本发明涉及的是一种测定血清白蛋白用的试剂 及其制备方法。
它是由白皂素、溴甲酚绿钠、柠檬酸、柠檬酸钠和 迭氮钠组成的固体试剂。其配比为(按每升溶液中含 固体试剂各成分量):白皂素10-12g/l、溴甲酚绿 钠80-100mg/l、柠檬酸4-5g/l、柠檬酸钠 4-5g/l、迭氮钠0.1-0.3g/l。柠檬酸、柠檬酸钠预 先研磨成细粉,至少过60目筛,然后按给定配方按比 例混合,搅拌均匀即可分装备用。
本固体试剂运输、储存容易,制备、使用方便。

Description

本发明涉及的是一种测定血清白蛋白用的试剂及其制备方法。
血清白蛋白浓度测定对人体疾病诊断或辅助诊断具有重要价值。临床上白蛋白浓度降低最为常见,可由于白蛋白合成障碍、蛋白质营养缺乏、吸收障碍、肝脏病变、晚期癌症等引起的创伤,感染和甲亢等消耗过多蛋白和肾脏病大量蛋白尿也可造成白蛋白低下。严重失水使血浆浓缩可导致白蛋白浓度增高。目前溴甲酚绿液体试剂是国内外广泛应用的测定血清白蛋白试剂。溴甲酚绿是一种酸碱指示剂,在PH4.2的环境中可与白蛋白形成兰绿色复合物,颜色之深浅与白蛋白浓度成正比,因此可用于血清白蛋白的定量测定。溴甲酚绿液体试剂主要成分有溴甲酚绿或其钠盐,Brij35(聚氧乙烯十二烷基醚)柠檬酸或琥珀酸缓冲液等。这种液体试剂配制过程十分复杂,试剂厂以液态试剂供应用户,体积大、重量大,包装困难,运输不容易。
本发明目的在于克服上述现有技术的缺点而提供一种测定血清白蛋白用的固体试剂及其制备方法,以使其运输、储存容易,配制使用方便。
本发明目的是这样实现的,在进行了大量的实验筛选后,选用了成分为白皂素、溴甲酚绿钠、柠檬酸、柠檬酸钠和迭氮钠的固体试剂,其配比为〔按配成溶液后每升溶液中含固体试剂各成分量〕:
白皂素  10~12克/升
溴甲酚绿钠  80~100毫克/升
柠檬酸  4~5克/升
柠檬酸钠  4~5克/升
迭氮钠  0.1~0.3克/升
其中白皂素是一种白色无定形粉末,干燥,不易吸收空气中的水分,具有发泡作用和乳化作用,配制溶液过程中没有强烈的刺激性气味,它是由皂素经提炼精制而成。迭氮钠是防腐剂。柠檬酸和柠檬酸钠是缓冲液,起缓冲作用。溴甲酚绿钠的作用则如前述,是作为一种指示剂。
其制备方法为,将柠檬酸、柠檬酸钠研磨成细粉状,至少过60目筛,然后与其它原料白皂素、溴甲酚绿钠、迭氮钠按上述配比混合,在搅拌器内混匀分装待用,使用时放入容器配加蒸馏水,使其满足上述溶液配比要求,混匀,即可使用。
本发明固体试剂的重量和体积都远远低于液体试剂,便于包装运输、储存;因采用了白皂素,使此固体试剂干燥、不结块,均匀性好;因本固体试剂酸碱度已调整合适,只要加蒸馏水至规定量,即可达PH4.2,无需再调整酸碱度,因此使用方便:本发明产品稳定,固体状态至少可保存一年,配制成溶液后保质期半年,(原液体试剂资料介绍保存期为一年),这大大方便了用户。
实施例:〔按配成溶液后每升溶液中含固体试剂各成分量〕:
选用:
白皂素  10.80克
溴甲酚绿钠  80毫克
柠檬酸  4.5克
柠檬酸钠  4.80克
迭氮钠  0.2克
将柠檬酸、柠檬酸钠研磨成细粉,过60目筛,然后与白皂素、溴甲酚绿钠、迭氮钠混合,在搅拌器内混均匀,按每瓶500毫升量分装。
取1000毫升三角烧瓶加约400毫升蒸馏水煮沸,将配制500毫升溶液的固体试剂量倒入三角烧杯中,搅拌至全溶,冷却至室温,以蒸馏水补足体积至500毫升,充分混匀,即可使用。
本发明固体试剂质量指标为:
精密度(批内)n=20  SD0.033  CV0.78%
精密度(批间)n=30  SD0.065  CV1.40%
准确度(回收率)97~102%
线性范围  2~6g/dL(r=0.9997)
稳定性  固体状态保质期一年(室温密封保存)
配成液体后保质期半年(室温密封保存)
测定血清白蛋白浓度的操作过程为:
将测试管中放入血清10微升,标准管中放入标准液10微升,并在上述测试管、标准管和另取一空试管中各放入试剂3毫升,各管充分混匀,室温置放3分钟,于波长628nm用空白管调零点,1厘米比色杯测各管O.D值。

1、一种测定血清白蛋白用的试剂,其特征在于:它是由白皂素、溴甲酚绿钠、柠檬酸、柠檬酸钠、迭氮钠组成的固体试剂,其配比为[按配成溶液后每升溶液中含固体试剂各成分量]:
白皂素      10~12克/升
溴甲酚绿钠  80~100毫克/升
柠檬酸      4~5克/升
柠檬酸钠    4~5克/升
迭氮钠      0.1~0.3克/升
2、一种如权利要求1所述的测定血清白蛋白用的固体试剂的制备方法,其特征在于:
(1).将柠檬酸、柠檬酸钠研磨成细粉状,至少过60目筛,
(2).将细粉状的柠檬酸、柠檬酸钠与白皂素、溴甲酚绿钠、迭氮钠按比例混合,搅拌均匀。
