本发明属于食品领域,具体涉及一种具有乌发、生发功能的组合物及其制备方法。
背景技术
头发,是指生长在头部的毛发,头发不是器官,不含神经、血管,但含有细胞,头发除了使人增加美感之外,主要作用是保护头脑。浓密蓬松的头发具有弹性,可以抵挡较轻的碰撞,寒冷时保暖、炎热时帮助头部汗液蒸发。毛发是皮肤的附属器,它同身体其它各部位的器官、组织一样,需要充足的营养。人种不同,发色也不同,白种人发色金黄、棕种人发色红黑、黄种人发色褐黑色或黑色。乌黑、浓密而具有光泽的美发一直是黑发人种群的美的追求;而早生的白发则给人以疲惫、衰老的印象。有关研究表明白发产生的主要原因是由于头发髓质和皮质里的黑色素颗粒减少或被空气填空。正常情况下,毛乳头内有丰富的血管,为毛乳头、毛球部提供充足的营养,黑色素颗粒顺利合成,头发呈黑色。当黑色素颗粒在毛乳头、毛球部的形成发生障碍,或者形成的黑色素但因某种原因,不能顺利运送到毛发中去,使毛发髓质、皮质部分的黑色素颗粒减少、消失,则头发变成白发。
人们随着年龄的增加,头发由黑变白是一种必然的生理趋势。老年头发灰白或变白是一种正常生理现象。灰发中黑素细胞数目正常,但黑素减少,而白发中黑素细胞也减少。青少年白发即头发过早变白,俗称少白头。中医学认为,下列因素与白发有关,一是精虚血弱,肾精不足,不能化生阴血,阴血亏虚,导致毛发失其濡养,故而花白。二是血热偏盛,情绪激动,致使肝旺血燥,血热偏盛,毛根失养。三是肝郁脾湿,肝气郁滞,损及心脾,气血生化无源,故而白发。这与现代医学研究的机体内分泌与免疫紊乱失调、精神创伤与过度紧张、血管及血液循环障碍等因素,导致毛发营养障碍而脱发、白发的观点一致。
人体的皮肤和毛发的颜色由许多因素决定,最重要的是黑色素的形成的程度和分布。黑色素是由专门的色素生成细胞形成的,叫做黑素细胞。黑色素是一类有着复杂结构、非均质的类多酚聚合体,其决定人体皮肤和毛发的颜色。黑色素具有吸收紫外辐射、清除自由基、抗氧化、吸附重金属离子、清除酚类化合物等生理功能。毛发黑素由毛囊黑素细胞合成,其合成代谢可分为黑素生成、转移和融化三个阶段,任何一个环节出现障碍均会影响黑素代谢,从而形成白发。酪氨酸酶对黑色素的生成起着关键作用,它的活性减低则使得头发的发干中缺乏黑色素而变成八法。故酪氨酸酶的激活作用可以反映乌发功效的强弱。一般认为,激活酪氨酸酶的以促进黑色素的产生,乌发过程就自然而然的完成了。
从生物化学角度以及酪氨酸酶的作用机理来看,酪氨酸酶在黑色素形成过程中专一,特定催化两个反应,1、酪氨酸的羟基化,生成多巴(DOPA),2、多巴的氧化,生成多巴醌。由于反应途径不同,主要生成两种黑色素,真黑色素和类黑色素。酪氨酸酶催化黑色素形成是开始的最关键一步,即酪氨酸酶转化为多巴醌,一般认为,酪氨酸酶的活性程度对色素沉积起着主要的作用。
很多患有脱发、早老性白发的人选择使用防脱、乌发产品染发而使其变黑。一般的染发剂会使头发发生氧化-还原反应而使头发的某些角质蛋白和色素成分发生明显的氧化,现在流行的漂染剂更是一种化学性脱色剂,在脱色的同时改变头发的颜色,这种成分对头发的毛鳞片有伤害,受损的毛鳞片会变得脆弱易断;同时经脱色剂处理后的头发各氨基酸含量可发生显著变化,如胱氨酸、蛋氨酸、酪氨酸含量会明显降低,对头发的质量和颜色有很大的负面影响,并且随着人的新陈代谢不断进行,新生成的头发仍然为白色,并未从根本上解决使头发变黑的问题。
发明内容
本发明的目的在于针对于现有技术的不足,根据中医传统理论的基础,结合药食同源类产品在健康食品领域的应用,提供一种具有乌发、生发功效的组合物。
为实现上述的发明目的,本发明提供如下的技术方案。
