本发明公开了一种无血清纤维芽母细胞培养基,配方包括:L‑精氨酸、L‑门冬酰胺、L‑门冬氨酸、L‑胱氨酸二盐酸盐、L‑谷氨酸、甘氨酸、L‑组氨酸、L‑羟脯氨酸、L‑亮氨酸、L‑赖氨酸盐酸盐、L‑甲硫氨酸、L‑苯丙氨酸、L‑脯氨酸、L‑丝氨酸、L‑苏氨酸、L‑色氨酸、L‑酪氨酸、L‑缬氨酸等共计50种药物。本培养基中没有加入动物血清,这样大大提高了细胞在培养过程中的稳定性,使得培养的细胞扩增倍数较高,同时增加了细胞活率。本培养基还是目前国际上独家拥有的皮肤纤维芽母细胞培养基,使纤维芽母细胞加快合成和分泌弹性纤维和胶原蛋白,增加皮肤的张力进而填平皱纹。

技术领域
本发明涉及一种较大范围,具体是一种无血清纤维芽母细胞培养基。
背景技术
培养基(Medium)是供微生物、植物组织和动物组织生长繁殖盒合成代谢产物和维持用的人工配制的按一定比例配制的养料,一般都含有碳水化合物、含氮物质、无机盐(包括微量元素)以及维生素和水等。不同培养基可根据实际需要,添加一些自身无法合成的化合物,即生长因子。培养基的要求有:营养物质的成分比例合适,组成比较丰富,浓度恰当,能够满足菌种发芽和生长繁殖成有生理功能的菌丝体的需要,更重要的是能显示出产物合成的潜力;在一定条件下,各组分彼此间不产生化学反反应,理化性质相对稳定;粘度适中,具有适当的渗透压;要考虑所选用的原材料的品种和浓度与代谢产物生物合成过程中的调节关系,要利于主要产物的生物合成并能维持较长时间的最高生产速率;生产过程中既不影响通气和搅效果,又不影响产物的分离精制和废物处理。
不过大部分培养基中都要添加动物血清,这样就会降低细胞在培养过程中的稳定性。培养的细胞扩增倍数会比较低,同时会减少细胞存活率。
发明内容
本发明的目的在于提供一种无血清纤维芽母细胞培养基,以解决上述背景技术中提出的问题。
为实现上述目的,本发明提供如下技术方案:
一种无血清纤维芽母细胞培养基,配方包括:L-精氨酸、L-门冬酰胺、L-门冬氨酸、L-胱氨酸二盐酸盐、L-谷氨酸、甘氨酸、L-组氨酸、L-羟脯氨酸、L-亮氨酸、L-赖氨酸盐酸盐、L-甲硫氨酸、L-苯丙氨酸、L-脯氨酸、L-丝氨酸、L-苏氨酸、L-色氨酸、L-酪氨酸、L-缬氨酸、L-谷氨酰胺、丁二酸、丙酮酸钠、对氨基苯甲酸、硝酸钙、硝酸钾、硝酸铵、无水硫酸镁、无水磷酸二氢钠、碳酸氢钠、氯化钾、氯化钠、氯化镁、无水氯化钙、葡萄糖、核糖、还原谷胱甘肽、酚红、生物素、D-泛酸钙、叶酸、i-肌醇、烟酰胺、氯化胆碱、盐酸吡哆醇、核黄素盐酸、酒石酸胆碱、硫胺素、维生素B12、2-巯基乙醇、铁、钴。
与现有技术相比,本发明的有益效果是:本培养基中没有加入动物血清,这样大大提高了细胞在培养过程中的稳定性,使得培养的细胞扩增倍数较高,同时增加了细胞活率。本培养基还是目前国际上独家拥有的皮肤纤维芽母细胞培养基,使纤维芽母细胞加快合成和分泌弹性纤维和胶原蛋白,增加皮肤的张力进而填平皱纹。
