Abstract
本发明公开一种毛钩藤碱的应用,将所述毛钩藤碱用于制备治疗肺癌的药物。本发明通过体外肺癌培养实验,结果证明毛钩藤碱对肺癌细胞具有浓度及时间依赖性的抑制其生长的作用,通过线粒体损伤途径诱导肺癌细胞的凋亡;裸鼠肺癌荷瘤模型实验表明,毛钩藤碱可抑制裸鼠肿瘤的生长;同时细胞与动物实验都表明毛钩藤碱毒性低。

本发明涉及医药领域,主要涉及毛钩藤碱的应用。
背景技术
肺癌是目前全世界范围内发病率、病死率最高的恶性肿瘤之一。目前临床上药物的治疗主要是采用化疗及外科手术治疗,但对于该疾病控制和治疗还依赖于更多有效药物的发现。传统的化疗药物是细胞毒性药物,在杀伤肿瘤细胞的同时也作用于正常细胞,因此毒副作用大。线粒体是细胞产生能量的主要细胞器,机体内肿瘤细胞的异常增殖依赖于高能量的获取,因此通过选择性地诱导肿瘤细胞线粒体损伤导致细胞凋亡,抑制肿瘤的生长是治疗肿瘤最直接、最有效的途径。
毛钩藤碱是一种钩藤属植物中的生物碱成分,现有研究表明其在心血管保护、抗高血压、抗心律失常方面具有很好的作用。至今,尚未见毛钩藤碱在通过线粒体损伤途径诱导肺癌细胞的凋亡,进而抑制肿瘤生长的报道。
发明内容
鉴于上述现有技术的不足,本发明的目的在于提供一种毛钩藤碱的应用,具体在于毛钩藤碱通过线粒体损伤途径诱导肺癌细胞的凋亡,进而对肿瘤产生抑制作用,旨在提供毛钩藤碱在肿瘤治疗方面的新用途。

一种毛钩藤碱的应用,其中,将所述毛钩藤碱用于制备治疗肺癌的药物。
所述的毛钩藤碱的应用,其中,所述药物的剂型为注射剂型。
所述的毛钩藤碱的应用,其中,所述药物中还包括药物辅料,药物辅料为能溶解难溶性药物的药物辅料。
所述的毛钩藤碱的应用,其中,所述药物辅料为包括丙二醇、β环糊精中的一种或几种。
所述的毛钩藤碱的应用,其中,注射剂的剂量为1~3mg·kg-1/day。
有益效果:本发明中提供了毛钩藤碱及其制剂在肺癌治疗方面的新用途,毛钩藤碱为一种副作用少,毒性低的天然抗肺癌药物。本发明通过体外肺癌培养实验,结果证明毛钩藤碱对肺癌细胞具有浓度及时间依赖性抑制其生长的作用,可通过诱导细胞线粒体损伤途径促进肺癌细胞凋亡;裸鼠肺癌荷瘤模型实验表明,毛钩藤碱可抑制裸鼠肿瘤的生长;同时细胞与动物实验都表明毛钩藤碱毒性低。所述的毛钩藤碱可作为治疗肺癌的新候选药物。

本发明中通过体外肺癌细胞培养实验,结果证明毛钩藤碱对肺癌细胞具有浓度及时间依赖性抑制其生长的作用,可通过线粒体损伤途径诱导肺癌细胞的凋亡;裸鼠肺癌荷瘤模型实验表明,毛钩藤碱可抑制裸鼠肿瘤的生长;同时细胞与动物实验都表明毛钩藤碱毒性低。因此,在本发明中提供毛钩藤碱的应用,将所述毛钩藤碱用于制备治疗肺癌的药物。进一步地,所述药物的剂型为注射剂型。所述药物中还包括药物辅料,药物辅料为能溶解难溶性药物的辅料,具体可以包括由丙二醇、β环糊精等中的一种或几种组成。注射剂的浓度,换算到人体应为D=1~3 mg·kg-1/day。
以下通过实施例对本发明作进一步说明。
实施例1:毛钩藤碱可抑制肿瘤细胞的生长
1.细胞培养
人肺癌A549细胞培养在DMEM+10%胎牛血清(FBS)完全培养基中进行。细胞培养在温度37℃、5%浓度的CO2及饱和湿度的细胞培养箱中。
2.实验方法及结果
将A549肿瘤细胞系状态稳定后,将细胞铺于96孔板中,每孔5000个细胞,每孔含完全培养基90μL。人肺癌A549细胞按以下方式分组并处理:
(1)对照组:完全培养基溶液10 μL处理;
(2)毛钩藤碱组:用DMSO溶解配制成不同的浓度,每孔中加10μL,使培养基的终浓度为100、80、60、40、20、0 μM(μmol/L)的毛钩藤碱溶液,处理不同时间(1、3、6、9、12、24 h)后,每孔中加入20μL磷酸盐缓冲溶液(PBS溶液)配制的5mg/mL的噻唑蓝(MTT)溶液继续培养4小时。吸走上层培养基,每孔中加入150μL 二甲亚砜(DMSO)进行溶解,振荡器上溶解10分钟后,用酶标仪在A570处检测吸光度值,细胞存活率根据以下公式计算:
细胞存活百分率=(处理组吸光度-空白孔吸光度)÷(对照组吸光度-空白孔吸光度)×100%。
实验结果见图1、图2和图3。图1中,纵坐标表示细胞存活率,即处理组相对于对照组的细胞存活率;图1横坐标为不同浓度的毛钩藤碱处理24h后的细胞平均存活率。图2中,纵坐标表示细胞存活率,横坐标为毛钩藤碱80μM处理作用不同时间。图柱对应的值为各组的平均值。图3为毛钩藤碱80μM处理作用不同时间人肺癌A549细胞的形态图。
从图1中可见,随着毛钩藤碱药物浓度的增加,肺癌细胞的生存率降低;图2中可见,随着作用时间的增加,肺癌细胞的生存率也降低。表明毛钩藤碱对肺癌A549细胞的作用成浓度和时间依赖性。
同时在图3中,还能直观地观察到,随着毛钩藤碱对肺癌细胞作用时间增长,人肺癌A549细胞的形态发生了明显的变化,24h后成聚集抱团生长的细胞分散生长,细胞完整形态消失。
实施例2:毛钩藤碱可诱导肿瘤细胞凋亡
1.细胞培养:同实施例1。
2.实验方法及结果
将A549肿瘤细胞系状态稳定后,将细胞铺于6孔板中,每孔100000个细胞,每孔含完全培养基2mL。按以下方式分组并处理:
(1)对照组:完全培养基溶液10 μL处理;
(2)毛钩藤碱组:用DMSO溶解配制成不同的浓度,每孔中加10μL,使培养基的终浓度为100、80、60、40、20、0 μM(μmol/L)的毛钩藤碱溶液,处理不同时间(1、3、6、9、12、24h)后,根据需要进行不同操作。
采用流式细胞技术检测不同浓度及不同作用时间后细胞的凋亡情况,收集细胞、PBS漂洗、加入Annexin V-FITC及碘化丙啶避光孵育15min,流式细胞仪检测;同时采用蛋白免疫印迹技术检测不同浓度及不同作用时间后对细胞凋亡蛋白的影响,细胞刮收集细胞、裂解细胞、测蛋白浓度,将得到的已知蛋白浓度的样品进行电泳、转膜、封闭、一抗孵育、洗膜、二抗孵育、显影、定影拍照。
图4为不同浓度的毛钩藤碱处理细胞24h的细胞凋亡率,图5为毛钩藤碱80μM处理不同时间细胞凋亡率,图6为不同浓度的毛钩藤碱处理细胞24h的细胞凋亡蛋白的表达,图7为毛钩藤碱80μM处理不同时间细胞相关凋亡蛋白的表达。从图4、图5、图6及图7可以清楚的观察到,随着毛钩藤碱作用浓度和作用时间的增加,细胞的凋亡数量增加,凋亡蛋白C-PARP、C-Caspase-3的表达量也显著增加。这就从不同的手段进一步验证毛钩藤碱可浓度和时间依赖性增加凋亡蛋白的表达,促进细胞凋亡,从而降低肺癌细胞A549的生存率。
实施例3:毛钩藤碱可能通过线粒体损伤途径诱导细胞凋亡
1.细胞培养:同实施例1。
2.实验方法及结果
