本发明公开了一种医用天然海洋生物敷料及其制备方法,包括基布层和贮药层,基布层和贮药层通过粘结剂粘接。贮药层包括以下重量份的原料:壳聚糖15‑25份,葡聚糖10‑20份,海藻酸盐15‑25份,透明质酸钠5‑15份,纳米银无机抗菌剂1‑5份,环丙沙星2‑8份,增塑剂2‑10份。本发明提供的敷料适用于医用小范围止血、抑菌和促进伤口愈合等方面的应用。

一种医用天然海洋生物敷料
技术领域
本发明涉及一种医用敷料,具体涉及一种医用天然海洋生物敷料及其制备方法。
背景技术
壳聚糖是由自然界广泛存在的几丁质经过脱乙酰作用得到的。自1859年,法国人Rouget首先得到壳聚糖后,这种天然海洋生物高分子的生物官能性和相容性、血液相容性、安全性、微生物降解性等优良性能被各行各业广泛关注。
近年来在医药和生物科技领域有着特别广泛的应用,原因是壳聚糖具有良好的生物活性、生物相容性、生物降解性,它可以促进伤口愈合和组织修复,在新型伤口敷料和组织工程方面的应用备受人们的关注。壳聚糖对格兰仕杨、阴性菌均有明显的抑制效果。Allam 等人的研究表明壳聚糖对绿脓杆菌、金黄色葡萄球菌、酿脓链球菌、表皮葡萄球菌、大肠杆菌及热带白色念珠菌均具有明显的抑制作用。另外,壳聚糖也具有止血和止痛的作用,这对于创面的愈合都具有积极的意义。
目前市售的天然海洋生物类止血敷料普遍存在止血效果不理想,和皮肤的贴合程度差等问题, 因此仍有待于进一步改善其止血效果和与皮肤组织的贴合性等问题。
发明内容
本发明的第一个目的在于提供一种医用天然海洋生物敷料,包括基布层和贮药层,基布层和贮药层通过粘结剂粘接。
上述贮药层包括以下重量份的原料:壳聚糖 15-25 份,葡聚糖 10-20 份,海藻酸盐 15-25 份,透明质酸钠 5-15 份,纳米银无机抗菌剂 1-5 份,环丙沙星 2-8 份,增塑剂2-10 份。
上述基布层为无纺布或PU防水透气膜。
上述粘结剂为压敏胶。
上述海藻酸盐为海藻酸钾、海藻酸钠或海藻酸钙。
上述增塑剂为甘油、山梨醇、甘露糖或聚乙二醇。
本发明的第二个目的在于提供一种医用天然海洋生物敷料的制备方法:按比例称取壳聚糖、葡聚糖、海藻酸盐、透明质酸钠、纳米银无机抗菌剂和环丙沙星,置于反应器中,加入 1.0%-2.0%的乙酸水溶液,搅拌至充分溶解;加入增塑剂,继续搅拌并恒温加热;将该混合溶液倒入制膜模具上,刮平,干燥充分后,脱膜,制得贮药层;将制备得到的贮药层和基布通过压敏胶粘接复合;利用密集针刺微孔机将粘结复合材料打孔;将打孔后的密集复合材料切割成所需形状,杀菌后包装。
上述恒温加热温度为 55-70℃,加热时间为 15-25 min。
上述粘结复合材料表面的微孔直径为 3 mm,每 100 cm2 粘结复合材料表面有90-100 个微孔。
本发明产品的制备方法简单,小范围止血、抑菌和促进伤口愈合效果显著。
具体实施方式:
下面结合具体实施案例对本发明作进一步详细地描述:
实施例1:
按比例称取壳聚糖 25 kg,葡聚糖 20 kg,海藻酸盐 25 kg,透明质酸钠 15 kg,纳米银无机抗菌剂 5 kg,环丙沙星 8 kg,置于反应器中,加入 1.0%-2.0%的乙酸水溶液,搅拌至充分溶解;加入山梨醇 10 kg,继续搅拌并在 70℃下恒温加热 25 min;将该混合溶液倒入制膜模具上,刮平,干燥充分后,脱膜,制得贮药层;将制备得到的贮药层和无纺布通过压敏胶粘接复合;利用密集针刺微孔机将粘结复合材料打孔;将打孔后的密集复合材料切割成所需形状,杀菌后包装。
实施例2:
按比例称取壳聚糖 20 kg,葡聚糖 15 kg,海藻酸盐 20 kg,透明质酸钠 12 kg,纳米银无机抗菌剂 5 kg,环丙沙星 5 kg,置于反应器中,加入 1.0%-2.0%的乙酸水溶液,搅拌至充分溶解;加入聚乙二醇 10 kg,继续搅拌并在 65℃下恒温加热 20 min;将该混合溶液倒入制膜模具上,刮平,干燥充分后,脱膜,制得贮药层;将制备得到的贮药层和PU防水透气膜通过压敏胶粘接复合;利用密集针刺微孔机将粘结复合材料打孔;将打孔后的密集复合材料切割成所需形状,杀菌后包装。
实施例3:
按比例称取壳聚糖 15 kg,葡聚糖 10 kg,海藻酸盐 15 kg,透明质酸钠 10 kg,纳米银无机抗菌剂 3 kg,环丙沙星 5 kg,置于反应器中,加入 1.0%-2.0%的乙酸水溶液,搅拌至充分溶解;加入山梨醇 10 kg,继续搅拌并在 55℃下恒温加热 15 min;将该混合溶液倒入制膜模具上,刮平,干燥充分后,脱膜,制得贮药层;将制备得到的贮药层和PU防水透气膜通过压敏胶粘接复合;利用密集针刺微孔机将粘结复合材料打孔;将打孔后的密集复合材料切割成所需形状,杀菌后包装。