一种组合物,包括深色类食品原料和药食同源组分,深色类食品进一步地是指黑色类食品。
一种组合物,其特征在于包括由深色类食品原料和药食同源组分组成的组合物,深色类食品进一步地是指黑色类食品,可以是黑豆、黑木耳、黑芝麻、黑米、核桃中的一种或多种,该组合为具有乌发的功效和刺激头发的生长活性。
进一步地,深色类食品原料包括黑豆、黑木耳、黑芝麻、黑米、核桃。
黑色食品营养成分丰富,含有丰富氨基酸、微量元素和亚油酸等营养素,黑色食品能够调节人体生理功能,刺激内分泌系统,促进唾液分泌,有益肠胃消化,益肾强肾,增强人体免疫功能,延缓衰老,补而不腻,食而不燥。
然而仅依靠食品进补,达到乌发的效果并不十分理想,本发明选择药食同源组分和本领域中具有一定乌发功效的原料与黑色类食品组合,以达到乌发的效果。
一种具有乌发功效的组合物,由以下组分制成:黑豆20-50份、黑木耳25-60份、黑芝麻10-35份、黑米10-40份、核桃25-50份、花生红衣10-30份、红枣10-20份、桑果25-40份、枸杞10-20份、山药15-25份、百合10-30份、黄精15-25份、覆盆子15-30份、蛹虫草10-60份、阿胶5-15份、益智仁10-35份、茯苓25-40份、西红花10-20份、马齿苋10-20份、紫草40-60份、葡萄糖酸锌0.1-10份。
进一步地,所述的组分配比可以为黑豆30-50份、黑木耳45-60份、黑芝麻20-35份、黑米10-30份、核桃25-40份、花生红衣20-30份、红枣10-15份、桑果25-30份、枸杞10-15份、山药15-20份、百合10-25份、黄精15-20份、覆盆子15-25份、蛹虫草10-25份、阿胶5-10份、益智仁10-30份、茯苓25-35份、西红花10-15份、马齿苋10-15份、紫草40-50份、葡萄糖酸锌0.1-8份。
在一些实施方案中,组分可以为:黑豆35-50份、黑木耳45-50份、黑芝麻25-35份、黑米18-30份、核桃30-40份、花生红衣24-30份、红枣12-15份、桑果14-30份、枸杞10-17份、山药15-19份、百合10-20份、黄精15-18份、覆盆子15-28份、蛹虫草10-15份、阿胶5-8份、益智仁10-20份、茯苓25-30份、西红花10-12份、马齿苋10-13份、紫草45-50份、葡萄糖酸锌0.1-0.5份。
上述组合物在制备具有改善乌发、生发效果的食品中的用途。
进一步地,该组合物可以制备为成品,在上述的原料基础上,还包括有效量的可接受的添加剂,该组合物剂型可以是片剂、粉剂、丸剂、胶囊剂、颗粒剂、口服液。
本发明还涉及到组合物的制备方法。
本发明可以使用常规的处理方式,如将原料混合后进行粉碎,过筛后经过常规的压片工艺制得。
本发明优选下述的加工方式,一种组合物的制备方法,如下
(1)将黑豆、黑芝麻、黑米、核桃烘干,烘干温度为85-95℃;然后将烘干后的原料进行粉碎,过100-300目筛,取筛下物,获得原料A;将阿胶在85-95℃温度下烘干,粉碎,将西红花直接粉碎。
(2)将花生红衣、红枣、桑果、枸杞、山药、百合、黄精、覆盆子、蛹虫草、益智仁、茯苓、马齿苋、紫草、黑木耳进行超声波处理,超声波温度为45-55℃,时间为30-50min,设置功率为150w进行;然后进行超微粉碎,通过300目筛,得原料B;
(3)将得到的原料A加水煎煮,煎煮时间为30-90min,将滤渣重复煎煮一次,合并得到煎煮液;将原料B加水煎煮,煎煮时间为30-60min,将滤渣重复煎煮2次,合并得到煎煮液;将原料A和B的煎煮液合并,加入葡萄糖酸锌和粉碎后的阿胶、西红花至合并后的煎煮液中并充分分散于煎煮液中,将煎煮液浓缩为稠膏,烘干干燥后粉碎,过150-200目筛网,得到原料C;
