Background
Type II diabetes is the most common form of diabetes, accounting for 90% of the cases of diabetes. Over a hundred million people worldwide suffer from type II diabetes (about one seventy million in the united states) and the incidence is dramatically increasing in both developed and developing countries. Type II diabetes is a lifelong disease that usually begins in middle age or later in life, but can begin at any age. Patients with type II diabetes cannot respond properly to insulin, which, as a hormone, generally allows the body to convert blood glucose into energy or store it in cells for later use. A problem with type II diabetes is a state known as insulin resistance, in which the body produces normal or even higher amounts of insulin, but certain mechanisms render insulin unable to transfer glucose into the cells. Because the body cannot use insulin properly, the glucose in the blood rises to unsafe levels, a condition known as hyperglycemia.
Over time, persistent hyperglycemia leads to glucotoxicity, which worsens insulin resistance and contributes to pancreatic beta cell dysfunction. The extent of persistent hyperglycemia is directly related to diabetic microvascular complications and may also contribute to macrovascular complications. Hyperglycemia thus circulates the harmful effects that exacerbate the control and complications of type II diabetes.
It is now widely accepted that glycemic control varies among type II diabetic patients. The goal of current diabetes therapy is to achieve and maintain as close to normal blood glucose as possible to prevent long-term microvascular and macrovascular complications associated with elevated blood glucose. Oral therapeutic options for the treatment of type II diabetes include the following known compounds: sulfonylureas, biguanides (metformin), thiazolidinediones and alpha-glucosidase inhibitors. Each type of active agent is typically administered to a patient separately. However, when monotherapy becomes inappropriate, combination therapy is an attractive and reasonable course of action in terms of treating hyperglycemia despite the known side effects associated with sulfonylurea and thiazolidinone treatment, i.e. weight gain.
A new class of antidiabetic agents has recently been discovered, which are known as sodium-glucose transporter-2 (SGLT2) inhibitors. SGLT2 inhibitors prevent reabsorption of glucose into the blood via the kidneys. The kidney continues to filter glucose from the glomerulus into the bladder, however, almost all of the filtered glucose is reabsorbed. SGLT2 is a protein responsible for most of the glucose reabsorption and helps the body retain glucose to meet its energy needs. For patients with diabetes, the excess glucose retained by this pathway contributes to persistent hyperglycemia. Inhibition of SGLT2 activity inhibits renal glucose reabsorption in the body, thereby resulting in the excretion of glucose in the urine.
Accordingly, the present invention provides a bilayer tablet comprising metformin and an SGLT2 inhibitor for oral administration for the treatment of a disease or disorder associated with SGLT2 activity without the occurrence of weight gain associated with other treatments. The first layer of the bilayer tablet is a metformin extended release (XR) formulation or a reduced mass metformin XR formulation. The second layer is an SGLT2 inhibitor formulation. The metformin/SGLT 2 inhibitor bilayer tablet of the present invention provides a patient with an antidiabetic treatment which is convenient and effective in terms of controlling blood glucose levels.
Disclosure of Invention
The present invention provides a bilayer tablet comprising (1) a metformin XR formulation or a reduced mass metformin XR formulation as a first layer, (2) an SGLT2 inhibitor formulation as a second layer, and (3) optionally a film coating covering both layers. Metformin may be in the form of a pharmaceutically acceptable salt, with metformin hydrochloride (HCl) being preferred. Preferred SGLT2 inhibitors are dapagliflozin (dapagliflozin) or a pharmaceutically acceptable salt thereof, dapagliflozin (S) propylene glycol hydrate (1:1:1), or dapagliflozin (R) propylene glycol hydrate (1:1: 1). The most preferred SGLT2 inhibitor is dapagliflozin or dapagliflozin (S) propylene glycol hydrate (1:1: 1). The SGLT2 inhibitor formulation is an immediate release or extended release formulation, with an immediate release formulation being preferred.

