技术领域
本发明涉及用于细胞保存的产品和方法,诸如用于随后的科学或者医学用途的任何类型的细胞的细胞存库(banking)。更接近地,本发明涉及使用设有冷冻介质/低温贮藏液体的真空管的用于细胞的低温贮藏的产品和方法。
背景技术
当今,存在存储细胞-例如人类胎盘或脐带干细胞-以用于未来的医学用途的若干细胞库。还存在存储在例如生物反应器中培养的细胞用于科学目的的细胞库。所有细胞库的共同点是细胞通过低温贮藏通常存储在处于-196°C的液体氮中。
低温贮藏是非常手动和耗时的工作,通常在分层式气流(LAF)下执行。此外,其包括若干具有开放式操纵的步骤,开放式操纵会增大污染的风险。由于传统的细胞存库期间的步骤的数量,还可发生批与批的变动。在细胞的低温贮藏期间,时间和温度是消除应力,诸如窒息的关键因素。
真空下的容器,诸如VacutainerTM,是已为大家接受的在医院和护理中心使用以用于快速的血液取样的产品。该容器包括真空管,其无菌地抽取血液通过带套筒的(sleeved)被遮盖的针。真空管以多种多样的容积可用,且通常预先涂覆有多种多样的复合物来防止例如血液凝结。真空管插入接近装置中,且当套筒保护的针刺穿真空管上的硅帽时,血液自动地将从患者被抽取。
将合乎需要的是具有这样的产品,利用该产品,细胞存库可在不使细胞离开生物反应器的情况下执行,且该产品将显著地缩短从生物反应器到冷冻机的时间。这将提供强有力的工具来用非常少的工作建立大的细胞库。
发明内容
本发明提供了用于快速细胞存库的产品和方法,与现有技术相比,其减小了可能的污染和操纵时间以及工作。
因此,在第一方面,本发明涉及用于细胞的低温贮藏的产品,包括设有冷冻介质的真空管,和所述管上的可穿透的和优选地可移除的帽。
优选地,冷冻介质是DMSO,甘油或甲基纤维素,且真空管填充有5-20%体积/体积,优选地10%体积/体积的冷冻介质。
在一个实施例中,提供了接近装置用于接收所述真空管。该接近装置设有将被插入到真空管中的中空针,且还设有用于连接到细胞源-诸如生物反应器中的细胞培养物-的连接器。
在第二方面,本发明涉及用于细胞的低温贮藏的方法,包括将细胞取样或收获到设有冷冻介质的真空管中;利用可穿透的帽闭合所述管;以及冷冻所述管。
在用于细胞的低温贮藏的优选方法中,该方法包括:在生物反应器中培养细胞;从所述生物反应器将细胞收获或取样到设有冷冻介质的真空管,其中,细胞与冷冻介质立即混合;从所述生物反应器移除所述管;以及冷冻所述管及其内容物。
优选地,冷冻介质是DMSO,甘油或甲基纤维素,且真空管填充有5-20%体积/体积,优选地10%体积/体积的冷冻介质。
在一优选的实施例中,细胞作为灌注培养来培养。优选地,细胞培养至10-200MVC/mL。
如果希望,细胞在冷冻之前被培育。
该方法的一个或者更多个步骤可为自动的。
本发明的方法将几乎消除从细胞收获至低温贮藏的时间,因为其将在一个步骤中完成。优选地,低温贮藏管在程序开始之前冷地存储。

具体实施方式
现在将关于非限制性的实例和附图描述本发明。
图1A显示了以浓缩的低温贮藏液体(2)准备的真空管(1)的示意图。该管还设有可穿透的和可移除的帽(3),以及硅或其它软材料制成的可穿透的隔膜(4)。该管可为施加有真空的适用于低温贮藏的任何器皿,例如管可为玻璃或者塑料。
图1B显示了接近装置(5)的示意图,其包括用于接收真空管的保护壳体(5)且具有针(6)以用于穿透真空管的盖。针设有橡胶或硅制成的保护套筒(7)。接近装置设有对生物反应器的鲁尔(Luer)或其它连接,用于细胞取样直接进入真空管中。
图1C显示了接近装置(5)的示意图,其连接到真空管(1)且准备好经由连接器(8)被插入到生物反应器中,以进行细胞取样。低温器皿沿着箭头的方向被推入保护帽(3)中。针将穿透软材料隔膜。真空将从生物反应器抽取细胞液,以便与低温贮藏液体(冷冻介质)直接混合。此后,管被放置在冷冻机中用于细胞存库。
示例:
传统的5ml真空管使用注射器填充有0.5ml的浓缩DMSO(冷冻介质)。使得CHO(中国仓鼠卵巢)培养达到直至10-100MVC/mL,优选地使用灌注培养。具有套筒保护的针(6)的接近装置通过连接器(8)附连到生物反应器。根据本发明的低温液体准备的真空管用于从生物反应器抽取细胞样本。真空管插入接近装置中,且真空通过接近装置的针将细胞液抽吸到管中。细胞液和冷冻介质立即混合。
此后,经填充的低温准备的真空管被直接传送到细胞存库冷冻机。如果必要,管中的细胞在冷冻之前简单地培育。
从生物反应器抽取细胞样本的程序并不会破坏细胞,并且非常迅速,这使得能够以最小的开放式操纵产生大的细胞库。如果需要,该过程可为自动的。

Claims (13)

1.一种用于细胞的低温贮藏的产品,包括:设有冷冻介质(2)的真空管(1);和所述管上的可穿透的帽(3);以及接近装置(5),所述接近装置(5)包括用于接收所述真空管(1)的保护壳体且设有将被插入到所述真空管中的针(6)以及设有连接器(8),所述连接器(8)用于通过鲁尔连接来连接到细胞源。
2.根据权利要求1所述的产品,其特征在于,所述冷冻介质是DMSO,甘油或甲基纤维素。
3.根据权利要求1或2所述的产品,其特征在于,所述真空管填充有5-20%体积/体积的冷冻介质。
4.根据权利要求1或2所述的产品,其特征在于,所述真空管填充有10%体积/体积的冷冻介质。
5.根据权利要求1或2所述的产品,其特征在于,所述细胞源是生物反应器中的细胞培养物。
6.一种用于细胞的低温贮藏的方法,包括:
在生物反应器中培养细胞;
通过权利要求1的产品将细胞取样或收获到设有冷冻介质的真空管中,其中所述接近装置(5)通过将该接近装置(5)的针(6)插入所述真空管(1)中而将所述真空管(1)接收到所述保护壳体中,且将所述连接器(8)通过鲁尔连接来连接到细胞源,使得所述细胞与所述冷冻介质立即混合;
从所述生物反应器移除所述管;
利用可穿透的帽闭合所述管;以及
冷冻所述管。
7.根据权利要求6所述的方法,其特征在于,所述冷冻介质是DMSO,甘油或甲基纤维素。
8.根据权利要求6所述的方法,其特征在于,所述真空管填充有5-20%体积/体积的冷冻介质。
9.根据权利要求6所述的方法,其特征在于,所述真空管填充有10%体积/体积的冷冻介质。
10.根据权利要求6所述的方法,其特征在于,所述细胞作为灌注培养来培养。
11.根据权利要求6所述的方法,其特征在于,所述细胞被培养至10-200MVC/mL。
12.根据权利要求6所述的方法,其特征在于,一个或更多个步骤是自动的。
