Abstract

技术领域
本发明涉及动物的基因修饰以产生特定特征的可控变化,例如,皮肤和/或皮毛着色中的变化。
背景技术
大多数动物物种中合成的唯一色素及所有哺乳动物物种中合成的唯一色素是黑色素。黑色素分为两类:褐黑素,其产生金色或红色,及真黑素,其产生深棕色或黑色。两种黑色素都由酪氨酸合成,但其合成途径在多巴醌产生后不同。
控制特定的黑素细胞产生褐黑素或真黑素的主要开关是黑素皮质激素受体(MC1R)。MCIR多态性还可能是红色或金色褐黑素的主要决定因素。
黑素皮质激素受体可以通过任何的黑素细胞刺激激素(MSH)激活,最通常的是α-MSH,还可以是β-防御素103。相反地,MC1R的激活可能通过刺鼠信号蛋白(agoutisignaling protein,ASP)的表达被抑制。β-防御素103信号优于ASP信号且ASP信号优于MSH信号。此外,所有这些加强或降低其活性的基因中存在突变和多态性。毛色进一步地由黑色素产生及转运的多个调节剂调节,其包括酪氨酸酶(TYR)、酪氨酸酶转运体OCA2及泛素基因HERC2等。
植物产生多种其他色素,包括叶绿素、类胡萝卜素、花青素和甜菜红。在这些色素中,类胡萝卜素最容易转移到动物体内,因为植物中类胡萝卜素色素的产生依赖于动物中也能发现的前体。香叶基香叶基焦磷酸是HMG-CoA还原酶途径中的中间体,并用作合成类固醇和固醇的前体。随着4-5个植物蛋白的添加,香叶基香叶基焦磷酸可替换用作合成胡萝卜素的黄色和橙色或类胡萝卜素红色的前体。植物色素转移至动物已发生于自然界:豌豆蚜虫A具有几个从真菌转移的基因。类胡萝卜素已在转基因酵母中产生,所述酵母属于动物界。
在多细胞动物中通过基因干预曾尝试的表面色素的唯一修饰是动物由浅色到深色或由深色到浅色的不加选择的变化。还未创建更详细的模式。在动物中天然发现的胡萝卜素色素不比蚜虫更复杂,且未被设计进入任何多细胞动物中。在本发明之前,还未产生动物物种的皮肤或皮毛的定制颜色或图案。
发明简述
本发明提供了创建动物中定制特征的材料和方法。例如,本发明提供了创建动物的皮肤和/或皮毛中定制图案的材料和方法。在其他实施方案中,所述定制特征可以包括动物皮肤或皮毛的长度和/或纹理。可选地,本发明还可用于产生动物指甲、爪和/或角的纹理、结构强度和/或长度的可控变化。
在一个具体的实施方案中,本发明的方法包括将基因构建体导入动物的细胞中,所述基因构建体包括角蛋白-14特异性启动子、loxp盒中的红色荧光蛋白、色素盒中的显性黑(△G23)β防御素103,及SV40(带有内含子)多聚腺苷酸化序列。当将包括Cre重组酶(或HTNCre)的组合物施用于具有基因构建体的动物皮肤时,施用所述组合物的动物的皮毛将变黑。以这种方式,所述皮毛被永久地基因修饰而以所需形状转变颜色。
因此,在一个实施方案中,本发明提供了在动物的皮肤和/或皮毛中创建定制的永久图案的方法。
在另一实施方案中,本发明提供了在动物物种的皮肤和/或皮毛中产生多颜色图案的方法。
在进一步的实施方案中,本发明提供了在动物物种的皮肤和/或皮毛中创建定制图案的方法,以使所述动物能继续生长皮毛以在其整个生命期保持这些颜色。
本发明还提供了创建定制的预定义的条纹图案的方法,所述图案在该动物中是可遗传的。
在一个实施方案中,本发明提供了一个或多个活化因子的经皮施用以引起本发明的转基因动物中的重组和永久的转基因表达。在某些具体的实施方案中,根据本发明使用的活化因子包括,但不限于,重组酶蛋白质、能够诱导重组酶表达的小分子(如,强力霉素,cumate、蜕皮激素等),能够诱导重组酶表达的病毒,及驱使重组酶表达的核酸(如DNA)构建体。
