本发明涉及药物领域,具体涉及预防或治疗脑卒中的多肽化合物。硒蛋白S激动剂多肽1,其特征在于:其序列为GGADRAAAAVEPDVVVKRQERGD。所述多肽1的制备方法,步骤包括(1)煎煮,(2)酶解,(3)蛋白质分离,(4)半制备RP‑HPLC分离纯化,(5)冻干。所述的酶解采用木瓜蛋白酶和碱性蛋白酶酶解。所述的蛋白质分离采用大孔树脂、葡聚糖凝胶。硒蛋白S激动剂多肽1在预防或治疗脑卒中中的应用。本发明中的硒蛋白S激动剂多肽1可以靶向促进硒蛋白S表达,提高脑组织中硒蛋白S的含量,达到预防或治疗脑卒中的作用,且安全性好。

一种硒蛋白S激动剂多肽及其制备方法、应用
技术领域:
本发明涉及药物领域,具体涉及用于预防或治疗脑卒中的多肽提取物。
背景技术:
脑卒中已继心脏病和癌症成为人类第三大死亡原因,也是全球成人致残的主要原因之一。现阶段,治疗脑卒中的方法多为药物治疗。多采用尿激酶,华法林和阿斯平林等抗凝血药物治疗,或采用活血化瘀的中药治疗,但往往因剂量不当反而引起脑出血,给治疗带来更大的危害。
近几年关于缺血性脑卒中的研究已逐渐发现炎症反应对卒中后脑组织损伤具有重要影响,脑缺血后持续发展的炎症可加重缺血损伤,所以抗炎治疗的研究逐渐受到重视并广泛开展。硒蛋白S是一种新发现的内质网和细胞膜驻留硒蛋白。以往的研究结果揭示硒蛋白S可以保护细胞拮抗氧化损伤及内质网应激诱导的细胞凋亡;参与脂蛋白代谢、精子发育过程、炎症反应及将错误折叠蛋白从内质网腔逆向转移到细胞质中然后降解的过程(即内质网相关蛋白降解)。曾有报道,硒蛋白S基因多态性与糖尿病、冠状动脉心脏病或先兆子痫等疾病密切相关。最新的研究发现,硒蛋白S与脑卒中密切相关。有实验证实体外培养的星形胶质细胞受到缺血损害时,硒蛋白S的表达增加,并且可以减轻星形胶质的缺血损伤,进而在疾病的发生发展中起到预防或治疗脑卒中的作用。
茶叶中除了富含硒以外,还富含具有硒蛋白S激动剂作用的多肽,但是过去的技术有限,没有从茶叶中提取制备硒蛋白S激动剂多肽的报道。
发明内容:
本发明目的是针对现有预防或治疗脑卒中药物的特点,设计一种从茶叶中天然提取的多肽提取物,能有效治疗或预防脑卒中。
技术方案
硒蛋白S激动剂多肽1,其特征在于:其序列为GGADRAAAAVEPDVVVKRQERGD。所述多肽1的制备方法,步骤包括(1)煎煮,(2)酶解,(3)蛋白质分离,(4)半制备RP-HPLC分离纯化,(5)冻干。所述的酶解采用木瓜蛋白酶和碱性蛋白酶酶解。所述的蛋白质分离采用大孔树脂、葡聚糖凝胶。硒蛋白S激动剂多肽1在预防或治疗脑卒中中的应用。
有益结果:
本发明中的硒蛋白S激动剂多肽1可以靶向促进硒蛋白S表达,提高脑组织中硒蛋白S的含量,达到预防或治疗脑卒中的作用,且安全性好。
具体实施方式
实施例1
多肽的化学合成方法
将茶叶加水煎煮2h后,冷却至室温;加入1%木瓜蛋白酶和0.5%碱性蛋白酶酶解4h;采用大孔树脂按照分子量分离蛋白质,再用半制备RP-HPLC分离纯化,得到的产物冻干后,即为硒蛋白S激动剂多肽1。应用上述方法提取的激动剂多肽1,经过质谱鉴定,序列为GGADRAAAAVEPDVVVKRQERGD,该序列为全新序列。也可以委托上海生工合成。
实施例2
硒蛋白S激动剂多肽1的体外细胞凋亡的测定
本实验研究了硒蛋白S激动剂多肽1抑制大鼠脑星形胶质细胞凋亡的作用。当细胞用10μmol/LTNF-α处理后,经TUNEL染色后发现与培养基对照组相比出现了很多绿色亮点,说明体系中存在凋亡的细胞。当20μmol/L硒蛋白S激动剂多肽1与TNF-α共孵育后,镜检看到比单纯加TNF-α的对照组较少的绿色亮点。对拍摄照片进行计数并统计后,发现用10μmol/LTNF-α处理的细胞凋亡率约为66.3%,而20μmol/L激动剂多肽1与TNF-α共同处理的细胞凋亡率减少到31.3%(p<0.05)。并且只用20μmol/L激动剂多肽1处理的细胞的凋亡情况与培养基对照组并无明显差别,说明激动剂多肽1显著减少了TNF-α诱导的细胞凋亡,但此浓度的激动剂多肽1自身对细胞没有毒性作用。
实施例3
硒蛋白S激动剂多肽1治疗大鼠脑卒中模型的作用
雄性8周龄的SD大鼠40只,随机分为4组,建立MCAO的模型,分别在造模同时给予药物干预,分别为:A组为空白对照组:生理盐水;B组为模型对照组:造模+生理盐水;C组为药物干预组:造模+硒蛋白S激动剂多肽1(10mg/kg/d);D为阳性药干预组:造模+尿激酶(10mg/kg)。连续10天。(1)观察大鼠局部脑缺血再灌注后梗死体积的大小;(2)采用神经行为学评分,观察MCAO动物的康复情况;(3)检测脑组织中硒蛋白S的含量。
结果:治疗性给予多肽1组的脑卒中模型大鼠与模型组相比,脑卒中面积减少(P<0.01),神经行为学评分降低(P<0.05),脑组织中硒蛋白S的含量升高(P<0.01),与模型对照组比较差异均有统计学意义(见表1)。