(4)将原料C按照常规压片方式得到片剂。
该组合物还可以制备为粉剂、丸剂、胶囊剂、膏剂、颗粒剂,采用本领域中常规的剂型加工方式即可。如为了得到膏剂,可以在步骤(3)浓缩为稠膏后即加工为膏剂。如为了得到胶囊剂,可以将原料C装入胶囊壳中得到胶囊剂。
上述的组合物及实施例优选采用该优选的制备方法进行处理得到组合物。
每种食品或药食同源类组分的活性成分是保健作用的物质基础,增加原料中活性成分的溶出度和提取率,促进人体对有效成分的消化吸收,能够增加组合物的效果。通过超微粉碎处理原料,能够增加细胞破壁率,细胞内有效成分可充分暴露出来,使细胞内的有效成分直接接触到提取的溶液,提高活性成分的溶出度,进而便于人体对有效物质的吸收。
组合物中,各原料相互配伍,如选择桑果、枸杞、益智仁、黄精用其须发、补肝肾、益精血;选择花生红衣利用其补气止血的功效;选择紫草具有补中益气的功效;选择西红花用其活血祛瘀、凉血解毒,促进血液循环;选择黑豆、黑米、黑芝麻、黑木耳、核桃进行食补,预防白发,促进黑色素的形成。选择山药、百合健脾、补肺、固肾益精、滋润皮肤。红枣、阿胶粉补气血。利用马齿苋散血泊肿、清热解毒和激活酪氨酸酶活性的功效。利用蛹虫草粉丰富的微量元素和矿物质元素,促进营养平衡,调节阴阳。选择覆盆子温肾功效。利用茯苓的健脾和胃的功效。利用葡萄糖酸锌补充人体微量元素,预防头发变黄,促进头发黑亮。以达到原料相互配伍、促进,补气血、调肝肾,实现乌发、生发的效果。
在生发方面,例如,由默克公司研发的名为Propecia的药物,在受试者中,可以防止68%的人脱发,并且这里面的人66%的头发可以再生。然后此药物只是在服用期间有效,在停止服用药物后,头发就会在几个月内脱落。而且这种药物十分的昂贵且会导致性功能的下降。
虽然现有技术中公开了部分文献具有乌发的效果,但并没有文献写到乌发患者可以完全治愈后并能够长期预防。比如CN201510837523.6公开了一种用于治疗脱发白发的保健食品,CN200710304587.5公开了一种补肾润发的中药组合物、制剂及其制备方法,CN201010585907公开了一种美发营养保健粉,CN201510978849公开了一种无添加剂含木耳养发乌发健康食品。然而作为乌发、生发的保健食品或药物,易导致身体其他部位生长多余的毛发,低血压、心律不齐或心跳过快等。
本发明原料的功效如下。
中医认为,花生能够调和脾胃,补血止血,降压降脂,其中补血止血的作用主要就是花生外层红衣的功劳,花生红衣能够补脾胃之气,所能花生红衣能够达到养血止血的作用,即补气止血,达到生发、乌发的效果。
桑果:味甘、酸,性寒。归心、肝、肾经。可补肝益肾、养血,用于头晕、耳鸣、头发早白、脱发。桑树的成熟果实,其性味甘寒,入肺、肝、肾、大肠经。每100克鲜桑葚含水分81 .8克,蛋白质1 .8克,脂肪0 .3克。纤维素4 .9克,碳水化合物10克,灰分1 .2克,胡萝卜素30微克,硫胺素0 .02毫克、核黄素0 .06毫克,维生素 E6 .95毫克,钾33毫克,锌0 .27毫克,铜0.08毫克,硒4 .8微克。此外,还含有鞣酸,苹果酸,维生素C、脂肪酸、矢车菊素、矢车菊甙、乌发素等。中医学认为桑葚具有补益肝肾、滋阴养血、生津润肠、息风、改善皮肤(包括头皮)血液供应、营养肌肤、乌发明目等功效。主治心悸失眠、头晕目眩、须发早白、耳鸣、盗汗、瘰疬、关节不利等病症。
