Abstract
本发明涉及Tie2对视网膜及其他组织中静脉血管的保护作用及应用,具体地,涉及Tie2调节化合物在制备用于治疗视网膜、皮肤、肝脏及肺等组织器官中静脉及相关血管病变的药物中的用途。本发明还涉及一种与视网膜等组织中静脉血管病变的程度密切相关的条件性酪氨酸激酶受体Tie2基因敲除非人动物模型和制备方法,及其在筛选Tie2调节化合物筛选中的用途。通过结合他莫昔酚不同剂量、及动物生长不同阶段进行处理,从而获得不同Tie2基因敲除效率的非人动物模型,以模拟不同程度的视网膜等组织中静脉血管病变,解决了视网膜等组织中静脉血管病变程度的可控性的难题。
Description
技术领域
本发明Tie2调节化合物在制备用于治疗视网膜、皮肤、及肝脏等组织器官中静脉及相关血管病变的药物中的用途。本发明还涉及一种非人转基因动物模型及其构建方法和应用,尤其是涉及一种由不同剂量他莫昔酚在动物不同生长阶段诱导的可控的视网膜及其他组织静脉血管病Tie2基因敲除转基因小鼠动物模型及其构建方法。
背景技术
静脉相关疾病包括静脉血栓、静脉曲张、与静脉炎症等。目前对于临床静脉血管病变的病理发生过程还了解较少,治疗手段也很有限。因此相关疾病模型的制备对于阐明静脉血管病变的发生机制、以及新药研发有重要的科学价值与应用意义。
小鼠视网膜血管生成在小鼠出生之后开始,因此用于静脉及其他血管生成及疾病发生模型较为可行。现有应用较广的小鼠视网膜模型包括氧诱导的血管增生型视网膜病变模型和糖尿病视网膜病变模型。但这两种模型的共同缺点在于不能有效调控血管异常的程度。而不同病变程度的可控性,对于药物的筛选、以及临床病理状况不一的准确模拟至关重要。
Tie2是促血管生成素家族(Angiopoietin 1-4)的共同受体,属于受体酪氨酸激酶家族成员。Tie2特异性表达在内皮细胞表面和某些造血祖细胞。Tie2介导的信号通路在胚胎期的血管发生和在成人阶段的血管新生中发挥重要作用。Tie2的主要生物学作用参与血管网络的生成、成熟与重塑;另外,Tie2介导的信号异常也参与多种疾病的病理发生过程,包括肿瘤转移、炎症、及视网膜病变等。
本发明利用Tie2条件性基因敲除小鼠模型,在小鼠出生后不同时间点诱导Tie2基因敲除,并结合基因敲除的不同剂量,以模拟静脉及相关血管的病理发生过程,并利用此疾病动物模型筛选静脉相关疾病的治疗药物。
本发明基于Cre-loxP系统建立的他莫昔芬诱导条件性基因敲除系统,利用条件性酪氨酸激酶受体Tie2基因敲除小鼠,在不同时间点和不同的方法诱导Tie2敲除,并分析Tie2的敲除效率与视网膜、皮肤、肝脏等组织器官中静脉及相关血管的表型,从而建立视网膜等组织中静脉血管病变与基因敲除剂量、以及敲除时间之间的关系,达到构建一种新型、病变程度可控的静脉血管疾病模型。
发明内容
本发明的目的之一在于提出了Tie2调节化合物在制备用于治疗视网膜、皮肤、肝脏、肺等组织器官中静脉及相关血管病变的药物中的用途。
在本发明的实施例中,所述Tie2调节化合物是Tie2受体激动剂或Tie2表达诱导剂。
在本发明的一些实施例中,所述受体激动剂包括激活Tie2的配体、多肽、抗体或小分子化合物。
在本发明的一个具体实施例中,所述配体是血管生成素。
在本发明的实施例中,所述Tie2表达诱导剂包括激活Tie2基因表达的酶、激素、生长因子、细胞因子或抗体。

