Abstract

背景技术
已知森林生物质是地球上最重要的生物质,并且随着木材工业产生大量的木屑,其可通过提取用作重要的蔬菜来源用于进一步处理和膳食成分的物价稳定措施。可通过森林生物质的溶剂提取获得的可提取的类黄酮尤其引人注意,因为它们容易从不同类型的森林和木材转化木屑中获得。类黄酮的一种最众人皆知的生物活性特性是它们的抗氧化剂和抗炎症活性。呈现对森林树木提取物的抗氧化剂能力最重要的结果并且与为来自富含生物活性类黄酮分子的健康食物提取物获得的那些比较[1]。类黄酮二氢槲皮素(紫杉叶素)是最有效的天然抗氧化剂和抗炎症化合物之一[2,3]。重点还放在松柏木材转化的木屑,比如根端圆木和树皮,因为这些材料代表大部分膳食纤维阿拉伯半乳聚糖的特别丰富来源[4,5,6,7,8]。更高的阿拉伯半乳聚糖含量通常伴随着更大量的类黄酮物质,比如二氢槲皮素(紫杉叶素)[9],其在落叶松硬木(包括茎干、树皮和根)中出现已经被文献[10-22]所记载。
本发明之前,已知松柏木材物种的不同部分的提取物包含各种化合物,比如天然抗氧化剂二氢槲皮素(紫杉叶素)、天然非淀粉多糖,其是来自硬木,主要来自植物落叶松属,尤其来自拉里克斯卡加德里(Larixcajanderi)、拉里克斯捷卡诺夫斯基(Larixczekanowskii)、拉里克斯达夫里卡(Larixdahurica)、拉里克斯梅林尼(Larixgmelinii)、拉里克斯卡茨恰蒂卡(Larixkamtschatica)、拉里克斯卢西卡(Larixrussica)、拉里克斯西比里卡(Larixsibirica)、拉里克斯苏卡捷威(Larixsukaczewii)的可溶的膳食纤维阿拉伯半乳聚糖,即落叶松阿拉伯半乳聚糖,其可定义为包含大量天然抗氧化剂,主要是天然与纤维基质相关的二氢槲皮素(紫杉叶素)的纤维,其具有下述具体特征:1.膳食纤维含量,大于70%干物质基。2.一克的膳食纤维落叶松阿拉伯半乳聚糖应具有基于ORAC值等于至少1,000umol TE/克的抑制脂质氧化的能力并且通常等于2,000-4,000umol TE/克。3.一克的膳食纤维落叶松阿拉伯半乳聚糖具有的基于细胞的抗氧化剂保护(CAP-e)以保护活细胞遭受氧化损伤的能力是,至少6CAP-e单位每克,其中CAP-e值以五倍子酸当量(GAE)单位计。4.抗氧化剂能力必须是固有特性,源自材料的天然成分(在消化液中可溶)而不是通过添加抗氧化剂或通过之前的化学或酶处理。本发明涉及落叶松木材提取物比如二氢槲皮素(紫杉叶素)、阿拉伯半乳聚糖和结合二氢槲皮素(紫杉叶素)的阿拉伯半乳聚糖在动物饲料产品应用中的用途,其中落叶松木材提取物建议用作膳食成分、天然抗氧化剂、食物添加剂和食物防腐剂。
传统的动物饲料膳食主要基于谷物和大豆粕。但是,最近这些年,可选产品比如豌豆、大豆、向日葵粕、菜籽粉、羽扇豆、谷类副产物和甜菜浆逐渐收到关注。在一些此类产品中,例如向日葵粕、菜籽粉、羽扇豆、谷类副产物和甜菜浆,低的消化率通常限制它们以显著的量包含在动物饲料膳食中。该低的消化率与这些产品中碳水化合物馏分的组成(主要由非淀粉多糖构成)相关。非淀粉多糖在小肠中不能被单胃动物的消化酶消化,并且因此不能为动物提供它们所有的能量潜能。已知这些多糖的水解解决了两个问题,一个是动物福利,另一个与改进的生产的经济性相关。
