本发明涉及一种检测混合气体中二氧化碳浓度的二氧化碳指示剂及制备方法,属临床医学及环境卫生监测领域。指示剂主要由氢氧化钾或氢氧化钠或氢氧化钙,乙醇,指示剂耐尔蓝A、酚酞,活性氧化铝组成,指示剂在接触到含有二氧化碳的混合氧气体时,由深红→紫红→紫灰→灰蓝→蓝,显色过程与CO2浓度成比例,指示剂在空气环境中相对稳定,遇1.0%以上浓度CO2时能迅速变色。从而可快速判定插入窒息患者气管内导管的正确位置,也可用于大气CO2分析及临床化学分析和坑道及密闭环境的卫生监测。
Description
本发明涉及一种检测混合气体中二氧化碳浓度的二氧化碳指示剂及制作工艺,属临床医学及环境卫生监测领域。
快速、准确判断气管导管位置在急救、复苏及麻醉过程中极为重要,如导管位置不正确或误入食道、且在短时间内未发现该错误,就有可能给病人造成严重危害甚至死亡,当前最有效的鉴别方法是检测呼出气中二氧化碳,以化学变色方法为手段检测呼出气中的二氧化碳浓度的方法在国外有运用,但由于其指示剂变色的反差小,不易观察。
本发明针对以上存在问题,其目的在于提供一种可快速检测混合气体中二氧化碳浓度的指示剂。
本发明利用氢氧化钠(钾、钙)做为二氧化碳吸收剂提供碱性环境,乙醇起到溶剂和吸湿剂作用,酸碱指示剂耐尔蓝A的pH域为10.1(蓝)-11.1(红),本发明的指示剂主要应用其酸色及蓝色;酚酞的pH域为8.2(无色)-10.0(深红),本发明的指示剂主要应用其碱色及深红色,两指示剂在鉴别二氧化碳时起到颜色互补作用,因颜色反差大,使终点观察更加容易;活性氧化铝做为指示剂载体提供指示剂与二氧化碳的作用界面;所配制的指示剂与含有二氧化碳的混合气体发生作用时,乙醇首先吸附水蒸汽使指示剂含有适量水分,在此情况下二氧化碳迅速生成碳酸并与氢氧化钠(钾、钙)反应生成盐和水,指示剂碱性环境随之减弱,当pH<10.0后,指示剂碱色的酚酞红色逐渐消退,而耐尔蓝A的酸色及蓝色逐渐显现,证实二氧化碳的存在,当接触到含有二氧化碳的混合氧气体时,由深红→紫红→紫灰→灰蓝→蓝,显色过程与CO2浓度成比例,指示剂在空气环境中相对稳定,遇1.0%以上浓度CO2时能迅速变色。从而可快速判定插入窒息患者气管内导管的正确位置,也可用于大气CO2分析及临床化学分析和坑道及密闭环境的卫生监测。
本发明取氢氧化钾或氢氧化钠或氢氧化钙溶入一定量的乙醇溶液中,制成所要求达到浓度的氢氧化钾或氢氧化钠氢氧化钙乙醇溶液,使溶液的pH为碱性,再加入指示剂耐尔蓝A和酚酞,使之配制成碱性指示剂溶液,再加入一定量的活性氧化铝(重量/体积),制成混悬液后移入真空干燥瓶内,置于干燥烤箱中抽真空过滤,并在烤箱内放入适量钠石灰和硅胶、使其压力保持一定,直至指示剂颗粒接近干燥时停止抽气,通入经钠石灰过滤的空气,将指示剂移入磨口瓶中避光保存。在水蒸气参于条件下(海平面肺泡水蒸气占6.2%),二氧化碳作用于吸附有指示剂的碱性固体,随着pH下降,指示剂颜色由深红→紫红→紫灰→灰蓝→蓝,显色过程与CO2浓度成比例。
现对本发明作详细描述。
本发明取氢氧化钾或氢氧化钠或氢氧化钙溶入50-100%乙醇溶液中,制成0.1-1.0mmol/L的氢氧化钾或氢氧化钠或氢氧化钙乙醇溶液,测pH为9.0-12.0,再加入指示剂耐尔蓝A和酚酞,使之浓度均为0.1-1.0%,配制成碱性指示剂溶液,再加入30-90%的活性氧化铝(重量/体积),制成混悬液后移入真空干燥瓶内,置130-150℃干燥烤箱中抽真空过滤,并在烤箱内放入200-500克钠石灰和100-400克硅胶、使其压力保持-200-400mmHg,直至指示剂颗粒接近干燥时停止抽气,通入经钠石灰过滤的空气,将指示剂移入磨口瓶中避光保存。
实施例:取氢氧化钾溶入100毫升70%的乙醇溶液中,制成0.8mmol/L的氢氧化钾乙醇溶液,取指示剂耐尔蓝A和酚酞加入到该溶液中,使指示剂浓度分别为0.4%和0.3%,加入活性氧化铝60克,充分混匀,制成混悬液后移入真空干燥瓶内,置130-150℃干燥烤箱中抽真空过滤,烤箱内放入适量钠石灰和硅胶,维持压力-200mmHg,直至指示剂颗粒接近干燥时停止抽气,通入经钠石灰过滤的空气,将指示剂移入磨口瓶中避光保存,此时的指示剂颜色为深红色。该指示剂具有在空气中相对稳定而遇高浓度二氧化碳时迅速变色的特点。具体表现在指示剂接触空气(含0.03-0.04%二氧化碳)时,5分钟后指示剂开始从深红变为紫红,10分钟后变为紫灰,120分钟后变为灰蓝;而在遇1.0%以上浓度二氧化碳时,指示剂迅速改变颜色。二氧化碳指示剂对较高浓度二氧化碳的反应如下表:(表见文后)

Claims (2)
1.一种二氧化碳指示剂,其特征在于:所述的二氧化碳指示剂每100克含有氢氧化钾0.5-5.6克或氢氧化钠0.4-4.0克或氢氧化钙0.74-7.4克、少量残留乙醇、指示剂耐尔蓝A0.1-1.0克、酚酞0.1-1.0克和30-90克的活性氧化铝组成。
2.权利要求1所述的二氧化碳指示剂的制备方法,该方法包括如下步骤:取氢氧化钾或氢氧化钠或氢氧化钙溶入50-100%乙醇溶液中,制成0.1-1.0mol/L的氢氧化钾或氢氧化钠或氢氧化钙乙醇溶液,测pH为9.0-12.0,再加入指示剂耐尔蓝A和酚酞,使之浓度均为0.1-1.0%,配制成碱性指示剂溶液,再加入30-90克的活性氧化铝(重量/体积),制成混悬液后移入真空干燥瓶内,置130-150℃干燥烤箱中抽真空过滤,并在烤箱内放入200-500克钠石灰和100-400克硅胶、使其压力保持-200-400mmHg,直至指示剂颗粒接近干燥时停止抽气,通入经钠石灰过滤的空气,将指示剂移入磨口瓶中避光保存。
