Abstract
促红细胞生成素衍生物在听力保护上的应用,本发明涉及听力保护技术领域;利用促红细胞生成素衍生物pHBSP对Cdh23erl/erl小鼠进行治疗;即,每天对7天龄的小鼠经腹腔注射pHBSP一次,为期8周,用药结束后,对实验小鼠的听功能进行检测。将该促红细胞生成素衍生物pHBSP应用在耳聋小鼠模型上,作为治疗小鼠遗传性耳聋的药物应用,为人类年龄相关性听力减退或者增龄性聋的治疗药物的制备提供新途径。
Description
技术领域
本发明涉及听力保护技术领域,具体涉及促红细胞生成素衍生物在听力保护上的应用。
背景技术
耳聋是最常见的感觉障碍性疾病之一,包括老年性耳聋(老年人群的35%),噪音性耳聋,先天性耳聋(新生儿的1~3‰),药物性耳聋和突发性耳聋。全球有超过3.6亿的人存在听力损失,随着中国人群老龄化、噪音和环境污染日趋严重,该病在中国的发病率将不断攀升,预计在2025-2030年间达到峰值并将持续相当长的时间。至今并没有有效的药物治疗耳聋,寻找和研发有效的听力保护药物对该病的治疗具有重大的经济价值和社会意义。基于耳聋的高度遗传异质性,其发病多样,治愈困难。需要一种可以广泛应用于治疗耳聋的手段,旨在通过阻断耳聋发生的下游分子通路,而非仅仅通过校正基因突变本身来达到改善听力的目的。一直以来,小鼠模型在疾病研究中发挥着重要作用。耳聋小鼠模型的应用对该病分子机制的理解,耳聋相关基因的鉴定,基因功能的探测和药物筛选方面发挥着重要作用。
钙黏蛋白-23(CDH23)是由69个外显子编码的大小约为365kDa的跨膜蛋白,结构组成包括27个重复的胞外钙黏蛋白结构域,一个跨膜结构域和一个胞内结构域。CDH23参与内耳毛细胞顶连接的形成,而顶连接与内耳毛细胞顶端机械-电传导离子通道(mechanoelectrical transduction,MET)的功能密切相关。
pHBSP(QEQLERALNSS)是一个由EPO衍生而来的包含11个氨基酸短肽,这个肽段去除了EPO的促红细胞生成功能,但保留了EPO的神经保护功能。
pHBSP的神经保护作用在癫痫、脑损伤等神经系统障碍疾病中已有报道,介导EPO/pHBSP的组织保护作用的细胞受体是由EPO受体和共同β受体(EPOR-βCR)组成的组织保护受体(tissue-protective receptor,TPR),TPR通过不同的信号通路介导EPO的组织保护作用。EPO/pHBSP与TPR的结合导致Janus激酶-2(JAK-2)的活化和磷酸化,继而激活其下游分子通路达到抗凋亡的作用。另外,有关EPO/pHBSP通过肿瘤坏死因子-α(TumorNecrosisFactor,TNF-α)信号通路介导抗凋亡、抗炎的报道说明TNF-α也是EPO/pHBSP发挥组织保护作用的靶向分子之一。然而,有关pHBSP在听力保护作用方面的研究还未见报道。
发明内容
本发明的目的在于针对现有技术的缺陷和不足,提供促红细胞生成素衍生物在听力保护上的应用,将该促红细胞生成素衍生物应用在耳聋小鼠模型上,作为治疗小鼠遗传性耳聋的药物应用,为人类年龄相关性听力减退或者增龄性聋的治疗药物的制备提供新途径。
为实现上述目的,本发明采用的技术方案是:利用促红细胞生成素衍生物对Cdh23erl/erl小鼠进行治疗;所述的促红细胞生成素衍生物为pHBSP;每天对7天龄的小鼠经腹腔注射pHBSP一次,为期8周,用药结束后,对实验小鼠的听功能进行检测;
其中,所述的听力功能检测的将实验小鼠内耳毛细胞铺片观察,得pHBSP治疗组的平均ABR阈值相比PBS治疗组或为治疗组明显降低,(N=10,年龄=4w、8w、12w);**P<0.01;
其中,所述的小鼠内耳毛细胞铺片采用免疫荧光和实时定量PCR的方法进行检测,发现TNF-α和βCR在Cdh23erl/erl小鼠内耳细胞中高表达,即pHBSP在内耳组织中通过与其组织保护受体βCR的结合发挥听力保护作用,TNF-α是其发挥作用的可能靶点。
所述的Cdh23erl/erl小鼠为渐进性耳聋小鼠模型,是由于CDH23基因的第三个外显子的一个点突变(208T>C)引起CDH23蛋白第一个细胞外钙黏蛋白结构域(extracellularcadherin,EC)中氨基酸的替换(S70P),导致其二级结构的改变和蛋白功能的缺失,该小鼠模型的特点是在出生后27天(P27)出现渐进性耳聋,到P90时听力几乎全部丧失,Cdh23erl /erl小鼠的发病周期为实验听力保护药物提供了一个理想的窗口时间,这使Cdh23erl/erl小鼠模型成为试验听力保护药物的理想动物模型。
本发明有益效果为:本发明所述的促红细胞生成素衍生物pHBSP在听力保护上的应用,将该促红细胞生成素衍生物pHBSP应用在耳聋小鼠模型上,作为治疗小鼠遗传性耳聋的药物应用,为人类年龄相关性听力减退或者增龄性聋的治疗药物的制备提供新途径。

本具体实施方式采用的技术方案是:利用促红细胞生成素衍生物对Cdh23erl/erl小鼠进行治疗;所述的促红细胞生成素衍生物为pHBSP;每天对7天龄的小鼠经腹腔注射pHBSP一次,为期8周,用药结束后,对实验小鼠的听功能进行检测;
其中,所述的听力功能检测的将实验小鼠内耳毛细胞铺片观察,详见图1;根据数据显示,pHBSP治疗组的平均ABR阈值相比PBS治疗组或为治疗组明显降低,(N=10,年龄=4w、8w、12w);**P<0.01;

1.促红细胞生成素衍生物在制备治疗小鼠遗传性耳聋、人类年龄相关性听力减退或增龄性聋的药物中的应用,其特征在于,所述促红细胞生成素衍生物为pHBSP,pHBSP在内耳组织中通过与其组织保护受体βCR的结合发挥听力保护作用,TNF-α也是其发挥作用的靶点。