与人类一样,农场动物在食用特定食物时最健康。母牛具有设计为消化草的胃。猪可消化草、玉米、谷物、大豆和其他植物。小鸡和火鸡可食用植物以及牧场出现的小虫和蠕虫。当给动物喂食可包括动物产品、抗生素和其他非天然物质(比如口香糖(chewing gum)和鸡粪)的常规的(或工业)饲料时,它们的健康处于危险中。并且当动物不健康时,由其制造的肉类和其他产品也较不健康。已经用膳食纤维治疗动物的胃肠不适。膳食纤维是包括许多物质(包括纤维素,半纤维素比如落叶松阿拉伯半乳聚糖、寡糖、果胶、树胶、蜡和木质素)的一般概念。更通用的定义是“膳食中植物材料的内源性组分”,其抵抗人或动物(肠道)酶的消化,即,主要是非淀粉多糖和木质素。膳食纤维可以是可溶的或不溶解的。
膳食纤维抵抗人和动物消化酶水解,但是可被结肠微生物群落发酵。一般而言,可溶的纤维比不溶解的纤维更容易发酵。这些物质的主要生理学作用在于胃排空和结肠传递时间,并且可导致改善的葡萄糖耐受和降低的淀粉消化。膳食纤维的发酵导致增加细菌生物质,增加排泄量,降低结肠内pH,原因是产生短链脂肪酸和产生作为代谢终产品的各种气体[24]。落叶松阿拉伯半乳聚糖在治疗门体静脉脑病变方面也可具有一些价值,因为其倾向于减少氨吸收且没有猛烈通便,而例如使用乳果糖则不这样[25]。
已经报道了源自植物的多酚(尤其类黄酮,例如,具有苯基-C3-苯基结构的化合物,其中苯环用一个或多个羟基基团官能化)显著降低人粪便中的产气荚膜梭菌和其他梭菌属物种(腐败细菌)的量,并且显著增加双歧杆菌(产酸菌)的量[26]。类黄酮和其他酚类化合物通过清除活性氧和氯物质在胃肠道(GI)中展示直接的保护作用。它们可抑制胃中血红素蛋白诱导的过氧化反应。它们能够通过源自HNO2的活性氮物质抑制DNA碱基脱氨基作用[27]。酚类可在胃肠道中上调毒素代谢或抗氧化剂防御酶[28]。它们可螯合氧化还原活性过渡金属离子并且降低它们的促氧化剂潜能[29,30]。膳食铁通常不能被完全吸收,尤其在高脂肪饲料膳食中。未吸收的膳食铁进入粪便,其中它可表示对结肠和直肠的促氧化剂挑战[31]。的确,高脂肪和低纤维的膳食可使该促氧化剂作用恶化[32]。通过螯合铁,酚类化合物可有助于减轻结肠铁的促氧化剂作用。现在已经发现一类类黄酮有效治疗寄生虫感染,对于治疗哺乳动物,尤其人、牛、绵羊、山羊、马、猪、家禽、狗和猫有显著益处[33]。尤其这些类黄酮具有期望的口服施用的品质。
US 6,558,693 B公开了包括下述酶的动物饲料添加剂,所述酶比如是阿拉伯半乳聚糖内切-1,4-β-牛乳糖苷酶和/或阿拉伯半乳聚糖内切-1,3-β-牛乳糖苷酶。
US 6241983公开了促进胃肠健康的组合物,其包含有效量的有益的人肠道微生物和有效量的膳食纤维。
US 6,087,092公开了包含结合多酚的半纤维素的组合物,制备该组合物的方法,和用组合物治疗人或动物的用于通过向动物施用有效量的组合物增加动物生长速度、改善饲料效率和减少断奶后腹泻的方法。
发明内容
所以,本发明的目的是提供便宜的饲料添加剂,尤其是落叶松木材提取物其作为天然化合物在动物饲料膳食中被认为具有许多益处,用于改善动物的饲料效率、消化的效率和总体健康,以改善动物健康、福利和表现/生产效率。
现在已经发现命名为落叶松木材提取物的特定木材提取物并入动物饲料尤其有益的,尤其当通过包括可溶的膳食纤维和类黄酮一起并入时。
因此,在其第一方面,本发明提供包括有效量的可溶的膳食纤维和类黄酮的动物饲料添加剂。
在另一方面,本发明提供改善动物膳食的饲料效率的方法,该方法包括将本发明的动物饲料添加剂补充至单胃动物。
在仍另一方面,本发明提供预先处理动物饲料的方法,通过该方法处理动物饲料以接受落叶松木材提取物的作用。
