Description

Composite peony seed oil self-emulsifying system composition and preparation method thereof
Technical Field
The invention belongs to the technical field of medicines, and particularly relates to a composite peony seed oil self-emulsifying system composition.
Background
The peony seed oil mainly contains fatty acids, contains a large amount of Unsaturated Fatty Acids (UFA), the total amount of UFA is about 80-92%, and is obviously higher than common vegetable oil such as olive oil, soybean oil and peanut oil which are called liquid gold, the peony seed oil is particularly worthy of being composed of α -linolenic acid, the content of UFA is generally 32-67%, the UFA is far higher than other common vegetable oil, and the UFA is similar to linseed oil, besides the remarkable advantages of the unsaturated fatty acids and the α -linolenic acid, the peony seed oil also contains monounsaturated fatty acid oleic acid which is easy to absorb by a human body and a large amount of polyunsaturated fatty acid linoleic acid which is necessary for the human body, the peony seed oil also contains certain sterol compounds (sitosterol, fucosterol and the like), fat-soluble vitamin E, squalene and the like, wherein the phytosterol is an important intermediate for synthesizing vitamin D3 and other medicaments, and has the functions of promoting metabolism, enhancing the metabolism of natural vitamin E, vitamin K, vitamin E, vitamin.
Modern researches show that the peony seed oil has higher nutrition and health care functions: reducing blood lipid, resisting oxidation, protecting liver, reducing blood sugar, improving immunity, preventing sunburn, and whitening skin. In 3 months in 2011, the Ministry of health issues a notice that the Ministry of health approves the peony seed oil as a new resource food (No. 9 in 2011), and the peony seed oil is approved as the new resource food.
The ganoderma lucidum spores are spores continuously released from the pileus of the ganoderma lucidum in the mature period, are hydrophobic oil substances extracted from the ganoderma lucidum spores, and mainly contain fatty acid, sterol, terpenes, vitamins and the like. Coix seed is dried mature seed of Coixlacryymajoib L.var.mayuen (Roman.) Stapf of Gramineae, is a hydrophobic oil substance extracted from Coix seed, and mainly contains fatty acid and Coix lacryma-jobi ester. The ganoderma lucidum spore oil and the coix seed oil are researched and developed more and are mature at present, and have great advantages in vegetable oil in the aspects of improving immunity and resisting tumors.
The peony seed oil applied in the current market is mainly provided in the form of liquid edible blend oil, for example, chinese patent CN103859064B discloses "a blend edible oil containing peony seed oil and its preparation method", although there are many patents on the combination of peony seed oil and other vegetable oils, there is no composite peony seed oil combining peony seed oil, ganoderma lucidum spore oil and coix seed oil. According to the composite peony seed oil, the ganoderma spore oil and the coix seed oil are combined into the composite peony seed oil, so that the advantages of the ganoderma spore oil and the coix seed oil in the aspects of improving immunity and resisting tumors can be fully exerted, and the effect of enhancing the immunity of the composite peony seed oil is greatly improved.
In recent years, research and development of peony seed oil are more popular, and more patents are applied. For example, the publication patent CN104106806A discloses a health product for stabilizing blood pressure and reducing blood lipid, and for example, the Chinese patent CN 103494199A discloses a peony seed oil soft capsule with blood lipid reducing effect and preparation thereof. However, in the existing literature, no report about a peony seed oil self-emulsifying system composition and a preparation method thereof is found, and no report about a composite peony seed oil self-emulsifying system composition and a preparation method thereof is found, and only the report about the application of peony seed oil as a health care product is reported, for example, patent CN 104666458A discloses "an american ginseng soft capsule containing peony seed oil and a preparation method thereof", and peony seed oil is mixed with a medicine to be directly pressed into a soft capsule containing peony seed oil. However, the existing soft capsule containing peony seed oil has the defect of low bioavailability. In addition, the peony seed oil is prepared into soft capsules, and the problems of unqualified disintegration in storage period, substance migration and leakage of oily components in the capsules, increased acid value and oxidation value and the like which are stability problems need to be overcome. Therefore, a new compound peony seed oil oral pharmaceutical dosage form needs to be developed.

s3, sieving polyethylene glycol 6000, hydroxypropyl- β -cyclodextrin and lactose with a 100-mesh sieve, uniformly mixing with the concentrated solution, sieving with a 18-mesh sieve to obtain granules, namely the self-emulsifying system composition L of the compound peony seed oil, and after inspection, filling the granules into aluminum plastic bags to obtain finished products.
Monitoring self-emulsifying effect: adding the self-emulsifying system composition A-L (such as 0.5mL for concentrated solution and 0.5g for granule) of the composite peony seed oil into L0mL artificial gastric juice, and stirring with glass rod to rapidly form emulsion with blue opalescence. The particle size of the emulsion was measured by a TSM ultrafine particle size analyzer, and the average particle size of the emulsion droplets was measured, and the results are shown in Table 13.
