Abstract
本发明涉及生物制品技术领域,具体涉及一种乙醇检测试剂盒质控品的制备方法,包括以下步骤:在成品人血浆中加入0.5%的氯化钙,37℃孵育2h后5000r/min离心10min,过滤去除纤维蛋白,在所得的上清液中加入5%的甘油,1%的吐温80和0.2%的叠氮钠,混合均匀,待用;将注射用大豆油加热至65℃,加入10%的改性大豆磷脂,混合均匀,按体积比1∶5趁热加入到上述加入过添加物的成品人血浆溶液中,混合均匀,加入0‑0.5%的乙醇,混合均匀即可。

本发明涉及生物制品技术领域,具体涉及一种乙醇检测试剂盒质控品的制备方法。
背景技术
近年来,酒后引起的刑事案件和交通事故案不断上升,2011年5月1日《中华人民共和国刑法修正案(八)》正式实施,醉酒驾驶作为危险驾驶罪被追究驾驶人刑事责任,有关酒驾的话题再次成为人们关注的焦点。目前,许多国家都制定了血液中乙醇浓度与交通事故责任认定的关系,建立了多种能够检测血液中乙醇含量的方法,为了保证这些方法对样本检测结果的准确性,经常对检测系统进行校准,及时进行质量控制就显得尤为重要。乙醇极易挥发,如何保证作为质控品的乙醇溶液浓度稳定一直是困扰乙醇检测方面的难题。
发明内容
本发明针对现有技术的不足,提供一种乙醇检测试剂盒质控品的制备方法,以保证乙醇测定的稳定性。
本发明的乙醇检测试剂盒质控品的制备方法,其特征在于包括以下步骤:
(1)在成品人血浆中加入0.5%(质量体积比)的氯化钙,37℃孵育2h后5000r/min离心10min,过滤去除纤维蛋白。
(2)在步骤(1)所得的上清液中加入5%(体积比)的甘油,1%(体积比)的吐温80和0.2%(质量体积比)的叠氮钠,混合均匀。
(3)将注射用大豆油加热至65℃,加入10%(质量体积比)的改性大豆磷脂,混合均匀。
(4)将步骤(3)所得溶液按体积比1∶5趁热加入到步骤(2)所得溶液中,混合均匀。
(5)在步骤(4)所得溶液中加入0-0.5%(体积比)的乙醇,混合均匀即可。
上述乙醇检测试剂盒质控品的制备方法,选用大豆油、改性大豆磷脂、甘油、吐温80与经氯化钙处理后的成品人血浆形成乳液体系,降低了作为质控品的乙醇的挥发性,保证了乙醇测定的稳定性。
具体实施方式
实施例1
(1)成品人血浆200ml,加入1g氯化钙,37℃孵育2h,5000r/min离心10min,过滤。
(2)取步骤(1)所得的上清液100ml,加入5ml甘油,1ml吐温80和0.2g叠氮钠, 混合均匀。
(3)取注射用大豆油20ml,加热至65℃,加入2g的改性大豆磷脂,混合均匀。
(4)量取步骤(3)所得溶液20ml趁热加入到步骤(2)所得溶液100ml中,混合均匀。
(5)取步骤(4)所得溶液100ml,加入0.1ml的乙醇,混合均匀。
实施例2
(1)成品人血浆200ml,加入1g氯化钙,37℃孵育2h,5000r/min离心10min,过滤。
(2)取步骤(1)所得的上清液100ml,加入5ml甘油,1ml吐温80和0.2g叠氮钠,混合均匀。
(3)取注射用大豆油20ml,加热至65℃,加入2g的改性大豆磷脂,混合均匀。
(4)量取步骤(3)所得溶液20ml趁热加入到步骤(2)所得溶液100ml中,混合均匀。
(5)取步骤(4)所得溶液100ml,加入0.3ml的乙醇,混合均匀。

1.一种乙醇检测试剂盒质控品的制备方法,其特征在于包括以下步骤:
(1)在成品人血浆中加入0.5%质量体积比的氯化钙,37℃孵育2h后5000r/min离心10min,过滤去除纤维蛋白;
(2)在步骤(1)所得的上清液中加入5%体积比的甘油,1%体积比的吐温80和0.2%质量体积比的叠氮钠,混合均匀;
(3)将注射用大豆油加热至65℃,加入10%质量体积比的改性大豆磷脂,混合均匀;
(4)将步骤(3)所得溶液按体积比1∶5趁热加入到步骤(2)所得溶液中,混合均匀;
(5)在步骤(4)所得溶液中加入0-0.5%体积比的乙醇,混合均匀即可。
