Description

用于创伤及外科的医用胶
本发明属于外科粘合剂或接合剂技术领域。其国际专利分类为A61L25/00。
现有的用于外科的粘合剂,主要可以有镇静、止痛及止血作用,但作用达到所需的时间较长,一般至少需要5分钟,且有一定的毒副作用,会给病人带来一些痛苦,说明其功能尚不够理想,不能满足使用要求,因此,现有的这种外科用粘合剂在用于创伤及外科时受到一定的限制。
本发明的目的是公开一种对伤口粘合速度快、使用方便、无毒副作用的医用胶。该医用胶为无色透明液体,是一种化学组合物。它是一种用于创伤及外科的粘合剂或接合剂。
本发明的目的是由以下技术方案实现的:用于创伤及外科的医用胶,其特征在于:以重量份计,它采用1份氰乙酸乙酯、1.5份异丁醇、0.5份多聚甲醛、0.3份硫酸和1份辅料为原料,经酯交换、酯聚合、酯解聚和精制步骤制备而成。
本发明产品制造的工艺路线、工艺过程及收率是这样的:
一、工艺路线:
第一步:酯交换,将原料进行酯交换反应,经冷却、水洗、中和及干燥制得粗酯;
第二步:酯聚合,将该粗酯经脱水、脱萃制得精品;
第三步:酯解聚,对该精品进行减压蒸馏,在加热及SO2作用下,制得单体酯;
第四步:精制,进一步进行减压蒸馏,在加热及SO2作用下,制得本发明的医用胶产品。
二、工艺流程:
1.原料配比(重量比):氰乙酸乙酯、异丁醇、硫酸、多聚甲醛=1∶1.5∶0.
   3∶0.5;
2、过程:将上述原料按配比加入反应釜内逐步加热进行酯交换,并
   加入混合液进行脱水,使聚合的粗酯进行解聚,此时适中控制温
   度,使单体酯精制成医用胶。
三、收率:45-48%
本发明产品为α-氰基丙烯酸酯,含氮(N)量应不低于8.68%(g/g),为无色或微黄色透明液体,有特臭。在丙酮、乙醚或氯仿中易溶。本发明产品的鉴别、检查、粘接强度及含量测定方法是:
(鉴别)(1)取本品6滴,滴入5ml乙醇中,30秒内应形成白色固体。此固体不溶于水,溶于丙酮;(2)取本品数滴,滴加高锰酸钾试液1-2滴,紫色逐渐消退,并发生固化。
(检查)成膜时间,取0.03%碳酸氢钠溶液(本液需临用新配)50ml置于直径为90mm的平皿中,用滴管吸取本品,在离液面约5mm外,滴下一滴,准确记录在液面周围开始聚合至完全聚合的时间,应不超过15秒,成膜不得有断裂。
(粘接强度)用磷酸盐缓中液(pH7.4)擦洗皮革试片(猪、羊或兔皮革,厚度为1-1.5mm,长×宽为50×10mm)的粘接表面,再用木棉签擦干。取本品1-2滴于皮革试片粘接表面,使其均匀铺开,将另一皮革试片搭接在该片的涂胶部位,搭接长度为5mm,用手稍加压力约20秒,使两试片的粘接部位表面均匀地紧密接触,10分钟后,将一试片固定在架子上,另一试片加400g砝码,粘接部位不应发生破坏。平行进行3次试验均不发生破坏者为合格。
(含量测定)取本发明产品约0.5g,精密称定,照氮测定法(中国药典1990年版二部附录63页第一法)测定即得。
本发明产品主要用于各种创伤伤口、外科整形、器官移植、计划生育等各种外科手术。具有粘合速度快、强度高、止痛止血、抑菌功能好、使用方便、无毒副作用等优点。一次使用,瞬间即愈,工作生活不受影响。可用于代替缝合,无需敷料包扎,无需换药,并能防止感染,使伤口不缝合即愈的神话传说成为现实。使用本发明产品后无缝合所产生的针疤痕,是外科整形术的重大革命,是战场救护、居家旅游的必备良药。

1.用于创伤及外科的医用胶,其特征在于:以重量份计,它采用1份氰乙酸乙酯、1.5份异丁醇、0.5份多聚甲醛、0.3份硫酸和1份辅料为原料,经酯交换、酯聚合、酯解聚和精制步骤制备而成。