Claims (8)

1.一种医用天然海洋生物敷料,其特征在于,包括基布层和贮药层,基布层和贮药层通过粘结剂粘接,所述的贮药层包括以下重量份的原料:壳聚糖 15-25 份,葡聚糖 10-20份,海藻酸盐 15-25 份,透明质酸钠 5-15 份,纳米银无机抗菌剂 1-5 份,环丙沙星 2-8份,增塑剂 2-10 份。
2.根据权利要求1所述的医用天然海洋生物敷料,其特征在于,所述的基布层为无纺布或PU防水透气膜。
3.根据权利要求2所述的医用天然海洋生物敷料,其特征在于,所述的粘结剂为压敏胶。
4.根据权利要求3所述的医用天然海洋生物敷料,其特征在于,所述的海藻酸盐为海藻酸钾、海藻酸钠或海藻酸钙。
5.根据权利要求4所述的医用天然海洋生物敷料,其特征在于,所述的增塑剂为甘油、山梨醇、甘露糖或聚乙二醇。
6.一种制备如权利要求5所述的医用天然海洋生物敷料的方法,其特征在于,按比例称取壳聚糖、葡聚糖、海藻酸盐、透明质酸钠、纳米银无机抗菌剂和环丙沙星,置于反应器中,加入 1.0%-2.0%的乙酸水溶液,搅拌至充分溶解;加入增塑剂,继续搅拌并恒温加热;将该混合溶液倒入制膜模具上,刮平,干燥充分后,脱膜,制得贮药层;将制备得到的贮药层和基布通过压敏胶粘接复合;利用密集针刺微孔机将粘结复合材料打孔;将打孔后的密集复合材料切割成所需形状,杀菌后包装。
7.根据权利要求6所述的制备医用天然海洋生物敷料的方法,其特征在于,所述的恒温加热温度为 55-70℃,加热时间为 15-25 min。
8.根据权利要求7所述的制备医用天然海洋生物敷料的方法,其特征在于,所述的粘结复合材料表面的微孔直径为 3 mm,每 100 cm2 粘结复合材料表面有 90-100 个微孔。