Magnesium stearate is a preferred lubricant and silicon dioxide or colloidal silicon dioxide is a preferred glidant. The bilayer tablets of the present invention can be administered to a mammal (preferably a human) to treat a variety of conditions and disorders associated with SGLT2 activity, including but not limited to treating or delaying the progression or onset of: diabetes (including type I and type II diabetes), impaired glucose tolerance, insulin resistance and diabetic complications such as nephropathy, retinopathy, neuropathy and cataracts, hyperglycemia, hyperinsulinemia, hypercholesterolemia, dyslipidemia, elevated blood levels of free fatty acids or glycerol, hyperlipidemia, hypertriglyceridemia, obesity, wound healing, tissue ischemia, atherosclerosis and hypertension. The formulations of the invention may also be used to increase the blood level of High Density Lipoproteins (HDL). Additionally, conditions, diseases and disorders collectively referred to as "syndrome X" or "metabolic syndrome" (see Johannsson, j.clin.endocrinol.meta., 82,727-34(1997)) can be treated using the formulations of the present invention.

the preparation of bilayer tablets with significantly different weight ratios between the two layers can lead to cracking at the layer interface, separation of the layers or even cross-contamination of the two layers. The large weight ratio difference between the metformin layers and the SGLT2 inhibitor layers of examples 4-15 provided the following challenges: the effectiveness and content uniformity of the lower weight layer, i.e., the SGLT2 inhibitor layer, is maintained. It was also found that various of the test formulations showed cracking in the SGLT2 inhibitor layer on the tablet surface in the region adjacent to but spaced from and parallel to the interface between the layers. For example, SGLT2 inhibitor layer formulations with increased microcrystalline cellulose levels allow cracking, separation, and cross-contamination of the two layers to be reduced or eliminated while maintaining the efficacy and uniformity of the second layer. In addition, pregelatinized starch, hydroxypropyl cellulose EXF, or mannitol may be used as a substitute for a portion of microcrystalline cellulose. Finally, a second layer having a total weight of about 300mg to about 400mg helps to reduce or eliminate cracking, separation, and cross-contamination of the two layers. The preferred weight of the second layer is 300 mg. Thus, the present invention provides a bilayer tablet that reduces or eliminates cracking, separation and cross-contamination of the metformin layer and the SGLT2 inhibitor layer and maintains or improves the efficacy and uniformity of the SGLT2 inhibitor layer.
The present invention provides a bilayer tablet comprising a metformin XR (1000mg) formulation, an SGLT2 inhibitor (2.5, 5.0 or 10.0mg) formulation and optionally a film coating. The metformin XR formulation comprises metformin hydrochloride (HCl), sodium carboxymethyl cellulose, hydroxypropyl methyl cellulose, and magnesium stearate. The SGLT2 inhibitor formulation comprises SGLT2 inhibitor, anhydrous lactose, microcrystalline cellulose, crospovidone, silicon dioxide, and magnesium stearate. For the second layer, pregelatinized starch, hydroxypropyl cellulose EXF, or mannitol (mannitol SD 200) may be used to replace a portion of the microcrystalline cellulose. Additionally, the total weight of the second layer is from about 300mg to about 400 mg. The preferred weight of the second layer is 300 mg. The optional film coating may be that in preferred embodiments, the SGLT2 inhibitor is dapagliflozin, dapagliflozin (S) PGS, or dapagliflozin (R) PGS.

It is to be understood that the above percentages (amounts) will be greater for dapagliflozin in the form of a hydrate of propylene glycol as compared to dapagliflozin in a non-solvated/hydrated form.
Metformin, an antidiabetic agent, is suitable for use in the bilayer tablets of the present invention in various forms including pharmaceutically acceptable salts thereof such as hydrochloride, hydrobromide, fumarate, succinate, p-chlorophenoxyacetate or pamoate. The fumarate and succinate salts are preferably metformin (2:1) fumarate and metformin (2:1) succinate. Metformin hydrochloride is preferred.
The present invention also encompasses a coated bilayer tablet wherein the coating comprises saxagliptin or a pharmaceutically acceptable salt thereof. Saxagliptin in free base form, monohydrate form or hydrochloride form is preferred. The coated tablet comprises a core, a first coating layer, a second coating layer, and optionally a third coating layer. The tablet core comprises metformin, preferably metformin hydrochloride. The first and second coating layers optionally contain saxagliptin, wherein at least one of the first and second coating layers contains saxagliptin. The third coating layer is an optional outer protective coating layer. Saxagliptin, shown below, may be prepared as described in U.S. patent 6,395,767, the disclosure of which is incorporated by reference into this application for any purpose. The first and second coating layers are prepared in a similar manner as described in WO2005/117841 for the preparation of the inner seal coating layer or the intermediate (drug) coating layer, the disclosure of which is incorporated by reference into the present application for any purpose. The third coating layer is prepared in a similar manner as described in WO2005/117841 for the preparation of the outer protective coating layer.
Saxagliptin
The first coating layer comprises at most 95% polymer (based on the weight of the first coating layer). The formulation will contain at least one coating layer polymer and a coating solvent, preferably water, which is used for processing and is removed by drying. The first coating layer polymer may be hydroxypropylmethyl cellulose, polyvinyl alcohol (PVA), ethyl cellulose, methacrylic acid (ester) polymer or hydroxypropyl cellulose, preferably PVA. The first coating layer optionally comprises saxagliptin in the range of about 0.5 to about 70% and preferably about 30 to about 50% (based on the weight of the second coating layer). The first coating layer may comprise: optional plasticizers such as triacetin, diethyl phthalate, tributyl sebacate or polyethylene glycol (PEG), preferably PEG; antiadherents or glidants such as talc, fumed silica or magnesium stearate; and opacifiers such as titanium dioxide. Accordingly, in one aspect, the present invention provides a coated bilayer tablet comprising: (1) a bilayer tablet core comprising two layers, wherein a first layer comprises metformin; and the second layer comprises an SGLT2 inhibitor; wherein the second layer is about 300 to about 400 mg; (2) a first coating layer coating the bilayer tablet core and optionally comprising saxagliptin; (3) a second coating layer coating the first coating layer and optionally comprising saxagliptin; and (4) optionally a third coating layer, said third coating layer coating said second coating layer; wherein at least one of the first coating layer and the second coating layer comprises saxagliptin.

