Abstract
与人MHC-I类分子结合的GRP78多肽表位,其特征在于:能与人MHC-I类分子的结合位点进行结合、可激活特异性的细胞毒性T淋巴细胞的多肽表位,为筛选自人葡萄糖调节蛋白78(GRP78),且与人GRP78具有同源性的下列氨基酸序列,为选自GRP788-16,GRP78236-244,GRP78337-345,GRP78415-423,GRP78416-424中的游离型多肽、融合型多肽和嵌合型多肽;以及以所述5条多肽之一为单体的各种形式的聚合体。本发明的优点:有效杀伤肝素酶阳性的肿瘤组织,达到治疗进展期肿瘤的目的。使用计算机模拟技术与实验相结合的方法,进一步提高了预测多肽表位的准确性,不单纯考虑分子结合位点对多肽表位的预测影响,同时从空间构象上,预测了多肽表位。体外容易合成,经静脉给药后容易被机体吸收,因此具有非常大的商业产业化价植。

技术领域
本发明涉及生物医药工程技术领域,包括筛选获得与人主要组织相容性复合物(MHC-I)类分子结合的GRP78多肽分子,以及编码这些多肽的DNA,RNA,特别涉及了与人MHC-I类分子结合的GRP78多肽表位。
背景技术
葡萄糖调节蛋白78(Glucose regulated protein,GRP78)属于热休克蛋白70家族,由内质网合成和分泌,其基本功能是:作为分子伴侣,在真核生物内质网膜上,通过与新生多肽表面的疏水片段以非共价键短暂结合,促进蛋白质的正确折叠、装配以及跨内质网膜转运,在生物机体的生长发育中起重要作用;作为应激蛋白,在低糖、低氧等应激状态下大量表达,参与未折叠蛋白反应以维持细胞的稳定。
研究表明,GRP78在人类肝癌、肺癌、结直肠癌、乳腺癌、前列腺癌、纤维肉瘤及骨肉瘤等恶性肿瘤细胞的GRP78表达显著增高,说明GRP78与多种肿瘤发生相关;GRP78在恶性肿瘤的侵袭和转移过程中也有重要作用,抑制GRP78基因的表达可抑制肿瘤细胞在体内的侵袭能力以及转移能力;GRP78的过度表达还可使许多恶性肿瘤细胞不同程度的对多种化疗药物耐药,而如果抑制GRP78的表达则可使化疗药物对这些恶性肿瘤细胞的杀伤作用提高。综上所述,GRP78作为肿瘤相关抗原,与肿瘤的发生、侵袭转移及药物敏感性密切相关。
随着对免疫应答分子机制研究的深入,现已逐渐认识到机体的免疫细胞并不是对应于各种各样的病原体或天然抗原的整体分子,而是针对各种各样抗原分子的抗原表位,即蛋白质抗原是通过其表位来体现其免疫特异性。目前的研究表明,CD8+T细胞所识别的靶抗原需先经抗原提呈细胞处理,之后以“抗原肽-MHC-I类分子”复合物的形式呈现在抗原提呈细胞或靶细胞表面,相应的与MHC-I类分子结合的抗原肽即为CTL表位。
已有研究显示,利用GRP78核酸疫苗免疫非小细胞肺癌小鼠后,小鼠生存期明显延长,小鼠免疫细胞分泌细胞因子肿瘤坏死因子(TNF-α)、γ干扰素(IFN-γ)的水平和对靶细胞的杀伤率明增加,提示GRP78核酸疫苗激活了特异性的CTL 反应。而将GRP78作为肿瘤相关抗原,采用生物信息学手段,研究其CTL表位,并将其作为肿瘤免疫治疗的一个新靶点,到目前为止,未见有文献报道。
发明内容
本发明的目的是提供能与人MHC-I类分子的结合位点进行结合、可激活特异性的细胞毒性T淋巴细胞的多肽表位;提供所述多肽表位在制备用于临床治疗和检测肿瘤性疾病疫苗中的用途。
本发明根据抗原肽与MHC-I类分子相互作用的机理,从肝素酶的全长基因序列中筛选出侯选多肽表位,最终得到能有效与MHC-I类分子结合并能激活特异性T淋巴细胞的多肽表位,该多肽表位长度仅9个氨基酸序列,体外容易合成,方便临床应用。
本发明的第一目的,即能与人MHC-I类分子的结合位点进行结合、可激活特异性的细胞毒性T淋巴细胞的多肽表位,筛选自人GRP78,为与人GRP78具有同源性的下列氨基酸序列,包括:

本发明所获得的多肽序列是通过超基序法与量化基序法结合的方法,从肽库中直接调取,保持了与MHC-I类分子结合特性。通过对这些多肽序列进行概率统计分析,获得多肽与MHC-I类分子的结合模型。
1.人GRP78氨基酸序列的获得
自国际开放共享基因库NCBI GeneBank中查找出天然人GRP78的全长氨基酸序列(共654个氨基酸)表示为:

2.多肽表位预测的超基序的获得
超基序法是基于在同一人白细胞抗原(HLA)家族,甚至不同家族的HLA同种异性分子接纳的抗原肽具有相同或相似的锚点残基构成的肽基序,超基础主要作用的锚点残基是位于一要作川的锚点残基是位于肽链2位(P2)氨基酸残基及羧基末端9位氨基酸残基。当P2、P9的残基为A、I、L、M、V、T时,且第二位为芳香族氨基酸,第九位为疏水性氨基酸,与HLA-A2分子有较高的亲和力。这些氨基酸的特定序列是多肽分子与MHC-I分子结合的活性位点或关键性氨基酸,决定了与MHC-I类分子结合多肽的活性特征。超基序预测CTL表位虽然克服了以往采用简单基序法易产生的假阴性结果,但由于此预测法与基序法具有相同的弱点,即在预测中仅仅考虑了二、九位残基的影响而忽略了其它位点的残基结合情况,易出现假阳性结果,故应联合量化基序法提高CTI.表位预测的准确性。
应用http://www.syfpeithi.de/bin/MHCServer.dll/EpitopePrediction.htm提供分析软件对上述人GRP78氨基酸序列进行分析,得到675个超基序,选择评分大于15的108个超基序用于下一步研究。

3.CTL表位预测的量化基序方案
量化基序法是基于IBS假设,即在抗原肽与HLA-A2分子的结合中,抗原肽的侧链效应(锚定、抑制或中性)主要依赖与相应的氨基酸残基在肽中所处的位置受相邻的氨基酸残基影响较小。这在一定的范围内可以假设在一个多肽序列中,每个氨基酸残基相互独立且影响整个肽与MHC-I类分子的结合,而该抗原肽的结合能就是每个位置氨基酸残基结合能的综合。基于这种假设,1993年Parker.等用肽结合实验分析154个九肽与HLA-A2分子的亲和力,并得到一个包含180个系数的结合矩阵,其中每一个系数反应了相应氨基酸残基在该位置时的相应结合能。结合能越大,多肽表位与HLA-A2分子结合越紧密。对于某测定多肽各位置相应氨基酸残基结合系数相乘再乘标准系数0.151,即为该肽与HLA-A2的结合力,当结合系数大于或等于5时,可以确定为与HLA-A2分子结合稳定,反之认定为结合不稳定。根据上述原理,应用http://www-bimas.cit.nih.gov/molbio/hla_bind/提供的分析软件对人GRP78氨基酸序列进行分析,选择评分大于5的30个基序用于进一步研究。

4.CTL表位预测蛋白酶解原理
基于抗原加工处理过程的CTL表位预测方法,主要针对抗原递呈中所涉及的细胞器,目前集中在蛋白酶体和转运相关蛋白上,又以蛋白酶体为主。蛋白酶体 进行酶切时有特定的裂解识别基序,该类预测方案通过计算蛋白酶体在抗原蛋白序列每个残基后酶切的概率,来判定该位点是否可以发生酶切而形成表位多肽,从而达到预测CTL表位的目的。根据上述原理,应用http://www-alt.mpiib-berlin.mpg.de/MAPPP/cleavage.html提供的分析软件对上述超基序法和量化基序法获得的24个基序进行进一步分析,筛选获得5个基序,其结果如下:

实施例2多肽表位与MHC-I类分子结合的计算机模拟
对上述所得多肽表位用ChemOffice软件包中的ChemDraw Ultra、ChemDraw3D Ultra分别建立多肽表位的二维和三维结构;用SYBYL-X软件对多肽表位和和HLA-A2.1的能量和结构进行修饰,模拟二者结合的三维结构,进行分子动力学结合模拟和生理活性和运用价值的预估,主要包括以下方法:(1)多肽表位分子模型构建。运用ChemOffice软件包中的ChemDraw Ultra构建多肽表位分子二维结构,再将多肽表位的二维结构导入ChemDraw3D Ultra得到三维模型(图1、3、5、7、9)并保存为Mol2格式,在对接模拟环节将三维结构导入SYBYL-X软件中,依次选择下拉菜单Compute-Minimize-Molecule,出现Minimize对话框进行多肽分子的能级优化,使之能级达到最小最适合与HLA-A2.1的复合物进行分子对接的能量和结构状态,然后将优化后的多肽表位导出为MOL2格式,以备和HLA-A2.1的结合槽凹槽进行三维对接。(2)HLA-A2.1的复合物进行分子动力学模拟前准备。HLA-A2.1的初始坐标来自于蛋白质晶体结构专属网站 http://www.rcsb.org/pdb/,将获得的HLA-A2.1结构数据导入SYBYL-X软件中,依次选择下拉菜单Applications-Docking Suite-Docking Ligands,出现Docking对话框,在Filename栏的右边选择Define对HLA-A2.1结构进行对接前结构修饰(修复侧链,脱水加氢),结构修饰完成后分析获得HLA-A2.1的结合槽凹槽三 维信息,保存以备与多肽表位进行对接。(3)对接模型的构建。在SYBYL-X软件中依次打开Applications-Docking Suite-Docking Ligands,出现Docking对话框,在Docking Mode和Filename栏选择获得的HLA-A2.1的结合槽凹槽三维信息,在Ligand Sourse栏中选择Mole2File并选择MOL2格式的各个已经优化并处于能级最低的多肽表位数据,分别进行对接并最终得到对接模拟的评分,通过对接模拟的评分值对多肽表位分子和HLA-A2.1的结合力做预估。由于抗原肽与MHC分子之间主要通过疏水作用、H键、盐键等弱相互作用进行分子间的识别,这些次级键作用越强,相应分子间的非键作用能就越低,结合则越紧密。计算机模拟结果显示,GRP788-16、GRP78415-423、GRP78236-244、GRP78337-345和GRP78415-4235条多肽表位与HLA-A2.1均能很好的结合(如图2、4、6、8、10)。
实施例3多肽表位的合成、纯化及分子量测定
多肽表位委托吉尔生化(上海)有限公司采用标准Fmoc方案合成,并利用HPLC进行纯化,质谱进行分子量的测定。其质谱分析图参见图11至图15,可见5条多肽的理论值均与实测值相近,在允许误差范围之内。HPLC分析图见图11至图15,可见且纯度均在95%以上,说明合成效果好。上述多肽经冻干处理后放于-70℃保存、备用。
实施例4本发明多肽表位与人MHC-I类分子亲和力检测
本实施例利用T2细胞系的特点,检测多肽表位与人MHC-I类分子的亲合力。T2细胞可表达MHC-Ⅰ类分子,但因其内源性抗原呈递途径中必需的抗原多肽转运蛋白(TAP)缺陷,故不能转运内源性抗原,只能将空载的HLA-A2分子呈递在细胞表面,而空载的HLA-A2分子在细胞表面不稳定,很快降解或返回细胞内部。但外源给与的抗原多肽与HLA-A2分子结合成复合物后,可增强细胞表面HLA-A2分子的稳定性,而留在细胞表面。抗原肽与HLA-A2的结合力越强,则T2细胞表面HLA-A2分子的表达量就越高。所以检测抗原多肽处理的T2细胞表面HLA-A2分子表达量的增加,可直观地反映外来抗原多肽与HLA-A2分子的结合力以及结合的稳定性,能够进一步证明本发明的多肽的有效性。
1.多肽表位与HLA-A2分子亲和力检测
实施例中,T2细胞经PBS洗涤2次,以无血清IMDM培养基重悬后,在6孔培养板上按1×106/孔的密度种植,各抗原多肽以二甲基亚砜(DMSO)溶解后,以50μg/ml的浓度加入到T2细胞的培养液中,并设置空白对照组及阳性对照组,阳性对照组加入30μg/ml流感病毒基质蛋白多肽。各孔均加入3μg/ml人β2微球蛋白。T2细胞在37℃,5%CO2、饱和湿度条件下培养24h后。孵育结束后,T2 细胞以1000rpm离心5min,沉淀细胞以冰PBS洗涤3次,然后以100μl冰PBS重悬,加入5μl鼠抗人HLA-A2.1-FITC单抗,4℃孵育15min,离心弃上清后以300μl冰PBS重悬,在流式细胞仪上检测平均荧光强度(MFI),结果见图16。
结果以荧光系数(FI)作为衡量指标,计算公式为:荧光系数(FI)=(多肽平均荧光强度-β2微球蛋白平均荧光强度)/β2微球蛋白平均荧光强度,判断标准:FI>1.5为多肽与HLA-A2.1分子结合力高,1.0<FI<1.5为中等结合力,FI>0.5为低结合力。
表6亲和力实验结果
实验结果表明:GRP788-16,和GRP78415-423两个多肽表位与MHC-I类分子有亲和力最高,其次是GRP78236-244和GRP78337-345,GRP78415-423亲和力较差。
实施例5多肽表位对人乳腺癌细胞MCF7、MBA-MD-231和人成骨肉瘤细胞MG-63、U-2OS的杀伤效果
采用钙黄素(Calcein-AM)释放试验进行。
1.效应细胞的制备
诱导成熟的人DC细胞,以PBS清洗后,无血清IMDM培养基重悬,加50μg/ml抗原多肽,37℃孵育过夜。DC细胞经PBS清洗、计数,按30μg/ml的 浓度加入丝裂霉素C,37℃孵育30min。DC细胞经冰PBS清洗、计数后,与繁殖期T细胞按1:20比例共培养,并加入50U/ml的IL-2,以刺激T细胞。1周刺激1次,共3次,将T细胞诱导成细胞毒性T细胞(CTL),即效应细胞。
2.靶细胞的制备
复苏乳腺癌细胞MCF7、MBA-MD-231和人成骨肉瘤细胞MG-63、U-2OS,正常培养、传代。其中MCF7、MG-63两细胞株为HLA-A2.1阴性,MBA-MD-231、U-2OS两细胞株为HLA-A2.1阳性。4株细胞均为GRP78蛋白表达阳性。
3.钙黄素释放试验
按10mM的浓度,以Calcein-AM标记靶细胞,清洗、重悬;效应细胞与标记后的肿瘤细胞按照1:1、10:1、20:1、40:1、80:1的效靶比例,37℃共培养4h;然后收集培养液,1000rpm离心5min,取100μl上清液测定平均荧光强度。以单纯靶细胞作为自发释放量,以单纯靶细胞加去垢剂作为最大释放量。
效应细胞对靶细胞的杀伤率以细胞溶解率表示:细胞溶解率=(实验释放量-自发释放量)/(最大释放量-自发释放量)。