在某些实施方案中,所述活化因子可单独地或在载体溶液中(例如,脂质体、溶剂、含有DMSO的混合物等)施用于动物皮肤的表面。在一个实施方案中,所述活化因子皮内(如使用纹身针)或皮下施用。
在一个实施方案中,所述转基因动物包含一个或多个外源核酸分子,其包括但不限于,着色-相关的基因,表皮/毛发特性基因(如控制动物毛发的长度和/或卷曲度的基因),与指甲/爪或角特性相关的基因(如编码交联角蛋白的核酸分子),及动物细胞中用于合成和/或表达植物色素的基因。
在某些实施方案中,根据本发明使用的启动子包括但不限于,皮肤-特异性启动子(例如,角蛋白特异性启动子),黑素细胞特异性启动子(例如,MCR启动子),组成型启动子(例如,β-球蛋白启动子、CMV启动子),及响应循环因子(如NF-kB、干扰素γ、雌激素和糖皮质激素类)的启动子。
在某些实施方案中,本发明提供了多种类型重组酶靶标的用途以允许通过施用不同重组酶选择性地特异性激活不同基因。在一个实施方案中,多元重组酶靶标用于允许动物出生后产生多种颜色。
在一个实施方案中,在具有天然金色皮毛的转基因狗中,Cre重组酶激活黑色狗皮毛的产生,且Flp重组酶激活红色狗皮毛的产生。
在一个实施方案中,本发明提供了自身启动子的用途以在不需要外源活化因子时引起表皮着色。在一个具体实施方案中,自身启动子涉及动物发育中限定体节边界。
在一个实施方案中,自身异源启动子用于创建不同物种的表皮图案,如,例如,使用猫中发现的Tabby或Ticked启动子以引起狗的表皮着色。
在某些实施方案中,本发明提供了具有在出生后永久可定制的表皮图案的转基因修饰的动物。
在某些实施方案中,本发明提供了具有不常发现于哺乳动物中的毛色的转基因修饰的动物。

在一个实施方案中,本发明提供了黑安格斯品种的转基因牛类动物,其具有白色或近白色的表皮或毛皮。

图6示出了转基因小鼠的毛色可以通过施用于小鼠皮肤的重组酶数量滴定。增加施用于小鼠的重组酶数量增加转基因表达的水平和真黑素激活。
图7是用于基因敲入牛的基因构建体的实施方案示意图,所述牛具有增强型绿色荧光蛋白(EGFP)的精子-特异性表达,及重组酶-介导的显性阴性Rab7的黑素细胞-特异性表达。编码新霉素耐药性生物标记物蛋白的核酸序列位于构建体的中心且所述构建体两侧为短的同源臂,其中所述标记物蛋白可通过PIGGYBACTM转座子被切除。

发明详述
本发明提供了创建动物中定制特征的材料和方法。例如,本发明提供了创建动物的皮肤和/或毛皮中定制图案的材料和方法。在其他实施方案中,所述定制特征可以包括动物皮肤或毛皮的长度和/或纹理。可选地,本发明还可用于产生动物毛发、指甲、爪和/或角的纹理、结构强度和/或长度的可控变化。
在一个具体的实施方案中,本发明的方法包括将基因构建体导入动物的细胞中,所述基因构建体包括角蛋白-14特异性启动子、loxp盒中的红色荧光蛋白、色素盒中的显性黑(△G23)β防御素103,及SV40(带有内含子)多聚腺苷酸化序列。当将包括Cre重组酶(或HTNCre)的组合物施用于具有基因构建体的动物皮肤时,施用所述组合物的动物的毛皮将变黑。以这种方式,所述毛皮永久地基因修饰而以所需形状转变颜色。

在一具体的实施方案中,将与DMSO载体混合的强力霉素(或蜕皮激素等)施用于具有金色毛皮的转基因动物,由此转基因动物的颜色变为红色;随后,将与DMSO混合的Cre或HTNCre施用于转基因动物以产生黑色。

创建具有“非黑色”表皮或毛皮的转基因牛类动物
在一个实施方案中,本发明提供了具有白色或近白色的表皮或毛皮的转基因牛动物,其中所述转基因动物(如在其基因组中)包括:

色素蛋白

