背景
已完善确立了研磨剂在口腔护理组合物中的使用。除清洁牙斑和污渍之外,口腔护理组合物(诸如洁牙剂)中的研磨剂还可以磨光牙釉质的表面。磨光牙釉质具有许多美容益处,包括增加牙齿的亮度和光亮、赋予牙齿光滑的质地、以及通过去除促使污渍附着的裂纹和粗糙表面来减少将来的变色。理想地,研磨剂或研磨剂的组合可提供牙釉质的清洁和磨光。
包含焦磷酸钙和二氧化硅的口腔护理组合物是本领域已知的(参见例如WO2008/157197、WO2009/134657和WO2010/068442)。然而,需要提供具有改进的磨光功能从而产生牙齿高光泽度或光亮而不明显增加牙釉质磨损的口腔护理组合物。
简要概述

本发明的口腔护理组合物还可含有调味剂和/或清新口气抗牙斑活性物。用于实践本发明的调味剂包括精油以及各种调味的醛、脂、醇和类似物质。精油的实例包括绿薄荷油、胡椒薄荷油、冬青油、黄樟油、丁香油、鼠尾草油、桉树油、马郁兰油、肉桂油、柠檬油、酸橙油、葡萄柚油和橙油。也适用的是诸如薄荷醇、香芹酮和茴香脑的化学物质。其中,最常使用的是胡椒薄荷油和绿薄荷油。口腔护理组合物可包含甜味剂,诸如糖精钠。
本发明的口腔护理组合物可包含一种或多种用于增加在刷涂口腔时产生的泡沫量的试剂。此类起泡剂是本领域技术人员已知的。增加泡沫量的试剂的说明性实例包括但不限于聚氧乙烯和某些聚合物,包括但不限于海藻酸盐聚合物。聚氧乙烯可增加泡沫量和由本发明的口腔护理载体组分产生的泡沫的厚度。聚氧乙烯通常也被称为聚乙二醇(″PEG″)或聚氧化乙烯。适于本发明的聚氧乙烯将具有200,000至7,000,000的分子量。在一个实施方案中,分子量将为600,000至2,000,000,且在另一个实施方案中将为800,000至1,000,000。口腔护理组合物中起泡剂的量可为0.01至0.9重量%、0.05至0.5重量%,且在另一个实施方案中可为0.1至0.2重量%。
本发明的口腔护理组合物可为洁牙剂(包括牙膏、牙粉和磨光糊剂(prophylaxispaste))或糖果(包括口香糖、珠粒和求斯糖)的形式。本发明的组合物不为薄膜。
组合物对于人,且通常其它动物的口腔使用是安全的。
如本文所用,术语“清洁”通常是指去除目标表面上的污物、污垢、杂质和异物。例如,在口腔表面的情形下,其中表面是牙釉质,清洁可去除至少一些膜或污渍,诸如牙斑生物膜、表膜或牙垢。如本文所用,“磨光”通常是指使表面更光滑和/或更有光泽的修整或精制过程。磨光和清洁也可提供表面的擦亮效果,其中发生污渍去除,例如,使牙齿表面增白。本发明中清洁和/或磨光和/或增白的口腔表面通常是牙釉质。
实施例
实施例1:1600下筛选方法的一般程序如下:
I.砂磨牛牙
选择无污渍的牛牙釉质样本,使用35粒度(grit)砂纸砂磨。在100rpm下砂磨牛牙釉质样本4分钟。砂磨后,将牛样本分类并选择具有大于4mm2的平面的那些样本。选择12颗牙齿用于研究。
II.准备牙齿
将牙齿浸于去离子水中的1%柠檬酸溶液3min。3min后用去离子水冲洗牙齿并拍干。使用热压化合物(thermal impression compound)在每个托盘中固定4颗牙齿。将6克含35%焦磷酸钙的无水牙膏涂覆于牙齿上并在每个托盘中加3克去离子水。使用重250克的牙刷以120下/min的速度刷涂牙齿1600下。冲洗牙齿上的牙膏浆,将牙齿从托盘移除,并拍干。用35%焦磷酸钙牙膏对照刷涂以使所有牙齿达到相同的磨光水平。然后在1%柠檬酸溶液中重复浸泡3min,用去离子水冲洗牙齿,并储存于去离子水中直到基线测量。
III.基线光泽度测量
按照制造商的说明书校准Novocurve光泽仪。将牙釉质样品从水中移除并拍干。然后将牙釉质表面集中在光泽仪上并在样本上做标记以明确定向。进行测量。旋转样品90°并进行测量。旋转样品90°并进行另一次测量。旋转样品90°并进行另一次测量。重复这种4次测量循环然后将样品放回到水中。取8个值的平均值以得到光泽度的平均基线值。
IV.产品评佑
使用热压化合物在每个托盘上重新固定4颗牙齿。将6克测试产品涂覆到牙齿上并在每个托盘中加3克去离子水。使用重250克的牙刷以120下/min的速度刷涂牙齿1600下。冲洗牙齿上的牙膏浆,将牙齿从托盘移除,并浸入水中。按照制造商的说明书校准Novocurve光泽仪。将牙釉质样品从水中移除并拍干。然后将牙釉质表面集中在光泽仪上并在样本上做标记以明确定向。进行测量。旋转样品90°并进行测量。旋转样品90°并进行另一次测量。旋转样品90°并进行另一次测量。重复这种4次测量循环然后将样品放回到水中。取将8个值的平均值以得到处理后的光泽度的平均值。报告处理后光泽度值和基线光泽度值的差异作为光泽度的变化。
V.循环
准备用下一产品进行测试所用的牙齿,重复酸蚀,用35%焦磷酸钙牙膏刷涂,最后酸蚀。此过程使牙齿恢复到统一的基线并使得下一测试产品可以在相同的牛样品上进行评估。