马齿苋,味酸、性寒,富含蛋白质、脂肪、多种维生素和氨基酸,还含有丰富的微量元素铜,而体内的铜离子是酪氨酸酶的重要组成部分,缺铜会导致黑色素的减少,马齿苋能够增加表皮中黑色素细胞和酪氨酸酶的活性,进而促进乌发的效果。
西红花,气为血之帅,血为气之母,气血相依,气行则血畅,气滞则血瘀,西红花具有活血化瘀的功效,利用西红花舒筋通络、活血化瘀的功效,与其他诸位成分配合使用,促进乌发的效果。
益智仁,辛温,归肾、脾经,具有暖肾固精的功效,与枸杞、桑果等配伍使用,增加补肝肾、益精血的效果。
紫草,归心、肝经,具有清热凉血、补中益气的功效,紫草具有较好地激活酪氨酸酶活性的作用,与花生红衣等配合使用实现较好的补气的功效。
葡萄糖酸锌,锌是头发是必不可少的微量元素,能使头发保持或再现其黑色,缺锌也是导致白发的原因。本发明在选择所述的药食同源类组分以达到补肝肾、益精血、补气等功效的同时,配伍使用葡萄糖酸锌,以促进乌发的效果。
枸杞:味甘,性平。归肝、肾、肺经。能滋阴补血、滋补肝肾、明目、润肺,增强免疫功能。
茯苓:味甘、淡,性平。归心、肺、脾、肾经。渗湿利水、健脾和胃、宁心安神,提高机体免疫力。
黑豆:味甘,性平;具有健脾利水,消肿下气,滋肾阴,润肺燥,制风热而活血解毒,止盗汗,生发黑发的功效。
枣具有补脾和胃,益气生津,养血安神,调营卫,解药毒。
黑芝麻:为胡麻科芝麻的黑色种子,含有丰富脂肪和蛋白质,以及糖类、维生素A、维生素E、卵磷脂、钙、铁、铬等营养成分及芝麻素(sesamin)、芝麻林酚素(sesamolin)、芝麻酚(sesamol)、胡麻甙(pedaliin)、车前糖(planteose)、芝麻糖(sesamose)、黑色素等特殊成分。另外黑芝麻脂肪中主要含有油酸、亚油酸、亚麻酸等人体必需脂肪酸。黑芝麻味甘、性平,具有滋补肝肾、益血润肠、通便、健胃、保肝、促进红细胞生长的作用。研究发现黑芝麻水提液能促使酪氨酸酶表达,提高黑色素的合成量,使白发转黑。
黑米性平、味甘,归脾、胃经,具有滋阴补肾、健脾开胃、养肝明目,益气补肺,活血补血、润肠等功效。现代医学研究证明黑米具有清除自由基、改善缺铁性贫血、抗应激反应以及免疫调节等多种生理功能;具有维持血管正常渗透压、减轻血管脆性、防止血管破裂、止血、抗菌,降血压、改善心肌营养、降低心肌耗氧量、抑制癌细胞生长等功效。
山药:为薯蓣科植物,山药的地下块茎。别名薯蓣、玉廷、修脆、儿草、九黄姜。性平、味甘。归肺、脾肾经。功能:健脾、补肺、固肾益精、滋润皮肤及毛发。
本发明以黑色食品和药食同源原料为主,达到生发乌发等目的。激活酪氨酸酶的活性以促进黑色素的产生,增加黑色素细胞酪氨酸酶和酪氨酸酶相关蛋白-1mRNA表达量。配伍合理,具有补益肝肾、健脾益肺、生发乌发、润肤养颜等功效;能够充分提高头皮的血流量;本产品能够激活头发毛囊,强健头皮育发;多种原料的配伍使用,能够促进细胞分裂,调节多种生物获活性物质,从而促进色素合成的作用,使萎缩的毛囊细胞恢复正常功能,促进乌发。

实施例2
该组合物由以下原料制成:黑豆40份、黑木耳60份、黑芝麻25份、黑米20份、核桃30份、花生红衣23份、红枣12份、桑果27份、枸杞14份、山药19份、百合20份、黄精20份、覆盆子15份、蛹虫草15份、阿胶7份、益智仁12份、茯苓25份、西红花10份、马齿苋10份、紫草50份、葡萄糖酸锌0.5份。
实施例3
该组合物由以下原料制成:黑豆50份、黑木耳45份、黑芝麻25份、黑米16份、核桃28份、花生红衣25份、红枣13份、桑果28份、枸杞12份、山药17份、百合18份、黄精18份、覆盆子20份、蛹虫草10份、阿胶5份、益智仁14份、茯苓26份、西红花13份、马齿苋13份、紫草45份、葡萄糖酸锌1份。