将A549肿瘤细胞系状态稳定后,将细胞铺于6孔板中,每孔100000个细胞,每孔含完全培养基2mL。按以下方式分组并处理:
(1)对照组:完全培养基溶液10 μL处理;
(2)毛钩藤碱组:用DMSO溶解配制成不同的浓度,每孔中加10μL,使培养基的终浓度为100、80、60、40、20、0 μM(μmol/L)的毛钩藤碱溶液,处理24h后,收集细胞,采用不同方式检测细胞线粒体的损伤情况,具体包括luminometer荧光分光光度计检测细胞ATP水平、流式细胞技术检测线粒体膜电位检测试剂盒(JC-1)检测细胞膜电位的变化、WesternBlot检测细胞浆中线粒体释放的细胞色素C的含量。相关实验步骤见试剂盒说明书。
细胞ATP水平实验结果见图8所示,图中可以看到随着毛钩藤碱浓度的增加,细胞ATP水平梯度降低,与对照组存在显著性差异。表明可能存在细胞线粒体的功能损伤或下降。
细胞线粒体膜电位结果如图9所示,随着毛钩藤碱浓度的增加,线粒体膜电位降低,与对照组存在显著性差异,同时也证明毛钩藤碱作用细胞后可能导致细胞线粒体损伤。
WesternBlot检测细胞浆中线粒体释放的细胞色素C的含量如图10所示。在正常细胞中,细胞色素C存在于线粒体中,当线粒体损伤后,细胞色素C将释放到细胞浆中。从实验结果可以看出,随着毛钩藤碱作用浓度的增加,细胞浆中细胞色素C的表达量增加,进一步表明随着毛钩藤碱作用浓度的增加,细胞线粒体的损伤严重。
实验从线粒体能量、膜电位的变化以及蛋白的表达不同的角度阐明了毛钩藤碱对肺癌A549细胞的抑制作用机理,可能是通过细胞线粒体损伤,诱导细胞凋亡。
实施例4:毛钩藤碱可选择性抑制肺癌细胞的生长,对人正常肝细胞及胚肺细胞作用小
1.细胞培养:同实施例1。另外,还包括肝人正常细胞LO2及人胚肺细胞WI38的培养。
2.实验方法及结果
待A549肿瘤细胞、LO2人正常肝细胞、WI38人胚肺细胞系状态稳定后,将细胞铺于6孔板中,每孔100000个细胞,每孔含完全培养基2 mL。按以下方式分组并处理:
(1)对照组:完全培养基溶液10μL处理;
(2)毛钩藤碱组:DMSO溶解,每孔中加10μL,使培养基的终浓度为80μM的毛钩藤碱溶液,流式细胞处理技术检测细胞的凋亡,结果见图11。
图11结果可见,毛钩藤碱能选择性的促进肿瘤细胞的凋亡,对正常肝细胞及胚肺细胞的影响较小,结果表明毛钩藤碱的毒副作用小。
为了更进一步证明毛钩藤碱对肺癌的作用,在细胞实验的基础上,通过裸鼠移植瘤模型实验从动物层面证明毛钩藤碱对肺癌的作用。
实施例5:毛钩藤碱可抑制裸鼠移植瘤模型肿瘤的生长
在本实验过程中,通过向成功移植肿瘤裸鼠腹腔注射毛钩藤碱,观察毛钩藤碱对肿瘤的抑制作用。剂量换算根据首先是根据中国医科大学肿瘤研究所刘晓东等人《体外测试肿瘤化敏感性指标的研究》换算出细胞与人临床剂量的关系,得到100μM的体外毛钩藤碱细胞作用浓度相当于毛钩藤碱人临床剂量D=1.5 mg·kg-1/day。
其后根据美国食品药品监督管理局(FDA)在《药物临床实验中健康成人受试对象安全给药剂量的估算》(《Estimating the Safe Starting Dose in Clinical Trials forTherapeutics in Adult Healthy Volunteers》)中给出的,达到相同生物学效应所需的人体与小鼠(即实验验证中的裸鼠)的注射剂量换算比(1∶12.3)。再依此换算比,对照具体实施方式二中所记载的剂量来换算出实验用裸鼠的用药剂量,即D小鼠≈19mg·kg-1