本发明的目的之四在于提供了一种构建视网膜等组织中静脉血管病变的程度密切相关的条件性酪氨酸激酶受体Tie2基因敲除小鼠模型的方法,其中,该方法的具体制备步骤为:利用Cre-LoxP系统,用Tie2+/-小鼠与表达UBC-Cre/ERT2或VE-Cadherin-Cre/ERT2的转基因小鼠交配得到Tie2+/-;Cre/ERT2小鼠,再用所述Tie2+/-;Cre/ERT2小鼠与Tie2flox /flox小鼠交配分别得到基因型为Tie2flox/-;UBC-Cre/ERT2或Tie2flox/-;VE-Cadherin-Cre/ERT2、和Tie2flox/+;UBC-Cre/ERT2或Tie2flox/+;VE-Cadherin-Cre/ERT2的小鼠模型。
发明的有益效果
本发明的有益效果在于提出了Tie2调节化合物在制备用于治疗视网膜、皮肤、及肝脏等组织器官中静脉及相关血管病变的药物中的用途。本发明的有益效果还在于所建立小鼠视网膜血管病变模型的严重程度是可控的,即利用Tie2条件性基因敲除小鼠模型与表达CRE重组酶的转基因小鼠,制备双重转基因小鼠模型,利用他莫昔酚诱导该基因在不同时空即不同发育时间与不同组织细胞的基因敲除,并利用多种生物学方法分析Tie2的敲除效果、以及视网膜等组织中静脉血管的表型,从而建立视网膜等组织中静脉血管病变与基因敲除剂量、以及敲除时间之间的关系,达到构建一种新型、病变程度可控的静脉血管疾病模型。

本发明所用的术语“受体激动剂”是指对受体有较强亲和力和内在活性,能通过受体兴奋发挥最大效应的药物。例如,本发明中,“受体激动剂”是指对Tie2受体有较强亲和力和内在活性,并激活Tie2受体。

如本文中所使用,术语“非人转基因动物”由本领域已知技术获得,本发明的非人转基因的动物可以是任何非人动物,所述非人动物通过对原基因组进行遗传修饰,并受外源启动子控制进行Tie2突变型等位基因的条件性表达。
如本文中所使用,术语“启动子”指代能够结合哺乳动物细胞内的RNA聚合酶并且起始可操作地连接于其上的下游(3'方向)编码序列的转录的DNA调控区,其包括诱导型启动子,诸如cre-lox启动子的条件性活性启动子。
如本文中所使用,术语“诱导性基因敲除(iKO)”或“基因敲除诱导”指代利用控制Cre表达的启动子的活性或所表达的Cre酶活性具有可诱导的特点,通过对诱导剂给予时间的控制或利用Cre基因定位表达系统中载体的宿主细胞特异性和将该表达系统转移到动物体内的过程在时间上的可控性,从而在1oxP动物的一定发育阶段和一定组织细胞中实现对特定基因进行遗传修饰之目的的基因敲除技术。常见的几种诱导性类型(诱导剂)如下:四环素诱导型;干扰素诱导型;激素诱导型;腺病毒介导型。
如本文中所使用,术语“血管表型分析”指代静脉及相关血管的病变程度,采用血管的分子标记物蛋白标记视网膜等组织中静脉血管,可视观察血管形态变化。
下述实施例中的实验方法,如无特殊说明,均为常规方法。
具体实施例
实施例1条件性基因敲除小鼠模型的建立
用Tie2+/-小鼠与表达UBC-CreERT2或VE-Cadherin-Cre/ERT2的转基因小鼠交配得到Tie2+/-;UBC-Cre/ERT2或Tie2flox/-;VE-Cadherin-Cre/ERT2的小鼠,再用此小鼠与Tie2flox/flox小鼠交配得到Tie2flox/-;UBC-Cre/ERT2或Tie2flox/-;VE-Cadherin-Cre/ERT2(本发明中又被称为Tie2-/iKO),此基因型小鼠作为实验鼠,同时得到的Tie2flox/+;UBC-Cre/ERT2或Tie2flox/+;VE-Cadherin-Cre/ERT2作为对照鼠(Control)。
实施例2Tie2基因敲除并检测其敲除效率