Human research
In a phase III clinical study at 24 weeks old, the combination of metformin and the SGLT2 inhibitor dapagliflozin reduced glycosylated hemoglobin levels (HbA1c) and Fasting Plasma Glucose (FPG) levels in type II diabetic patients. Metformin alone does not control glycosylated hemoglobin and fasting plasma glucose levels properly compared to placebo + metformin. The study also showed that individuals receiving dapagliflozin had a statistically greater average weight loss compared to individuals receiving placebo.
The study was designed to evaluate the effectiveness and safety of the combination of dapagliflozin and metformin over a 24 week period in patients with inadequate control of type II diabetes. The data represent results from a randomized, double-blind, placebo-controlled study of 546 individuals with type II diabetes who had HbA1c greater than or equal to 7.0% and less than or equal to 10% at baseline. After a two-week introduction period, individuals were randomized into one of four different treatment groups: 2.5mg dapagliflozin (n 137), 5mg dapagliflozin (n 137), 10mg dapagliflozin (n 135), or placebo (n 137). Patients in all groups also received metformin (greater than or equal to 1500 mg/d). The primary endpoint of the study was a comparison of the mean change in HbA1c from baseline in each dapagliflozin treated group to the placebo group after 24 weeks. Secondary endpoints included comparison of changes in FPG and body weight from baseline at week 24 with placebo and correction of the percentage of individuals treated with dapagliflozin and achieving HbA1c of less than 7% at week 24. The endpoint was probed for weight loss of greater than or equal to 5% or greater than or equal to 10% and percent change in weight from baseline.
After 24 weeks, individuals receiving 2.5mg, 5mg, and 10mg of dapagliflozin + metformin exhibited statistically significant corrected mean percent changes in HbA1c from baseline of-0.67%, -0.70%, and-0.84%, respectively, versus-0.30% in the placebo group. Individuals treated with dapagliflozin showed a statistically significant corrected mean change in FPG, the secondary endpoint, from baseline at week 24: 17.8mg/dL in the 2.5mg dapagliflozin group, -21.5mg/dL in the 5mg dapagliflozin group, and-23.5 mg/dL in the 10mg dapagliflozin group, and-6.0 mg/dL in the placebo group.
The study also evaluated the potential effect of dapagliflozin on weight loss. These results included data measured for changes in overall body weight over a 24-week study period. At week 24, the overall weight change in kg, the secondary endpoint, was-2.21 kg in the 2.5mg dapagliflozin group, -3.04kg in the 5mg dapagliflozin group, and-2.86 kg in the 10mg dapagliflozin group, and-0.89 kg in the placebo group. Overall, more patients receiving dapagliflozin achieved a weight loss greater than or equal to 5% (probed endpoint) compared to placebo.
These results indicate that SGLT2 inhibitors, in particular dapagliflozin or dapagliflozin (S) propylene glycol hydrate in combination with metformin, are effective in treating hyperglycemia in type II diabetic patients without causing weight gain.

dapagliflozin can be prepared using a method similar to that described in U.S. Pat. No. 6,515,117 or published International applications WO03/099836 and WO2008/116179, the disclosures of which are incorporated by reference into this application for any purpose. SGLT2EC50 is 1.1 nM.

