Description
技术领域
本发明涉及一种用于分析mRNA(信使核糖核酸)上调的癌细胞中亚精胺/精胺N1-乙酰基转移酶(SSAT)活性的方法,尤其涉及SSAT底物作为抗癌药物化合物以及在抗癌治疗中的用途。
背景技术
于2004年11月4日授予Sitar等人的专利号为6,811,967的美国专利,其完整公开内容在此被引入作为参考,该专利公开了通过检测SSAT底物的乙酰化形式来利用SSAT底物用于分析SSAT酶活性的方法。SSAT底物可包括金刚烷胺,其中金刚烷胺的代谢部分地由可诱导酶SSAT作用产生乙酰化的代谢产物N-乙酰基金刚烷胺。还公开了SSAT的活性与病理学状态的相关性。

具体实施方式
本发明公开了一种将亚精胺/精胺N1-乙酰基转移酶(SSAT)底物作为抗癌药物化合物的方法。
人体肿瘤细胞株HEK-293、Malme-3M、HeLa、PC-3和US-02中SSAT的相对表达水平是通过逆转录-定量聚合酶链反应分析法(RT-qPCR分析法)测定的,并且如图1和图2所示,观察到Malme-3M中SSAT的相对表达量最高,采用GAPDH归一化时,Malme-3M中SSAT的相对表达量比对照HEK-293细胞株多11倍,采用HPRT1归一化时,Malme-3M中SSAT的相对表达量比对照HEK-293细胞株多58倍。PC-3中SSAT的相对表达水平为第二高,分别用GAPDH和HPRT1归一化时,PC-3中SSAT的相对表达水平相对HEK-293分别相差大约3倍和7倍。HeLa和U2-OS中SSAT的相对表达水平均低于HEK-293。SSAT的表达水平还与N-乙酰基金刚烷胺代谢物的形成进行了比较,结果显示SSAT的表达与N-乙酰化代谢活性具有因果关系。

基于文献的SSAT表达数据,从肺癌、乳腺癌及前列腺癌中各选出三种细胞株,并用MTT分析法进行潜在的细胞毒性筛选。SSAT无表达的人体骨肉瘤细胞株U2-OS作为阴性对照细胞株。本发明使用了以下人体细胞株:

实验步骤
每种人体癌细胞株分别与一系列试验浓度的四种单胺试验药物共孵育。用半数最大抑制浓度或IC50表示的细胞毒性,根据(二甲基-2-噻唑基)-2,5-二苯基-2H-溴代四唑或MTT分析法测定。平行地,用RT-qPCR分析法测定上述细胞株中SSAT的表达水平。
人体细胞株的准备
分别将已经贴壁的10个细胞株的细胞用胰酶-EDTA消化收集,离心沉淀,再分别用适合的培养基重悬得到每个细胞株的细胞悬液。
MTT细胞毒性分析法评价IC50
准确称量各单胺试验药物(金刚烷胺、金刚烷乙胺、多巴胺和左旋多巴)以及阳性对照(亚精胺),溶解并进一步用无菌水稀释成一系列的溶液,其浓度为它们的目标孵育浓度的100倍。金刚烷胺、金刚烷乙胺和多巴胺的目标孵育浓度为0.03,0.1,0.3,1,3,10,30,100,300和1000μM。左旋多巴和阳性对照亚精胺的目标孵育浓度为0.1,0.3,1,3,10,30,100,300和1000μM。
每种受试细胞株的孵育在每个药物浓度下重复进行3次。与每个试验药物的处理相平行,每种细胞株同样用亚精胺作为阳性对照进行处理。
将等份的细胞悬液加入到96孔培养板的每个孔中,然后将培养板于37℃、含95%空气及5%二氧化碳的高湿度环境中过夜孵育。次日将每孔换为新鲜培养基,加入等份的合适的试验药物溶液,以细胞培养体积的1%加入以达到目标试验浓度。空白培养基用于代替底物溶液以作为空白对照(即0μM底物)。然后将培养板放回37℃、含95%空气及5%二氧化碳的高湿度环境中孵育三天。
