Abstract

本发明公开了一种利用PericoannosinA作为抗菌组分的牙膏,包括摩擦剂、保湿剂、粘合剂及发泡剂,牙膏中添加有抗菌组合物,所述的抗菌组合物含有化合物PericoannosinA。本发明的牙膏具有抗菌物,抗菌物由PericoannosinA与雅姆皂苷元混合组成,试验证实牙膏中含有这一抗菌组合物有着很好的抑菌作用,特别是最口腔中常见的四种病菌具有很好的抑制作用。

技术领域
本发明涉及一种利用PericoannosinA作为抗菌组分的牙膏。
背景技术
口腔疾病是影响人体健康的常见病和多发病。常见的口腔疾病主要有龋病、牙龈炎、牙周病以及口腔溃疡等。WHO把龋齿列为在世界内应重点防治的3大慢性非传染性疾病之一,排在心脑血管病和癌症之后的第3位。这表明慢性非传染性疾病对人群健康的危害性已引起人类的关注,口腔疾病的防治是未来生命科学研究的重要课题之一。大部分口腔疾病及口腔问题,主要是由口腔中存在的致病菌引起的。口腔细菌摄取糖后所产生的酸能够导致蛀牙;由口腔细菌所产生的毒素可导致牙龈出血;此外,这些细菌的代谢废物还可导致牙结石及口臭等。因此,控制口腔致病菌是预防口腔疾病的主要任务。口腔疾病大多起病隐匿,早期症状不明显,常常被忽略或被错误判断为“上火”等其他问题。而且,消费者对口腔保健的意识还比较薄弱,在口腔问题未发展到非常严重的程度之前,普遍不会太重视,不会去医院进行医治,而可能仅是自行吃一些抗生素或清火的药物,以缓解症状。事实上,如果通过良好和有效的口腔清洁卫生习惯,可大大避免或减少口腔问题的发生。而其中最简便和最直接的方法就是使用牙膏进行口腔清洁,清除口腔中有害的细菌,并控制其再生。
PericoannosinA,a Polyketide Synthase-Nonribosomal Peptide SynthetaseHybrid Metabolite with New Carbon Skeleton from the Endophytic FungusPericonia sp.Org.Lett.,2015,17(17),4304-4307。PericoannosinA具有全新骨架,具有很好的抗HIV活性,申请人发现PericoannosinA与雅姆皂苷元在一定比例下制备的牙膏中具有很好抑制口腔细菌的效果。
发明内容
针对上述现有技术存在的问题,本发明提供一种利用PericoannosinA作为抗菌组分的牙膏,很好抑制口腔细菌的作用。
为了实现上述目的,本发明采用的技术方案是:一种利用PericoannosinA作为抗菌组分的牙膏,包括摩擦剂、保湿剂、粘合剂及发泡剂,牙膏中添加有抗菌组合物,所述的抗菌组合物含有化合物PericoannosinA,所述的PericoannosinA化学结构式如下:
所述的抗菌组合物还含有雅姆皂苷元。
所述的抗菌组合物中,PericoannosinA与雅姆皂苷元重量份数之比为1∶1-20∶1。
所述的摩擦剂为碳酸钙、磷酸氢钙、二氧化硅中一种或几种,保湿剂为山梨醇、甘油、丙二醇中的一种或几种;粘合剂为羧甲基纤维素钠、黄原胶、羟乙基纤维素、瓜尔胶、卡拉胶中的一种或几种;发泡剂为十二烷基硫酸钠。
所述的各组分的重量百分比将抗菌组合物、摩擦剂、保湿剂、粘合剂及发泡剂混合,通过搅拌、真空脱气得牙膏膏体。
所述的质量比分别为,摩擦剂15%~50%;保湿剂15%~55%;粘合剂0.5%~2.0%;发泡剂2%~3%;抗菌组合物0.1%~2%;其余为去离子水。
本发明的牙膏具有抗菌物,抗菌物由PericoannosinA与雅姆皂苷元混合组成,试验证实牙膏中含有这一抗菌物有着很好的抑菌作用,特别是最口腔中常见的四种病菌具有很好的抑制作用,其PericoannosinA与雅姆皂苷元在7-10∶1的比例下,其抑菌环直径显著增大,可见对于口腔病菌具有很好的抑制作用。