Bilayer tablets containing metformin (1000mg) extended release formulation and dapagliflozin (S) PGS (5.00mg) immediate release formulation were prepared as follows.

metformin granulate (1000mg)
metformin hydrochloride, 0.5% magnesium stearate and sodium carboxymethyl cellulose were combined and mixed in a high shear granulator for one minute. Purified water was added using a nozzle while stirring for one minute. The wet granulated material was passed through a mill and then dried until the moisture content was 1.0% or less. The dry mass containing metformin hydrochloride, 0.5% magnesium stearate and sodium carboxymethylcellulose was passed through a mill and discharged into a polyethylene-lined cylinder to give 1g of milled metformin agglomerated granules.
Hydroxypropyl methylcellulose 2208USP (100,000 centipoise) (Methocel K100M Premium) was added to the box blender and mixed for 60 revolutions. The material was passed through a mill and discharged to give milled hydroxypropyl methylcellulose 2208 USP.
Metformin (ground 1g agglomerated particles), hydroxypropyl methylcellulose 2208USP (ground), hydroxypropyl methylcellulose 2208USP (unground), and magnesium stearate were added to the box blender and mixed for 60 revolutions. The mixed material was discharged into a polyethylene lined cartridge to give an extended release metformin 1g pellet.
Dapagliflozin pellet (2.5, 5.0 or 10.0mg)
Dapagliflozin (S) PGS is blended with microcrystalline cellulose, anhydrous lactose, a portion of crospovidone, and a portion of silicon dioxide in a suitable tumbler mixer and passed through a suitable conical mill. A portion of the magnesium stearate (sieved) is blended into the mixture and then compressed using a suitable roller compactor. The compressed mixture is reduced to form granules. The granules were blended with the remaining amounts of crospovidone and silica in a suitable tumbler mixer. The granules were then blended with the remaining amount of magnesium stearate in a suitable tumbler mixer.

The metformin XR formulation or reduced mass metformin XR formulation is placed in a first addition funnel and the SGLT2 inhibitor formulation, which is dapagliflozin (S) PGS granules (2.5mg, 5mg or 10mg in intensity), is placed in a second addition funnel in a bi-layer tablet press. The tablet press was set to obtain the target weight of the first layer (metformin XR or reduced mass metformin XR). The second addition funnel was opened and the tablet press was adjusted to obtain the target tablet weight for dapagliflozin and metformin XR or dapagliflozin and reduced mass metformin XR bilayer tablets. After the target weight is obtained, the tablet press is adjusted to obtain the target hardness. Once the hardness is obtained, the tablets prepared are periodically monitored for weight of the first layer and weight, hardness, gauge and friability of the entire tablet. The collected bilayer tablets were film coated with PVA (polyvinyl alcohol).

Metformin hydrochloride, 0.5% magnesium stearate and sodium carboxymethyl cellulose were combined and mixed in a high shear granulator for one minute. Purified water was added using a nozzle while stirring for one minute. The wet granulated material was passed through a mill and then dried until the moisture content was 1.0% or less. The dry mass containing metformin hydrochloride, 0.5% magnesium stearate and sodium carboxymethylcellulose was passed through a mill and discharged into a polyethylene-lined cylinder to give 1g of milled metformin agglomerated granules.
Metformin (ground 1g of agglomerated particles), hydroxypropyl methylcellulose 2208USP (100,000 centipoise) (Methocel K100M Premium) and silica were added to a box blender and mixed for 120 revolutions. Magnesium stearate was added and after 60 revolutions the material was discharged into a polyethylene lined cylinder to give a reduced mass of metformin extended release 1g agglomerated granules.
The dapagliflozin (S) PGS granulation and double layer compression operations were performed as described in example 4.

Bilayer tablets containing reduced mass metformin (1000mg) extended release formulation and dapagliflozin (S) PGS (2.5mg) immediate release formulation were prepared in a similar manner to that described in example 7.

Other SGLT2 inhibitor IR formulations described in table 2 can be used to prepare bilayer tablets of the present invention. The formulations in table 2 are compatible with the metformin XR formulations described herein or reduced mass metformin XR formulations. The bilayer tablets prepared from the SGLT2 inhibitor IR formulation in table 2 reduced or eliminated cracking, separation and/or cross-contamination with the metformin XR layer and with the reduced mass metformin XR layer.

