Abstract

本发明公开一种细叶云南松松针精油的应用,将所述松针精油作为大肠杆菌抑制剂。与其它松的松针精油相比,本发明的细叶云南松松针精油对大肠杆菌具有良好的抑制效果。

细叶云南松松针精油的应用
技术领域
本发明涉及细叶云南松松针精油的应用。
背景技术
松针药用历史悠久,具有止痒、明目安神、祛风活血、解毒等功效,用于治疗冻疮、流行性感冒、跌打肿痛、风湿性关节炎等。现代药理学研究表明,松针具有抑菌、降血压、降血脂、镇痛、抗突变、祛痰、抗感染等作用。松针中含有钙、钾、铁等无机盐和维生素A、维生素C,19种脂肪酸,17种氨基酸等40余种营养成分,而且还含有挥发油类、木脂素类、色素、黄酮类。因此松针的开发利用价值不可忽视。
发明内容
本发明要解决的技术问题是克服现有的缺陷,提供了一种细叶云南松松针精油的应用。
为了解决上述技术问题,本发明提供了如下的技术方案:
细叶云南松松针精油的应用,所述松针精油作为大肠杆菌抑制剂。
所述松针精油通过如下步骤得到:将松针切碎置于挥发油提取器中,按料液为1:4~1:20加入水,浸泡4-16小时,然后加热回流提取1-2小时。
进一步,料液比为1:12,浸泡4小时,回流提取2小时。
具体实施方式
以下对本发明的优选实施例进行说明,应当理解,此处所描述的优选实施例仅用于说明和解释本发明,并不用于限定本发明。
实施例1
称取100g阴干的细叶云南松松针,将针叶切成长度2cm左右,把切碎的松针置于挥发油提取器中,按料液比为1:12加入水,浸泡4h后,加热回流提取精油,提取时间为2h,提取完毕后读取精油的体积,并平行试验三次,提取率为0.62(mL·100g-1,提取率以干重计)。
实施例2
称取100g阴干的细叶云南松松针,将针叶切成长度2cm左右,把切碎的松针置于挥发油提取器中,按料液比为1:10加入水,浸泡4h后,加热回流提取精油,提取时间为0.5h,提取完毕后读取精油的体积,并平行试验三次,提取率为0.46(mL·100g-1,提取率以干重计)。
实施例3
称取100g阴干的细叶云南松松针,将针叶切成长度2cm左右,把切碎的松针置于挥发油提取器中,按料液比为1:15加入水,浸泡12h后,加热回流提取精油,提取时间为2h,提取完毕后读取精油的体积,并平行试验三次,提取率为0.44(mL·100g-1,提取率以干重计)。
抑菌活性测试
采用滤纸片扩散法测定拉雅松和细叶云南松松针精油(均按实施例1制得)的抑菌活性,以阿莫西林颗粒(对细菌)和克霉唑(对霉菌)作为阳性对照。具体实验过程如下:在标准净化台上,用无菌移液器吸取0.2mL的测试菌悬液,将其均匀涂布于固体培养基上,正置于培养箱1h(细菌于37℃恒温培养,真菌于28℃恒温培养)。1h后移于净化台中,将直径6mm的无菌双层滤纸片放置于培养皿中心,然后用无菌移液器分别移取10μL的待测样品(精油和阳性对照)垂直加到滤纸片上,培养皿先正置培养1h,然后将含细菌菌株的培养皿在37℃下倒置恒温培养24h,含真菌菌株的培养皿在28℃下倒置恒温培养48h。测量抑菌圈的直径,以此衡量松针精油的抑菌效果。
拉雅松和细叶云南松松针精油的抑菌活性
附注:a:NA没有效果;b:NT没有测试
细菌:大肠杆菌(Escherichia coli)、沙门氏菌(Salmonella sp.)、枯草芽孢杆菌(Bacillus subtilis)、金黄色葡萄球菌(Staphylococus aureus)。
霉菌:黑曲霉(Aspergillus niger)、橘青霉(Penicillium citrinum.)、根霉(Rhizopus sp.)、黄曲霉(Aspergillus flavus)。
酵母:热带假丝酵母(Candida tropicalis)、红酵母(Rhodotorula sp.)、酿酒酵母(Saccharomyces cerevisiae)。
如上可以看出,与拉雅松松针精油对大肠杆菌没有抑制效果相比,细叶云南松松针精油对大肠杆菌具有良好的抑制效果。

1.细叶云南松松针精油的应用,所述松针精油作为大肠杆菌抑制剂。
2.根据权利要求1所述细叶云南松松针精油的应用,其特征在于,所述松针精油通过如下步骤得到:
将松针切碎置于挥发油提取器中,按料液为1:4~1:20加入水,浸泡4-16小时,然后加热回流提取1-2小时。
3.根据权利要求2所述细叶云南松松针精油的应用,其特征在于:所述料液比为1:12,浸泡4小时,回流提取2小时。