以上实施例制备方法为:
(1)将黑豆、黑芝麻、黑米、核桃烘干,烘干温度为85-95℃;然后将烘干后的原料进行粉碎,过100目筛,取筛下物,获得原料A;将阿胶在85-95℃温度下烘干,粉碎,将西红花直接粉碎。
(2)将花生红衣、红枣、桑果、枸杞、山药、百合、黄精、覆盆子、蛹虫草、益智仁、茯苓、马齿苋、紫草、黑木耳进行超声波处理,超声波温度为50℃,时间为40min,设置功率为150w进行;然后进行超微粉碎,通过300目筛,得原料B;
(3)将得到的原料A加水煎煮,煎煮时间为60min,将滤渣重复煎煮一次,合并得到煎煮液;将原料B加水煎煮,煎煮时间为40min,将滤渣重复煎煮2次,合并得到煎煮液;将原料A和B的煎煮液合并,加入葡萄糖酸锌和粉碎后的阿胶、西红花至合并后的煎煮液中并充分分散于煎煮液中,将煎煮液浓缩为稠膏,烘干干燥后粉碎,过150-200目筛网,得到原料C;
(4)将原料C按照常规压片方式得到片剂。
通过以下方式进行实验效果的验证
酪氨酸酶对黑色素的生成起着关键的作用,它的活性减低或丧失会使得头发的发干中缺乏黑色素而变成白发,因此酪氨酸酶的激活作用可以反映产品的乌发功效的强弱。
试验方法
产品准备 实施例1-3的保健食品组合物,粉碎加水,加热回流提取30min,过滤,残渣再加水100ml,加热回流30min,过滤,合并两次提取液,浓缩至0.1g/ml,用0.45μm滤膜过滤,滤液置于冰箱贮存备用。
酪氨酸酶活性测定
酪氨酸酶既能催化左旋酪氨酸生成左旋多巴( L-DOPA),又能催化左旋多巴氧化为多巴醌,多巴醌为红色,在475 nm 可以测定其吸光度值,通过其吸光度值的变化可反映出产品对蘑菇酪氨酸酶活性的影响。
溶液配制 配制左旋多巴(L-DOPA) 溶液:称取左旋多巴0.0034g加PBS 7ml,超声波促溶15min。配制蘑菇酪氨酸酶液:称取蘑菇酪氨酸酶0.0033g 加PBS 1.2ml。
测定方法 取配制好的溶液100μl加入蘑菇酪氨酸酶25μl(100U)混合,于30℃恒温培养箱中温育5min后加入150μl左旋多巴(0.15%),继续温育5min,立即测其吸光度值A450。实验重复3次。
产品对酪氨酸酶的激活率(%)=[(C-D)-(A-B)]/( A-B) ×100
其中A:未加产品样品的加酶混合液所测的吸光度;B:未加产品样品亦未加酶的混合液所测的吸光度;C:加产品样品和酶的混合液所测的吸光度;D:加产品样品而未加酶的混合液所测的吸光度。
实验结果 经验证,实施例1-3的激活率(%)分别为356.3±21.3、347.6±19.5、367.4±28.4。结果显示,实施例1-3具有较好的激活酪氨酸酶的活性,即本发明的组合物具有比较突出的乌发效果。
对比试验
对比例1
该组合物由以下原料制成:黑豆50份、黑木耳45份、黑芝麻25份、黑米16份、核桃28份、花生红衣25份、红枣13份、桑果28份、枸杞12份、山药17份、百合18份、黄精18份、覆盆子20份、蛹虫草10份、阿胶5份、茯苓26份、马齿苋13份、紫草45份、葡萄糖酸锌1份。
对比例2:
黑豆50份、黑木耳45份、黑芝麻25份、黑米16份、核桃28份、花生红衣25份、红枣13份、桑果28份、枸杞12份、山药17份、百合18份、黄精18份、覆盆子20份、蛹虫草10份、阿胶5份、益智仁14份、茯苓26份、西红花13份。