Bilayer tablets containing metformin (500mg) extended release formulation and dapagliflozin (S) PGS (5.0mg) immediate release formulation were prepared as follows.

metformin hydrochloride, 0.5% magnesium stearate and sodium carboxymethyl cellulose were combined and mixed in a high shear granulator for one minute. Purified water was added using a nozzle while stirring for one minute. The wet granulated material was passed through a mill and then dried until the moisture content was 1.0% or less. The dry mass containing metformin hydrochloride, 0.5% magnesium stearate and sodium carboxymethylcellulose was passed through a mill and discharged into a polyethylene-lined cylinder to give 500mg of milled metformin agglomerated granules.
The milled metformin 500mg agglomerated particles, hydroxypropyl methylcellulose 2208, hydroxypropyl methylcellulose 2910, microcrystalline cellulose and silicon dioxide were added to a box blender and mixed for 240 revolutions. Magnesium stearate was added and after 60 revolutions the material was discharged into a polyethylene lined cylinder to give metformin extended release 500mg agglomerated granules.
The dapagliflozin (S) PGS granulation and double layer compression operations were performed as described in example 4.
Example 14
Bilayer tablets containing metformin (500mg) extended release formulation and dapagliflozin (S) PGS (10.0mg) immediate release formulation were prepared in a similar manner to that described in example 13.

Claims (4)

