the differences among JH1, JH2, JH3, JH0 and CK are not obvious; under the injection treatment, there was only a significant difference between JH3 and CK.
Then, the first four groups respectively adopt three methods of spraying, drying and injecting to treat 2-instar male larva populations of the cerambycis formosanus in the Anthriscus sylvestris area, and compare the 2-instar male larva populations with the individual wax excretion (CK) of the 2-instar male larvae in the natural state (as shown in figure 2). Under the spraying treatment, only a significant difference exists between JH2 and CK; under the condition of coating dry treatment, JH1 and JH2 have significant difference with CK; under the condition of spraying treatment, significant differences exist between JH0 and JH3 and CK, and the others are not significant.
In both Kunming and Sichuan Emei areas, JH2 showed significant differences from CK under spray treatment. The wax-up rates of individual wax insects in two regions under different treatments also showed differences in the increased yield under different concentrations of exogenous juvenile hormone analogue treatment (see table 3).
In Kunming areas, JH0, JH1 and JH3 show inhibition effects of different degrees on individual wax secretion amount of white wax worms under three treatment conditions of spraying, drying and injecting; JH2 shows promotion effect on individual wax secretion amount of the wax insect under three treatment conditions of spraying, drying and injecting, and the yield increase rate is 4.02% -41.50%, and JH2 shows the highest individual wax secretion amount increase rate of the wax insect under the spraying treatment. In the region of Szechwan Emei, JH0, JH1, JH2 and JH3 have promotion effect on individual wax secretion amount of white wax insect under three treatment conditions of spraying, drying and injection, and the yield is increased by 9.84-33.76%.
In the areas of Kunming and Sichuan Emei, JH2 has a promoting effect on the individual wax secretion amount of the white wax insects under the three treatment conditions of spraying, drying and injecting, and the yield increase rate of the individual wax secretion amount of the white wax insects in the areas of Kunming (41.50 mg/. holandra) and Emei (25.56 mg/. holandra) is higher than that under the treatment conditions of drying and injecting.
TABLE 3 increase of wax secretion yield of individual Ceriporiopsis chinensis treated at different concentrations in two regions
Figure DEST_PATH_IMAGE006
Exogenous juvenile hormone analogue is diluted into three different concentrations (25 mg.ml) of high, medium and low by using acetone aqueous solution with volume concentration of 1%-1、2.5mg•ml-1、1.25mg•ml-1) According to the nymph habit of the wax insect, three different methods of spraying, injecting and drying are selected to carry out exogenous juvenile hormone treatment in the 2-instar male larva stage. The test results show that medium and low concentration (2.5 mg.ml) is adopted-1And 1.25 mg.ml-1) The hormone analogue has obvious effect of promoting wax secretion of the wax insect individual, and is moderately and relatively superior to the juvenile hormone analogue with low concentration. For three different treatment modes, the wax secretion of the white wax insect is obviously promoted by spraying the juvenile hormone analogue with medium concentration. Therefore, the purpose of obviously increasing the wax can be achieved by spraying the juvenile hormone analogue with medium concentration (the average yield increase of 41.50% in the Kunming area: JH2 treatment and the average yield increase of 25.56% in the Emei area: JH2 treatment).