表2中的结果表明,特定量的calpyro结合特定量的高清洁性二氧化硅的选择提供优于(A)仅含高清洁性二氧化硅的牙膏和(E)仅含calpyro的牙膏的牙釉质的磨光效果。这些结果是意料之外的,因为用calpyro替换一定量的高清洁性二氧化硅会提供优于单独使用高清洁性二氧化硅或calpyro的协同效应是未预见到的。

广泛使用的牙釉质磨光潜能方法的一般程序如下:
切下五十六颗牛永久中切牙以获得5x5mm2的牙釉质样本。然后将这些牙釉质样本嵌入到自聚合的甲基丙烯酸酯树脂中以便仅暴露牙釉质表面。将这些块状物编号并用粘蜡以使其编号侧面对面的方式将其固定在Struers磨光盘上。用500粒度砂纸研磨底部,直到它完全平整。将这些样本从盘中移除并以牙釉质样本表面朝上的方式再次固定。使用500粒度新砂纸用5牛顿的压力将它们研磨10秒并用充足的D1水冲洗。然后将样本连续用1200-、2400-和4000-粒度砂纸各磨光20秒。用去离子水冲洗样本3分钟并晾干。通过使用磨光糊剂(pumice paste)(大约3g LPA/2ml蒸馏水)在安装于车床上的布制打磨机(cloth buffer)中历时5秒手动完成磨光/再磨光。然后将样本旋转90°并再磨光5秒。重复这个过程直到四个方向均被磨光且样本已被磨光共计20秒。
评分
使用Novo-Curve光泽仪对样本评分。将牙釉质表面集中在光泽仪上并根据线I、2、3和4在样本上做标记以对样本评分,然后将其旋转90°并再次评分。两次分数的平均值用于计算光泽仪数据。然后通过将样本在1%HCL(v/v)中脱钙2分钟将它们刻蚀,以提供黯面用于开始研究。重复光泽度测量。
程序
在基线(刻蚀)评分之后;将样本放置在横向刷涂机上。将刷涂张力调节至150克,用适当的洁牙剂浆(25克洁牙剂和40克去离子水)和中等刷子刷涂样本4,500下。然后将样本从刷涂机上移除,冲洗并再次针对磨光评分。然后刻蚀将再次使样本变暗且将整个过程重复额外次数以便在各牙齿组上测定每种产品。处理设计是改进的拉丁方设计,使得不会在处理之后始终接着另一处理。
计算
对每个样本计算基线分数和刷涂后分数之间的差异,且代表磨光增量。
研究1:4500下后的光泽度变化

(1)使用的高清洁性二氧化硅具有>85的PCR,5至15μm的平均粒度d50,和40至120cm3/100g二氧化硅的吸油值。
表4中的结果表明,包含12%高清洁性二氧化硅和10%Calpyro(焦磷酸钙)的组合物提供比包含22%高清洁性二氧化硅的组合物更好的牙釉质磨光效果。这些结果证实了筛选方法的结果。
研究2:4500下后的光泽度变化。