将实施例1中获得的实验鼠和对照组分别进行以下实验,即在以下几个阶段诱导Tie2基因敲除:
(1)出生第1天(P1)到第4天(P4),连续4天每天向每只小鼠注射(胃内注射)60ug的他莫昔酚;
(2)出生第5天(P5)到第8天(P8),连续5天每天向每只小鼠注射(胃内注射)100ug的他莫昔酚;
将实施例1中的小鼠出生后进行诱导基因敲除,第7天分析肺组织。摘取经过诱导的转基因小鼠的肺组织,加入含有蛋白酶抑制剂的组织裂解液进行充分匀浆裂解,离心、得到的上清溶液、加入上样缓冲液并煮沸15分钟,随后进行聚丙烯酰胺蛋白电泳,转膜和5%脱脂牛奶封闭1小时。采用稀释度为1:200的山羊抗小鼠Tie2抗体(R&D,AF762)37℃孵育2小时,TBST洗涤三次后加入对应的HRP标记的二抗,37℃孵育1小时,加入显色底物,暗室曝光,Western Blot实验结果如图9示出,基因敲除的小鼠(每组4只小鼠)组织中Tie2的含量相比对照组都明显下调。同时利用免疫印记方法检测肺组织中的β-肌动蛋白作为上样内参,其中抗体浓度选择1:5000的小鼠单抗(C4,Santa Cruz sc-47778)。如图1A所示,相比对照组该条件敲除小鼠模型中Tie2蛋白表达量显著降低。
另一方面,取下小鼠眼球在4%多聚甲醛里于冰上固定2小时;剥离出眼球内的视网膜放于PBS中漂洗10分钟,3次;将视网膜放置于含3%(W/V)脱脂奶的0.3%PBS-Tx中封闭过夜;一抗溶于含3%(W/V)脱脂奶的0.3%PBS-Tx中4℃过夜;一抗分别为稀释度为1:400的山羊抗小鼠Tie2抗体(R&D,AF762)和稀释度为1:500的大鼠抗小鼠PECAM-1抗体(BDPharmigen)。之后0.3%PBS-Tx洗30分钟,6次;相应的荧光标记二抗溶于0.3%PBS-Tx中,滴加在片子上4℃过夜;0.3%PBS-Tx洗30分钟,6次;封片;共聚焦拍摄染色结果。如图1B所示,免疫荧光图结果显示相比对照组该条件敲除小鼠模型中Tie2表达量显著降低。
实施例3Tie2基因敲除并在视网膜组织中检测视网膜静脉血管病变
将实施例1中的小鼠出生后进行诱导基因敲除,第7天分析视网膜血管。取下小鼠眼球,通过免疫荧光观察到,Tie2-/iKO小鼠与该同窝对照小鼠相比,视网膜静脉前的血管生长显著增加(图2A),血管密度在静脉端和动脉端的定量结果如图2所示,静脉端周围血管的血管密度上升(图2B),而视网膜动脉端血管周围的血管密度没有变化(图2C),。并且我们还观察到Tie2-/iKO小鼠得视网膜静脉血管的形态异常、及静脉特征退化的变化,并伴随着静脉周围血管新生增加(用分子标记物Dll4表示毛细血管的新生)(图3A),而对照小鼠没有发生这些变化(图3B)。
实施例4不同时期视网膜静脉血管表型分析及激活Tie2对减弱视网膜静脉血管病变的程度
将小鼠按照以下条件诱导并分析不同时期的小鼠视网膜:
(1)P1-P4,每只小鼠每天注射60ug他莫昔酚,P21分析
(2)P5-P8,每只小鼠每天注射100ug他莫昔酚,P21分析
取下小鼠眼球在4%多聚甲醛里于冰上固定2小时;剥离出眼球内的视网膜放于PBS中漂洗10分钟,3次;将视网膜放置于含3%(W/V)脱脂奶的0.3%PBS-Tx中封闭过夜;一抗溶于含3%(W/V)脱脂奶的0.3%PBS-Tx中4℃过夜;0.3%PBS-Tx洗30分钟,6次;二抗溶于0.3%PBS-Tx中4℃过夜;0.3%PBS-Tx洗30分钟,6次;封片;共聚焦拍摄染色结果。