各种色素蛋白的氨基酸序列;参与色素合成和/或转运的蛋白质;参与动物皮肤或毛皮的长度和/或纹理的蛋白质;参与动物指甲、爪和/或角的纹理、结构强度和/或长度的蛋白质是公开的,如通过GenBank数据库。本发明包括这些蛋白质的用途。
在本发明范围内的多核苷酸和多肽可以根据与本文中具体列举的那些序列的一致性定义。序列一致性通常大于60%,优选地大于75%,更优选地大于80%,甚至更优选地大于90%,且可大于95。与本文中所列举的序列相比,序列一致性可以是60、61、62、63、64、65、66、67、68、69、70、71、72、73、74、75、76、77、78、79、80、81、82、83、84、85、86、87、88、89、90、91、92、93、94、95、96、97、98或99%。

在一个实施方案中,根据本发明使用的诱导型系统是Cre-LoxP重组系统。所述转基因动物的基因组可包含受Cre/LoxP重组系统调控的外源核酸分子,其中Cre/LoxP重组系统阻止外源核酸分子的表达。在一具体的实施方案中,Cre/LoxP重组系统包含lox-stop-lox(LSL)序列。
所述Cre-LoxP重组系统是用于进行细胞或转基因动物的DNA中位点特异性删除、插入、易位及倒置的位点特异性重组技术。Cre重组酶蛋白(由最初称为“引起重组”的位点编码)由4个亚基和2个结构域组成:较大的羧基(C-末端)结构域和较小的氨基(N-末端)结构域。loxP(X-over PI位点)是噬菌体PI的位点且由34bp组成。Cre-重组酶-介导的重组结果取决于loxP位点的位置和方向,其可以顺式或反式定位。在顺式定位的情况下,loxP位点的方向可以是相同或相反的。在反式定位的情况下,所涉及的DNA链可以是线形的或环形的。Cre重组酶-介导的重组结果可以是在顺式loxP位点情况下切除(当loxP位点处于相同方向时)或倒置(当loxP位点处于相反方向时)间插序列,或在反式loxP位点情况下将两个分子(染色体)间的一个DNA插入另一个中或易位。所述Cre-LoxP重组系统在本领域是已知的,参见,例如Andras Nagy,Cre recombinase:the universal reagent for genometailoring,Genesis 26:99-109(2000)。
在没有Cre-介导重组的情况下Lox-Stop-Lox(LSL)盒阻碍转基因的表达。存在Cre重组酶、LoxP位点重组时,所述stop盒被去除。所述Lox-Stop-Lox(LSL)盒在本领域是已知的。参见,Allen Institute for Brain Science,Mouse Brain Connectivity Altas,Technical White Paper:Transgenic Characterization Overview(2012)。
在某些实施方案中,所述loxP位点进一步包含编码基因(例如,lacz、GFP)的报告基因和/或编码第二着色蛋白(例如,如果第一着色蛋白含有显性作用的MC1R,另一刺鼠基因(agouti gene)两侧可以是loxP位点)的核酸分子。
四环素(Tet)-调控转录激活是诱导型表达的方法,其中转录通过抗生素四环素或其衍生物(例如,强力霉素)之一的存在或缺失可逆地控制。基因表达因Tet-off或Tet-on蛋白与位于诱导型启动子内的四环素响应元件(TRE)结合而被激活。Tet-on和Tet-off蛋白都能激活基因。Tet-Off蛋白子在没有四环素衍生物-强力霉素(Dox)时激活基因表达,然而Tet-on蛋白在存在Dox时激活基因表达。