雅姆皂苷元,CAS号:512-06-1。
本发明所涉及化合物PericoannosinA的制备方法参见文献:PericoannosinA,aPolyketide Synthase-Nonribosomal Peptide Synthetase Hybrid Metabolite withNew Carbon Skeleton from the Endophytic Fungus Periconia sp.Org.Lett.,2015,17(17),4304-4307。
本发明涉及的比例均为重量比。
实施例1
二氧化硅32%;山梨醇21%;丙二醇12%;瓜尔胶1%;十二烷基硫酸钠3%,抗菌组合物1%,余量去离子水。
实施例2
磷酸氢钙25%;丙二醇30%;羟乙基纤维素1%;十二烷基硫酸钠3%,抗菌组合物1%,余量去离子水。
实施例3
碳酸钙30%;山梨醇30%;羧甲基纤维素钠1%;十二烷基硫酸钠3%,抗菌组合物0.8%,余量去离子水。
实施例4
二氧化硅35%;甘油20%;卡拉胶1%;十二烷基硫酸钠3%,抗菌组合物2%,余量去离子水。
以实施例1的组分为标准制备牙膏,通过添加不同比例的PericoannosinA与雅姆皂苷元,对各组的牙膏进行抑菌环试验,以确定牙膏对4种主要的口腔致病菌的抑制能力。
4.3牙膏抑菌环试验
1)原理
利用抑菌剂不断溶解,经琼脂扩散形成不同浓度梯度,以显示其抑菌作用。
2)试验菌株
牙龈卟啉单胞菌菌悬液、变形链球菌菌悬液、中间普氏菌菌悬液和具核梭杆菌菌悬液。
3)实验牙膏
抗菌组合物为不同比例的PericoannosinA与雅姆皂苷元,利用实施例1组分制备牙膏,。
4)试验方法
抑菌片的制备:采用牙膏与水1∶3稀释成均匀牙膏浆后,取无菌并干燥的滤纸片。每片滴加牙膏20μL,然后将滤纸片平放于清洁的无菌平皿内,开盖置温箱37℃中烤干,或置室温下自然干燥后备用。②阴性对照样片的制备:取无抑菌活性成分或无显著抑菌效果的牙膏与水1∶3稀释成均匀牙膏浆后,取无菌并干燥的滤纸片。每片滴加牙膏20μL,然后将滤纸片平放于清洁的无菌平皿内,开盖置温箱37℃中烤干,或置室温下自然干燥后备用。③试验菌的接种及抑菌剂样片贴放:参照《消毒技术规范(2002年版)》“抗(抑)菌试验”。④测量抑菌环时,应选均匀而完全无菌生长的抑菌环进行。测量其直径应以抑菌环外沿为界5)评价规定抑菌作用的判断:抑菌环直径大于7mm者,且试验组的抑菌环显著大于阴性对照组,判为有抑菌作用。抑菌环直径小于或等于7mm者,判为无抑菌作用。3次重复试验均有抑菌作用结果者,判为合格。空白组是不含有抗菌组合物的实施例1牙膏。
试验结果
空白牙膏的抑菌试验结果表明,空白牙膏对4种口腔常见致病菌的抑菌环平均直径均不大于7mm,即不具有抑菌作用。

上述实验数据表明,PericoannosinA与雅姆皂苷元在7-10∶1的比例下,其制备的牙膏抑菌环直径显著增大。显著高于其他比例组以及空白组,具有显著的抑菌效果,可见其在PericoannosinA与雅姆皂苷元在7-10∶1的比例下制备的牙膏,协同抑菌效果明显。

Claims (6)

1.一种利用PericoannosinA作为抗菌组分的牙膏,包括摩擦剂、保湿剂、粘合剂及发泡剂,其特征在于牙膏中添加有抗菌组合物,所述的抗菌组合物含有化合物PericoannosinA,所述的PericoannosinA化学结构式如下:
2.根据权利要求1所述的一种利用PericoannosinA作为抗菌组分的牙膏,所述的抗菌组合物还含有雅姆皂苷元。
3.根据权利要求1所述的一种利用PericoannosinA作为抗菌组分的牙膏,所述的抗菌组合物中,PericoannosinA与雅姆皂苷元重量份数之比为1∶1-20∶1。
4.根据权利要求1所述的一种利用PericoannosinA作为抗菌组分的牙膏,其特征在于,所述的摩擦剂为碳酸钙、磷酸氢钙、二氧化硅中一种或几种,保湿剂为山梨醇、甘油、丙二醇中的一种或几种;粘合剂为羧甲基纤维素钠、黄原胶、羟乙基纤维素、瓜尔胶、卡拉胶中的一种或几种;发泡剂为十二烷基硫酸钠。
5.根据权利要求1所述的一种利用PericoannosinA作为抗菌组分的牙膏,其特征在于,所述的各组分的重量百分比将抗菌组合物、摩擦剂、保湿剂、粘合剂及发泡剂混合,通过搅拌、真空脱气得牙膏膏体。
6.根据权利要求1所述的一种利用PericoannosinA作为抗菌组分的牙膏,其特征在于,所述的质量比分别为,摩擦剂15%~50%;保湿剂15%~55%;粘合剂0.5%~2.0%;发泡剂2%~3%;抗菌组合物0.1%~2%;其余为去离子水。
