本发明公开了一种牛肉汞含量的检测方法,包括以下步骤:取二苯碳酰二肼乙醚溶液与乳化剂OP水溶液按比例混合,中速定量滤纸浸渍,制成试纸;取1000μg/mL标准汞溶液,HCl溶液调节pH至6.0,加水稀释汞含量为0.5μg/mL的溶液,放入试纸,显色,拍照,处理后形成纸标准色阶。称取牛肉样品,破碎,加10%HCl,微波消解,离心,取上清液,调整PH值在6.0,减压浓缩上清液至牛肉样品重量的1/10,放入试纸,显色,拍照,将试纸显色深度和标准色阶比较,判断汞含量是否超标。发明的检测方法操作简单灵敏度高,作为汞含量是否超标的初筛,适合工厂生产质检的使用。

技术领域
本发明涉及肉制品重金属检验领域,具体为一种牛肉汞含量的检测方法。
背景技术
牛肉是全世界人都爱吃的食品,中国人消费的肉类食品之一,仅次于猪肉,它含有丰富的蛋白质,氨基酸组成比猪肉更接近人体需要,能提高机体抗病能力,寒冬食牛肉,有暖胃作用,为寒冬补益佳品。中医认为,牛肉有补中益气、滋养脾胃、强健筋骨、化痰息风、止渴止涎的功效。适用于中气下陷、气短体虚,筋骨酸软、贫血久病及面黄目眩之人食用。
随着人们对牛肉在需求量的增大,肉牛的养殖和屠宰业得以迅速的发展,由于养殖户的大小不一,加之环境的污染,使得屠宰企业的出产的肉制品的质量很不稳定。每个肉制品生成企业都在积极摸索和建立自己的检测系统,力求保障自己产品的安全,在牛肉制品的国家卫生标准中,对汞含量有严格的限定,牛肉中的汞含量不大于0.05mg/KG,因此对汞含量的检测尤为重要。
发明内容
本发明的目的是提供一种牛肉汞含量的检测方法,该发明的检测方法操作简单,适合工厂生产质检的使用。
本发明的目的是通过以下技术方案来实现的:
本发明的一种牛肉汞含量的检测方法,包括以下步骤:
(1)制备检测试纸:
取0.7%二苯碳酰二肼乙醚溶液与15%乳化剂OP水溶液按6∶1体积比混合均匀,加入中速定量滤纸浸渍15min,取出,避光、自然晾干,制成二苯碳酰二肼试纸,避光保存;
(2)制备汞含量为0.5mg/KG的试纸色卡:
取1000μg/mL标准汞溶液适量,加水适量并以HCl溶液调节pH至6.0,加水稀释并制成汞含量为0.5μg/mL的溶液作为标准汞对比溶液;取二苯碳酰二肼试纸,置于上述标准对比溶液中,计时5min,取出,置于白色水平背景上,立即拍照;照片经“画图”程序处理后形成二苯碳酰二肼纸标准色阶;
(3)称取牛肉样品,破碎,加入3倍量的10%HCl,微波消解,离心,取上清液,使用HCl和NaOH调整pH值在6.0,减压浓缩上清液至牛肉样品重量的1/10;
(4)将制备好的检测试纸放入步骤(2)中的浓缩后的上清液中,5min后,观察显色情况,拍照;
(5)将牛肉样品上清液的试纸显色深度和标准色阶比较,若牛肉样品上清液的试纸显色深度明显小于标准色阶,则判断为所抽检牛肉的汞含量低于国家标准;若牛肉样品上清液的试纸显色深度相当或大于标准色阶,则判断所抽检牛肉的汞含量有问题,继续使用更为精密的检测方法再行检测汞含量,判断汞含量是否超标。
优选的,所述步骤(1)中,二苯碳酰二肼试纸裁剪成1cm×1cm的小方片。
本发明的优点:

以下结合具体实施例对本发明作进一步说明:
实施例
制备检测试纸,供日常抽检检测使用:
取0.7%二苯碳酰二肼乙醚溶液与15%乳化剂OP水溶液按6∶1体积比混合均匀,加入中速定量滤纸浸渍15min,取出,避光、自然晾干,制成二苯碳酰二肼试纸,避光保存;
为了方便使用,可以将试纸裁剪成1cm×1cm的小方片;
通常根据5个月的试纸使用量来制备一次试纸。
制备汞含量为0.5mg/KG的试纸色卡:
取1000μg/mL标准汞溶液适量,加水适量并以HCl溶液调节pH至6.0,加水稀释并制成汞含量为0.5μg/mL的溶液作为标准汞对比溶液;取二苯碳酰二肼试纸,置于上述标准对比溶液中,计时5min,取出,置于白色水平背景上,立即拍照。照片经“画图”程序处理后形成二苯碳酰二肼纸标准色阶,保持标准色阶,供长期检测实验对比使用。
牛肉抽检检测实验:
(1)称取牛肉样品100g,破碎,加入3倍量的10%HCl,微波消解,离心,取上清液,使用HCl和NaOH调整pH值在6.0,减压浓缩上清液至10mL;为了更好取得显色效果,特将标准色阶的汞含量浓度设定为0.5μg/mL,相对于国标中规定的牛肉汞含量标准的10倍,提高汞含量的浓度,显色效果易于观察;同时在取样提取检测液时,也将检测液浓缩至10mL,与标准色阶的汞含量浓度相对应;
(2)将制备好的检测试纸放入步骤(1)中的浓缩后的上清液中,5min后,观察显色情况,拍照;
(3)将牛肉样品上清液的试纸显色深度和标准色阶比较,若牛肉样品上清液的试纸显色深度明显小于标准色阶,则判断为所抽检牛肉的汞含量低于国家标准;若牛肉样品上清液的试纸显色深度相当或大于标准色阶,则判断所抽检牛肉的汞含量有问题,继续使用更为精密的检测方法再行检测汞含量,判断汞含量是否超标。

1.一种牛肉汞含量的检测方法,其特征在于:包括以下步骤:
(1)制备检测试纸:
取0.7%二苯碳酰二肼乙醚溶液与15%乳化剂OP水溶液按6∶1体积比混合均匀,加入中速定量滤纸浸渍15min,取出,避光、自然晾干,制成二苯碳酰二肼试纸,避光保存;
(2)制备汞含量为0.5mg/KG的试纸色卡:
取1000μg/mL标准汞溶液适量,加水适量并以HCl溶液调节pH至6.0,加水稀释并制成汞含量为0.5μg/mL的溶液作为标准汞对比溶液;取二苯碳酰二肼试纸,置于上述标准对比溶液中,计时5min,取出,置于白色水平背景上,立即拍照;照片经“画图”程序处理后形成二苯碳酰二肼纸标准色阶;