在Tet-off系统中,四环素反式激活(tTA)蛋白,其通过融合TetR(四环素阻遏)蛋白(由大肠杆菌细菌获得)和VP 16蛋白(由单纯疱疹病毒获得)而创建,在TetO操纵子处结合DNA。一旦结合TetO操纵子激活与TetO操纵子连接的启动子,从而激活附近基因的转录。四环素衍生物结合tTA并使其不能与TRE序列结合,从而阻止了靶基因的反式激活。
在Tet-On系统中,当tTA蛋白与强力霉素结合时,强力霉素-结合tTA能够与TetO操纵子结合。因此,将强力霉素导入系统中启动了基因产物的转录。Tet-on系统有时优选地用于更快的响应。
反向tTA(rtTA)是用于通过加入Dox(Tet-on)的快速基因激活的互补基因模件。参见Kistner等,Doxycycline-mediated quantitative and tissue-specific control ofgene expression in transgenic mice,Proc.Natl.Acad.Sci.U.S.A.Vol.93,pp.10933-10938(1996)。
Tet-on高等反式激活子(也称为rtTA2s-M2)是Tet-On的另一版本,其示出了比Tet-on降低的基础表达及功能,低10倍的Dox浓度。此外,认为其表达在真核细胞中更稳定,因为其为优化的人类密码子及利用3个最小转录激活域。Tet-on 3G(也称为rtTA-VIO)与高等Tet-on相似,且为优化的人类密码子并由3个最小VP 16激活域组成。Tet-on 3G与原始Tet-on相比低100倍的Dox敏感。
在四环素-响应调控表达元件的一实施方案中,四环素-调控的逆向反式激活子(rtTA)包括tetR(例如,来自大肠杆菌的Tnl0);哺乳动物转录因子VP 16,其可反式激活作为效应子的结构域;及组织-特异性启动子,其可控制rtTA效应子转录。存在强力霉素时,rtTA与位于CMV启动子上游的(TeTO)7操纵子(7个串联重复的TetO序列)结合,所述CMV启动子引起转基因的表达。因此,可通过施用及戒断强力霉素开启或关闭转基因表达。
表达构建体
本发明还提供了用于产生转基因动物的表达构建体、载体及宿主细胞。
本文中使用的术语“表达构建体”是指核酸序列的组合,其用于可操作连接的核酸序列的转录。本发明的表达构建体还通常包括调控元件,其在预计的宿主细胞中发挥作用,所述表达构建体在所述宿主细胞中表达。因此,本领域普通技术人员可选择宿主细胞中使用的调控元件,例如,细菌宿主细胞、酵母宿主细胞、植物宿主细胞、昆虫宿主细胞、哺乳动物宿主细胞和人类宿主细胞。调控元件包括启动子、转录终止序列、翻译终止序列、增强子和多聚腺苷酸元件。
在某些实施方案中,本发明提供了用于定制动物的颜色和/或图案的表达构建体,包括着色构建体和图案构建体。
图1示出了着色构建体的实施方案,其包括启动子、loxP盒、编码目的色素蛋白或参与目的色素合成和/或转运的蛋白质的核酸分子及多聚腺苷酸化序列。在着色构建体的一实施方案中,通过施用Cre重组酶激活目的色素蛋白或参与目的色素合成和/或转运的蛋白质的表达。
在另一实施方案中,本发明提供了图案构建体。图2示出了图案构建体的实施方案。在一个实施方案中,所述图案构建体的启动子源自特异于体节边界规格的基因。在一个实施方案中,所述启动子选自Ripply 2启动子、Tabby启动子和Ticked启动子。在一个实施方案中,所述转基因动物包含着色构建体和图案构建体。在一个实施方案中,所述转基因动物在背侧真皮具有定制的组成型垂直条纹。
在一个实施方案中,所述转基因动物的基因组包含:
1)如图1所示的着色构建体,其中启动子是黑素细胞-特异性启动子且核酸分子编码参与红色素(如ASIP或MC1R)合成的蛋白质;

本发明的表达构建体可以包含与编码本发明中肽的多核苷酸序列可操作地连接的启动子序列。可以使用本领域已知的标准技术将启动子并入多核苷酸。在本发明的表达构建体中可使用启动子的多个拷贝或多个启动子。在一优选实施方案中,启动子可被定位在与其天然遗传环境中距离转录起点大约相同的位置。启动子活性没有大幅下降的情况下,可允许该距离的一些改变。转录起点通常包括在表达构建体中。
本文中使用的术语“可操作连接”是指所述组分的并列,其中所述组分的关系是允许其以预计的方式发挥作用。通常,可操作连接的组分是连续的关系。通常,与编码序列可操作连接的序列能够引起编码序列的复制、转录和/或翻译。例如,当启动子能够直接转录编码序列时,该编码序列可操作地连接启动子。
“编码序列”或“编码区域”是转录为mRNA和/或翻译成多肽的多核苷酸序列。例如,编码序列可以编码目的多肽。编码序列的边界由5'-端的翻译起始密码子和3'-端的翻译终止密码子确定。

术语“polyA”或“p(A)”或“pA”是指转录终止及mRNA多腺苷酸化信号的核酸序列。polyA序列的特征是六核苷酸基序AAUAAA。常用的多腺苷酸化信号是SV40pA、人生长激素(hGH)pA、β-肌动蛋白pA和β-球蛋白pA。所述序列的长度范围为32至450bp。可以使用多个pA信号。

具体实施方式
以下是用于解释实践本发明过程和实施方案的实施例。这些实施例不构成对本发明的限制。
实施例1-动物皮肤或毛皮着色的定制
该实施例提供了用于定制动物皮肤或毛皮着色的基因构建体的实施方案
具有定制的皮肤或毛皮着色或图案的动物
具有棕色(“Agouti”染色)肤色的C3H/HeJ鼠经转基因表达以下三个构建体:1)构建体,其包括角蛋白-14特异性启动子、loxp盒(其包括编码红色荧光蛋白的核酸)、色素盒(其包括编码显性黑色(ΔG23)β防御素103蛋白的核酸)和(具有内含子的)SV40多聚腺苷酸化序列;2)构建体,其包括编码反向四环素反式激活子(rtTA)的核酸分子和SV40多聚腺苷酸化序列,所述核酸分子可操作地与角蛋白-14特异性启动子连接,所述启动子启动编码rtTA的核酸的转录;和3)构建体,包括刺鼠信号蛋白(ASP)和牛生长激素(BGH)多聚腺苷酸化序列,ASP可操作地与四环素敏感型启动子(TetO)7连接,所述启动子启动编码ASP的核酸分子的转录。
当将强力霉素施用到基因修饰的小鼠时,小鼠毛皮变成金色(这是出生后毛发颜色的遗传修饰的第一例证)。
利用载体(例如,蛋白质载体,如脂质双层)向转基因的小鼠被刮开的皮肤施用Cre或HTNCre,诱导在Cre和/或HTNCre施用的区域毛皮出现遗传上永远的黑色颜色。根据本发明,出生时具有棕色毛皮的小鼠可以被改变为带任意形状黑色标记的具有金色毛皮的小鼠。例如,可以在金色背景的小鼠毛皮上创建黑色名字或黑色标志。
为了定制动物的皮肤着色,动物可以仅表达着色构建体,并不需要表达图案构建体(如图2所示)。着色和图案的定制可以直接由施用Cre和/或HTNCre来激活。
在一个实施方案中,利用基因修饰动物从而表达图案构建体来创建可遗传的图案。图2示出了图案构建体的一个实施方案。启动子可以是Ripply2启动子或来自特异于体节边界范围的基因的启动子。可替代的启动子可以是Tabby或Ticked启动子。
在一个实施方案中,基因修饰的动物(其基因组包括着色构建体和图案构建体)在背部真皮具有组成型垂直条纹。
复合图案/彩色图案