观察显微镜拍摄的结果发现不同时期诱导Tie2敲除可以导致视网膜静脉血管发生不同程度的血管病变(如图4-6所示)。从P1-P4对每个小鼠每天注射60ug他莫昔酚(胃内注射),分析P11、P15和P21的小鼠视网膜血管表型,发现P11时沿着视网膜静脉出现持续的异常血管生成(图4A),P15时视网膜静脉形态退化(图4B),P21时静脉血管变性伴随着血管束形成,即严重的血管瘤样病变(图4C)。
另外,P1-4诱导Tie2基因敲除2天后,小鼠(P6)通过肌肉注射重组腺病毒过表达Tie2配体血管生成素-1(Angiopoietin-1),可减弱视网膜静脉血管病变的程度(图5)。
从P5-P8对每个小鼠每天注射100ug他莫昔酚(胃内注射),分析P21的小鼠视网膜血管表型,发现在P5-8诱导Tie2基因敲除相比图6B所示的对照小鼠而言,也导致了视网膜静脉血管形成类似的缺陷,但血管瘤样病变的程度较轻(图6A),其中Tie2的敲除效率如图6C所示。
实施例5Tie2基因敲除并在肝脏组织中检测血管病变
将实施例1中的小鼠出生后1周后或成年(2月龄)进行诱导基因敲除,2-3周后分析肝脏组织。即采肝脏组织在4%多聚甲醛里于4度固定过夜,石蜡包埋、切片后利用H&E方法对切片组织进行染色。如图7所示,出生后诱导敲除Tie2导致肝脏血管病变性出血。
实施例6Tie2基因敲除并在肺组织中检测血管病变
实时定量RT-PCR实验结果显示,诱导敲除Tie2基因导致静脉血管标志物包括APJ(如图8B所示)与EphB4(如图8C所示)等的表达水平下降,而动脉或毛细血管内皮细胞标志物Dll4的表达水平上升(如图8D所示),其中Tie2基因敲除效率如图8A所示。
将小鼠中的肺组织取出,样品在离体后应迅速冷冻在液氮中,从液氮罐中取出样本后,把组织块放入已预冷的研钵中进行研磨,边研磨边加液氮,研磨到组织样品成粉末状后,在液氮基本挥发完时,在每个研钵中加Trizol试剂(Ambion)。再进一步匀浆。按照Trizol试剂的标准操作抽提总RNA,用反转录试剂盒(Thermo Scientific)将肝组织RNA反转录为cDNA,采用生物染料法荧光定量试剂盒(SYBR premix Ex Taq kit,TaKaRa)进行实时定量RT-PCR,荧光结果用ABI PRISM 7500进行分析。
其中引物序列为:
GAPDH:5'-GGTGAAGGTCGGTGTGAACG-3',5'-CTCGCTCCTGGAAGATGGTG-3';
Tie2:5'-GATTTTGGATTGTCCCGGGTCAAG-3',5-CACCAATATCTGGGCAAATGATGG-3。
APJ:5’-CAGTCTGAATGCGACTACGC-3',5'-CCATGACAGGCACAGCTAGA-3';
Ephb4:5'-CTGGATGGAGAACCCCTACA-3',5'-CCAGGTAGAAGCCAGCTTTG-3';
Dll4:5'-TGCCTGGGAAGTATCCTCAC-3',5'-GTGGCAATCACACACTCGTT-3'
实施例7Tie2基因敲除并在皮肤组织中检测静脉血管病变
将实施例1中的小鼠出生后进行诱导基因敲除,在成年鼠(2月龄及以上)分析耳皮肤血管。取小鼠耳皮肤组织,通过免疫荧光观察到,Tie2-/iKO小鼠(图9A)与该同窝对照小鼠(图9B)相比,皮肤静脉血管的形态变得曲折。
以上,基于本发明的实施方式进行了说明,但本发明不限定于此,本领域的技术人员应该明白,在本发明的主旨的范围内能够以进行变形和变更的方式实施,这样的变形和变更的方式,理应属于本发明的保护范围。