细胞与底物孵育三天以后,将等份的浓度为5mg/mL的MTT加到每个孔中,然后孵育1-3小时。孵育之后,用DMSO(二甲基亚砜)代替培养基使甲瓒溶解。取自每个孔的等份在96孔平底板上用酶标仪测量550nm或555nm处的吸光度,DMSO用作背景吸光度校正。
用RT-qPCR检测SSAT表达水平
用QIAshredderTM试剂盒和RNeasyTM微量试剂盒进行RNA提取,这两种试剂盒均购自Qiagen公司,地址为美国加利福尼亚州瓦伦西亚特恩贝里街27220号200室。每种受试细胞株的重悬细胞用RNeasyTM裂解缓冲液裂解。然后用RNeasyTM微吸附柱从裂解物中提取RNA。经Nanodrop分光光度测量确认每个提取的样品中有足够的总RNA浓度。
SSAT的RT-qPCR使用同样购自Qiagen公司的QuantiTect SYBR Green RT-PCR试剂盒进行测量。每个样品的反应混合物包括QuantiTect SYBR Green RT-PCR预混液、无RNA酶的水、SSAT的PCR引物以及从每个样品中提取的RNA。
上述每个样品使用相应的PCR引物平行测定管家基因GAPDH和HPRT1的表达水平。SSAT的表达水平使用GAPDH或HPRT1作为内参进行归一化,并表达为相对于A549的SSAT表达水平的倍数差异。
仪器和RT-qPCR程序:
仪器:Applied Biosystems Cycler
反转录:20分钟,50℃;20℃/秒
起始步骤:15分钟,95℃;20℃/秒
循环步骤:
变性:15秒,94℃;20℃/秒
退火:20秒,55℃;20℃/秒
延伸:30秒,72℃;20℃/秒
循环次数:40
结果与讨论
图4中示出了每个SSAT单胺试验药物对每种受试人体癌细胞株的表示为IC50的细胞毒性的总结。细胞毒性基于将一系列浓度的每个试验药物对每种细胞株进行处理来测定。三天孵育期结束后,用MTT分析法测定细胞毒性。以抑制百分率表示的细胞毒性水平对试验浓度作图推导出IC50值,如图5至14所示。与每个药物的处理相平行,每种细胞株同样用亚精胺作为阳性对照进行处理,并且测量亚精胺的IC50值用于比较。
数据显示,在评价的所有细胞株中,四个单胺试验药物的IC50值在34.1μM至1605μM之间,大多数在100μM至500μM之间。总体而言,最有效的单胺试验药物是作用于阴性对照细胞U2-OS骨肉瘤细胞株的多巴胺,其IC50值为34.1μM。紧随其后的是作用于乳腺癌细胞株BT-549的多巴胺,其IC50值为52.0μM。效果最差的试验药物是作用于NCI-H23(肺癌)和BT-549(乳腺癌)的金刚烷胺,其IC50值分别为1605μM和1158μM。
这四种单胺试验药物的IC50值与阳性对照亚精胺比相对较高,从3.72μM到32.7μM,表明单胺试验药物的细胞毒效力低于亚精胺。对于每种细胞株,注意到大致上单胺试验药物的IC50值(细胞毒性)的排序表现为与多胺类亚精胺阳性对照的排序相关联。这种细胞毒性排序上明显的相似可能揭示了单胺药物与多胺药物之间共同的机制。
图4、图15和图16示出了在受试人体细胞株中SSAT的相对表达水平的总结。受试细胞株中SSAT的相对表达水平是基于RT-qPCR分析法测得的。每个样品测得的SSAT的循环阈值以管家基因GAPDH或HPRT1的循环阈值作为内参进行归一化,以校正不同样品间RNA数量和质量可能存在的差异。结果由SSAT表达水平相对于A549中表达水平的倍数差异来表示。