图3示出了用于在动物皮肤或毛皮中创建复合或彩色图案的基因构建体的某些实施方案。在一个实施方案中,利用包含诱导系统(如具有诱导性机制的启动子)的构建体创建复合图案。如图3所示,仅在组织特异性rtTA启动子中存在强力霉素时,Cre被激活。在动物中,组织特异性启动子的使用维持了体节边界,例如转基因能够仅在发育期被激活。可诱导系统还可以用于创建彩色图案。
根据本发明使用的可诱导系统包括但不限于四环素、蜕皮激素和三苯氧胺(tamoxifen)可诱导系统;FLP-FRT重组系统;和Cre-LOX重组系统。
实施例2-具有定制毛皮颜色和图案的小鼠
该实施例示出了基因修饰的小鼠的创建,其毛皮颜色可通过经皮施用易于跨过细胞膜的HTNCre重组酶而永久地被改变。
图4示出了用于创建小鼠皮肤或毛皮中定制的图案和颜色的基因构建体。所述构建体包含角蛋白l4启动子,其在所有皮肤成纤维细胞中都表达,以引起信号分子β-防御素l03的显性黑色的形式。β-防御素l03的表达被编码环指蛋白(RFP)的LoxP位点切割核酸阻断,RFP用作标记物。
刺鼠(Agouti)小鼠经转基因以表达编码“显性黑色”信号分子β-防御素l03的转基因,通过施用重组酶激活转基因表达。
图5A和B是示出了两种经转基因的小鼠,其中通过皮肤或皮肤内施用HTNCre的载体溶液激活显性黑色素蛋白的表达。在本发明之前,重组酶从未被施用到活的动物体内。
图6示出了向转基因小鼠施用重组酶可能导致毛皮颜色的剂量依赖性改变。刺鼠小鼠毛发的顶端通常的特征在于细胞包括黄褐黑素(A)。如图6所示,通过增加重组酶的剂量,转基因的活化增加,小鼠毛皮色素可能从褐黑素和黑真黑素(B)的混合物转变成纯的真黑素(C),其转化程度为打破黑色素划分的结构(D)。该实施例示出了重组酶的经皮使用更能够导致在转基因的小鼠内更精细地控制毛发颜色。
实施例3-定制肤色和图案用于牛的鉴定
在养牛业中,对于所有权证明、牧业管理、动物移动追踪以及可能是最重要的动物疾病可追溯性来说,用于个体识别的分娩处理的可靠方法变得越来越重要。
该实施例提供了用于牛识别的具有可以用作代码(例如,条形码)的定制肤色和图案的转基因牛。可以使用实施例2中描述的方法来创建转基因牛。如实施例2所示,向转基因小鼠(其基因组包括Cre-loxP重组系统)经皮或皮内施用重组酶,诱导出生后毛色出现定制的变化。
图7示出了用于在牛中创建定制的图案或颜色标志的构建体设计。所述构建体包括编码显性阴性Rab7的核酸分子,其可操作地与MCIR启动子连接并受loxP-STOP-loxP序列的调控。通过施用Cre重组酶可诱导Rab7的表达。TAL核酸酶的使用使得位点特异性基因可以敲入短的同源臂。构建体上的顶体蛋白启动子/EGFP臂允许流式分选基因修饰的精子,从而确保在F1代为100%基因修饰的子代。用于构建体的重要的元件是MC1R(黑素皮质激素受体)启动子,其仅在被重组酶激活时,驱动显性阴性Rab7基因的表达。构建体中各种元件的组合创建了用位点特异性基因敲入改性的牛,该位点特异性敲入允许在牛身上创建永久识别信息。
图8示出了转基因的黑安格斯牛在皮肤中表达定制的识别图案的描述。
分娩处理期间,通过将Cre重组酶施用到基因组包含图7所示的构建体的牛中,可以很容易地创建个体识别信息。同样地,转基因仅表达于动物的皮肤,这可在食品加工过程中除去;因此,在动物皮中创建的牛的识别不应成为政府机构(如美国农业部)的监管问题。
在某些实施方案中,可以使用常规的技术创建位点特异性基因敲入的动物,所述技术包括但不限于精原干细胞(SSC)、利用转座元件的piggyBacTM流动DNA技术、黄单胞菌属转录激活子类(TAL)核酸酶(XTN)[aka TAL-效应子核酸酶(TALENs)]和其组合。