优选的待喂养的动物包括,但不限于母牛、猪、家禽、小牛、马和家养宠物。该组合物优选地作为饲料添加剂施用至动物,剂量水平在按饲料重量计0.1至30%之间,优选地按饲料重量计0.1和10%之间。优选地,组合物作为添加至饲料配方或添加至饮品的粉末施用至动物。

在公开和描述本组合物和使用方法之前,应当理解本发明不限于具体的实施例、方法步骤,和本文公开的材料,因为这些方法步骤和材料可稍微改变。也应理解,本文使用的术语用于仅仅描述具体实施方式的目的并且不旨在是限制性的,因为本发明的范围仅仅受所附的权利要求和其等价物限制。
本发明提供了动物饲料添加剂,其包括有效量的落叶松阿拉伯半乳聚糖和二氢槲皮素(紫杉叶素)。在优选的实施方式中,该落叶松阿拉伯半乳聚糖是定义为膳食纤维、益生元的非淀粉可溶性多糖并且主要提取自植物落叶松属,尤其提取自拉里克斯卡加德里(Larixcajanderi)、拉里克斯捷卡诺夫斯基(Larixczekanowskii)、拉里克斯达夫里卡(Larixdahurica)、拉里克斯梅林尼(Larixgmelinii)、拉里克斯卡茨恰蒂卡(Larixkamtschatica)、拉里克斯卢西卡(Larixrussica)、拉里克斯西比里卡(Larixsibirica)、拉里克斯苏卡捷威(Larixsukaczewii)。在另一优选的实施方式中,饲料添加剂包括二氢槲皮素(紫杉叶素),一种定义为抗氧化剂、抗炎症、抗微生物化合物的类黄酮,并主要提取自如上述植物属落叶松。在第三种优选的实施方式中,动物饲料添加剂包括有效量的定义为膳食纤维的结合二氢槲皮素(紫杉叶素)的落叶松阿拉伯半乳聚糖,其包含大量的天然与纤维基质相关的天然抗氧化剂,其中落叶松阿拉伯半乳聚糖结合主要提取自如上述植物落叶松属的二氢槲皮素(紫杉叶素)。
在本发明的背景下,动物饲料添加剂是落叶松木材提取物制剂,其包括类黄酮二氢槲皮素(紫杉叶素)和/或非淀粉多糖阿拉伯半乳聚糖和/或结合二氢槲皮素(紫杉叶素)的阿拉伯半乳聚糖和适当的载体和/或赋形剂,并以适于添加至动物饲料的形式提供该落叶松木材提取物制剂。可根据本领域已知的方法制备本发明的动物饲料添加剂并且可为干燥或液体制剂的形式。
在具体的实施方式中,本发明的动物饲料添加剂是颗粒状落叶松木材提取物,其可容易与饲料组分混合,或更优选地,形成预混合料的组分。颗粒状落叶松木材提取物可以有包衣或没有包衣。落叶松木材提取物颗粒的颗粒尺寸优选地与饲料和预混合料组分兼容。这提供安全的和方便的将落叶松木材提取物并入饲料的方式。
在另一具体的实施方式中,本发明的动物饲料添加剂是稳定的液体组合物,其可以是水性或油基浆液。在使动物饲料组合物粒化之后,液体组合物可任选地添加至该组合物。

在其最优选的实施方式中,本发明提供动物饲料添加剂,其仅仅包括落叶松阿拉伯半乳聚糖和二氢槲皮素(紫杉叶素)。在动物饲料中,类黄酮比如二氢槲皮素(紫杉叶素)附着至落叶松阿拉伯半乳聚糖,其是阿拉伯半乳聚糖基质的主要成分。
二氢槲皮素(紫杉叶素)是具有分子结构的化合物,其基于由通过三个碳连接结合的两个芳环组成的C6-C3-C6骨架,没有C2–C3双键并且在第2和3位具有两个手性碳原子。见图1。类黄酮结构的A环是乙酸酯衍生的(3x C2)并且C和B环源自肉桂酸衍生物(类苯基丙烷通路)。从而,B环可以为(2S)-或(2R)-构造。二氢黄酮醇二氢槲皮素(紫杉叶素)的C-3原子具有氢原子和羟基基团,并且所以是另外的不对称中心。因此,对于每个二氢黄酮醇结构可能有四个立体异构体,(2R,3R)、(2R,3S)、(2S,3R)和(2S,3S)。天然存在的二氢黄酮醇中已经发现了所有四个构型,但是(2R,3R)-构型是迄今最常见的。