1. A coated bilayer tablet comprising: (1) a bilayer tablet core comprising two layers, wherein the first layer comprises a metformin hydrochloride extended release formulation; and the second layer comprises a sodium-dependent glucose transporter (SGLT2) inhibitor selected from dapagliflozin or dapagliflozin (S) propylene glycol hydrate; wherein the second layer is 300 to 400 mg; (2) a first coating layer coating the bilayer tablet core and optionally comprising saxagliptin; (3) a second coating layer coating the first coating layer and optionally comprising saxagliptin; and (4) optionally a third coating layer, said third coating layer coating said second coating layer; wherein at least one of the first coating layer and the second coating layer comprises saxagliptin;
Wherein the weight ratio between the first and second layers is 4.4: 1;
wherein (1) the first layer comprises 64-82% metformin hydrochloride, 3-5% sodium carboxymethylcellulose, based on the total weight of the first layer; 15-30% hydroxypropyl methylcellulose; 0.1-0.75% magnesium stearate; and 0-2% silica or 0-1.5% colloidal silica; wherein the sum of the percentages of the components is 100%; (2) (ii) said second layer comprises 0.5-4% dapagliflozin or dapagliflozin (S) propylene glycol hydrate, based on the total weight of the second layer; 14-18% anhydrous lactose; 50-80% microcrystalline cellulose; 0-20% of pregelatinized starch; 0-20% mannitol; 0-15% hydroxypropyl cellulose; 2-6% of crospovidone; 0.5-2.5% silica; and 0.5-2% magnesium stearate; wherein the sum of the percentages of the components is 100%.
2. The coated bilayer tablet according to claim 1 wherein (1) the first layer comprises 64-82% metformin hydrochloride; 3-5% sodium carboxymethylcellulose; 15-30% hydroxypropyl methylcellulose; 0.1-0.75% magnesium stearate; and 0-2% silica or 0-1.5% colloidal silica, said percentages being based on the total weight of the first layer, wherein the percentages of the components sum to 100%; and the second layer comprises 0.5-4% dapagliflozin or dapagliflozin (S) propylene glycol hydrate; 14-18% anhydrous lactose; 50-80% microcrystalline cellulose; 0-20% of pregelatinized starch; 0-20% mannitol; 0-15% hydroxypropyl cellulose; 2-6% of crospovidone; 0.5-2.5% silica; and 0.5-2% magnesium stearate, said percentages being based on the total weight of the second layer, wherein the percentages of the components sum to 100%; (2) the first coating layer comprises a polyvinyl alcohol-based polymer; (3) the second coating layer comprises saxagliptin and a polyvinyl alcohol-based polymer; and (4) the third coating layer comprises a polyvinyl alcohol-based polymer.
3. The coated bilayer tablet according to claim 1 wherein (1) the first layer comprises 67-71% metformin hydrochloride; 3-5% sodium carboxymethylcellulose; 25-29% hydroxypropyl methylcellulose; and 0.1-0.75% magnesium stearate, said percentages based on the total weight of the first layer, wherein the percentages of the components sum to 100%; and the second layer comprises:
(A) 0.5-4% dapagliflozin or dapagliflozin (S) propylene glycol hydrate; 14-18% anhydrous lactose; 72-80% microcrystalline cellulose; 2-6% of crospovidone; 0.5-2.5% silica; and 0.5-1.5% magnesium stearate, said percentages being based on the total weight of the second layer, wherein the percentages of the components sum to 100%;
(B) 0.5-4% dapagliflozin or dapagliflozin (S) propylene glycol hydrate; 14-18% anhydrous lactose; 50-70% microcrystalline cellulose; 10-22% of pregelatinized starch; 2-6% of crospovidone; 0.5-2.5% silica; and 0.5-1.5% magnesium stearate, said percentages being based on the total weight of the second layer, wherein the percentages of the components sum to 100%;
(C) 0.5-4% dapagliflozin or dapagliflozin (S) propylene glycol hydrate; 14-18% anhydrous lactose; 60-70% microcrystalline cellulose; 5-15% hydroxypropyl cellulose; 2-6% of crospovidone; 0.5-2.5% silica; and 0.5-1.5% magnesium stearate, said percentages being based on the total weight of the second layer, wherein the percentages of the components sum to 100%; or
(D) 0.5-4% dapagliflozin or dapagliflozin (S) propylene glycol hydrate; 14-18% anhydrous lactose; 55-65% of microcrystalline cellulose; 10-20% mannitol; 2-6% of crospovidone; 0.5-2.5% silica; and 0.5-1.5% magnesium stearate, said percentages being based on the total weight of the second layer, wherein the percentages of the components sum to 100%;
(2) The first coating layer comprises polyvinyl alcohol, polyethylene glycol, talc and titanium dioxide; (3) the second coating layer comprises saxagliptin, polyvinyl alcohol, polyethylene glycol, talc and titanium dioxide; and (4) the third coating layer comprises polyvinyl alcohol, polyethylene glycol, talc and titanium dioxide.
4. The coated bilayer tablet according to claim 1 wherein (1) the first layer comprises 72-82% metformin hydrochloride; 3-5% sodium carboxymethylcellulose; 15-22% hydroxypropyl methylcellulose; 0.75-1.25% silica or 0.25-0.75% colloidal silica; and 0.1-0.6% magnesium stearate, said percentages based on the total weight of said first layer, wherein the percentages of said components sum to 100%; the second layer comprises:
(A) 0.5-4% dapagliflozin or dapagliflozin (S) propylene glycol hydrate; 14-18% anhydrous lactose; 72-80% microcrystalline cellulose; 2-6% of crospovidone; 0.5-2.5% silica; and 0.5-1.5% magnesium stearate, said percentages being based on the total weight of the second layer, wherein the percentages of the components sum to 100%;
(B) 0.5-4% dapagliflozin or dapagliflozin (S) propylene glycol hydrate; 14-18% anhydrous lactose; 50-70% microcrystalline cellulose; 10-22% of pregelatinized starch; 2-6.00% of crospovidone; 0.5-2.5% silica; and 0.5-1.5% magnesium stearate, said percentages being based on the total weight of the second layer, wherein the percentages of the components sum to 100%;
(C) 0.5-4% dapagliflozin or dapagliflozin (S) propylene glycol hydrate; 14-18% anhydrous lactose; 60-70% microcrystalline cellulose; 5-15% hydroxypropyl cellulose; 2-6% of crospovidone; 0.5-2.5% silica; and 0.5-1.5% magnesium stearate, said percentages being based on the total weight of the second layer, wherein the percentages of the components sum to 100%; or
(D) 0.5-4% dapagliflozin or dapagliflozin (S) propylene glycol hydrate; 14-18% anhydrous lactose; 55-65% of microcrystalline cellulose; 10-20% mannitol; 2-6% of crospovidone; 0.5-2.5% silica; and 0.5-1.5% magnesium stearate, said percentages being based on the total weight of the second layer, wherein the percentages of the components sum to 100%;
(2) The first coating layer comprises polyvinyl alcohol, polyethylene glycol, talc and titanium dioxide; (3) the second coating layer comprises saxagliptin, polyvinyl alcohol, polyethylene glycol, talc and titanium dioxide; and (4) the third coating layer comprises polyvinyl alcohol, polyethylene glycol, talc and titanium dioxide.