在一个实施方案中,黑色安格斯毛色是用杂合敲入而进行后天修饰的,该杂合敲入使用黑素细胞特异性显性阴性Rab7,其需要关键的黑素生成基因Tyrpl的细胞内转运。
实施例4-用于牛疾病检测的定制肤色和图案
疾病是几乎所有的牛肉和奶制品生产商关注的问题,许多常见疾病可以显著地影响生产,而不必具有明显的临床症状。猝死往往是梭菌属疾病的第一个也是唯一的标志。亚临床乳腺炎是奶制品生产中常见的、花费颇高的问题。甚至有传言说有几乎没有明显的临床症状的疯牛病(BSE)可能会导致牛肉产业严重的经济损失。除了传染病,牛代谢性疾病也可能导致经济损失:1/3的肉牛由于在它们的饮食中存在螯合剂,而患有亚临床铜缺乏症。根据本发明的毛皮或皮肤颜色的变化(例如,使用重组酶激活表皮色特异性标记)可用于识别牛是否患病。
在一个实施方案中,用于检测牛疾病的构建体与图7所示的构建体相同,不同之处在于用可选的重组酶系统(例如,Flp-Frt重组系统)和/或疾病特异性启动子(例如响应NFkB,Ifnγ或铜缺乏靶标的启动子)替代了MC1R启动子。
在一个实施方案中,分娩处理期间,牛涂有重组酶以创建所需的符号。一旦牛出现炎症或代谢应力(由疾病特异性启动子调控的色素蛋白的表达),涂抹区域会改变颜色。牛的疾病检测应用可以通过使用不同的重组酶和重组目标与牛识别应用组合使用。
实施例5-具有定制表皮的狗
作为不同组合的色素基因突变的结果,不同品种的狗具有不同的毛色。哈巴狗具有完整的黑素皮质激素受体,而没有内源性表达“显性黑色”的信号分子β-防御素03。
具有定制毛色和图案的狗可以通过使用如实施例3和4所述用于牛的方法来创建,或者通过如实施例2所示用于小鼠的方法来创建。在一个实施方案中,经转基因的哈巴狗(其基因组包括如图7所示的构建体)具有定制颜色和图案的毛皮,包括虎纹、图形、文字和心形。
由于犬科动物的卵细胞发育系统的特异性,所以狗通常难以被转基因。目前,转基因的狗不能通过体外受精(IVF)而获得。可以利用精原干细胞(SSC)、利用转座元件的piggyBacTM流动DNA技术、黄单胞菌属转录激活子类(TAL)核酸酶(XTNs)[aka TAL-效应子核酸酶(TALENs)]和其组合来创建位点特异性基因敲入的转基因狗。
实施例6-具有“白色”毛色的转基因安格斯牛的创建
在牛中,热应力是引起发病率和死亡率的主要原因,并对养牛业具有不利的经济影响。黑色表皮色大幅度地削弱动物耐受热应力的能力,而黑色表皮的牛显示出其核心体温度的增加超过白色牛的五倍。
黑色安格斯牛是美国最流行的肉牛品种。部分原因是因为它的黑色表皮,黑色安格斯品种耐热应力很差,不适合生活在炎热的气候中。在黑安格斯牛耐热性差的问题一直是养牛业的一个长期存在的问题。
现有的通过育种计划创建白色或浅色毛色的安格斯品种的尝试都失败了,部分原因是因为黑安格斯品种的黑色表皮色的特征在于显性作用的MCIR的组成型活性等位基因,且在牛中不存在下游的显性白色等位基因。通过杂交繁殖创建“白色”表皮安格斯品种需要显性下游等位基因。酪氨酸酶和oca突变偶尔发生,并且创建白化病黑安格斯牛对于创建“白色”表皮安格斯品种是没有用的,因为这种战略需要同系繁殖。
本实施例提供了通过向牛中引入异源、非天然显性白色等位基因而创建具有“白色”表皮安格斯牛的方法。
在一个实施方案中,鸡中的Pmel17的显性白色等位基因(也称为GP100和Silv)被转移到牛中。Pmell7参与黑素体黑色素的组装。具有Pmel17隐性突变的夏洛莱牛有白色毛色,具有显性白色等位基因的牛是优选的。在小鼠、狗和马中发现了显性作用的Pmel17等位基因。可通过将显性突变敲入到隐性Pmel17等位基因,或通过在不同于天然Pmel17等位基因的位点引入非天然显性白色Pmel17来将这些显性白色等位基因转移到牛中。