表5中的结果表明,10%高清洁性二氧化硅和10%焦磷酸钙的组合也提供比含有20%高清洁性二氧化硅的牙膏更好的磨光效果。将二氧化硅负载增加至12%进一步提高磨光性能。
通过标准方法测量牙釉质耐磨性。
样本制备
在由ADA概述的受控条件下使八(8)个人牙釉质样本经受中子撞击使得在样本内形成放射性磷(32P)。将样本固定在甲基丙烯酸甲酯中因此它们在V-8横向刷涂机中点燃。将样本刷涂5000下,用含有于50ml0.5%CMC甘油溶液中的10g ADA参照物质的浆液进行预处理。所用的刷子是由ADA指定的具有150g刷涂张力的那些。
程序
在预处理之后,使用以上参数(150g和5000下)在″三明治设计(sandwichdesign)″中进行测试。在用测试产品(25g产品/40ml水)刷涂前后,用ADA参考物质(10gCa2P2O7/50ml 0.5%CMC)刷涂各个牙齿组。重复此程序额外次数以便在各牙齿组上测定每种产品。处理设计是改进的拉丁方设计,使得不会在处理之后始终接着另一处理。
计算
取1ml样品,称重(约lg),并加入5ml“Ultima Gold”闪烁混合物中。将样品充分混合并立即置于液体闪烁计数器上进行辐射探测。计数后,将每分钟的净计数(CPM)值除以样品的重量以计算净CPM/克浆液。

Claims (26)
1.一种口腔护理组合物,基于所述组合物的总重量,所述组合物包含3至12重量%的焦磷酸钙和10至19重量%的沉淀二氧化硅,其中所述沉淀二氧化硅具有大于85的表膜清洁率。
2.根据权利要求1所述的口腔护理组合物,其中所述沉淀二氧化硅具有5至15μm的平均粒度d50和40至120cm3/100g二氧化硅的吸油值。
3.根据权利要求1或权利要求2所述的口腔护理组合物,其中基于所述组合物的总重量,所述组合物包含5至10重量%的焦磷酸钙和12至17重量%的沉淀二氧化硅。
4.根据任一前述权利要求所述的口腔护理组合物,其中基于所述组合物的总重量,所述组合物包含8至10重量%的焦磷酸钙和12至14重量%的沉淀二氧化硅。
5.根据任一前述权利要求所述的口腔护理组合物,其中所述组合物还包含选自三聚磷酸钠、焦磷酸四钾、焦磷酸四钠、六偏磷酸钠、gantrezTM聚合物或其任意组合的防污渍剂。
6.根据权利要求5所述的口腔护理组合物,其中基于所述组合物的总重量,所述防污渍剂包含1至5重量%的三聚磷酸钠和1至5重量%的焦磷酸四钾。
7.根据任一前述权利要求所述的口腔护理组合物,其中所述组合物包含氟化物来源。
8.根据权利要求7所述的口腔护理组合物,其中所述氟化物来源是选自氟化亚锡、氟化钠、氟化钾、单氟磷酸钠、氟硅酸钠、氟硅酸铵、氟化胺和氟化铵中的至少一种。
9.根据权利要求7所述的口腔护理组合物,其中所述氟化物来源是单氟磷酸盐。
10.根据权利要求7所述的口腔护理组合物,其中所述氟化物来源是氟化钠。
11.如任一前述权利要求所述的口腔护理组合物,其还包含表面活性剂。
12.如权利要求11所述的口腔护理组合物,其中所述表面活性剂选自月桂基硫酸钠、椰油酰胺丙基甜菜碱及其组合。
13.如任一前述权利要求所述的口腔护理组合物,其还包含保湿剂。
14.如权利要求13所述的口腔护理组合物,其中所述保湿剂选自甘油、山梨糖醇及其组合。
15.根据任一前述权利要求所述的口腔护理组合物,其还包含增稠剂。
16.如权利要求15所述的口腔护理组合物,其中所述增稠剂选自聚乙二醇、纤维素衍生物、多糖胶、二氧化硅及其组合。
17.如权利要求16所述的口腔护理组合物,其中所述纤维素衍生物是羧甲基纤维素钠。
18.如权利要求16或权利要求17所述的口腔护理组合物,其中所述多糖胶是黄原胶。
19.根据任一前述权利要求所述的口腔护理组合物,其中所述组合物还包含一种或多种选自以下的试剂:稀释剂、pH调节剂、泡沫调节剂、甜味剂、矫味剂、色素、防腐剂、抗细菌剂、抗龋齿剂、抗牙垢剂或牙垢控制剂及其混合物。
20.根据任一前述权利要求所述的口腔护理组合物,基于所述组合物的总重量,所述组合物包含以下成分(以重量%计):
21.根据任一前述权利要求所述的口腔护理组合物,其为洁牙剂或糖果。
22.一种清洁和/或磨光和/或增白口腔表面的方法,其包括使所述口腔表面与如任一前述权利要求中所定义的口腔护理组合物接触。
23.根据权利要求22所述的方法,其中所述口腔护理组合物为洁牙剂,并且所述接触包括用所述口腔护理组合物刷涂牙齿。
24.根据任一前述权利要求所述的口腔护理组合物,其中基于所述组合物的总重量,研磨剂的总量为至多25重量%。