对比例1、2的制备方法(省略的组分相应地省去制备工艺)与实施例3相同,取与实施例3相同量的组合物,按照上述的试验方法测定对酪氨酸酶活性的激活影响。结果显示,对比例1、2的激活率(%)分别为269.1±18.4、251.2±20.4。由该结果可以看出,省略的这部分原料与其他的组分配伍具有相互促进的效果。相同用量的情况下,实施例3取得的效果要明显优于对比例1、2。
同时,选择实施例3的组合物进行人群测试,选择年龄在30-50岁年龄段的人群,该部分人群在受试前白发较多,尤其是头顶、鬓角较多的显现白发,选择100人,每日2片,连续服用30天、60天。根据回访显示,在30天时白发无继续增加的现象,60天时大部分白发转为黑色,无继续增加的白发。
由上述结果可以看出,本发明的组合物显示出比较优越的乌发的效果。
以上所述仅是本发明的优选实施方式,应当指出,对于本领域普通技术人员来说,在不脱离本发明原理的前提下,还可以作出若干改进和润饰,这些改进和润饰也视为本发明的保护范围。
Claims (7)
1.一种组合物,其特征在于包括由深色类食品原料和药食同源组分组成的组合物,深色类食品进一步地是指黑色类食品,可以是黑豆、黑木耳、黑芝麻、黑米、核桃中的一种或多种,该组合为具有乌发的功效和刺激头发的生长活性。
2.根据权利要求1所述的组合物,其特征在于深色类食品原料包括黑豆、黑木耳、黑芝麻、黑米、核桃。
3.一种具有乌发功效的组合物,由以下组分制成:黑豆20-50份、黑木耳25-60份、黑芝麻10-35份、黑米10-40份、核桃25-50份、花生红衣10-30份、红枣10-20份、桑果25-40份、枸杞10-20份、山药15-25份、百合10-30份、黄精15-25份、覆盆子15-30份、蛹虫草10-60份、阿胶5-15份、益智仁10-35份、茯苓25-40份、西红花10-20份、马齿苋10-20份、紫草40-60份、葡萄糖酸锌0.1-10份。
4.根据权利要求3所述的组合物,其特征在于组分配比为黑豆30-50份、黑木耳45-60份、黑芝麻20-35份、黑米10-30份、核桃25-40份、花生红衣20-30份、红枣10-15份、桑果25-30份、枸杞10-15份、山药15-20份、百合10-25份、黄精15-20份、覆盆子15-25份、蛹虫草10-15份、阿胶5-10份、益智仁10-30份、茯苓25-35份、西红花10-15份、马齿苋10-15份、紫草40-50份、葡萄糖酸锌0.1-8份。
5.权利要求3所述的组合物在制备具有改善乌发、生发效果的食品中的用途。
6.权利要求3所述的组合物,其特征在于包括有效量的可接受的添加剂,该组合物剂型可以是片剂、粉剂、丸剂、胶囊剂、膏剂、颗粒剂。
7.权利要求3所述的组合物的制备方法,(1)将黑豆、黑芝麻、黑米、核桃烘干,烘干温度为85-95℃;然后将烘干后的原料进行粉碎,过100-300目筛,取筛下物,获得原料A;将阿胶在85-95℃温度下烘干,粉碎,将西红花直接粉碎;
(2)将花生红衣、红枣、桑果、枸杞、山药、百合、黄精、覆盆子、蛹虫草、益智仁、茯苓、马齿苋、紫草、黑木耳进行超声波处理,超声波温度为45-55℃,时间为30-50min,设置功率为150w进行;然后进行超微粉碎,通过300目筛,得原料B;
(3)将得到的原料A加水煎煮,煎煮时间为30-90min,将滤渣重复煎煮一次,合并得到煎煮液;将原料B加水煎煮,煎煮时间为30-60min,将滤渣重复煎煮2次,合并得到煎煮液;将原料A和B的煎煮液合并,加入葡萄糖酸锌和粉碎后的阿胶、西红花至合并后的煎煮液中并充分分散于煎煮液中,将煎煮液浓缩为稠膏,烘干干燥后粉碎,过150-200目筛网,得到原料C;
(4)将原料C按照常规压片方式得到片剂。