用于创建具有白色表皮动物(如安格斯牛)的基因包括,但不限于,参与黑素体组装的显性等位基因(如Pmel17、MLPH、显性阴性Rab7等位基因);和编码抑制黑色素向黑素体转运的蛋白的等位基因。
同样地,可通过在编码蛋白的基因中引入显性突变来创建具有白色毛色的转基因动物(如牛),由此得到缺少黑素细胞的白色动物(如牛)。所述蛋白参与发育过程中来自神经嵴的黑素细胞的迁移,
此外,通过向动物中引入siRNA构建体(如miRNA构建体)能够来创建具有白色毛色的转基因动物(如牛),所述构建体抑制编码用于转运黑色素或对于黑素细胞迁移所必须的蛋白的核酸表达。
本发明还提供了具有白色毛色的转基因动物(如牛),其中可以通过真皮或经皮使用重组酶蛋白来创建遗传性黑色标记。
使用上述方式中的任何的组合,牛的非天然基因可用于创建显性超过在黑安格斯中发现的组成型活性MC1R“白色”等位基因,从而在转基因公牛和正常的黑安格斯母牛之间的远系繁殖产生的后代中出现白色或银色毛发。

Claims (10)
1.一种具有白色或近白色表皮或毛皮的非人转基因动物的细胞,其中所述转基因动物包含:
外源核酸分子,其可操作地与启动子连接,其中所述外源核酸分子编码显性白色Pmel17蛋白质;和/或
编码目的序列的外源抑制性RNA,其可操作地与启动子连接,其中所述编码目的序列的外源抑制性RNA干扰野生型动物核酸分子的表达,其中所述野生型动物核酸分子编码黑素皮质激素受体(MC1R)蛋白质。
2.根据权利要求1所述的细胞,其中所述动物是狗、猫、小鼠、大鼠、豚鼠、仓鼠、马、牛、猪、绵羊、山羊、鸭子、鹅、鸡、灵长类动物、鱼、蝾螈、蛇、蜥蜴、狐狸、黄鼠狼、兔子、河狸、貂、水獭、海豹、草原狼、灰鼠、鹿、麝鼠和负鼠。
3.根据权利要求2所述的细胞,其中所述貂是水貂或黑貂。
4.根据权利要求1所述的细胞,其中所述动物是牛科动物。
5.根据权利要求4所述的细胞,其中所述动物是黑安格斯品种的牛。
6.根据权利要求1所述的细胞,其中表达所述显性白色Pmel17蛋白质的等位基因来自原鸡。
7.根据权利要求1所述的细胞,其中所述启动子选自通用启动子、组成型启动子、组织特异性启动子和诱导型启动子。
8.根据权利要求1所述的细胞,其中所述启动子选自巨细胞病毒(CMV)启动子、CMV-鸡β肌动蛋白启动子、泛素启动子、JeT启动子、SV40 启动子、β球蛋白启动子、延伸因子1 α(EF1-α)启动子、RSV 启动子、Ripply 2启动子、Ticked启动子、Tabby启动子、Mo-MLV-LTR 启动子、Rosa26 启动子、角质形成细胞特异性启动子、黑素细胞特异性启动子、基质细胞特异性启动子和毛乳头特异性启动子。
9.根据权利要求1所述的细胞,其中所述外源核酸分子的表达或外源抑制性RNA编码序列的表达受以下诱导型系统的调控,所述诱导型系统选自Cre-LoxP重组系统、FLP-FRT 重组系统、四环素(Tet)-调控转录激活系统、蜕皮激素诱导型系统、热冲击开/关系统、lacO/IPTG系统、cumate阻遏蛋白CymR系统、硝基还原酶系统、香豆霉素/新生霉素调控系统、RheoSwitch配体RSL1系统、双向嵌合核受体表达系统、GAL4 系统、固醇诱导/抑制系统、类固醇诱导/抑制系统、合成类固醇诱导/抑制系统;及其组合。
10.根据权利要求9所述的细胞,其中所述诱导型系统包括Cre-LoxP重组系统。