1.裸鼠的选择
取6~8 周龄无特异病原微生物(SPF)级裸鼠6只,体重范围为20±2g,在昼夜交替、室温(20~23℃)环境饲养,自由饮水、进食。
2.实验分组及给药剂量(共2组)
所有裸鼠随机分为两组后,每只右大腿外侧接种1×107个/ml人肺癌A549细胞所制悬液0.2mL,待瘤体形成后开始按以下方式给药。
(1)对照组:与处理组体积相同的生理盐水腹腔注射,1次/日;
(2)毛钩藤碱组:按裸鼠体重19 mg/kg的剂量,腹腔注射毛钩藤碱,1次/日(按照0.1ml/10g腹腔注射,配制成1.9mg/ml,所用的溶剂可用作注射剂使用并且可溶解难溶性药物的辅料)。
3.测量与记录
(1)每7天后测量、记录一次各组裸鼠的体重;
(2)每7天后用游标卡尺测量肿瘤的长度和宽度、记录一次各组裸鼠肿瘤体部分的体积;肿瘤体积按照以下公式计算:
肿瘤体积(mm3)=0.5×肿瘤长度(mm)×肿瘤宽度的平方(mm2)。
体重记录结果如图12所示,纵坐标表示各组裸鼠的平均体重;横坐标表示测量时间点;各种线条节点表示各处理组;各点的数值为各组平均体重。由图12可见,毛钩藤碱组裸鼠体重与对照组比较未见显著差异。表明毛钩藤碱对裸鼠的毒性小,不会影响裸鼠的正常生长,这与前面细胞实验的结果一致。
肿瘤体积记录结果如图13所示,纵坐标表示各组裸鼠的肿瘤体积;横坐标表示测量时间点;各种线条节点表示各处理组;各点的数值为各组肿瘤体积的平均值。结果表明毛钩藤碱可抑制肿瘤的生长。
图14为给药6周处死剥离肿瘤后的实物图。图14更加直观地观察到:毛钩藤碱组与对照组相比较,毛钩藤碱组肿瘤体积在治疗4周后显著低于对照组。6周后,剥离肿瘤组织可见肿瘤体积明显减小。
在上述实施例1~5中,为证明上述验证结论是准确而可靠的,验证时同时还对样本进行了统计学的计算与分析。图中“**”表示p<0.01,“*”表示p≤0.05。其中“p”为统计学上可接受错误的边界符号,其值为p≤0.05。在计算出p<0.01的情况下,即可认定上述结果之间的差异出现错误概率小,同时说明了验证结果是准确而可靠的。
综上,通过体外肺癌培养实验,结果证明毛钩藤碱可通过线粒体损伤途径浓度及时间依赖性诱导肺癌细胞的凋亡,抑制肿瘤细胞的生长;裸鼠肺癌荷瘤模型实验表明,毛钩藤碱可抑制裸鼠肿瘤的生长;同时细胞与动物实验都表明毛钩藤碱毒性低。因此,毛钩藤碱可作为治疗肺癌的新候选药物。
应当理解的是,本发明的应用不限于上述的举例,对本领域普通技术人员来说,可以根据上述说明加以改进或变换,所有这些改进和变换都应属于本发明所附权利要求的保护范围。

1.一种毛钩藤碱的应用,其特征在于,将所述毛钩藤碱用于制备治疗肺癌的药物。
2.根据权利要求1所述的毛钩藤碱的应用,其特征在于,所述药物的剂型为注射剂型。
3.根据权利要求2所述的毛钩藤碱的应用,其特征在于,所述药物中还包括药物辅料,药物辅料为能溶解难溶性药物的药物辅料。
4.根据权利要求3所述的毛钩藤碱的应用,其特征在于,所述药物辅料为包括丙二醇、β环糊精中的一种或几种。
5.根据权利要求2所述的毛钩藤碱的应用,其特征在于,注射剂的剂量为1~3 mg·kg-1·day-1。