提取类黄酮化合物二氢槲皮素(紫杉叶素)的方法建议在真空系统中进行提取,使用能量以加热溶剂混合物(两个或更多个接触木材颗粒的极性试剂的混合物),以便从木材颗粒提取类黄酮二氢槲皮素(紫杉叶素)。提取条件包括固体/液体比、提取时间、提取温度和真空度,这些条件可在通过本领域已知的方法分离所述类黄酮之后进行优化。
落叶松阿拉伯半乳聚糖是一类长的、高度分枝的低分子和高分子多糖,MW:3,000-120,000。已经集中研究了来自不同硬木物种的水溶性阿拉伯半乳聚糖的分子结构。落叶松阿拉伯半乳聚糖由b-D-(1fi3)-吡喃半乳糖单元(b-D-(1fi3)-Galp)主链组成,其中大部分主链单元在C-6[fi3,6)-Galp-(1fi]上携带侧链。几乎一半的这些侧链是b-D-(1fi6)-Galp二聚体,并且约四分之一是单个Galp单元[图2]。剩下的包含三个或更多个单元。阿拉伯糖以吡喃糖(Arap)和呋喃糖(Araf)形式存在,作为阿拉伯二糖基基团[b-L-Arap-(1fi3)-LAraf-(1fi]或作为末端a-L-Araf例如单GeL-阿拉伯呋喃糖单元或3-O-(β-L-阿拉伯吡喃糖基)-α-L-阿拉伯呋喃糖基单元连接至侧链。
提取多糖物质或落叶松阿拉伯半乳聚糖和结合二氢槲皮素(紫杉叶素)的落叶松阿拉伯半乳聚糖的方法建议在真空系统中进行提取,使用能量加热溶剂混合物(两个或更多个极性试剂的混合物),接触木材颗粒以便从木材颗粒提取多糖物质。提取条件包括固体/液体比、提取时间、提取温度和真空度,这些条件可在通过本领域已知的方法分离所述多糖物质之后进行优化。
与利用其他来源相比,从落叶松木材提取落叶松阿拉伯半乳聚糖和结合类黄酮二氢槲皮素(紫杉叶素)的落叶松阿拉伯半乳聚糖是非常有利。结果,根据上述方法获得的约70-80%的提取物可以是多糖试剂(一种或多种),表明多糖试剂由类黄酮试剂构成。另一感兴趣的特征是特定物质可以是源自多糖基团物质的主要化合物。例如,阿拉伯半乳聚糖可以是约90-98%的多糖且二氢槲皮素(紫杉叶素)可以是约75-85%的源自落叶松木材的类黄酮。
根据本发明考虑的木材提取物可源自任何来自松柏木材物种的可用资源,尤其来自松科的那些。在优选的实施方式中,木材提取物源自植物落叶松属。优选地植物落叶松属是拉里克斯卡加德里(Larixcajanderi)、拉里克斯捷卡诺夫斯基(Larixczekanowskii)、拉里克斯达夫里卡(Larixdahurica)、拉里克斯梅林尼(Larixgmelinii)、拉里克斯卡茨恰蒂卡(Larixkamtschatica)、拉里克斯卢西卡(Larixrussica)、拉里克斯西比里卡(Larixsibirica)、拉里克斯苏卡捷威(Larixsukaczewii)。在更优选的实施方式中,植物落叶松属是拉里克斯达夫里卡(Larixdahurica)、拉里克斯梅林尼(Larixgmelinii)、拉里克斯卢西卡(Larixrussica)、拉里克斯西比里卡(Larixsibirica),更具体地是拉里克斯达夫里卡(Larixdahurica)、拉里克斯西比里卡(Larixsibirica)。
落叶松木材提取物可作为饲料添加剂添加至动物饲料。动物饲料包括但不限于牛饲料、家禽饲料、猪饲料、马饲料、早期断奶小牛的饲料,以及狗和猫食品。包括落叶松木材提取物,即,类黄酮二氢槲皮素(紫杉叶素)和/或非淀粉多糖阿拉伯半乳聚糖和/或结合二氢槲皮素(紫杉叶素)的阿拉伯半乳聚糖的饲料添加剂的例子是在食品/饲料的制造或准备用于消费时添加至其中的任何组合物/剂型。