实验结果表明:GRP788-16,、GRP78415-423、GRP78236-244、GRP78337-345和GRP78416-4245条多肽对GRP78+和HLA-A2+双阳性的人骨肉瘤细胞U-2OS和人乳腺癌细胞MBA-MD-231具有杀伤作用,其中GRP788-16,和GRP78415-423杀伤作用最强,GRP78236-244和GRP78337-345次之,GRP78416-424杀伤作用最弱;5条多肽对GRP78+和HLA-A2-的人骨肉瘤细胞MG-63和人乳腺癌细胞MCF-7不具有杀 伤作用。
Claims (3)
1.与人MHC-I类分子结合的GRP78多肽表位,其特征在于:所述的与人MHC-I类分子结合的GRP78多肽表位,能与人MHC-I类分子的结合位点进行结合、可激活特异性的细胞毒性T淋巴细胞的多肽表位,为筛选自人葡萄糖调节蛋白78(GRP78),且与人GRP78具有同源性的下列氨基酸序列,包括:
GRP788-16,Ala Met Leu Leu Leu Leu Ser Ala Ala;
GRP78236-244,Tyr Ile Asp Asn Gly Val Phe Glu Val;
GRP78337-345,Ser Tyr Met Lys Pro Val Gln Lys Val;
GRP78415-423,Val Leu Leu His Val Cys Pro Leu Tyr;
GRP78416-424,Leu Leu His Val Cys Pro Leu Tyr Leu。
2.按照权利要求1所述的与人MHC-I类分子结合的GRP78多肽表位,其特征在于:为选自GRP788-16,GRP78236-244,GRP78337-345,GRP78415-423,GRP78416-424中的游离型多肽、融合型多肽和嵌合型多肽;以及以所述5条多肽之一为单体的各种形式的聚合体。
3.按照权利要求1所述的与人MHC-I类分子结合的GRP78多肽表位,其特征在于:所述的与人MHC-I类分子结合的GRP78多肽表位,通过原核细胞或真核细胞表达纯化获得,或人工合成。