Claims (11)
1.一种制备组织静脉血管病变的程度密切相关的条件性酪氨酸激酶受体Tie2基因敲除小鼠模型及其后代的方法,其中,该方法的具体制备步骤为:
i)利用Cre-LoxP系统,用Tie2+/-小鼠与表达UBC-Cre/ERT2或VE-Cadherin-Cre/ERT2的转基因小鼠交配得到基因型为Tie2+/-;UBC-Cre/ERT2或Tie2+/-;VE-Cadherin-Cre/ERT2小鼠;
ii)再用所述Tie2+/-;UBC-Cre/ERT2小鼠与Tie2flox/flox小鼠交配分别得到基因型为Tie2flox/-;UBC-Cre/ERT2小鼠和Tie2flox/+;UBC-Cre/ERT2小鼠;或
用所述Tie2+/-;VE-Cadherin-Cre/ERT2小鼠与Tie2flox/flox小鼠交配分别得到基因型为Tie2flox/-;VE-Cadherin-Cre/ERT2小鼠和Tie2flox/+;VE-Cadherin-Cre/ERT2小鼠模型;
iii)在小鼠出生1-8天或2周至成年,使用不同剂量的基因敲除诱导剂诱导小鼠,使得小鼠受外源启动子UBC或VE-Cadherin控制地进行Tie2等位基因的条件性敲除,制备严重静脉血管瘤样病变的模型或较轻静脉血管病变的模型;
其中Tie2flox/-;UBC-Cre/ERT2小鼠或Tie2flox/-;VE-Cadherin-Cre/ERT2小鼠为组织静脉血管病变的程度密切相关的条件性酪氨酸激酶受体Tie2基因敲除小鼠模型;Tie2flox/+;UBC-Cre/ERT2小鼠或Tie2flox/+;VE-Cadherin-Cre/ERT2小鼠为对照模型,
其中在步骤iii)中,在小鼠出生后1-4天,使用剂量为50-70μg/天的他莫昔酚诱导小鼠,获得的小鼠模型是静脉血管病变程度严重的模型;所述模型中视网膜静脉血管退化并伴随有血管新生的严重血管瘤样病变;
其中在步骤iii)中,在小鼠出生后5-8天,使用剂量为90-100μg/天的他莫昔酚诱导小鼠,获得的小鼠模型是静脉血管病变程度轻微异常的模型;所述模型中血管样病变程度较轻。
2.如权利要求1所述方法,其中所述组织静脉血管病变包括视网膜静脉血管病变、皮肤静脉血管病变、肝脏静脉血管与肝组织病变、肺静脉血管病变中的至少一种。
3.如权利要求2所述的方法,其中所述视网膜静脉血管病变包括糖尿病视网膜病变、湿性年龄相关黄斑变性、视网膜静脉阻塞或炎症、及早产儿视网膜病变中的至少一种。
4.如权利要求2所述的方法,其中所述皮肤静脉血管病变包括静脉曲张、静脉阻塞或炎症中的至少一种。
5.如权利要求2所述的方法,其中所述肝脏血管与肝组织病变包括肝脏静脉及相关血管出血性血管病变、静脉阻塞或炎症中的至少一种。
6.如权利要求2所述的方法,其中所述肺静脉血管病变包括肺静脉血栓、静脉炎症中的至少一种。
7.如权利要求1至6任一项所述的方法制备的小鼠模型及其后代在制备组织器官中不同程度血管病变模型中的用途。
8.如权利要求7所述的用途,其中所述组织器官为视网膜、皮肤、肝脏或肺。
9.如权利要求7所述的用途,其中所述血管为静脉血管。
10.如权利要求7所述的用途,其中组织器官中血管病变的模型为严重静脉血管瘤样病变的模型或较轻静脉血管病变的模型。
11.如权利要求1至6任一项所述的方法制备的小鼠及其后代在制备用于治疗视网膜静脉血管病变、皮肤静脉血管病变、肝脏血管病变或肺血管病变的药物的体内筛选、药效检测、疗效评估、病理学或行为监测的模型中的用途。