典型的剂量范围是按动物饲料重量计的0.1至30%之间,优选地按动物饲料重量计的0.1和10%之间。
包括宠物食品组合物的功能饲料的例子是旨在提供必要的膳食需求,以及调剂物(treats)(例如,狗饼干)或其他饲料添加剂的饲料。包括根据本发明的组合物的动物饲料可以为干燥组合物(例如,粗磨食物)、半潮湿组合物、潮湿组合物的形式,或其任何混合形式。可选地或另外,动物饲料是补剂,比如肉汤、饮用水、酸奶、粉末、悬浮液、咀嚼物、调剂物(例如,饼干)或任何其他递送形式。
本发明进一步方面涉及饲料添加剂或添加剂组合物,比如待添加至一个或多个可食用饲料物质(一种或多种)或成分(一种或多种),例如制备饲料组合物或用于补充至现有饲料以形成饲料组合物。
所谓的预混合料是本发明动物饲料添加剂的例子。预混合料指优选地一个或多个微成分与稀释剂和/或载体的均匀混合物。预混合料用于促进微成分在更大混合物中的分散。
预混合料可以为颗粒(granule)或粒料(pellet)的形式。
在具体的实施方式中,形式为其被添加至饲料或当被包括在饲料添加剂中时的结合二氢槲皮素(紫杉叶素)的落叶松阿拉伯半乳聚糖被明确定义了。术语明确定义意思是落叶松阿拉伯半乳聚糖制剂是至少50%纯。在其他具体的实施方式中,明确定义的落叶松阿拉伯半乳聚糖制剂是至少60、65、70、75、80、85、88、90、92、94或至少95%纯。
通常脂溶性和水溶性维生素以及痕量矿物质形成用于添加至饲料的所谓的预混合料的一部分,而大量矿物质通常单独添加至饲料。因为膳食纤维的摄入可能不利影响某些哺乳动物中维生素和矿物质的吸收,可期望将组合物与维生素和/或矿物质补充或预混合料结合。
根据该方法,本发明的动物饲料添加剂在进食之前或同时补充至单胃动物。优选地,本发明的动物饲料添加剂与进食同时补充至单胃动物。在更优选的实施方式中,动物饲料添加剂以颗粒状或稳定的液体形式添加至食物。
在另一方面中,本发明提供预处理动物饲料的方法,通过该方法处理动物饲料以接受落叶松木材提取物的作用。优选地落叶松木材提取物是结合二氢槲皮素(紫杉叶素)的落叶松阿拉伯半乳聚糖。
参考图3和4中显示的两个具体的实施方式进一步阐释本发明,其不旨在以任何方式限制如所要求的本发明的范围。图3中显示的实施方式1,显示具体的组合物,其用作作为狗的预混合料的功能饲料。图4中显示的实施方式2,显示具体的组合物,其用作作为猫的预混合料的功能饲料。

Claims (13)
1.动物饲料组合物,其包括:
(a)落叶松阿拉伯半乳聚糖,其量在所述动物饲料组合物的0.1%和30%之间;
(b)二氢槲皮素(紫杉叶素),其量在所述动物饲料组合物的0.1%和10%之间;和
(c)至少一种动物饲料载体和赋形剂。
2.根据权利要求1所述的动物饲料组合物,其中所述二氢槲皮素(紫杉叶素)是提取自下述至少一种植物的类黄酮:拉里克斯卡加德里、拉里克斯捷卡诺夫斯基、拉里克斯达夫里卡、拉里克斯梅林尼、拉里克斯卡茨恰蒂卡、拉里克斯卢西卡、拉里克斯西比里卡和拉里克斯苏卡捷威。
3.根据权利要求1所述的动物饲料组合物,其中所述落叶松阿拉伯半乳聚糖是提取自下述至少一种植物的膳食纤维:拉里克斯卡加德里、拉里克斯捷卡诺夫斯基、拉里克斯达夫里卡、拉里克斯梅林尼、拉里克斯卡茨恰蒂卡、拉里克斯卢西卡、拉里克斯西比里卡和拉里克斯苏卡捷威。

6.改善动物饲料效率、动物消化的效率、动物的总体健康和动物饲料生产效率的方法,所述方法包括下述步骤:
向动物施用有效量的饲料添加剂组合物,该饲料添加剂组合物包括膳食纤维和类黄酮的组合。