结果显示,LNCaP被观察到具有最高的SSAT相对表达水平,用GAPDH归一化时,大约比A549的SSAT相对表达水平多5倍,用HPRT1归一化时,大约比A549的SSAT相对表达水平多3倍。T-47D具有第二高的表达水平,其SSAT表达相对A549大约有2倍差异。SSAT无表达的细胞株U2-OS(阴性对照)具有如预期的最低的SSAT表达水平。
将RT-qPCR结果与每个试验药物对每种细胞株的IC50值相比较,以使SSAT的表达水平与单胺试验药物的细胞毒性相关联。如图17所示,从SSAT表达水平相对以1/IC50表示的药效的相关性来看,可以有趣地发现,大致上高SSAT表达被观察到与高细胞毒性效力相关,然而对有的肿瘤细胞株,低SSAT表达被观察到与高细胞毒性相关。过表达SSAT的细胞株例如LNCaP(>5倍变化)和T-47D(>2倍变化),当用金刚烷胺和金刚烷乙胺处理时,显示出高细胞毒性(低IC50值),但是当使用其它单胺处理时,没有观察到一致的细胞毒性。与此相反,不表达SSAT的阴性对照细胞株U2-OS,当用多巴胺和左旋多巴处理时,表现出低IC50值(高细胞毒性)。该结果因而揭示了,所选的单胺试验药物对SSAT的竞争性抑制表现为只在具有SSAT敏感表型的特定肿瘤上有效。还注意到SSAT不敏感的肿瘤表型也在评价的10种人体肿瘤细胞株当中。
本发明评价了单胺试验药物金刚烷胺、金刚烷乙胺、多巴胺和左旋多巴对于肺癌、乳腺癌和前列腺癌中各选出的三种SSAT过表达的人体癌细胞株的细胞毒性。在所有九种受试肿瘤细胞株中,观察到单胺试验药物的细胞毒性效力与作为多胺阳性对照的亚精胺相比为低。大致上,单胺试验药物的细胞毒性的排序表现为与多胺亚精胺的细胞毒性的排序相关联,揭示了单胺药物与多胺药物之间共同的机制。因此得到结论,所述的单胺试验药物及其它SSAT底物可作为抗癌药物化合物使用并用于抗癌治疗。
本领域技术人员能够理解,上述提供的诸多细节仅是举例说明,并非要限制本发明的保护范围。本发明的保护范围应参考所附权利要求确定。
Claims (12)
1.一种包含亚精胺/精胺N1-乙酰基转移酶底物的抗癌药物化合物。
2.如权利要求1所述的抗癌药物化合物,其中所述亚精胺/精胺N1-乙酰基转移酶底物为单胺。
3.如权利要求1所述的抗癌药物化合物,其中所述亚精胺/精胺N1-乙酰基转移酶底物为金刚烷胺。
4.如权利要求1所述的抗癌药物化合物,其中所述亚精胺/精胺N1-乙酰基转移酶底物为金刚烷乙胺。
5.如权利要求1所述的抗癌药物化合物,其中所述亚精胺/精胺N1-乙酰基转移酶底物为多巴胺。
6.如权利要求1所述的抗癌药物化合物,其中所述亚精胺/精胺N1-乙酰基转移酶底物为左旋多巴。
7.亚精胺/精胺N1-乙酰基转移酶底物用于治疗癌症的用途。
8.如权利要求7所述的亚精胺/精胺N1-乙酰基转移酶底物的用途,其中所述亚精胺/精胺N1-乙酰基转移酶底物为单胺。
9.如权利要求7所述的亚精胺/精胺N1-乙酰基转移酶底物的用途,其中所述亚精胺/精胺N1-乙酰基转移酶底物为金刚烷胺。
10.如权利要求7所述的亚精胺/精胺N1-乙酰基转移酶底物的用途,其中所述亚精胺/精胺N1-乙酰基转移酶底物为金刚烷乙胺。
11.如权利要求7所述的亚精胺/精胺N1-乙酰基转移酶底物的用途,其中所述亚精胺/精胺N1-乙酰基转移酶底物为多巴胺。
12.如权利要求7所述的亚精胺/精胺N1-乙酰基转移酶底物的用途,其中所述亚精胺/精胺N1-乙酰基转移酶底物为左旋多巴。
